This work studied the combined use of gliadins and SSRs to analyse inter- and intra-accession variability of the Spanish collection of cultivated einkorn (Triticum monococcum L. ssp. monococcum) maintained at the CRF-INIA. In general, gliadin loci presented higher discrimination power than SSRs, reflecting the high variability of the gliadins. The loci on chromosome 6A were the most polymorphic with similar PIC values for both marker systems, showing that these markers are very useful for genetic variability studies in wheat. The gliadin results indicated that the Spanish einkorn collection possessed high genetic diversity, being the differentiation large between varieties and small within them. Some associations between gliadin alleles and geographical and agro-morphological data were found. Agro-morphological relations were also observed in the clusters of the SSRs dendrogram. A high concordance was found between gliadins and SSRs for genotype identification. In addition, both systems provide complementary information to resolve the different cases of intra-accession variability not detected at the agro-morphological level, and to identify separately all the genotypes analysed. The combined use of both genetic markers is an excellent tool for genetic resource evaluation in addition to agro-morphological evaluation.  相似文献

This article describes a fast and simple methodology for the extraction and determination of organic acids in tissues and root exudates of maize, lupin, and chickpea by LC/MS/MS. Its main advantage is that it does not require sample prepurification before HPLC analysis or sample derivatization to improve sensibility. The results obtained showed good precision and accuracy, a recovery close to 100%, and no significant matrix effect. Moreover, the sensibility of the method is in general better than that of previously described methodologies, with detection limits between 15 and 900 pg injected.  相似文献

Information on temporal and spatial patterns of N mineralization is critical in designing tree-crop mixed systems that could maximize N uptake while minimizing N loss. We quantified N mineralization rates in a pecan (Carya illinoensis K. Koch)–cotton (Gossypium hirsutum L.) alley cropping system in northwestern Florida with (non-barrier) and without tree-crop belowground interactions (barrier separating the root systems of pecan and cotton). Monthly rates of mineralization were estimated using buried bag incubations over a 15-month period. In addition, seasonal mineralization rates and cotton lint yield on soils supplied with two sources of N—inorganic fertilizer and organic poultry litter—were assessed. Results indicated that temporal variations in net NH4 and NO3 accumulation and mineralization rates were driven primarily by environmental factors and to a lesser degree by initial soil NH4 and NO3 levels. Mineralization varied by belowground interaction treatment during the initial growing season, when the non-barrier treatment exhibited a higher mineralization rate than the barrier treatment, likely due to reduced nutrient uptake by cotton in the non-barrier or a higher degree of immobilization in the barrier treatment. Mineralization during the second growing season was similar for both treatments. Source of N had no effects on N transformation in the soil. Lint yield reductions were observed in the non-barrier treatment during both years compared to the barrier treatment, likely due to interspecific competition for water. Yield differences between treatments in the second growing season were likely compounded by a diminishing pre-study fallow effect. Source of N was found to have a significant effect on cotton yield, with inorganic fertilizer resulting in 39% higher lint compared to poultry litter in the barrier treatment.  相似文献

Sheep production is the main agricultural activity in Patagonia. Since the middle of the 20th century, sheep numbers have declined steadly. We used historical records of stock numbers in four ranches to analyze the importance of regional factors so as to explain the decline of the Patagonian sheep flocks. We found that the stocks of all the four ranches declined with a similar trend but fluctuated independently, thus reflecting a complex interaction between regional and local factors. Aboveground net primary production (ANPP) and vegetation physiognomy explained most of the differences in the flocks declining rates. We estimated demographic parameters for two ranches differing in their average annual growth rates. From these demographic parameters, we constructed deterministic and stochastic matrix models to establish the relative contribution of demographic processes to the observed decline. Matrix models projected a faster decline than that observed in the ranch used to calibrate the model. This suggests that the recorded demographic parameters could drive most stocks to extinction in less than 100 years. We concluded that the observed dynamics would be generated by demographic processes, but extinction is delayed or avoided by a continuous intake of animals. Ewe survival was the most important parameter in controlling the growth rate of the flocks. The environmental stochastic model showed that the growth of the stocks was highly sensitive to increases in the frequency of good years (those that produce a positive growth) and in the transition from normal years to bad years. All these evidences point out the existence of biological constraints to sheep production in Patagonia: ANPP and vegetation structure would control flock population dynamics throughout its effects on key demographic parameters, ewe survival and marking rate (a recruitment measure). Our model results suggest that the decline in sheep numbers, and hence the sustainability of the activity, is driven, to a large extent, by the demographic characteristics of the flocks.  相似文献

Sample plots were established in the principal forest types in the the Nevado de Toluca National Park, Mexico including those dominated by Pinus hartwegii, Abies religiosa, Quercus laurina and Alnus jorullensis. The vertical structure was defined by three strata in the coniferous forests and two strata in the broadleaved forests. Timber harvesting in Abies religiosa and Quercus laurina forests and fires generated by humans in Pinus hartwegii forests impeded the recruitment of saplings. Mature trees were also heavily impacted by logging in Pinus hartwegii forests. On the contrary, Alnus jorullensis forests were increasing due to the disturbance of Pinus and Quercus forests, as well abandoned crop lands within the park. A combination of logging, uncontrolled fire, and grazing appears to be compromising the recruitment of important tree species in this national park. These factors, together with human settlements, have also increased the proportion of early successional species. Changes in forest structure from human disturbance indicate a need to control these activities if conservation goals are not to be compromised.  相似文献

Ever since the Kyoto Protocol, agroforestry has gained increased attention as a strategy to sequester carbon (C) and mitigate global climate change. Agroforestry has been recognized as having the greatest potential for C sequestration of all the land uses analyzed in the Land-Use, Land-Use Change and Forestry report of the IPCC; however, our understanding of C sequestration in specific agroforestry practices from around the world is rudimentary at best. Similarly, while agroforestry is well recognized as a land use practice capable of producing biomass for biopower and biofuels, very little information is available on this topic. This thematic issue is an attempt to bring together a collection of articles on C sequestration and biomass for energy, two topics that are inextricably interlinked and of great importance to the agroforestry community the world over. These papers not only address the aboveground C sequestration, but also the belowground C and the role of decomposition and nutrient cycling in determining the size of soil C pool using specific case studies. In addition to providing allometric methods for quantifying biomass production, the biological and economic realities of producing biomass in agroforestry practices are also discussed.  相似文献

There is growing interest in using switchgrass (Panicum virgatum L.) as a biofuel intercrop in forestry systems. However, there are limited data on the longevity of intercropped bioenergy crops, particularly with respect to light availability as the overstory tree canopy matures. Therefore, we conducted a greenhouse study to determine the effects of shading on switchgrass growth. Four treatments, each with different photosynthetically active radiation (PAR) levels, were investigated inside the greenhouse: control (no shade cloth, 49 % of full sunlight), low (under 36 % shade cloth), medium (under 52 % shade cloth), and heavy shade (under 78 % shade cloth). We determined the effect of shading from March to October 2011 on individually potted, multi-tillered switchgrass transplants cut to a stubble height of 10 cm. In the greenhouse, there was a reduction in tiller number, tiller height, gas exchange rates (photosynthesis and stomatal conductance), leaf area, above- and belowground biomass and light-use efficiency with increasing shade. Total (above- and belowground) biomass in the control measured 374 ± 22 compared to 9 ± 2 g pot?1 under heavy shade (11 % of full sunlight). Corresponding light-use efficiencies were 3.7 ± 0.2 and 1.4 ± 0.2 g MJ?1, respectively. We also compared PAR levels and associated aboveground switchgrass biomass from inside the greenhouse to PAR levels in the inter-row regions of a range of loblolly pine (Pinus taeda L.) stands from across the southeastern United States (U.S.) to estimate when light may limit the growth of intercropped species under field conditions. Results from the light environment of loblolly pine plantations in the field suggest that switchgrass biomass will be significantly reduced at a loblolly pine leaf area index between 1.95 and 2.25, which occurs on average between ages 6 and 8 years across the U.S. Southeast in intensively managed pine plantations. These leaf area indices correspond to a 60–65 % reduction in PAR from open sky.  相似文献
