ABSTRACT The aggressiveness of an extensive collection of strains of Erwinia amylovora was analyzed using immature fruit and detached pear flower assays under controlled environmental conditions. The analysis was performed by means of a quantitative approach based on fitting data to mathematical models that relate infection incidence to pathogen dose and time. Probit and hyperbolic saturation models were used for disease-dose relationships and provided information on the median effective dose (ED(50)). Values of ED(50) ranged from 10(3) to 10(6) CFU/ml (10 to 10(4) CFU per site of inoculation). A modified Gompertz model was used for disease-time relationships and provided information on the rate of infection incidence progression (r(g)) and time delayed to start of the incidence progress curve (t(0)). Values of r(g) ranged from near 0 to 1.90, and t(0) varied from 1.3 to more than 10 days. The more aggressive strains showed high r(g), low ED(50) values, and short t(0), whereas the less aggressive strains showed low r(g), high ED(50), and long t (0). The aggressiveness was dependent on plant material type and pear cultivars and was significantly different between strains of E. amylovora. Infectivity titration and kinetic analysis of progression of incidence of infections using the immature pear test and a standardized scale are proposed for assessment of strain aggressiveness. The implications of r(g), ED(50), and t(0) for the epidemiology and management of fire blight are discussed, particularly the wide range of aggressiveness among strains, the degree of host specificity observed in pear isolates, the very high infective potential of this pathogen, the independent action of pathogen cells during infection, and the possible advantage of including aggressiveness parameters into fire blight risk forecasting systems.  相似文献

Vegetative propagation techniques such as grafting can be used, in conjunction with field studies, to decouple the relative effects of age and size on tree metabolism and growth. Despite interest in this approach, little attention has been paid to the best metrics for assessing the growth performance of grafted plants over time. Based on an analysis of the grafting literature and our own data, we show that the choice of metrics to assess tree growth can entirely change the conclusions reached about the relative importance of age versus size. We recommend that absolute as well as relative rates of growth are calculated and that scion size be standardized as much as possible at the start of the experiment. Once proper metrics are chosen, all of the available published evidence is largely concordant with two concepts: (1) age-mediated controls of tree growth are likely to be important during the first few years of a tree's life (before phase change); and (2) after the first few years of a tree's life, size-mediated factors largely prevail over age-mediated factors in determining tree growth rates. We found no support for theories invoking age-mediated sink limitations in old trees.  相似文献

A field study was conducted to estimate seasonal differences in follicular development in weaned sows and to evaluate the implication of these differences on seasonal infertility. A total of 110 sows were selected at weaning during winter–spring (WS, n = 58) and summer–autumn (SA, n = 52). Ovaries were scanned once daily from weaning to the onset of oestrus and twice daily from then until ovulation. Six sows during WS were removed from study for not showing growing follicles at weaning. Oestrus was evaluated twice daily from day 1 after weaning to day 14 post-weaning. One of 52 (1.9%) sows in WS and 9/52 (17.3%) in SA showed no signs of oestrus within 14 days of weaning (P < 0.05). The diameters of the follicles at weaning, at the onset of oestrus and just before ovulation were smaller (P < 0.01) in SA sows than in WS sows. There were fewer follicles in SA sows than in WS sows just before ovulation (P < 0.05). Fifty of 51 (98.0%) sows in WS and 31/43 (72.1%) sows in SA experienced a weaning-to-oestrus interval (WOI) of 3–6 days (P < 0.05). Fifty-one of 52 (98.1%) sows in WS and 43/52 (82.7%) sows in SA were inseminated; the percentage of pregnant sows that failed to farrow was lower in WS (1/51, 2.0%) than in SA (5/43, 11.6%; P < 0.05). The percentage of farrowed sows was greater in WS (46/51, 90.2%) than in SA (32/43, 74.4%; P < 0.05). Sows in WS had on average 1.5 more piglets than sows in SA (P < 0.05). Sows with a WOI of 3–6 days had lower rates of pregnancy losses (P < 0.05) and higher farrowing percentages (P < 0.01) than those with a WOI > 6 days, irrespective of season.  相似文献

This study shows that 4- and 6-month-old infants can discriminate languages (English from French) just from viewing silently presented articulations. By the age of 8 months, only bilingual (French-English) infants succeed at this task. These findings reveal a surprisingly early preparedness for visual language discrimination and highlight infants' selectivity for retaining only necessary perceptual sensitivities.  相似文献

The level of irrigation restriction to apply in a deficit irrigation (DI) programme for sustainable peach (‘Baby gold 6’) production was investigated. The experiment involved four irrigation treatments over five consecutive seasons (2007–2011). They were full irrigation (control), reducing irrigation by 20 % during the first half of stage III (DI-80 %), withholding irrigation until reaching a light stress level (DI-L) and withholding irrigation until reaching a moderate stress level (DI-M). The withholding of irrigation in both DI-L and DI-M was applied only during stage II and postharvest periods and was based on midday stem water potential thresholds (Ψ stem). For the DI-L treatment ?1.5 MPa was used in both periods, and for DI-M ?1.8 and ?2.0 MPa were used during stage II and postharvest, respectively. Average Ψ stem values during DI periods were approximately ?1.4 and ?1.2 MPa for DI-M and DI-L, respectively. The pre-defined thresholds required to trigger irrigation were rarely reached. No significant differences between treatments were found in terms of yield in any experimental year. However, DI-M and to a lesser extent DI-L had lower final fruit fresh mass at harvest related to lower Ψ stem after three consecutive years of the experiment (during 2010 and 2011). Therefore, in terms of fruit size, DI was not sustainable. Rather than lowering Ψ stem thresholds, we recommend discontinuing DI after 3-year application.  相似文献

In this paper, accelerated solvent extraction (ASE) for the analysis of ochratoxin A (OTA) is applied for the first time. Optimization of the method is given for the extraction of OTA from rice samples. Several parameters such as type of solvent, temperature, pressure, static time, and cell size were investigated thoroughly to find the optimal extraction conditions. The optimum ASE operating conditions were methanol as extraction solvent, 1500 psi, 40 degrees C, 5 min of static time, 50% flush volume, 60 s of purge, 1 cycle, and 11 mL cell size. The total extraction time was approximately 15 min. OTA was determined by liquid chromatography coupled with a fluorescence detector and confirmed by methyl ester derivatization. The analytical performance of the method was monitored with quality control parameters. Finally, the optimized method was used to evaluate 12 rice samples, 1 of which was positive with an OTA content of 4.17 ng/g. The proposed method offers the possibility of a fast and simple process to obtain a quantitative extraction of OTA.  相似文献

Growth and production in the first year, as additional selection criteria, were assessed for nine poplar clones to be used as short rotation woody crops (SRWC) in the production of biomass for energy purposes. In order to identify the most promising clones in terms of growth and yield and also to assess their stability, trials were established at different locations in Spain. The majority of these clones, which form part of the European list of base materials, have frequently been used in plantations aimed at timber production but not for biomass in Mediterranean conditions. Others, such as those selected in Italy specifically for biomass production (currently provisionally admitted), are being tested for the first time under different soil and climatic conditions in Southwest Europe. The early selection of clones for rapid juvenile growth provides a valuable additional input to the clonal selection process, especially where very short rotations are desired (no more than 3 years). In any case, determining clonal stability in terms of growth is of great use not only when deciding on the clones to be used in plantations but also when developing breeding programs. ANOVA and Genotype plus Genotype × Environment (GGE) biplot analyses were used to analyse the growth and stability of the clones, which were then ranked according to mean performance and stability. Differences were detected between clones as well as between the different environments tested. The biplot analysis allowed different groups of clones to be identified according to their performance and degree of interaction displayed, thus providing useful information for the selection process. The production of aboveground biomass in the first vegetative period ranged from 1.7 to 8.0 Mg DM ha−1 at the different sites. ‘Monviso’, ‘Guardi’, ‘AF2’ and ‘2000 verde’ were the most productive clones whereas ‘Unal’, ‘Pegaso’ and ‘USA 49-177’ were the least productive. The stability analysis identified ‘AF2’, ‘Guardi’, ‘I-214’ and ‘MC’ as more stable clones while ‘Monviso’, ‘2000 verde’, ‘Unal’, ‘Pegaso’ and ‘USA 49-177’ were found to be specifically adapted to certain environments. This implies that where information on site conditions is not available, the ‘AF2’ and ‘Guardi’ clones offer greater assurance of successful establishment and higher initial growth. The growth of ‘Monviso’ ‘2000 verde’ ‘Unal’ ‘Pegaso’ and ‘USA 49-177’ clones is highly dependent on site conditions during the establishment phase. Similarly, the SH (Shore Henares river) and LT (La Tallada) sites were identified as the most highly discriminative environments for the set of clones while CS (Cubo de la Solana) and AR (Atarfe) were identified as those where performance levels were average.  相似文献

A five-year experiment (2002–2006) was conducted to determine apple (cv ‘Golden Smoothee’) and pear (cv ‘Conference’) crop coefficients (Kc) using two large weighing lysimeters. Daily reference evapotranspiration (ETo) and crop evapotranspiration (ETc) were evaluated. Midday canopy light interception of both crops planted in hedgerows, 4 × 1.6 m, was determined on a weekly basis from bud-break until leaf fall from year 2002 (fourth after planting) to year 2006 of both plantations. Relationships between canopy light interception and calculated Kc (ETc/ETo) were evaluated from bud-break until harvest. There were differences in Kc values between apple and pear trees. When daily Kc values from bud-break until harvest were adjusted to hyperbolic functions each year, adjusted curves for pear trees were very similar regardless of year (maximum Kc around 1.0). In apple trees, the maximum values of Kc increased over time, from 0.49 in 2002 to 1.04 in 2006. Midday light interception in both apple and pear trees increased during the 5 years of experiment from 29.0 to 45.6% in apples and from 27.5 to 41.6% in pears in midsummer. Although there was a significant positive correlation between midday canopy light interception and Kc in apple and pear trees, in different times within a specific year, these relationships were different between crops. While the apple data fitted into the same equation regardless of the year, different equations were needed to fit the pear data in different years. This discrepancy may have been related to differences in the canopy properties between apple and pear trees. Pear canopies had higher porosity than apple canopies and thus improved light penetration. Apple trees were more vigorous and produced taller and denser canopies. Pear Kc values were greatly influenced by the evaporative demands of different years and consequently differences in midday canopy light interception did not adequately reflect the differences in Kc across the two species.  相似文献

Fruit thinning in pear is feasible for mitigation of water stress effects. However, it is not well known how fruit quality at harvest and after cold storage is affected by pre-harvest water stress. Even less is known about the effects of fruit thinning on quality under these circumstances. To elucidate these, we applied deficit irrigation (DI) and fruit thinning treatments to ‘Conference’ pear over the growing seasons of 2008 and 2009. At the onset of Stage II (80 and 67 days before harvest in 2008 and 2009, respectively), two irrigation treatments were applied: full irrigation (FI) and DI. FI trees received 100% of crop evapotranspiration (ETc). DI trees received no irrigation during the first three weeks of Stage II to induce water stress, but then received 20% of ETc to ensure tree survival. From bud-break until the onset of Stage II and during post-harvest, FI and DI trees received 100% of ETc. Each irrigation treatment received two thinning levels: no thinning leaving commercial crop load (∼180 fruits tree−1), and hand-thinning at the onset of Stage II leaving a light crop load (∼85 fruits tree−1). Under commercial crop loads, DI trees were moderately water-stressed and this had some positive effects on fruit quality. DI increased fruit firmness (FF), soluble solids concentrations (SSC) and acidity at harvest while no changes were observed in fruit maturity (based on ethylene production). Differences in FF and acidity at harvest between FI and DI fruit were maintained during cold storage. DI also reduced fruit weight loss during storage. But fruit size was reduced under DI. Fruit thinning under DI resulted in better fruit composition with no detrimental effect on fresh-market yield compared to un-thinned fruit. Fruit size at harvest and SSC values after five months of cold storage were higher in fruit from thinned trees than fruit from un-thinned trees. Fruit thinning increased fruit ethylene production, indicating advanced maturity. This may lead to earlier harvest which is desirable in years with impending drought. Fruit thinning is therefore a useful technique to enhance pear marketability under water shortage.  相似文献
