Analysis was made of dsRNA in 37 cultivars and species of Ribes, that were healthy, naturally affected with the virus-like diseases, blackcurrant yellows, blackcurrant infectious variegation, gooseberry veinbanding or blackcurrant reversion, or graft-inoculated with scions from such diseased plants. Various dsRNA species, differing in size (from ca. 2 to 11kbp), number and staining intensity in gels, were detected in some or all assays of all plants, including those held as virus-tested stock. In different plant tissues from individual plants, the dsRNA species were usually similar in size and number but, in some sources, the dsRNA profile from flowers and/or bark differed greatly from the profiles of dsRNA obtained from leaves. No dsRNA species were associated consistently with any of these diseases. A 499kbp cDNA probe was obtained that in Northern blot analysis was specific to a ca. 5kbp dsRNA species present in the blackcurrant cv. Baldwin. It also detected a similarly sized dsRNA species in plants of many other blackcurrant cultivars, but it did not react with a similarly sized dsRNA species in redcurrant and gooseberry tissues. The 156 amino acid sequence encoded by the cDNA was very similar to sequences in the RNA-directed RNA polymerases of virus species in the family Totiviridae, especially Saccharomyces cerevisiae viruses L-1 and L-A. The significance of these findings and the possible origin of these dsRNA species are discussed.  相似文献

The clinical, radiographic, ultrasonographic, computed tomographic, surgical and histopathological findings in a Boxer dog with retroperitoneal hemangiosarcoma are described in this study. A seven-year-old, male, castrated Boxer dog was referred for evaluation of chronic hindlimb lameness. The physical examination revealed muscle atrophy and sciatic nerve deficits. Radiography and ultrasonography revealed a caudodorsal abdominal mass. Computed tomography revealed that the mass involved the left margin of the L7 vertebra, lumbosacral canal, and lumbosacral plexus. At surgery, a large retroperitoneal haematoma was removed. Histopathology of amorphous tissue found near the haematoma was consistent with haemangiosarcoma. The owner declined any further treatment. Ten weeks after discharge, the dog was euthanatized due to collapse and haemo-abdomen.  相似文献

Objective: To report the manifestations, history, and pathophysiologic basis of disease in 2 dogs with Amanita muscaria toxicosis. Case summaries: Two dogs were evaluated for an acute onset of gastroenteritis and seizures. A. muscaria toxicosis was suspected in each dog after confirmation of environmental exposure and visualization of ingested mushrooms in vomitus. The diagnosis was confirmed following identification of toxic Amanita metabolites in the urine and serum of each dog. Administration of supportive and symptomatic therapies resulted in the complete recovery of each animal. Unique information provided: Ingestion of the mushroom, A. muscaria, by dogs can result in acute gastrointestinal distress that precedes a potentially life‐threatening central neurologic syndrome characterized by seizures, tremors, and somnolence. Central nervous system dysfunction results primarily from the actions of ibotenic acid and its decarboxylation product, muscimol, which are analogues of the neurotransmitters glutamate and γ‐aminobutyric acid (GABA), respectively. Identification of these toxins in the urine and serum of affected dogs using high‐performance liquid chromatography (HPLC) provides a definitive diagnosis.  相似文献

This study investigated the potential of seed transmission of Cape St. Paul wilt disease (CSPWD) in coconuts. PCR amplification was used to assess the distribution of phytoplasmas in parts of West African Tall (WAT) palms infected with CSPWD. Employing phytoplasma universal primer pair P1/P7 in standard PCR, or followed with a nested PCR using CSPWD–specific primer pair G813f/AwkaSR, phytoplasma infection was detected in the trunks, peduncles, spikelets, male and female flowers of four infected WAT coconut palms. Through nested PCR, phytoplasma was also detected in four of 19 embryo DNA samples extracted individually from fruits harvested from three of the four infected palms and was confirmed as CSPWD by cloning and sequencing. Subsequently, CSPWD phytoplasma was again detected in five of 33 embryos from nine infected palms, and in one of eight fruits from two symptomless palms. Fruits from infected palms recorded higher percentage germinations in two field nurseries (average of 71·0%) compared to fruits from healthy palms (average of 57·6%), and matured fruits that had dropped from infected palms showed the same levels of germination as those harvested directly from the palms. This indicates that infected fruits retain the ability to germinate whether harvested or dropped. No phytoplasmas were detected in any of the resulting seedlings and plantlets obtained through embryo in-vitro culture. Therefore, although phytoplasma DNA can be detected in embryos, there is as yet no evidence that the pathogen is seed transmitted through to the seedling to cause disease in progeny palms.  相似文献

The southwest Australian floristic region (SWAFR) is an internationally recognized 'hot spot' of global biodiversity and has an endangered flora. It represents a unique interface between an ancient ecosystem and a recent agroecosystem, providing the opportunity to investigate encounters where the recipient of the virus is an introduced crop and the donor a native plant and vice versa. Phylogenetic analysis of the virus coat-protein genes was used to study isolates of three potyviruses representing different 'new encounter' scenarios at this interface. The incidence, symptomatology, host range, non-persistent aphid transmission and considerable genetic diversity of the first indigenous virus described from the SWAFR, where it infects the native legume Hardenbergia comptoniana , and its potential to damage lupin, a locally important, newly introduced cultivated grain legume, was studied. The name Hardenbergia mosaic virus is proposed for this virus. Two other examples of 'new encounter' scenarios involving other legume-infecting potyviruses studied were: Passion fruit woodiness virus , which has been found only in Australasia, where it damages recently introduced species of Passiflora and legumes; and Bean yellow mosaic virus, which is not indigenous to Australia and was introduced recently to the SWAFR, where it infects a number of introduced legumes, but also damages the local native legume Kennedia prostrata . Isolates of the former had considerable genetic diversity consistent with the virus being indigenous, while isolates of the latter virus from K. prostrata had a low genetic diversity consistent with recent arrival. This research illustrates how introduced viruses can damage indigenous plants and indigenous viruses can damage introduced cultivated plants within this unique ecosystem, and how human activities can facilitate damaging 'new encounters' between plants and viruses.  相似文献

The postoperative analgesia and sedation in cats given carprofen (4·0 mg/kg bodyweight by subcutaneous injection preoperatively) was compared to that in cats given pethidine (3·3 mg/kg bodyweight by intramuscular injection postoperatively) in a controlled, randomised, blinded, multicentre clinical trial. Further dosing with the particular analgesic was allowed if a cat was exhibiting unacceptable pain. In total, 57 carprofen cases and 59 pethidine cases were evaluated. Significantly fewer cats in the carprofen group required additional doses of analgesic, and mean pain scores were significantly lower from four hours after ovariohysterectomy, and at 18 to 24 hours after castration, compared to the pethidine group. In conclusion, carprofen provided as good a level of postoperative analgesia as pethidine, but of a longer duration (at least 24 hours) and was well tolerated. It thus provides an option for 'pre-emptive analgesia' in cats about to undergo surgery.  相似文献

AIM: The purpose of this study was to obtain information on aspects of dogs' lifestyle and diet that may be related to flatulence, whether the dogs were flatulent or not and if the owners were concerned about flatulence of their pet. METHODS: The owners of 110 pet dogs were randomly selected from the Massey University clinic tiles and asked to complete a questionnaire relating to their dog's flatulence, lifestyle and diet. RESULTS: Flatulence was detected by 47 owners and occurred more often in less active inside dogs than those exercised more often. No individual food or dietary association was identified. Nineteen of the 47 owners of flatulent dogs would alter their dog's diet if that change would reduce flatulence. CONCLUSION: Flatulence occurs in pet dogs and most owners accept flatulence and were unconcerned about its consequence.  相似文献
