Asian bears face major threats due to the impact of human activities as well as a critical lack of knowledge about their status, distribution and needs for survival. Once abundant in northern Pakistan, the Himalayan brown bear (Ursus arctos isabellinus) has been exterminated in most of its former distribution range. It presently occurs sparsely, in small populations, the Deosai National Park supporting the largest isolate. This decline might imply a reduction in genetic diversity, compromising the survival of the population. Using a combination of fecal DNA analysis and field data, our study aimed at assessing the size and genetic status of the Deosai population and give guidelines for its conservation and management. Using fecal genetic analysis, we estimated the population to be 40-50 bears, which compares well with the field census of 38 bears. The northern Pakistani brown bear population may have undergone an approximate 200-300-fold decrease during the last thousand years, probably due to glaciations and the influence of growing human population. However, in spite of the presence of a bottleneck genetic signature, the Deosai population has a moderate level of genetic diversity and is not at immediate risk of inbreeding depression. Gene flow might exist with adjacent populations. We recommend careful monitoring of this population in the future both with field observations and genetic analyses, including sampling of adjacent populations to assess incoming gene flow. The connectivity with adjacent populations in Pakistan and India will be of prime importance for the long-term survival of Deosai bears. 相似文献
Abstract In an attempt to identify appropriate feeding rates for multispecies of fish raised in fertilized earthen ponds, the present work was conducted over a 19‐week experimental period to establish the growth performance, production and body composition of Nile tilapia, common carp and silver carp fed 0.0%, 0.5%, 1.0%, 3.0%, 5.0% biomass and to apparent satiation (treatments). Twelve ponds were stocked with a similar number and weight of each fish species. Two ponds were assigned to each of the treatments, and a 25% protein pelleted fish feed was used to feed fish at the specified rate of feeding. At the end of the experiment, growth, weight gain, survival, yield and body composition of fish groups were affected by the treatments. The economic effectiveness also varied among treatments. The most conspicuous attribute of the feeding rates was its lack of influence on growth (g day?1), weight gain (g per fish), yield (kg ha?1) or body composition of silver carp. The results of whole‐body proximate analysis indicated that various feeding rates had either an irregular pattern or no effects on the protein and ash gain per 100 g of fish body weight (bw) gain. The most notable exceptions were significant (P < 0.05) increases in body fat and gross energy gains in Nile tilapia, common carp and silver carp accompanied by decreases in percentages of moisture (but not in silver carp) as feeding rate increased. Among the six different feeding levels, feeding to apparent satiation (feed amount was equivalent to 2.67% of fish bw day?1) appeared to be optimal, as it significantly (P < 0.05) supported the highest fish production, income and net profit compared with all other treatments except for the 3% feeding level, for which the differences in those measurements were comparable. 相似文献
We investigate Sardinella aurita and Sardinella maderensis recruitment success relative to the variability of oceanographic conditions in Senegalese waters using generalized additive models (GAM). Results show that recruitment of both species is marked by a strong intra‐annual (seasonal) variation with minimum and maximum in winter and summer, respectively. Their interannual variations are synchronous until 2006 (recruitment decreasing), while from 2007 there is no synchrony. The model developed shows that sardinella recruitment variability is closely related to the tested environmental variables in the study area. However, the key environmental variables influencing the recruitment success are different for both species: the Coastal Upwelling Index and the sea surface temperature for S. aurita and S. maderensis, respectively. We report that recruitment success of S. aurita and S. maderensis are associated with distinct ranges of sea surface temperature, upwelling intensity, wind‐induced turbulence, concentration of chlorophyll‐a and north Atlantic oscillation index. Considering food security and socio‐economic importance of both stocks, we recommend that consideration is given to the environmental variability in the small pelagic fish national management plans, particularly in the context of climate change. 相似文献
Olive leaf is known to have the high polyphenol content of 6–9% in dry weight. We investigated the effects of olive leaf powder (OLP) supplemented to fish feed on muscle protein of red sea bream (Pagrus major). Fish reared with feed containing 8% OLP for 40 days had 1.4 times higher myofibril content and 2.2 times higher acid-soluble collagen content than fish reared with control feed for the same period. On the other hand, sarcoplasmic protein content and collagenase activity of the muscle were almost the same between the control fish and OLP-diet fish. Microstructure observation of fish muscle showed that OLP-diet fish has more rigid endomysium structure than that of the control-diet fish. Since collagen fiber in endomysium is responsible for the texture of the muscle, feeding OLP to aquaculture fish will lead to a harder muscle texture. The present study suggests that OLP is a useful feed additive to enhance the texture of aquaculture red sea bream muscle through strengthening of the collagen structure in the muscle. 相似文献
Knowledge of crop production in suboptimal environmental conditions not only helps to sustain crop production but also aids in the design of low-input systems. The objective of this study was to evaluate the effects of water stress imposed at low-sensitive growth stages (vegetative, reproductive, and both vegetative and reproductive) and level of nitrogen (N) supply (100 and 200 kg ha−1) on the physiological and agronomic characteristics of two hybrids of maize (Zea mays L.). A two-site field experiment was carried out using a randomized complete block design with three replications and a split-factorial arrangement. A water deficit (WD) was induced by withholding irrigation at different stages of crop development. The results showed that proline content increased and the relative water content, leaf greenness, 100-kernel weight and grain yield decreased under conditions of WD. The highest IWUE was obtained when maize endured WD at vegetative stage at two sites. The limited irrigation imposed on maize during reproductive stage resulted in more yield reduction than that during vegetative stage, compared with fully irrigated treatment. The 100-kernel weight was the most sensitive yield component to determine the yield variation in maize plant when the WD treatments were imposed in low-sensitive growth stages. The results of the statistical regression analysis showed liner relationships between RGR during a period bracketing the V8 or R3 stages and 100-kernel weight in all the WD treatments. The increase of N supply improved yield and IWUE when maize plant endured once irrigation shortage at vegetative stage. But, the performance of high N fertilizer reduced and eliminated when water deficit imposed once at reproductive stage and twice at vegetative and reproductive stages, respectively. Arid areas are highly sensitive to climate change and are ideal model systems to study the potential impact of climate change on species' community structure. Biological soil crust (BSC) formation plays an ecological role in a number of key processes in the development of dry ecosystems. It was hypothesized that BSC succession and function are affected by aridity level and limited by rainfall. Furthermore, it is possible to infer the direction of the BSC succession based on aridity level, and the latter can imitate future climate change scenarios. The objectives of this study were to investigate the microbial biomass and diversity of the BSC structure in three sites differing in aridity level (semiarid, arid and hyper-arid), by combining physical and biophysiological measurements with 16S rRNA gene fragment and phospholipid fatty acid (PLFA) analyses. Physical and biophysiological parameters of the BSC were significantly influenced by aridity level. Total protein and polysaccharide contents were strongly correlated with total PLFA-based microbial biomass. Gram-positive biomarkers and microbial biomass were significantly higher in the wettest (semiarid) site than in the driest (hyper-arid) one. Multivariate-analysis based ordination of the PLFA data segregated the cluster of semiarid data from that of the hyper-arid site, while data from the arid site were dispersed between the two. The phylogenetic distribution of prominent 16S rRNA bacterial gene sequences along the aridity levels was in agreement with the PLFA analysis: the hyper-arid site was dominated by the cyanobacterium Microcoleus vaginatus, while diverse populations of cyanobacteria and soil bacteria were found in the other sites. These complementary tools allowed a simple and sensitive measurement of the influence of aridity levels on BSC successional stage. The results demonstrate that different aridity levels correspond to different BSC successional stages and those differences can be used as parameters for global change scenarios. 相似文献
Background: Spinal cord has a limited capacity to repair; therefore, medical interventions are necessary for treatment of injuries. Transplantation of Schwann cells has shown a great promising result for spinal cord injury (SCI). However, harvesting Schwann cell has been limited due to donor morbidity and limited expansion capacity. Furthermore, accessible sources such as bone marrow stem cells have drawn attentions to themselves. Therefore, this study was designed to evaluate the effect of bone marrow-derived Schwann cell on functional recovery in adult rats after injury. Methods: Mesenchymal stem cells were cultured from adult rats’ bone marrow and induced into Schwann cells in vitro. Differentiation was confirmed by immunocytochemistry and RT-PCR. Next, Schwann cells were seeded into collagen scaffolds and engrafted in 3 mm lateral hemisection defects. For 8 weeks, motor and sensory improvements were assessed by open field locomotor scale, narrow beam, and tail flick tests. Afterwards, lesioned spinal cord was evaluated by conventional histology and immunohistochemistry. Results: In vitro observations showed that differentiated cells had Schwann cell morphology and markers. In this study, we had four groups (n = 10 each): laminectomy, control, scaffold and scaffold + Schwann cells. Locomotor and sensory scores of cell grafted group were significantly better than control and scaffold groups. In histology, axonal regeneration and remyelination were better than control and scaffold groups. Conclusion: This study demonstrates that bone marrow-derived Schwann cells can be considered as a cell source for Schwann cells in SCI treatment. Bluetongue virus (BTV), a member of Orbivirus genus (family Reoviridae), is a non-contagious infection of domestic and wild ruminants. The current study was designed to detect various serotypes of BTV in small ruminants of Khyber Pakhtunkhwa (KPK) province of Pakistan, along with their effects on hemato-biochemical parameters. A total of 408 serum samples in four districts (Mansehra, Abbottabad, Swabi, and Kohat) of KPK from small ruminants were screened based on competitive ELISA (cELISA). A total of 204 (50%) samples were found positive for BTV group–specific antibodies. The seropositive samples were processed for the detection of BTV serotypes through real-time polymerase chain reaction (qPCR). Out of 204 cELISA-positive samples, 60 (29.41%) were found positive through qPCR. Three serotypes [6, 8, 9] were detected from Mansehra District and two from Kohat [2, 8] and Abbottabad [6, 8], while only one from Swabi [8]. The serotype “8” was found consistently in all the four study districts. A significant (p?<?0.05) increase in the level of blood urea nitrogen (BUN) and alkaline phosphatase (ALP) was recorded in goats, whereas aspartate aminotransferase (AST) in sheep infected with BTV, compared to healthy animals. The hematological parameters showed significantly (p?<?0.05) raised total leucocyte count (TLC) in both sheep and goats, whereas only hematocrit (HCT) value was increased significantly (p?<?0.05) in infected sheep. This is the first report on serotyping of BTV among small ruminants in Pakistan.
