BACKGROUND: Recombinant feline interferon-omega (rFeIFN-omega) was tested as a treatment option for cats with fibrosarcoma to assess safety and feasibility. HYPOTHESIS: Treatment with rFeIFN-omega in cats with fibrosarcoma is safe and feasible. ANIMALS: Twenty domestic cats. METHODS: In an open-labeled uncontrolled clinical trial 12 injections of 1 x 10(6) U/kg rFeIFN-omega were administered over a 5-week period: the 1st through 4th injections were given intratumorally, and the 5th through 12th injections were administered subcutaneously at the tumor excision site. Wide surgical excision of the tumors was carried out after the 4th injection and before the 5th injection of rFeIFN-omega. A Common Terminology Criteria for Adverse Events (CTCAE) analysis was conducted. Flow cytometry of fibrosarcoma cells after incubation with rFeIFN-omega and recombinant feline interferon-gamma was performed to assess the biological effect of rFeIFN-omega. RESULTS: Changes in blood cell count, increases in serum aspartate-amino-transferase activity, serum bilirubin concentration, serum creatinine and serum electrolyte concentrations, weight loss, anorexia, increased body temperature, and reduced general condition were observed but were mostly minor (grade 1 and 2) and self limiting. Eosinophilia (P = .025), neutropenia (P = .021), and weight loss (P < .001) were statistically correlated with rFeIFN-omega-treatment (analysis of parameters before treatment and after 3 injections of rFeIFN-omega). Flow cytometry of 5 unrelated feline fibrosarcoma cell lines showed increased expression of major histocompatibility complex (MHC) class I molecules (P = .026) in response to in vitro incubation with rFeIFN-omega, whereas expression of MHC class II molecules was not affected significantly. CONCLUSIONS AND CLINICAL IMPORTANCE: RFeIFN-omega for the treatment of feline fibrosarcoma is safe, well tolerated, and can be easily performed in practice. We measured a series of physiological and physical indicators and compared them to xylem sap flow, to identify the most sensitive and reliable plant water status indicator. In the growing season of 1998, 4-year-old peach trees (Prunus persica Batsch cv. 'Suncrest', grafted on 'GF 677' rootstock) were studied under two irrigation treatments, 25 l dayу and no irrigation, and during recovery. Trials were conducted near Pisa (Italy) in a peach orchard situated on a medium clay loam soil and equipped with a drip-irrigation system (four 4 l hу drippers per tree). Measurements of leaf water potential (W), stem water potential (S), and leaf temperature (Tl) were taken over 5 days (from dawn to sunset) and analyzed in conjunction with climatic data, sap flow (SF), trunk diameter fluctuation (TDF) and soil water content (SWC). Physiological indicators showed substantial differences in sensitivity. The first indication of changes in water status was the decrease of stem radial growth. TDF and SF revealed significant differences between the two irrigation treatments even in the absence of differences in pre-dawn leaf water potential (pdW), up until now widely accepted as the benchmark of water status indicators. Irrigated trees showed a typical trend in SF rate during the day, while in non-irrigated plants the maximum peak of transpiration was anticipated. Measurements of water potential showed S to be a better indicator of tree water status than W. Tl was found to have poor sensitivity. In conclusion, we found the sensitivity of the indicators from the most to the least was: TDF >SF rate >SF cumulated = pdW=S>mdW>Tl. 相似文献

Recent community based actions to ensure the sustainability of irrigation and protection of associated ecosystems in the Murrumbidgee Irrigation Area (MIA) of Australia has seen the implementation of a regional Land and Water Management Plan. This aims to improve land and water management within the irrigation area and minimise downstream impacts associated with irrigation. One of the plan objectives is to decrease current salt loads generated from subsurface drainage in perennial horticulture within the area from 20 000 tonnes/year to 17 000 tonnes/year. In order to meet such objectives Controlled Water table Management (CWM) is being investigated as a possible ‘Best Management Practice’, to reduce drainage volumes and salt loads.During 2000–2002 a trial was conducted on a 15 ha subsurface drained vineyard. This compared a traditional unmanaged subsurface drainage system with a controlled drainage system utilizing weirs to maintain water tables and changes in irrigation scheduling to maximize the potential crop use of a shallow water table. Drainage volumes, salt loads and water table elevations throughout the field were monitored to investigate the effects of controlled drainage on drain flows and salt loads.Results from the experiment showed that controlled drainage significantly reduced drainage volumes and salt loads compared to unmanaged systems. However, there were marked increases in soil salinity which will need to be carefully monitored and managed. 相似文献

A field experiment comparing different irrigation frequencies and soil matric potential thresholds on potato evapotranspiration (ET), yield (Y) and water-use efficiency (WUE) was carried out in a loam soil. The experiment included five treatments for soil matric potential: F1 (-15 kPa), F2 (-25 kPa), F3 (-35 kPa), F4 (-45 kPa) and F5 (-55 kPa) and six treatments for irrigation frequency: N1 (once every day), N2 (once every 2 days), N3 (once every 3 days), N4 (once every 4 days), N6 (once every 6 days) and N8 (once every 8 days). Results indicate that both soil matric potential and drip irrigation frequency influenced potato ET, Y and WUE. Potato ET increased as irrigation frequency and soil matric potential increased. Comparing soil water potential, the highest ET was 63.4 mm (32.1%) more than the lowest value. Based on irrigation frequency treatments, the highest ET was 36.7 mm (19.2%) more than the lowest value. Potato Y and WUE were also found to increase as irrigation frequency increased. Potato Y increased with an increase in soil water potential then started to decrease. The highest Y and WUE values were achieved with a soil matric potential threshold of -25 kPa and an irrigation frequency of once a day.Communicated by J. Ayars 相似文献

Timing of supply of irrigation water is an important measure of water management performance. Yet despite this widely accepted importance of timeliness, few studies have attempted to quantify this performance indicator. This paper suggests a methodology for quantifying timeliness. The indices thus generated are used to compare the performance of some types of smallholder irrigation schemes in Zimbabwe. Results indicate that applying measures of timeliness helps to assess water management practices across scheme types.The methodology given in this paper differentiates between timely irrigation deliveries which meet Crop Water Requirements (CWR) and surplus water supplies due to poor timeliness which cannot be used by the crop, hence denoting wastage. The timeliness indices provide more information regarding management practices than simple measures of total water applications over a given season. 相似文献

Oral stimulation with high-tastant concentrations that are alternared with low-tastant concentrations or water rinses (pulsatile stimulation) results in taste intensity ratings that are higher than continuous stimulation with the same average tastant concentration. This study tested the combined effects of taste pulsation rate and viscosity on pulsation-induced taste enhancement in apple juice. According to a tastant-kinetics hypothesis, less pulsation-induced taste enhancement is expected at enhanced pulsation rates in the high-viscous proximal stimulus compared to lower viscous stimuli. High-concentration sucrose apple juice pulses and low-concentration sucrose apple juice intervals were alternated at different pulsation periods (pulse + interval in seconds) every 2.5 s (period length = 5 s) or every 1.25 s (period length = 2.5 s). Pulsed stimuli were presented at two viscosity levels by the addition of pectin (0 and 10 g/L). Sweetness intensities of pulsed stimuli were compared to a continuous reference of the same net but nonalternating sucrose concentration. Sweetness ratings were higher for pulsatile stimuli than for continuous stimuli. In low-viscous stimuli, enhancement depended on the pulsation period and peaked at 5 s periods. In high-viscous stimuli, the same enhancement was observed for both pulsation periods. These results contradict a tastant-kinetics hypothesis of viscosity-induced taste suppression because impaired tastant kinetics by viscosity would predict the opposite: lower pulsation-induced taste enhancement for viscous stimuli, especially at higher pulsation rates. Instead, these observations favor an explanation based on perceptual texture-taste interactions, which predict the observed independence between viscosity and pulsation rate. 相似文献

Purpose
The low conductivity of sediments for mass and electron transport is the most severe limiting factor in sediment microbial fuel cells (SMFCs), so that sediment ameliorations yielded more remarkable effects than electrode improvements. The objective of this research was to enhance the electricity generation of SMFCs with amendments of biochar to freshwater sediments for conductivity enhancement.Materials and methods
Laboratory-scale SMFCs were constructed and biochars were produced from coconut shells at different temperatures. Variations in the power output, electrode potential, internal resistance, total organic carbon (TOC) content, and microbial communities were measured.Results and discussion
Amending with biochar reduced the charge transfer resistances of SMFCs and enriched the Firmicutes (mainly Fusibacter sp.) in the sediment, which improved the SMFC power generation by two- to tenfold and enhanced the TOC removal rate by 1.7- to fourfold relative to those without the amendment.Conclusions
The results suggested that biochar amendment is a promising strategy to enhance SMFC power production, and the electrical conductivity of biochar should be considered important when interpreting the impact biochar has on the electrical performance of soil or freshwater sediment MFCs.78.

Nutrients constrain the soil carbon cycle in tropical forests, but we lack knowledge on how these constraints vary within the soil microbial community. Here, we used in situ fertilization in a montane tropical forest and in two lowland tropical forests on contrasting soil types to test the principal hypothesis that there are different nutrient constraints to different groups of microorganisms during the decomposition of cellulose. We also tested the hypotheses that decomposers shift from nitrogen to phosphorus constraints from montane to lowland forests, respectively, and are further constrained by potassium and sodium deficiency in the western Amazon. Cellulose and nutrients (nitrogen, phosphorus, potassium, sodium, and combined) were added to soils in situ, and microbial growth on cellulose (phospholipid fatty acids and ergosterol) and respiration were measured. Microbial growth on cellulose after single nutrient additions was highest following nitrogen addition for fungi, suggesting nitrogen as the primary limiting nutrient for cellulose decomposition. This was observed at all sites, with no clear shift in nutrient constraints to decomposition between lowland and montane sites. We also observed positive respiration and fungal growth responses to sodium and potassium addition at one of the lowland sites. However, when phosphorus was added, and especially when added in combination with other nutrients, bacterial growth was highest, suggesting that bacteria out-compete fungi for nitrogen where phosphorus is abundant. In summary, nitrogen constrains fungal growth and cellulose decomposition in both lowland and montane tropical forest soils, but additional nutrients may also be of critical importance in determining the balance between fungal and bacterial decomposition of cellulose. 相似文献

The integrated modelling of coupled socio-ecological systems in land-change science requires innovative model concepts capable of grasping the interrelations between socioeconomic and natural components. Here, we discuss the integrated socio-ecological model SERD (Simulation of Ecological Compatibility of Regional Development) that was developed for the municipality of Reichraming in Upper Austria in a participative 2-year process involving local stakeholders. SERD includes three main components: (1) an agent-based actors module that simulates decisions of farmsteads, the municipal administration and other important actors; (2) a spatially explicit (GIS based) land-use module that simulates land-use change at the level of individual parcels of land and (3) an integrated socio-ecological stock-flow module that simulates carbon and nitrogen flows through both socioeconomic and ecological system compartments. We report on outcomes of a scenario analysis that outlines possible future trajectories depending on both external (e.g. agricultural subsidies and prices) and internal (e.g. innovation, willingness to co-operate) factors. We find that both external and internal factors can affect the behaviour of the integrated system considerably. Local and regional policies are found to be able to counteract adverse global socioeconomic conditions to some extent, but not to reverse the trend altogether. We also find strong interdependencies between socioeconomic and ecological components of the system. Fully evaluating these interdependencies is, however, not possible at the local scale alone and will require explicit consideration of higher-level effects in future research. 相似文献

Florida is the largest producer of fresh-market tomatoes in the United States. Production areas are typically intensively managed with high inputs of fertilizer and irrigation. The objectives of this 3-year field study were to evaluate the interaction between N-fertilizer rates and irrigation scheduling on yield, irrigation water use efficiency (iWUE) and root distribution of tomato cultivated in a plastic mulched/drip irrigated production systems. Experimental treatments included three irrigation scheduling regimes and three N-rates (176, 220 and 230 kg ha−1). Irrigation treatments included were: (1) SUR (surface drip irrigation) both irrigation and fertigation line placed right underneath the plastic mulch; (2) SDI (subsurface drip irrigation) where the irrigation line was placed 0.15 m below the fertigation line which was located on top of the bed; and (3) TIME (conventional control) with irrigation and fertigation lines placed as in SUR and irrigation being applied once a day. Except for the “TIME” treatment all irrigation treatments were controlled by soil moisture sensor (SMS)-based irrigation set at 10% volumetric water content which was allotted five irrigation windows daily and bypassed events if the soil water content exceeded the established threshold. Average marketable fruit yields were 28, 56 and 79 Mg ha−1 for years 1-3, respectively. The SUR treatment required 15-51% less irrigation water when compared to TIME treatments, while the reductions in irrigation water use for SDI were 7-29%. Tomato yield was 11-80% higher for the SUR and SDI treatments than TIME where as N-rate did not affect yield. Root concentration was greatest in the vicinity of the irrigation and fertigation drip lines for all irrigation treatments. At the beginning of reproductive phase about 70-75% of the total root length density (RLD) was concentrated in the 0-15 cm soil layer while 15-20% of the roots were found in the 15-30 cm layer. Corresponding RLD distribution values during the reproductive phase were 68% and 22%, respectively. Root distribution in the soil profile thus appears to be mainly driven by development stage, soil moisture and nutrient availability. It is concluded that use of SDI and SMS-based systems consistently increased tomato yields while greatly improving irrigation water use efficiency and thereby reduced both irrigation water use and potential N leaching. 相似文献
