Varroa destructor is considered a major reason for high loss rate of Western honey bee (Apis mellifera) colonies. To prevent colony losses caused by V. destructor, it is necessary to actively manage the mite population. Beekeepers, particularly commercial beekeepers, have few alternative treatments other than synthetic acaricides to control the parasite, resulting in intensive treatment regimens that led to the evolution of resistance in mite populations. To investigate the mechanism of the resistance to amitraz detected in V. destructor mites from French and U.S. apiaries, we identified and characterized octopamine and tyramine receptors (the known targets of amitraz) in this species. The comparison of sequences obtained from mites collected from different apiaries with different treatment regimens, showed that the amino acid substitutions N87S or Y215H in the OctβR were associated with treatment failures reported in French or U.S. apiaries, respectively. Based on our findings, we have developed and tested two high throughput diagnostic assays based on TaqMan technology able to accurately detect mites carrying the mutations in this receptor. This valuable information may be of help for beekeepers when selecting the most suitable acaricide to manage V. destructor.

This paper describes a conceptual framework that was developed to integrate livestock, human, cultivation and forest constraints to model community fuelwood consumption over a 25-year planning horizon. This framework was constructed as an energy balance based on human caloric requirements in order to examine the effects of household-level decisions for nutrition, fuelwood and land use. A scenario from a virtual community in Uganda is presented to illustrate the utility of this model to allow rapid policy and scenario evaluation. User-defined inputs combined with published research data are used in simulating resource responses and energy consumption rates. This model is a potential tool to monitor fuelwood consumption and to understand the implications of various land-use practices.

This paper reviews the scientific literature addressing the environmental fate and nontarget effects of the Cry protein toxins from Bacillus thuringiensis (Bt), specifically resulting from their expression in transgenic crops. Published literature on analytical methodologies for the detection and quantification of the Cry proteins in environmental matrices is also reviewed, with discussion of the adequacy of the techniques for determining the persistence and mobility of the Bt proteins. In general, assessment of the nontarget effects of Bt protein toxins indicates that there is a low level of hazard to most groups of nontarget organisms, although some investigations are of limited ecological relevance. Some published reports on the persistence of the proteins in soil show short half-lives, whereas others show low-level residues lasting for many months. Improvements in analytical methods will allow a more complete understanding of the fate and significance of Bt proteins in the environment.  相似文献

Field studies using open-top chambers were conducted at USDA-BARC involving the growth of soybeans ('89 & '90), wheat ('91 & '92), and corn ('91), under increased concentrations of atmospheric CO2 and O3. Treatment responses were compared in all cases to plants grown in charcoal-filtered (CF) air (seasonal 7-h mean = 25±3 n mol O3 mol–1) having 350 or 500 mol CO2 mol–1. Elevated seasonal O3 levels for the soybean, wheat, and corn studies averaged 72.2±4, 62.7±2, and 70.2 n mol O3 mol–1, respectively. Results presented were obtained for plants grown in silt loam soil under well-watered conditions. Grain yield increases in response to elevated CO2 in the absence of O3 stress averaged 9.0, 12.0, and 1.0% for soybean, wheat, and corn; respectively. Reductions in grain yields in response to the elevated O3 treatments at 350 mol CO, mol–1 averaged 20.0, 29.0 and 13.0% for soybean, wheat, and corn, respectively. Reductions in grain yields in response to elevated O3 at 500 mol CO2 mol–1 averaged 20.0, 8.0, and 7.0% for soybean, wheat, and corn, respectively. Dry biomass and harvest index in wheat were significantly reduced by O3 stress at 350 mol mol 1 CO2 but not at 500 u mol mol–1 CO2. Seed weight 1000–1 for scybeans and wheat was significantly increased by CO2 enrichment and decreased by O3 stress. Over the last decade, the ecosystem services (ESs) framework has been increasingly used to support mapping and assessment studies for sustainable land management purposes. Previous analysis of practical applications has revealed the significance of the spatial scale at which input data are obtained. This issue is particularly problematic with soil data that are often unavailable or available only at coarse scales or resolutions in various part of the world. In this context, four soil-based ecosystem services, namely biomass provision, water provision, global climate regulation, and water quality regulation, are assessed using three conventional soil maps at the 1:1,000,000, 1:250,000 and 1:50,000 scales. The resulting individual and joint ES maps are then compared to examine the effects of changing the spatial scale of soil data on the ES levels and spatial patterns. ES levels are finally aggregated to landforms, land use, or administrative levels in order to try to identify the determinants of the sensitivity of ES levels to change in the scale of input soil data. Whereas the three soil maps turn out to be equally useful whenever ESs levels averaged over the whole 100 km2 territory are needed, the maps at the 1:1,000,000 and 1:250,000 induced biases in the assessment of ESs levels over spatial units smaller than 100 and 10 km2, respectively. The simplification of the diversity and spatial distribution of soils at the two coarsest scales indeed resulted in local differences in ES levels ranging from several 10 to several 100%. Identification of the optimal representation of soil diversity and distribution to obtain a reliable representation of ESs spatial distribution is not straightforward. The ESs sensitivity to scale effect is indeed context-specific, variable among individual ESs, and not directly or simply linked with the soil typological diversity represented in soil maps. Forested and natural lands in the study area appear particularly sensitive to soil data scales as they occupy marginal soils showing very specific ESs signatures.  相似文献

Zygophyllum xanthoxylum is a salt‐accumulating xerophytic species with excellent adaptability to adverse environments. Previous studies demonstrated that Z. xanthoxylum absorbs a great quantity of Na+ as an osmoregulatory substance under arid conditions. To investigate the nutritional status of Z. xanthoxylum in comparison with a typical glycophyte, Arabidopsis thaliana, seedlings were exposed to NaCl (50 mM for Z. xanthoxylum and 5 mM for A. thaliana), osmotic stress (–0.5 MPa), and osmotic stress combined with the NaCl treatment. Compared to the control, NaCl treatment or osmotic stress significantly increased Na+ concentration in leaves and roots of Z. xanthoxylum, but not of A. thaliana. Under osmotic stress, the addition of NaCl significantly increased Na+ concentration in leaves and roots of Z. xanthoxylum, resulting in improved biomass and tissue water content. However, such changes were not observed in A. thaliana. Compared to the control, K+ concentrations in leaves and roots remained unchanged in Z. xanthoxylum when exposed to osmotic stress, with or without additional 50 mM NaCl. In contrast, significant reductions in shoot K+ concentrations of A. thaliana were observed under osmotic stress alone or when combined with 5 mM NaCl. Moreover, NaCl alone or when combined with osmotic stress enhanced the accumulation of N, P, Fe, Si, Ca2+, and Mg2+ in Z. xanthoxylum, but did not cause such nutritional changes in A. thaliana. Compared to the glycophyte A. thaliana, Z. xanthoxylum could accumulate Na+ and maintain the stability of nutritional status at a relatively constant level to cope with drought stress.  相似文献

A 12-year-old Morgan gelding was examined for colic of 3 days duration. Signs of depression, colic, diarrhea, and endotoxemia persisted despite aggressive medical therapy and surgical exploration. Culture results from gastric fluid and feces yielded many colonies of Clostridium perfringens. This organism also was recovered from peritoneal fluid 10 days after admission; consequently, the horse was euthanized. At necropsy, a localized gas-filled, necrotic stomach wall was found; many mucosal and submucosal gas blebs were visible. Culture of this tissue yielded Clostridium perfringens. Emphysematous gastritis is a fulminant infection of the stomach wall caused by gas-forming organisms that gain access to the submucosa via mucosal defects such as ulcers. This condition has been reported infrequently in people, and the case reported herein represents the first instance of emphysematous gastritis in the horse.  相似文献

Skeletal deformities, a major contributing factor to economic losses in market-age turkeys, may be associated with the poult malabsorption syndrome. A study was performed to determine whether the naturally occurring malabsorption syndrome produces skeletal lesions in turkeys at market age. Poults were placed on litter on which poults had previously developed malabsorption. Exposed poults developed enteric disease with 21% mortality during the first 3 weeks. Controls had no enteric disease and no mortality. At the end of the study, 20-week-old exposed turkeys weighed 0.621 kg (5%) less than controls and had a higher incidence of angular limb deformities with significantly higher angulation scores. Exposed turkeys also had a greater incidence of rotated tibias and bowed tibias. The mechanical property of tibial shear strength was significantly lower in turkeys that had survived the earlier poult malabsorption syndrome.  相似文献
