Headspace solid-phase microextraction coupled to gas chromatography/ion trap mass spectrometry-65 microm polydimethylsiloxane/divinylbenzene (PDMS/DVB) was used to identify and monitor the emission patterns of biogenic volatile organic compounds from leaves of Eucalyptus dunnii, Eucalyptus saligna, and Eucalyptus citriodora in situ. Short extractions (1 min) were performed every 30 min for periods of 8-10 h during 24 days taking advantage of the high capacity of this porous polymer coating. Forty-two compounds were detected and 20 identified in the headspace of E. saligna leaves, and 19 of 27 compounds were identified in the headspace of E. dunnii leaves. The emission pattern of (E)-beta-ocimene and rose oxide suggests that they may play a bioactive role in Eucalyptus. 相似文献

Headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography and ion-trap mass spectrometry has been used to identify biogenic volatile organic compounds present in the headspace of chopped leaves of Eucalyptus (E.) dunnii, E. citriodora, and E. saligna. A simple HS-SPME method entailing 30 min of extraction at 30 degrees C was developed for this purpose. Thirty compounds were identified in the headspace of 60 juvenile chopped Eucalyptus leaves, and another 30 were tentatively identified. The presence of compounds such as (E)-4,8-dimethyl-1,3,7-nonatriene (DMNT), (E,E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene (TMNT), (E,E)-alpha-farnesene, (E,E,E)-3,7,11,15-tetramethyl-1,3,6,10,14-hexadecapentaene (TMHP), beta-caryophyllene, alpha-humulene, germacrene D, and beta-cubebene in the headspace of the leaves but not in the essential oils from the same Eucalyptus trees and information about the infochemical roles of some of these compounds in other living plant systems suggest they might play a bioactive role in Eucalyptus leaves. 相似文献

All commercial gasoline fuels build up deposits on the spark plugs, injectors, oxygen sensors, catalytic converter, and inside the combustion chamber, which will lower the engine's performance and increase air pollution. As a result, fuel-based detergents have been developed to prevent and remove unwanted deposits. Unfortunately, many of the detergents use high amounts of aromatic solvents, which result in a greater risk of exposure to aromatic compounds like benzene. In this study, car exhaust was analyzed for benzene, toluene, ethylbenzene, and xylenes (BTEX), as well as formaldehyde and acetaldehyde during engine cleaning service using different chemical cleaners. A special device was designed for sampling from car exhaust using solid phase microextraction. The extracted compounds were analyzed using a gas chromatograph with flame ionization detector. The cleaning products were rated with regard to the amount of pollutants produced during the cleaning service. 相似文献

The aim of this study was to evaluate the effect of the level of fat and selected fatty acids found in the milk of sows on the rearing of native breed piglets. Simultaneously, in order to improve the accuracy of the performed analyses, atomic absorption spectrometry was employed in the applied analytic methodology. The experimental animal material comprised 60 sows of the indigenous White Z?otnicka breed. Colostrum and milk were collected on the first and 14th days of lactation. In all, 240 samples were collected. The following parameters were determined in the course of the experiment: number and weight of piglets, body weight gains as well as deaths of piglets. A total of 1270 born piglets was subjected to investigations. The performed experiments demonstrated that, with the progress of the lactation period, the content of fat and saturated fatty acids (SFA) turned out to be statistically significant and showed a growing tendency. Fat increased by about 2% and palmitic acid (C16:0) increased most, that is by 5%. Linolic (C18:2) and linolenic (C18:3) acids revealed decreasing trends. Irrespective of the day of lactation, the level of unsaturated fatty acids (UFA) determined in sows' colostrum and milk was higher in comparison with that of SFA, and the UFA to SFA ratio ranged from 1.84% to 1.33%. Proportions of n‐6 to n‐3 fatty acids were determined at the level of about 1.6:1.0 in the colostrum and 1.3:10 in milk. The highest daily body weight gains were recorded in the case of piglets derived from sows with the highest fat level – 294 g, while in the case of stearic acid (C18:0), the smaller its concentration in the colostrum and milk of the experimental sows, the better body weight gains of piglets – 262 g. At the same time, stearic acid (C18:0) was found to exert a statistically significant effect on piglet mortality at the level of P ≤ 0.05. Its highest concentration caused the highest proportion of deaths among piglets ‐ 16.23%. The performed analysis of correlations that occurred between fat, fatty acids and traits associated with piglet rearing confirmed that linolic acid (C18:2; n‐6) was highly significantly correlated with piglets' body weights (r = 0.456**) and was negatively correlated with piglets' deaths (r = ?0.312). On the other hand, fat revealed correlation with body weight gains of piglets (r = 0.333*_ and a negative correlation with deaths of piglets (r = ?0344*). Recapitulating, the results of the performed experiments revealed that differences in the levels of fat and fatty acids found in sows' colostrum and milk influenced results of piglet rearing. Together with the increase in the content of fat and UFA in sows' colostrum and milk, piglets were characterized by the best body weight, growth rate, as well as by small mortality. 相似文献

Background
The aim of the study was to demonstrate the immunohistochemical expression of proteins that affect the metastatic potential of a tumour, including matrix metalloproteinase 2 (MMP-2) and E-cadherin. Another objective was to determine their correlation with the expression of the Ki-67 antigen in metastasizing and non-metastasizing mammary carcinomas in female dogs. The study was conducted on 32 canine mammary carcinomas (12 metastatic and 20 non-metastatic), classified as simple tubular and tubulopapillary carcinomas. Immunohistochemistry was performed to evaluate the expression of MMP-2, E-cadherin and Ki-67 antigen.Results
MMP-2 was expressed in 85 % of the non-metastatic tumours and in all the metastatic tumours, while E-cadherin was expressed in 85 % of the non-metastatic tumours and in 66 % of the metastatic tumours. The Ki-67 antigen was expressed in 65 % of the non-metastatic tumours and in 91 % of the metastatic tumours. The mean Ki-67 expression was slightly higher in tumours that had metastasized (1.5?±?0.90 vs 1.1?±?0.94; p?=?0.22). A similar relationship was found in terms of the intensity of the MMP-2 expression (2.9?±?1.9 vs 2.7?±?2.4; p?=?0.50). A decrease in the expression of E-cadherin (2.8?±?2.5) was found in metastatic tumours compared to the expression in non-metastatic tumours (3.2?±?2.3). However, these differences were not statistically significant (p?=?0.63).Conclusion
We did not show significant differences in MMP-2, E-cadherin and Ki-67 expression between metastatic and non-metastatic tumours due to low number of cases studied, however further experiments are necessary to assess the role of these antigens in the process of canine mammary tumours metastasis.66.

Wild ruminants are thought to serve as natural hosts for Rift Valley fever virus (RVFV) but the role of these animals as reservoirs for RVFV during inter-epidemic periods and as amplifiers during epidemics is not well understood. An indirect enzyme-linked immunoassay (I-ELISA) based on the recombinant nucleocapsid protein (rNp) of RVFV was validated for the detection of specific IgG antibodies in African buffalo. Data sets derived from testing buffalo sera from Kenya (n=405) and South Africa (n=618) were dichotomised according to the results of a virus neutralisation test. The assay characteristic performance was analysed using threshold values optimised by the two-graph receiver operating characteristics (TG-ROC) analysis, and by mean plus two, as well as by mean plus three standard deviations derived from I-ELISA PP values in uninfected animals. Among 1023 buffalo sera tested, 77 (7.5%) had detectable virus neutralising antibodies. The assay had high intra- and inter-plate repeatability in routine runs. At a cut-off optimised by the TG-ROC at 95% accuracy level, the diagnostic sensitivity of the I-ELISA was 98.7% and diagnostic specificity 99.36% while estimates for the Youden's index (J) and efficiency (Ef) were 0.98 and 99.31%. When cut-off values determined by traditional statistical approaches were used, the diagnostic sensitivity was 100% but estimates of J, Ef and other combined measures of diagnostic accuracy were lower compared to those based on cut-off value derived from the TG-ROC. Results of the study indicate that the I-ELISA based on the rNp would be useful for seroepidemiological studies of RVFV infections in African buffalo. 相似文献

Context
Waterlogging is predicted to become more common in boreal forests during winter and early spring with climate change. So far, little is known about the waterlogging tolerance of boreal tree species during their winter dormancy.Aim
The aim was to quantify the degree of waterlogging tolerance of 1-year-old dormant Norway spruce (Picea abies (L.) Karst.) seedlings.Methods
The seedlings were exposed to waterlogging in a growth chamber at temperature of 2 °C for 4 weeks and then allowed to recover for 6 weeks during the growth stage. Shoot and root responses were monitored by physiological and growth measurements.Results
No effect was found in the seedling biomass, but root mortality increased slightly during the early growth stage following waterlogging. The water potential of the needles became less negative at the end of the waterlogging and the early growth stage. The ratio of apoplastic to symplastic electrical resistance (R e/R i) of the needles was lower after waterlogging, indicating changes in the proportions of symplastic and apoplastic space. No differences were found between the treatments in the dark-acclimated chlorophyll fluorescence (F v/F m) of the needles. Slightly greater accumulation of starch and temporary reductions of some mineral nutrients in needles were found after waterlogging.Conclusions
We conclude that in late winter and early spring, Norway spruce seedlings potentially tolerate short periods of waterlogging. 相似文献68.
We determined the effects of removal of leaves, stem axillary buds, or the entire shoot on root suckering (adventitious shoot formation by roots) and basal stem sprouts in 3- and 4-year-old potted seedlings of aspen (Populus tremuloides Michx.). The greatest number of root suckers (67.9 +/- 8.5 per plant) emerged after excision of the entire shoot. Defoliated and debudded stems were the major source of inhibitory agents for root suckering, although axillary buds and developing new leaves also exerted a significant inhibitory effect. Removal of mature leaves had only a minor effect on root suckering. Removal of a continuous band of bark (girdling) at the base of the stem consistently stimulated growth of adventitious shoots from the stem below the girdle and occasionally promoted root suckering. Exogenous application of indole-3-acetic acid to excised stumps inhibited root suckering and basal stem sprouting. Naphthylphthalamic acid (NPA), an auxin polar transport inhibitor, had no effect on root suckering or stem sprouting when it was applied to the bark of the basal stem. However, NPA significantly increased root suckering when it was applied to the exposed surface of xylem after girdling. These results suggest that polar transport of auxin in the xylem parenchyma is an important inhibitor of root suckering. On decapitated stems, vacuum extraction of xylem sap from the root system lowered the frequency of root suckering compared with decapitation alone, indicating that substance(s) originating in the root system also play a significant role in controlling root suckering. 相似文献

Arbuscular mycorrhizal fungi(AMF) are known to facilitate the growth and vigour of many plants, particularly in arid ecosystems. In a survey of AMF in a date palm plantation and two natural sites of a desert in Oman, we generated many single spore-derived cultures of AMF. We identified a number of these isolates based on spore morphotyping and molecular phylogenetic analysis using the sequence of the LSU-rDNA. Here, we presented the characteristics of four species of AMF recovered, namely Claroideoglomus drummondii, Diversispora aurantia, Diversispora spurca and Funneliformis africanum. The four species have been described previously, but for the Arabian Peninsula they are reported here for the first time. Our endeavor of isolation and characterization of some AMF habituated to arid sites of Arabia represents a first step towards application for environmental conservation and sustainable agriculture in this region. 相似文献
