To determine whether glucocorticoids affect the function of the bovine corpus luteum (CL) during the estrous cycle and early pregnancy, we examined the effects of exogenous cortisol or reduced endogenous cortisol on the secretion of progesterone (P4) and on pregnancy rate. In preliminary experiments, doses of cortisol and metyrapone (an inhibitor of cortisol synthesis) were established (n=33). Cortisol in effective doses of 10 mg blocked tumor necrosis factor-induced prostaglandin F(2α) secretion as measured by its metabolite (PGFM) concentrations in the blood. Metyrapone in effective doses of 500 mg increased the P4 concentration. Thus, both reagents were then intravaginally applied in the chosen doses daily from Day 15 to 18 after estrus (Day 0) in noninseminated heifers (n=18) or after artificial insemination (n=36). Pregnancy was confirmed by transrectal ultrasonography between Days 28-30 after insemination. Plasma concentrations of P4 were lower in cortisol-treated heifers than in control heifers on Days 17 and 18 of the estrous cycle (P<0.05). However, the interestrus intervals were not different between control and cortisol-treated animals (P>0.05). Moreover, metyrapone increased P4 and prolonged the CL lifespan in comparison to control animals (P<0.05). Interestingly, in inseminated heifers, cortisol increased the pregnancy rate (75%) compared with control animals (58%), whereas metyrapone reduced the pregnancy rate to 16.7% (P<0.05). The overall results suggest that cortisol, depending on the physiological status of heifers (pregnant vs. nonpregnant), modulates CL function by influencing P4 secretion. Cortisol may have a positive influence on CL function during early pregnancy, leading to support of embryo implantation and resulting in higher rates of pregnancy in heifers.  相似文献

In order to investigate the genetic variation between Tritrichomonas foetus from bovine and feline origins, cysteine protease 8 (CP8) coding sequence was selected as the polymorphic DNA marker. Direct sequencing of CP8 coding sequence of T. foetus from four feline isolates and two bovine isolates with polymerase chain reaction successfully revealed conserved nucleotide polymorphisms between feline and bovine isolates. These results provide useful information for CP8-based molecular differentiation of T. foetus genotypes.  相似文献

Group B streptococci (GBS) were considered a major cause of mastitis in cattle until preventive measures succeeded in controlling the disease in the 1970s and 1980s. During the last 5-6 years an increasing number of cases have been observed in some Scandinavian countries. A total of 187 GBS isolates from mastitis cases were collected from 119 animals in 34 Norwegian farms in the period from April 2007 to November 2010. 133 (71%) of the isolates were from farms with automated milking systems. The strains underwent typing of capsular polysaccharides (CPS) and surface proteins, and were analyzed by multi-locus variable repeat assay (MLVA) to investigate the epidemiological relationship of strains within and between farms. The GBS strains were differentiated into 12 types by CPS and surface protein analysis, with CPS types V (54%) and IV (34%) predominating. MLVA was superior to CPS and protein typing for strain differentiation, resolving the 187 strains into 37 types. In 29 of 34 farms all GBS strains had identical MLVA profiles specific for each farm. However, in one farm represented with 48 isolates, four MLVA variants with differences in one repeat locus were observed during the almost 3-year long collection period. Similar variations were observed at four other farms. This might reflect the stability of repeat loci under in vivo conditions. Farms with automated milking systems were overrepresented in this material. In conclusion, the five-loci MLVA allowed rapid high-resolution genotyping of the bovine GBS strains within and between farms.  相似文献

The aim of this study was to evaluate the association between serum methylmalonic acid (MMA), a proposed marker of cellular cobalamin deficiency, and serum cobalamin concentrations in dogs. Serum samples from 555 dogs were grouped according to their serum cobalamin concentrations (<150 ng/L to 1000 ng/L). Additionally, serum samples were collected from 43 healthy dogs to calculate a reference interval for canine serum MMA. MMA was measured using a GC/MS method. Groups were compared using a Kruskal-Wallis test with Dunn's post test. Proportions of dogs above the upper limit of the reference interval were calculated and a χ2-test for trend was performed to evaluate the association between serum cobalamin and MMA concentrations. The reference interval for serum MMA was calculated to be 414.7-1192.5 nmol/L. Dogs with serum cobalamin concentrations <251 ng/L had significantly higher MMA concentrations (P<0.05) and the χ2-test for trend showed a trend for increasing serum MMA concentrations with decreasing serum cobalamin concentrations (P<0.0001). Additionally, a number of dogs with normal serum cobalamin concentrations had increased serum MMA concentrations, suggesting that some of these dogs may have cobalamin deficiency on a cellular level. Further studies are warranted to determine if these dogs should receive cobalamin supplementation.  相似文献

In in vitro studies, the acyclic nucleoside phosphonate 9-(2-phosphonylmethoxypropyl)-2,6-diaminopurine (PMPDAP) inhibited the replication of feline immunodeficiency virus (FIV). No information about its clinical efficacy is available so far. The aim of this prospective placebo-controlled, double-blinded study was to evaluate the antiviral efficacy of PMPDAP in cats naturally infected with FIV. Twenty cats were randomly assigned to two treatment groups receiving either PMPDAP (25 mg/kg) or placebo twice per week subcutaneously for 6 weeks. The general health status (Karnofsky's score), clinical signs, laboratory, immunological, and surrogate parameters were evaluated. No significant differences were found between PMPDAP- and placebo-treated cats, although cats treated with PMPDAP showed a tendency for improvement in their Karnofsky's score and clinical signs. Haematological side effects were noted in the PMPDAP-treated cats. Thus, PMPDAP may be an option in treating cats if it becomes available for veterinarians, but side effects have been monitored.  相似文献

Wheat bran‐derived arabinoxylan‐oligosaccharides (AXOS) recently have been shown to potentially exert prebiotic effects. In this study, 15 bran samples obtained by milling different wheat cultivars were treated with xylanases from Hypocrea jecorina (XHJ), Aspergillus aculeatus (XAA), and Pseudoalteromonas haloplanktis (XPH) to assess the effect of bran source and xylanase properties on the AXOS yield and structure. The total arabinoxylan (AX) extraction yield was higher with XHJ (8.2–10.7%) and XAA (8.2–10.8%) than with XPH (6.9–9.5%). Irrespective of the enzyme, a significant negative correlation was observed between extraction yield and arabinose to xylose (A/X) ratio of bran AX (r = –0.7), but not between yield and bran AX level. The A/X ratio of the extracted material was 0.27–0.34 for all bran samples and all enzymes, which combined with yield data and microscopic analysis, indicated primary hydrolysis of aleurone and nucellar epidermis AX. The average degree of polymerization (avDP) of the extracted AX was very low for all enzymes (2–3), owing to the release of high levels of monomeric arabinose and xylose. The release of these monosaccharides could be ascribed to 1) the activity of wheat bran‐associated enzymes (arabinofuranosidases and xylosidases); 2) the hydrolytic properties of the xylanases themselves; and 3) the presence of xylosidases as contaminations in enzyme preparation, in that order of importance. Heat treatment of bran before xylanase treatment significantly decreased the levels of monomeric arabinose and xylose in the extract, without affecting the extraction yield, resulting in a higher avDP of 3–7, thus yielding true AXOS. Overall, for AXOS production, wheat cultivars with a low bran A/X ratio of the AX are preferable as starting materials, and inactivation of bran‐associated enzymes before incubation is desirable. The XHJ xylanase was the best enzyme for wheat bran‐derived AXOS production.  相似文献

OBJECTIVE: To determine the effect of two doses of fentanyl, administered transdermally, on the minimum alveolar concentration (MAC) of isoflurane in cats. STUDY DESIGN: Prospective, randomized study. ANIMALS: Five healthy, spayed, female cats. METHODS: Each cat was studied thrice with at least 2 weeks between each study. In study 1, the baseline isoflurane MAC was determined in triplicate for each cat. In studies 2 and 3, isoflurane MAC was determined 24 hours after placement of either a 25 or 50 microg hour(-1) fentanyl patch. In each MAC study, cats were instrumented to allow collection of arterial blood and measurement of arterial blood pressure. Twenty-four hours prior to studies 2 and 3, a catheter was placed and secured in the jugular vein and either a 25 or 50 microg hour(-1) fentanyl patch was placed in random order on the left thorax. Blood samples for plasma fentanyl determination were collected prior to patch placement and at regular intervals up to 144 hours. After determination of MAC in studies 2 and 3, naloxone was administered as a bolus dose (0.1 mg kg(-1)) followed by an infusion (1 mg kg(-1) hour(-1)) and MAC redetermined. RESULTS: The baseline isoflurane MAC was 1.51 +/- 0.21% (mean +/- SD). Fentanyl (25 and 50 micro g hour(-1)) administered transdermally significantly reduced MAC to 1.25 +/- 0.26 and 1.22 +/- 0.16%, respectively. These MAC reductions were not significantly different from each other. Isoflurane MAC determined during administration of fentanyl 25 micro g hour(-1) and naloxone (1.44 +/- 0.16%) and fentanyl 50 micro g hour(-1) and naloxone (1.51 +/- 0.19%) was not significantly different from baseline MAC (1.51 +/- 0.21%). CONCLUSIONS AND CLINICAL RELEVANCE: Fentanyl patches are placed to provide long-lasting analgesia. In order to be effective postoperatively, fentanyl patches must be placed prior to surgery. Plasma fentanyl concentrations achieved intraoperatively decrease the need for potent inhalant anesthetics in cats.  相似文献

To determine if bedding has any influence on the time horses spend recumbent, 8 horses kept on straw and 8 kept on wood shavings were observed from 10:00 to 5:30 for two successive nights. Observations were conducted using time-lapse video recordings. Lying down and rising behavior, as well as frequency and duration of bouts spent in lateral and sternal recumbency, was registered. The results showed that horses on straw were lying in lateral recumbency three times longer than horses on shavings (P < .001), whereas the time horses spent in sternal recumbency did not differ. The longest period of noninterrupted lateral recumbency was longer for horses on straw than for those on shavings. Because horses must lie down, preferably in lateral recumbency, to achieve paradoxical sleep, the reduced time spent in lateral recumbency in horses on wood shavings may affect their welfare and performance. Independent of the bedding, we further observed that, as the horses got up from recumbency, most of them made attempts to roll over before rising. This behavior appeared to be caused by some difficulty in rising, possibly due to the box size, and might have a connection with the fact that horses sometimes get stuck against the box wall.

Introduction

Many riding horses spend the majority of their life in an artificial environment. Horse owners keep their horses under certain conditions because of tradition, because they want to make the horse feel comfortable from a human point of view, or to reduce the amount of work involved in horse husbandry. Often the choice of bedding substrate is made from a subjective point of view without assessing both short-term and long-term effects of the bedding. Part of the reason is that only few studies have analyzed horses' preferences for different bedding substrates and their effect on the time horses spend recumbent. In one study comparing straw and wood shavings, no significant preference was found.[1] In another study comparing plastic, wheat straw, and wood shavings, the time horses spent standing, sleeping, or lying down was not affected significantly by the bedding substrates. [2] Mills et al [3] found that horses, given a choice between straw and wood shavings, spent significantly more time on straw. Whereas the substrates had no significant effect on behaviors such as eating, lying, and standing alert, horses spent more time performing bedding-directed behaviors on straw but more time dozing on shavings. Finally, it has been reported that the use of nonstraw bedding may increase the risk of abnormal behaviors such as weaving. [4]As far as bedding properties are concerned, Airaksinen et al[5] concluded that air quality in the stable and utilization of manure can be improved by selecting a good bedding material. According to Reed and Redhead, [6] both straw and shavings are economical and easy to obtain, and they make a bright, comfortable bed. Straw bales are convenient to store, but may be eaten by the horse, are labor intensive, and may be dusty or contain fungal spores. Wood shavings are not eaten by the horse and are good for respiratory problems but need to be kept very clean because they are porous. In addition, they are not as warm as straw because they do not trap air the way straw does.Electroencephalographic (EEG) studies in cats have demonstrated that sleep can be divided into two stages of differing electrocorticographic (EcoG) patterns, ie, slow-wave-sleep (SWS) and paradoxical sleep (PS).[7] During PS, bursts of rapid eye movements (REM) can be seen at irregular intervals. [8] In humans, dreaming occurs during this stage. [9 and 10] Horses are able to sleep while standing, [11] but in this position they only go into SWS. [14, 15 and 16] During PS there is a complete abolition of muscular tone of antigravity muscles and of neck muscles, as shown in cats. [17] In horses, there is a gradual loss of muscular tone until the middle of the recorded SWS period, whence it decreases to a negligible amount during PS. [15] Consequently, muscular tone disappears entirely at the onset of PS. [18] Horses are unable to complete a sleeping cycle without lying down to enter PS. [8, 19 and 20] They normally fall asleep while standing and, when they feel confident about their environment, lie down in sternocostal recumbency. [8] Thereafter, they proceed to lateral recumbency and enter PS. [14 and 19] Dallaire and Ruckebusch [18] demonstrated that the SWS state was infrequent in the standing animal and most often occurred during sternocostal recumbency with the head resting or not on the ground. PS occurred in both sternocostal and lateral recumbency, although the animal frequently had to readjust its position into sternocostal recumbency due to the disappearance of neck muscular tone.The sleep pattern of horses depends on many circumstances, such as age,[21, 22 and 23] diet, [16] and familiarity with the environment. When horses are put outdoors it may take some days before they lie down. If one horse that is familiar with the environment lies down, the others usually follow. [8 and 13] Dallaire and Ruckebusch [16] subjected three horses to a four-day period of perceptual (visual and auditive) deprivation. After this period total sleep time increased due to an augmentation of both SWS and PS. Finally, there is large individual variation between horses in the time they spend recumbent and sleeping. [15]Horses spend 11% to 20% of the total time in recumbency.[11 and 15] Lateral recumbency represents about 20% of total recumbency time, and uninterrupted periods of lateral recumbency vary from 1 to 13 minutes (mean, 4.6 min). [14 and 16] Steinhart [11] found that the mean length of uninterrupted lateral recumbency periods was 23 minutes, the longest period being one hour. Total sleeping time in the stabled horse averages 3 to 5 hours per day or 15% of the total time. [8, 13 and 16] Keiper and Keenan [24] found similar time budgets in feral horses that were recumbent approximately 26% of the night. PS is about 17% to 25% of total sleeping time, and the mean length of a single PS period is 4 to 4.8 minutes. [13 and 18]In stabled horses sleep is mainly nocturnal and occurs during three to seven periods during the night.[8, 13 and 16] Ruckebusch [13] observed that neither sleep nor recumbency occurred during daytime in three ponies observed for a month and, in another experiment conducted on horses, PS occurred only during nighttime. [15] A group of ponies observed for more than a month between 8:45 and 4:45 spent only 1% of the daytime recumbent.[25] The maximum concentration of sleep occurs from 12:00 to 4:00 .[8, 16, 18 and 24]The purpose of this study was to examine two groups of horses in a familiar environment, one group kept on a bedding consisting of straw, and the other kept on wood shavings, and to determine if there was any difference between the two groups in the time they spend recumbent.

Materials and methods

Housing. The study was conducted in one of the biggest riding clubs in Denmark, housing about 150 horses. The 18 horses used in the study stood in three different parts of the stable. They were all stabled in boxes measuring 3 × 3 m and subjected to the same feeding and management routine. They were unable to see their next-door neighbor because of a tall wooden board, but they were able to see the horses stabled on the opposite side of the corridor through bars. Nine horses were stabled on wheat straw (15 cm long, dry matter content 87-88%) and nine on oven-dried wood shavings (80% spruce and 20% pine, dry matter content 82%).Animals. All horses used in the study were privately owned. They had been kept in the boxes in which they were observed a minimum of three weeks. Three of the horses were mares and 15 were geldings. Most of them were Danish Warmblood used for dressage riding. Their ages ranged from 5 to 18 years (mean, 10.6 y) and their height ranged from 1.60 to 1.76 m (mean, 1.68 m). All horses wore a blanket.
