OBJECTIVE: To investigate the physiologic endocrine effects of food intake and food withholding via measurement of the circulating concentrations of acylated ghrelin, growth hormone (GH), insulin-like growth factor-I (IGF-I), glucose, and insulin when food was administered at the usual time, after 1 day's withholding, after 3 days' withholding and after refeeding the next day in healthy Beagles. ANIMALS: 9 healthy Beagles. PROCEDURES: Blood samples were collected from 8:30 AM to 5 PM from Beagles when food was administered as usual at 10 AM, after 1 day's withholding, after 3 days' withholding, and after refeeding at 10 AM the next day. RESULTS: Overall mean plasma ghrelin concentrations were significantly lower when food was administered than after food withholding. Overall mean plasma GH and IGF-I concentrations did not differ significantly among the 4 periods. Circulating overall mean glucose and insulin concentrations were significantly higher after refeeding, compared with the 3 other periods. CONCLUSIONS AND CLINICAL RELEVANCE: In dogs, food withholding and food intake were associated with higher and lower circulating ghrelin concentrations, respectively, suggesting that, in dogs, ghrelin participates in the control of feeding behavior and energy homeostasis. Changes in plasma ghrelin concentrations were not associated with similar changes in plasma GH concentrations, whereas insulin and glucose concentrations appeared to change reciprocally with the ghrelin concentrations. 相似文献

Pear leaf scorch, the only Xylella fastidiosa-induced disease reported from Taiwan, was found in area where the variety Hengshan (Pyrus pyrifolia) was grown. Strains of pear leaf scorch Xyl. fastidiosa (XF-PLS) shared similarities to strains of other host origins in the requirement of complex medium and the exhibition of
rippled cell walls, however, recent serological and molecular biology studies showed difference among them. Five strains of
XF-PLS were compared with 20 other strains originally isolated from almond, oleander, pecan, plum, peach, mulberry, grapes,
citrus, coffee, and sycamore by sequence analyses of the 16S rRNA gene and 16S–23S rRNA internal transcribed spacer region
(ITS). When sequences of 16S rRNA gene based on fragment size of 1,537–1,540 bp were compared, the similarity index among
5 XF-PLS strains was 99.3–99.8%, whereas it was 97.8–98.6% between XF-PLS strains and strains from other hosts. When sequences
of 16S–23S rRNA ITS based on fragment size of 510–540 bp were compared, the similarity index among 5 XF-PLS strains was 99.0–100%,
whereas it was 80.7–82% between XF-PLS strains and strains from other hosts. Multiple sequence alignments led to the identification
of 5 polymorphic nucleotides in the 16S rRNA gene among the 25 Xyl. fastidiosa strains, and there were considerable variations in the nucleotide sequences of 16S–23S rRNA ITS between XF-PLS and the other
20 Xyl. fastidiosa strains. The phylogenetic trees revealed that XF-PLS strains were separated from strains of other hosts. Strains of other
hosts were divided into four subgroups: strains from (1) oleander, (2) grape, almond M23 and mulberry, (3) citrus and coffee,
and (4) pecan, peach, plum, sycamore and almond M12. Results indicate that XF-PLS strains were not closely related to the

Loose smut fungi of barley and wheat (Ustilago nuda and U. tritici, respectively) colonize the plant without causing obvious disease symptoms before heading. The availability of diagnostic methods to detect and follow the growth of these pathogens in the plant would therefore be highly advantageous for both resistance breeding and the development of effective seed treatments. Using seed lots of barley and wheat highly infected with loose smut, we studied the early establishment of the loose smut pathogens in the plant by fluorescence microscopy. In hand-cut sections stained with the fluorochrome Blankophor?, fungal hyphae were observed to invade the shoot apical meristem and leaf primordia during the first days after the onset of germination. At the first node stage the ear and leaf primordia were generally extensively colonized. Hyphae of U. nuda were also regularly observed in high density in the nodes. A protocol was developed for the specific amplification of U. nuda and U. tritici DNA extracted from infected plant tissue. PCR screening of U nuda in seedlings from infected and healthy seed lots was compared to ELISA, microscopy and ultimately head infection of mature plants derived from tillers of the tested seedlings. The results indicated that a prediction of loose smut infection by real-time PCR is possible at the second leaf stage, and that the assay is equally suited for use with spring and winter varieties of barley and wheat. 相似文献

During the last few decades, the physiological and genetic background of dormancy, and correlated pre-harvest sprouting (PHS) have been intensively investigated. Special attention has often been paid to genetic factors that may explain and predict PHS susceptible behaviour. A major candidate is the Vp1 gene which is involved in embryo development and maturation as well as in dormancy establishment. In this study, Vp1 gene expression during kernel development was studied in wheat, triticale and rye as a potential biomarker for selecting PHS tolerant varieties in cereal breeding programs. Plants of known PHS tolerant and PHS susceptible varieties were grown under controlled conditions from flowering until harvest ripeness. During that period, kernels were regularly harvested for RNA extraction and cDNA synthesis. Calibrated and normalized relative Vp1 expression levels were obtained in an RT-qPCR assay. During kernel development, Vp1 expression levels generally showed a typical peak during the soft dough stage, after which they decreased and remained low until harvest maturity. Differences in Vp1 expression levels could be observed between the PHS susceptible and PHS tolerant varieties of wheat, with the PHS tolerant variety showing higher levels of relative Vp1 expression compared to the PHS susceptible variety. In triticale, however, this difference was only seen once and could not be confirmed in further experiments. Life-span theories explain successful aging with an adaptive management of emotional experiences like regret. As opportunities to undo regrettable situations decline with age, a reduced engagement into these situations represents a potentially protective strategy to maintain well-being in older age. Yet, little is known about the underlying neurobiological mechanisms supporting this claim. We used a multimodal psychophysiological approach in combination with a sequential risk-taking task that induces the feeling of regret and investigated young as well as emotionally successfully and unsuccessfully (i.e., late-life depressed) aged participants. Responsiveness to regret was specifically reduced in successful aging paralleled by autonomic and frontostriatal characteristics indicating adaptive shifts in emotion regulation. Our results suggest that disengagement from regret reflects a critical resilience factor for emotional health in older age. 相似文献

Mortality of sows is a major problem for pig production worldwide. In this study, we used hierarchical multivariable logistic analyses to investigate different risk factors for mortality at the sow and herd level in herds with group-housed pregnant sows. Data included 3652 pregnant and 1266 lactating sows from 34 sow herds. A clinical examination for 16 clinical signs was carried out for each sow, and information about 16 herd related factors was obtained by interviews. Farm records were used to obtain information about whether or not sows died suddenly or were euthanized within 3 months after the clinical examination. Factors increasing the risk of sow mortality in the gestation unit were solid pen floors (OR = 1.87), presence of vulva bites (OR = 1.73) and unwillingness to stand when approached (OR = 1.62). Factors increasing the risk of sow mortality in the lactation unit were pale vulva color (OR = 12.69), body leanness (OR = 4.11), and presence of shoulder ulcers (OR = 2.89). The estimated between herd variation was small. Thus, the findings for the sow level variables may be generally applicable for sows in herds with group housed systems. 相似文献

Concerns exist that some uses of antimicrobials in cattle may lead to the emergence, proliferation, dissemination and persistence of resistant pathogenic bacteria in animal agriculture, which in turn can infect humans via the food supply. The degree of perceived risk varies with the clinical indication for which the antimicrobial in question is used. In this study, four uses of antimicrobials are considered, including in acutely sick, chronically sick, at-risk, and high-risk cattle, contrasting the degree of risk among these uses. Using a random sample of 103 feedlot cattle veterinarians and variables drawn from the theory of planned behavior, we predict differences in risk perception by clinical indication with differences in perceived efficacy of antimicrobials, social pressures to use antimicrobials, and moral obligations to use antimicrobials. In most models, veterinarians who perceived that others in the feedlot industry (i.e., other feedlot veterinarians, nutritionists, feedlot clients, and retained owners of cattle) were more likely to expect them to use antimicrobials in one situation versus another, the less likely those veterinarians perceived the risk of antimicrobial risk to be greater in the former versus the latter situation. Only two of these contrasts contained influences outside the immediate feedlot relationships. This exception involves the 'downstream' public: meat packers, retailers, and consumers. Veterinarians who believe that using antimicrobials for acutely sick cattle is more beneficial than using antimicrobials for chronically sick cattle were more likely to believe that antimicrobial resistance was a less probable outcome in acutely sick cattle than in chronically sick cattle. 相似文献
