The objective of this study was to validate the assessment of bull sperm morphology done by veterinary practitioners. Out of 1606 bulls, 1400 (87.2%) and 1344 (83.7%) were designated by practitioners and an experienced andrologist, respectively, as having > 70% morphologically normal sperm. Most organic carbon (C) in soils eventually turns into CO2 after passing through microbial metabolic pathways, while providing cells with energy and biosynthetic precursors. Therefore, detailed insight into these metabolic processes may help elucidate mechanisms of soil C cycling processes. Here, we describe a modeling approach to quantify the C flux through metabolic pathways by adding 1-13C and 2,3-13C pyruvate and 1-13C and U-13C glucose as metabolic tracers to intact soil microbial communities. The model calculates, assuming steady-state conditions and glucose as the only substrate, the reaction rates through glycolysis, Krebs cycle, pentose phosphate pathway, anaplerotic activity through pyruvate carboxylase, and various biosynthesis reactions. The model assumes a known and constant microbial proportional precursor demand, estimated from literature data. The model is parameterized with experimentally determined ratios of 13CO2 production from pyruvate and glucose isotopologue pairs. Model sensitivity analysis shows that metabolic flux patterns are especially responsive to changes in experimentally determined 13CO2 ratios from pyruvate and glucose. Calculated fluxes are far less sensitive to assumptions concerning microbial chemical and community composition. The calculated metabolic flux pattern for a young volcanic soil indicates significant pentose phosphate pathway activity in excess of pentose precursor demand and significant anaplerotic activity. These C flux patterns can be used to calculate C use efficiency, energy production and consumption for growth and maintenance purposes, substrate consumption, nitrogen demand, oxygen consumption, and microbial C isotope composition. The metabolic labeling and modeling methods may improve our ability to study the biochemistry and ecophysiology of intact and undisturbed soil microbial communities. 相似文献

The objective of this research was to evaluate comparative pharmacokinetics of doramectin in alpacas, after subcutaneous administration of 0.2 mg/kg dose. Six healthy adult alpacas, mean age of 5 years ± 1, (three female and three gelded males) of mean bodyweight of 62 kg ± 16 kg with an average body condition scored 2.8 ± 1 out of five, were used in this study. Serial blood samples were collected from the jugular vein before the administration until day 21 afterwards to establish the pharmacokinetics of doramectin after its subcutaneous administration at 0.2 mg/kg dose. The blood samples were analysed using high-performance liquid chromatography (HPLC), fluorescence detection method with precolumn derivatisation, validated for alpacas. The pharmacokinetic parameters were calculated using a noncompartmental model, and results showed Cmax (6.05 ± 5.34 ng/ml), Tmax (3.83 ± 2.48 days), AUC (62.12 ± 18.86 ng/ml × d), terminal half-life (6.2 ± 4.9 days) and MRT (11.56 ± 4.43 days). The results of this study showed that the Cmax and AUC were much lower than in cattle and sheep at the same dosage. Tmax remained similar to cattle and sheep. This study presents valuable information about pharmacokinetics of doramectin in alpacas, which can be utilised in its future efficacy studies. 相似文献

The 2020 Student Debates of the Entomological Society of America (ESA) were live-streamed during the Virtual Annual Meeting to debate current, prominent entomological issues of interest to members. The Student Debates Subcommittee of the National ESA Student Affairs Committee coordinated the student efforts throughout the year and hosted the live event. This year, four unbiased introductory speakers provided background for each debate topic while four multi-university teams were each assigned a debate topic under the theme ‘Technological Advances to Address Current Issues in Entomology’. The two debate topics selected were as follows: 1) What is the best taxonomic approach to identify and classify insects? and 2) What is the best current technology to address the locust swarms worldwide? Unbiased introduction speakers and debate teams began preparing approximately six months before the live event. During the live event, teams shared their critical thinking and practiced communication skills by defending their positions on either taxonomical identification and classification of insects or managing the damaging outbreaks of locusts in crops. 相似文献

The efficiency of composting to remove the pollutants DEHP, PAHs and LAS from organic waste products was investigated. Source separated MSW and sewage sludge, respectively, were composted at temperatures ranging from 35 to 70°C using a laboratory-scale reactor composting system. At regular intervals, compost samples were taken from the reactors for chemical analysis. DEHP disappeared rapidly during composting of MSW; 96 to 99% of the initial content was degraded within 25 days of composting regardless of process temperature. In sewage sludge, the highest degradation of 91% was reached at a composting temperature of 65°C. LAS degradation in sewage sludge was fastest at 55°C, but at both process temperatures tested 99% of the initial content had been removed within 25 days. Degradation of PAHs was less complete. However, in all composting experiments, with MSW as well as sewage sludge, the final PAH concentration was less than in the starting material. Further experiments suggested that less than 0.1% of the pollutants had evaporated. Thus, composting proved to be a promising technique to promote degradation of organic pollutants — especially DEHP and LAS — in MSW and sewage sludge. 相似文献

Two diazotrophic enterobacterial strains, Pantoea agglomerans D5/23 and Klebsiella pneumoniae CC12/12, were observed in sterile and non-sterile hydroponic and soil experiments in order to determine, by means of an immunological
detection method (double antibody sandwich enzyme linked immunosorbent assay), their colonization sites, their migration within
individuals of different plant species, and their ability to compete with indigenous organisms. To investigate the interaction
between bacteria and plants, root and shoot samples were analysed using electron microscopy. Field experiments were performed
to determine the growth-promoting abilities of the bacterial strains. In field experiments, inoculation with P. agglomerans led to an increase in the grain yield of different wheat (Triticum aestivum) cultivars. The same strain was also able to colonize the rhizosphere and the phyllosphere of different cereals due to its
ability to migrate within the plant. Roots and media were colonized 10–100 times more intensively than shoots, with about
106 cells g–1 root and 104 cells g–1 shoot. We found that P. agglomerans colonized the root and plant-growth medium of wheat to a greater extent than those of rye (Secale cereale) and barley (Hordeum vulgare), whereas the colonization of shoots was higher in rye and barley compared to wheat. Furthermore, while cell numbers of K. pneumoniae in media and roots were 10 times higher than cell numbers of P. agglomerans, only the latter markedly increased root growth. We were able to detect significant differences in colonization numbers between
treatments even if the data were not normally or log-normally distributed or the variances were not homogenous.
