Detailed studies were carried out on the influence of corn size distribution on the values obtained for diastatic power (DP) of commercially malted barley. The presence of the recombinant cp4 epsps gene from Roundup Ready (RR) corn and RR soybean was quantified using real-time PCR in soil samples from a field experiment growing RR and conventional corn and soybean in rotation. RR corn and RR soybean cp4 epsps persisted in soil for up to 1 year after seeding. The concentration of recombinant DNA in soil peaked in July and August in RR corn and RR soybean plots, respectively. A small fraction of soil samples from plots seeded with conventional crops contained recombinant DNA, suggesting transgene dispersal by means of natural process or agricultural practices. This research will aid in the understanding of the persistence of recombinant DNA in agricultural cropping systems.  相似文献

The purpose of this study was to determine the diurnal composition and concentration of volatile fatty acids (VFA) and to determine VFA composition and concentration differences between stomach compartment 1 (C1) and caecum of alpacas fed grass and alfalfa hay. The study was divided into two experiments. In Experiment 1 (EXP 1), 10 male alpacas (3+ years old, 65 kg BW) were divided into two groups, housed in drylot pens, provided ad libitum water and fed alfalfa (AH) or grass hay (GH) for 30 days. The alpacas were slaughtered and the digestive tract collected, divided into sub‐tract sections, weighed and digesta sampled for pH, dry matter (DM) and NDF. Volatile fatty acid composition and concentration were determined on C1 and caecal material. Four adult male (3+ years old, 60 kg BW), C1 fistulated alpacas were housed in metabolism crates and divided into two forage groups for Experiment 2 (EXP 2). Alpacas were fed the forages as in EXP 1. Diurnal C1 VFA samples were drawn at 1, 3, 6, 9, 12, 18 and 24 h post‐feeding. There were no differences between forages for tract weight, C1 and caecum digesta DM or NDF. Differences were noted (p < 0.05) for pH between forages and sub‐tract site. Volatile fatty acids concentrations were different (p < 0.05) for forage and site, and total VFA was higher for AH than GH (110.6 and 79.1 mm ) and C1 than caecum (40.7 and 27.6 mm ). Proportion of VFA was significant (p < 0.05) for forage and site, C1 acetate highest for GH (84.8 vs. 74.0 mm ) and caecum acetate 83.7 and 76.2 mm for GH and AH respectively. These data demonstrate the level of VFA produced in C1 and the caecum of alpacas and the diurnal VFA patterns. Composition of VFA is similar to other ruminant species.  相似文献

Paddlefish are gaining increasing acceptance as an aquaculture species worldwide. Commercial trout feeds, containing high protein and lipid levels, are currently used in intensive culture; however, nutritional requirements of paddlefish are not currently known. A study was conducted examining the effects on growth, survival and fillet composition of juvenile paddlefish when fed commercial feeds differing in protein and lipid levels. Paddlefish larvae were first stocked in 14.0 m3 round tanks and fed trout starter feeds for 43 days until trained to accept a 1.6 mm pellet. Paddlefish juveniles of mean weight (±SE) 20±0.27 g were randomly stocked into six0.02 ha ponds at 12 500 ha?1 and fed floating commercial trout or catfish (lower protein and lipid) feeds, twice daily (08:00 and 15:30 hours) for 92–97 days. At harvest, there were no significant differences in final weight, percent survival, specific growth rate , relative growth and feed conversion ratio between treatments, which averaged 223.6 g, 96.2%, 2.5% day?1, 10.2 and 1.98 respectively. Surface feeding activity index was significantly higher in ponds supplied with catfish feed than in ponds supplied with trout feeds. Relative pellet buoyancy was not a factor in feeding activity. Fulton's condition factor averaged0.238, was not significantly different, and was similar to a reported value for extensively cultured paddlefish (zooplanktivore). There was no significant difference in liver somatic index between treatments, which averaged 1.91%. Percent protein and moisture of fillets averaged 14.9% and 80.9%, respectively, and were not significantly different between treatments. However, lipid content of fillets was significantly higher in paddlefish fed the trout feed (4.45%), compared with paddlefish fed the catfish feed (2.42%). Fillet lipid content for both treatments was higher than reported values for extensively cultured paddlefish. Percent abdominal fat was significantly higher (0.82%) in paddlefish fed the trout feed compared with paddlefish fed the catfish feed (0.52%). Results from this study indicate that paddlefish can be fed a commercial catfish feed labeled to contain 32% protein and 4.5% lipid without adverse effects on growth, survival and fillet composition, lowering production costs.  相似文献

Plant protein sources were evaluated in 32% protein grow-out feeds for channel catfish (initial weight: 180 g/fish) stocked at high densities (24,700 fish/ha) in 0.04 ha earthen ponds. Each of the eight practical-type feeds was assigned for five replicate ponds. The fish were fed to satiation once daily for 170 d. Specifically, cottonseed meal and cottonseed meal plus supplemental lysine were evaluated as replacements for soybean meal. Soybean meal, a combination of soybean meal and cottonseed meal, or a combination of soybean meal and cottonseed meal plus supplemental lysine were evaluated as a substitute for animal protein sources. Based on weight pin, feed conversion ratio, body composition, percentage visceral fat, and dressed yield, the data indicated that cottonseed meal plus lysine can be used as a total substitute for soybean meal in catfish feeds. However, it is not recommended that more than 30% cottonseed meal be used in catfish feeds until additional data are available on the effects of gossypol on reproduction in catfish. Also, data indicated that plant proteins can be used as a total replacement for animal protein without detrimental effects. Reduced weight gain was observed in fish fed a feed that contained 68% of the established available lysine requirement. However, fish fed feeds estimated to contain only 76 or 82% of the available lysine requirement did not show reduced weight gain. This suggests that lysine may be more highly available from cottonseed meal than previously estimated, or that natural food organisms in the pond contributed nutrients including lysine, or that fish were able to consume enough of the marginally deficient feeds to meet their requirement for lysine. This study was conducted with large catfish fed a 32% protein feed to satiation once daily. If smaller fish, a lower protein fed, or a restricted feeding regimen had been used, the results may have been different.  相似文献

The nucleotide sequences of DNA fragments amplified by polymerase chain reaction (PCR) from four different genomic regions of nine red sea bream iridoviruses (RSIVs) isolated from different species of fish, different areas and in different years in Korea were compared with the reported reference sequences. One isolate, RSIV Namhae, showed 100% homology to the reference sequences, while the other eight isolates, which appeared to contain identical nucleotide sequences, showed 96.6–98.9% homology with reference sequences depending upon the target regions of PCR gene amplification. However, differences in nucleotide sequences were not apparent between the RSIVs isolated in different locations, in different years or in different host species. We also cloned and sequenced the 3′ end flanking region (K1) of the DNA polymerase (DPOL) gene using the cassette ligation-mediated PCR method. This sequence was 4436-bp long and possessed two open reading frames (ORF-1 and ORF-2) oriented in opposite directions. The putative proteins encoded by these two ORFs could not be characterized by comparison with the proteins of other species in the data banks. The presence of the ribonucleotide reductase small subunit (RNRS) gene at the 3′ end of the K1 region allowed us to determine that these two genes, RNRS and DPOL, are separated 5508 bp and oriented in the same direction in the genome of RSIV. Moreover, it is of interest that a PstI-restriction fragment, of which the sequence but not the location within the RSIV genome had previously been reported, is located at nucleotide positions from 1096 to 2054, extending from within the ORF-1 region, spanning the intervening sequence between ORF-1 and ORF-2, and extending into the ORF-2 region. Various repeating sequences up to 86 bp were present at the 3′ ends of ORFs, especially within the nucleotide sequences at the 3′ terminus of ORF-2. No similarities were detected when the DNA sequences of the K1 region were compared to the DNA sequences of a repetitive element in the genome of other iridoviruses.  相似文献

Previous soil-disc force prediction models have considered spherical and concave blades, as used on disc ploughs and harrows, with many simplifying assumptions. This study proposes an approach applying the fundamental equation of earth moving mechanics for calculating the soil passive reaction acting on a rotating fiat disc blade as used on a zero-till single disc seeder. The study considers the effects of disc variable depth of cut, sweep and tilt angles, free rotation and a scrubbing reaction at the beveled edge. This paper outlines the modeling approach and prediction results for a fixed circular blade operating at 90° sweep angle over a range of speeds and at two tilt angles. To account for the varying depth across the circular disc shape width, elemental wide blade force reactions acting on wide blade segments of elemental width were integrated across the disc working width. By including inertia forces due to speed as well as additional bulldozing forces due to the loose soil accumulation in front of the blade the draught and upward vertical forces acting on the vertical disc blade were predicted with a deviation of 6%-19% and 1.5%-14% from measured data, respectively. The model was able to predict the effect of increasing the tilt angle from 0° to 20° on reducing both draught and upward vertical forces. Further development and validation of the model will be described in subsequent papers, reflecting a step by step approach of increasing complexity to model a disc blade as used on a zero-till single disc seeding system.  相似文献
