Relationship between fish growth rate and progression of artificially induced erythrocytic inclusion body syndrome in coho salmon, Oncorhynchus kisutch (Walbaum)

Abstract. Studies were conducted to clarify the relationship between growth rate and disease progression of erythrocytic inclusion body syndrome (EIBS) in artificially infected coho salmon, Oncorhynchus kisutch (Walbaum). In the diseased state, the haematocrit values decreased and the number of erythrocytes with inclusions was higher in faster growing fish. Rapid growth was accompanied by an abundance of immature erythrocytes, which had the greatest incidence of inclusion bodies.     

Comparation on UV curing reactivity of Allyl abietate and allyl isopimarate北大核心CSCD

The UV curing reaction of the difference between allyl abietate(AA) and allyl isopimarate(AI) was studied. The conversion rate of the vinyl double bond was characterized and detected by FT-IR analysis. Results showed that conversion of double bonds are firstly increased then decreased with the increases of the dosage of initiators. Conversion of double bonds firstly increased and then leveling off with the increases of the illumination time, and AA is higher before the 200 s. AA is higher then AI under the same illumination intensity. Surface drying time of AA is shorter than AI, and relative curing speed become faster at the same conditions. Under the conditions of 2-hydroxy-2-methylpropiophenone(Darocure-1173)as photo initiator and dosage 6%, illumination time 200 s, illumination distance 4.5 cm and illumination intensity 100%, number average molecular weight of cure products of AA and AI are 4 176 and 9 407 with polymerisation degrees of 12 and 28, respectively. The obtained UV cured coating all have smooth and transparent appearance. It represented some superior properties in flexibility, mechanical impact strength, adhesive power, hardness, acid resistance, alkali resistance, salt resistance, water resistance, etc.. The hardness of AA and AI cured products are H and HB, respectively.     

Assessing the use of a dietary probiotic/prebiotic as an enhancer of spinefoot rabbitfish Siganus rivulatus survival and growth

The use of prebiotics and probiotics as feed supplements that improve efficiency of intestinal bacteria is becoming de rigueur in animal husbandry in many regions worldwide. We tested the effects of a commercial probiotic (Biogen^®) containing allicin, high unit hydrolytic enzyme, Bacillus subtilis spores and ginseng extracts on survival, growth, carcass composition and feed cost/benefit in rabbitfish Siganus rivulatus. Fifteen net cages (100 × 100 × 40 cm; L × W × H) were stocked with 10 juvenile rabbitfish (10.3 g per fish) each and placed in a large rectangular tank and offered feed at 4% body weight daily. Cages were offered one of five isonitrogenous and isocaloric diets containing 0, 1, 2, 3 and 4 g kg⁻¹ probiotic at three replicates per treatment for 98 days. Fish in all cages were weighed at 2‐week intervals and feed regimen was adjusted accordingly. Rabbitfish offered the control diet exhibited lower growth and feed utilization than all experimental treatments. There was no effect of probiotic inclusion level on survival but growth was better at all inclusion levels than in the control. No significant differences (P > 0.05) in growth were observed among fish groups fed various levels of the probiotic. Carcass composition was not affected by dietary probiotic inclusion. Ultimately, when all variables are considered, Biogen^® inclusion to diets appears to reduce feed cost per unit growth of rabbitfish.

Determination of allogeneic and xenogeneic markers in the genus of Tilapia. II. Identification of T. aurea,T. vulcani and T. nilotica by electrophoretic analysis of their serum proteins

Sera from Israeli stocks of T. nilotica, T. vulcani and T. aurea were analysed, by polyacrylamide gel electrophoresis, for the existence of species-specific markers. Markers situated in the fast region of the electropherogram were found to be male sex and species specific. Interspecies polymorphism in markers in the post albumin area (transferrins) permit the distinction between these related species of Tilapia. No significant polymorphism was found in the electropherogram of T. vulcani. T. aurea presented some slight variations in the transferrin zone, and T. nilotica displayed a high polymorphism everywhere along the electropherogram. Two other species specific bands were found in the prealbumin zone; bands 1 (specific for T. aurea) and 2 (specific for T. vulcani). A selection of fish stocks of T. aurea and T. vulcani is currently done now on the basis of this electrophoretic criteria and on the ability to produce a high percentage of male hybrids. It was concluded that the stock of T. nilotica which exist in Israel do not represent the pure species of T. nilotica. The failure to obtain 100% males in all paired interspecific mating (Pruginin et al., 1975) was explained on this basis.

Isolation of pituitary glycoprotein gonadotropins from the grass carp (Ctenopharyngodon idell)

Gonadotropins (GTH) were isolated from pituitary glands of grass carp and their properties investigated. Acetone-dried pituitaries were extracted with ammonium acetate-ethanol and were then passed through cation exchanger, anion exchanger, and gel filtration column. Two glycoprotein gonadotropins (DE I and DE II) were isolated of MW 56,000 and 53,000 daltons, respectively, as determined by gel filtration technique. Isoelectric points of the two GTHs also differed from each other. Sialic acid contents were 3.64% and 1.47% for DE I and DE II, respectively. Both GTH's stimulated, in a similar manner, testicular androgen and ovarian estradiol synthesisin vitro.     

Purification and characterization of a cysteine-like protease from the body wall of the sea cucumber <Emphasis Type="Italic">Stichopus japonicus</Emphasis>

The sea cucumber (Stichopus japonicus) is able to undergo autolysis in response to a variety of environmental and mechanical cues. Within the framework of a long-term study of this phenomenon we have purified a protease from the body wall of the sea cucumber by means of ion-exchange chromatography with DE-52 cellulose and gel filtration chromatography with Sephadex G-100. The final enzyme preparation was nearly homogeneous on polyacrylamide gel electrophoresis, and its molecular weight was estimated to be approximately 35.5 kDa. The purified enzyme exhibited a maximum activity for the hydrolysis of casein at pH 7.0 and 50°C and a remarkable stability at pH 4.0–7.0 and 40–60°C. Based on the inhibition and activation profiles obtained with numerous specific protease inhibitors and an activator, the protease purified from the body wall of the sea cucumber was defined to be a cysteine-like protease.     

Purification of anaphase promoting complex/cyclosome from goldfish oocytes

Destruction of cyclin B is required for exit from mitosis and meiosis. A cyclin-specific ubiquitinating system, including anaphase-promoting complex/cyclosome (APC/C) is thought to be responsible for cyclin B destruction. To learn more about the molecular mechanism of cyclin B degradation, a molecular study of the ubiquitinating system in goldfish has been undertaken. For biochemical preparation of APC/C, we first conducted the cloning, sequencing and expression analysis of goldfish, Carassius auratus, cdc27 that encodes a subunit of APC/C from goldfish ovary. The deduced amino acid sequence is highly homologous to cdc27 from other species. Then recombinant goldfish Cdc27CT (C-terminal half of Cdc27) was expressed in Escherichia coli, and an antibody was raised against purified recombinant protein. Polyclonal antiserum cross-reactive with Cdc27 was obtained. By the assay using the antibody, APC/C was purified by column chromatographs.
