The aim of this study was to investigate the effects of excess leucine (Leu) vs. its metabolites α‐ketoisocaproate (KIC) and β‐hydroxy‐β‐methyl butyrate (HMB) on Leu metabolism, muscle fibre composition and muscle growth in growing pigs. Thirty‐two pigs with a similar initial weight (9.55 ± 0.19 kg) were fed 1 of 4 diets for 45 days: basal diet, basal diet + 1.25% L‐Leu, basal diet + 1.25% KIC‐Ca, basal diet + 0.62% HMB‐Ca. Results indicated that relative to the basal diet and HMB groups, Leu and KIC groups exhibited increased Leu concentrations and decreased concentrations of isoleucine, valine and EAAs in selected muscle (< 0.05) and had lower mRNA levels of MyHC I and higher expression of MyHC IIx/IIb (< 0.05), and there was no significant difference between the basal and HMB‐supplemented groups. Moreover, the mRNA expression levels of AMPKα and UCP3 were higher but the myostatin mRNA levels were lower in the soleus muscle of the HMB group than those from other groups (< 0.05). These findings demonstrated that doubling dietary Leu content exerted growth‐depressing effects in growing pigs; dietary KIC supplementation induced muscular branched‐chain amino acid imbalance and promoted muscle toward a more glycolytic phenotype; while dietary HMB supplementation promoted the generation of more oxidative muscle types and increased muscle growth specially in oxidative skeletal muscle, and these effects of HMB might be associated with the AMPKα‐Sirt1‐PGC‐1α axis and mitochondrial biogenesis.  相似文献

Antigens and molecular mass diversity of the Haemophilus somnus major outer membrane protein (MOMP) were investigated. The molecular mass of the MOMP of 53 strains of H. somnus varied from 43 to 33 kDa and four MOMP MAb reactivity patterns were detected in immunoblot analysis and immunodot assay. The molecular mass and MAb reactivity data were used for preliminary grouping of H. somnus strains. Disease strains fell into groups 1 and 3, including two of three Group 3 subgroups, whereas strains from asymptomatic carriers were found in all the four groups and three subgroups. Immunoblot analysis with convalescent phase serum showed strain specific reactivity with MOMPs from three isolates used to reproduce disease in cattle. The reaction with the MOMP was only detectable at dilutions of 1:100 or less, whereas the same convalescent sera showed strong reactivity at dilutions of 1:1000 (or more) with other H. somnus antigens. The data suggest that the bovine immune response to the MOMP during infection is weak and is directed to antigenically variable determinants in a strain-specific manner. This may be important in evaluating the role of the antibody response to MOMPs in protective immunity.  相似文献

A single grain (~3 micrograms) returned by the Hayabusa spacecraft was analyzed by neutron activation analysis. This grain is mainly composed of olivine with minor amounts of plagioclase, troilite, and metal. Our results establish that the Itokawa sample has similar chemical characteristics (iron/scandium and nickel/cobalt ratios) to chondrites, confirming that this grain is extraterrestrial in origin and has primitive chemical compositions. Estimated iridium/nickel and iridium/cobalt ratios for metal in the Itokawa samples are about five times lower than CI carbonaceous chondrite values. A similar depletion of iridium was observed in chondrule metals of ordinary chondrites. These metals must have condensed from the nebular where refractory siderophile elements already condensed and were segregated.  相似文献

[目的]研究Cu~(2+)和Pb~(2+)对石蚕蛾幼虫的急毒性效应和安全浓度。[方法]从广西十万大山保护区采集石蚕蛾(Stenopsyche marmorata)幼虫,采用静水生物毒性试验法进行重金属离子Cu~(2+)和Pb~(2+)对其的急毒性试验。采用概率单位法分别计算Cu~(2+)和Pb~(2+)对石蚕蛾幼虫的半致死浓度和安全浓度。[结果]Cu~(2+)对石蚕蛾幼虫的24、48、72、96 h的半致死浓度(LC50)分别为123.651、112.975、84.536和70.509 mg/L,Pb~(2+)对石蚕蛾幼虫的24、48、72、96 h的半致死浓度(LC50)分别为49.138、41.878、30.735和29.245 mg/L。Cu~(2+)和Pb~(2+)对石蚕蛾幼虫的安全浓度分别为7.051和2.925 mg/L。[结论]研究结果可为将石蚕蛾幼虫用作水体重金属污染指示生物提供科学依据。  相似文献

Murine T10 and T22 are highly related nonclassical major histocompatibility complex (MHC) class Ib proteins that bind to certain gammadelta T cell receptors (TCRs) in the absence of other components. The crystal structure of T22b at 3.1 angstroms reveals similarities to MHC class I molecules, but one side of the normal peptide-binding groove is severely truncated, which allows direct access to the beta-sheet floor. Potential gammadelta TCR-binding sites can be inferred from functional mapping of T10 and T22 point mutants and allelic variants. Thus, T22 represents an unusual variant of the MHC-like fold and indicates that gammadelta and alphabeta TCRs interact differently with their respective MHC ligands.  相似文献

Summary The results showed that the F1 genotype from the cross (Brassica napus cv. Zheshuang 758 × cv. Z-4115) had good response to embryogenesis, and their embryo yield and rate of plant regeneration reached 69.8 embryo/bud and 46.9%, respectively. Characters from the doubled haploid (DH) populations in B. napus were analyzed and it was showed that the means of each agronomic trait were between their parents, but they were nearer to the paternal in 6 agronomic traits (plant height, branch position, number of pods in the main raceme, length of pod, number of pods/plant and number of seeds/pod). The number of genes controlling each agronomic trait was analyzed based on the DH populations. The results showed that the number of genes controlling number of pods in the main raceme was the highest (15.6), and the least number of genes was involved for stem width (only 7.9). According to estimated coefficients of skewness and kurtosis of the traits tested, gene interaction was found to be absent for stem width, plant height, length of main raceme, number of primary and secondary branches, pod density in the main raceme and seed weight/plant. Complementary interaction was also observed in five agronomic traits (number of pods in the main raceme, length of pod, number of pods/plant, number of seeds/pod and 1000-seed weight). A significantly positive correlation was observed between seed yield/plant and four agronomic traits (length of main raceme, length of pod, number of pods/plant and 1000-seed weight). The experiment also showed that the erucic acid, glucosinolate, oil and protein contents of DH populations were 34.23%, 87.09 μmol/g, 44.09% and 42.67%, respectively. The numbers of genes controlling each quality trait were 7.8, 9.7, 9.4 and 8.7, respectively. Partial correlations between the seed quality traits and the agronomic characters of DH populations were analyzed. In this experiment, the partial correlations among seed quality traits were also analyzed and it was found that the oil content had a negative correlation with the other three seed quality traits.  相似文献

A growth trial was conducted to estimate the optimum concentration of dietary calcium (Ca) for grass carp (Ctenopharyngodon idella). Triplicate groups of grass carp (4.52 ± 0.02 g) were fed diets containing graded levels (2.75, 4.51, 6.24, 7.99, 9.66 and 11.5 g kg?1) of Ca for 8 weeks. Weight gain, feed efficiency and protein efficiency ratio were linearly increased up to the 7.99 g kg?1 dietary Ca and then maintained stable beyond this level (P < 0.05). Dietary Ca levels higher than 7.99 g kg?1 significantly increased the ash contents of whole body, vertebrae and scales. Ca contents in whole body, vertebrae and scales were linearly increased up to the 7.99 g kg?1 dietary Ca and then maintained stable beyond this level (P < 0.05). In contrast, dietary Ca levels higher than 9.66 g kg?1 significantly decreased Mg contents in whole body, vertebrae and scales. Dietary Ca levels higher than 7.99 g kg?1 significantly increased plasma alkaline phosphatase activity. However, plasma Ca, P and Mg contents were not significantly affected by dietary Ca supplements (P > 0.05). Polynomial regression analysis indicated that 10.4 g kg?1 dietary Ca was required for maximal tissue storage and mineralization as well as optimal growth.  相似文献

Long‐term monoculture of watermelon results in inhibited growth and decreased crop yields, possibly because of imbalance in microbial ecology caused by accumulation of the pathogen in soil. This results in serious problems in the economics of watermelon production. We investigated the build‐up of Fusarium in soil under watermelon cultivation and changes over 3 yr of fallow in a microcosm. We focused on changes in the microbial community of Fusarium‐infected soil, including the diversity of the microfloral species composition, and species abundance. Long‐term monoculture of watermelon leads to changes in microbial diversity and community structure. The microbes most readily cultured from infested soil were suppressed by watermelon wilt pathogen Fusarium oxysporum f. sp. niveum (FON). Of 52 isolated and identified culturable microbes, 83.3% of bacteria, 85.7% of actinomycetes, 31.6% of fungi and 20.0% of Fusarium sp. were inhibited by FON on bioassay plates. Prior to fallowing, infested soil was a transformed ‘fungus‐type’ soil. After 3 yr of fallow, the infested soil had remediated naturally, and soil microbial diversity recovered considerably. Abundance of dominant bacterial populations was increased by 118–177%, actinomycetes, fungi and FON were decreased by 23–32, 33–37 and 50%, respectively. The ratio of bacteria: actnomycetes: fungus: Fusarium sp. in infested soil changed from 24 000:100:4:1 prior to fallow to 57 000:100:3.5:1 after fallowing, nearer to the 560 000:400:8:1 ratio of healthy soil not used for watermelon cultivation. This suggests the ‘fungus‐type’ soil was converting to ‘bacteria‐type’ soil and that disrupted microbial communities in infested soil were restored during fallow.  相似文献
