A novel application of a hybrid biosensor is here employed as an analytical method for the detection and presumptive identification of beta-lactam residues in milk. The method is based on measurements of carbon dioxide (CO2), the production of which is related to the microbial growth of the test microorganism Bacillus stearothermophilus var. calidolactis. The presence of beta-lactams in milk inhibits microbial growth and, consequently, the CO2 production rate. The analysis is based on the variation of CO2 between a milk sample spiked with beta-lactams and a twin milk sample containing beta-lactams plus a broad spectrum beta-lactamase, using an electrochemical device of biosensor. A blank milk sample is included as control. The result is obtained starting from the first 120 min. Moreover, the ability to recognize all of the beta-lactams speeds the total time of analysis when chemical identification and quantification are required. The analytical method appears to be adequate for milk control for qualitative screening purposes, complying with the requirements stated in Decision 2002/657/EC. 相似文献

Trees are essential in the urban environment not only because of their aesthetic and social values, but also for their effects on air quality. Data of the present experiment show some of the integrated mechanisms that may confer sensitivity/tolerance in Tilia americana saplings exposed to ozone treatment (120 ppb, 5 h day?1 for 45 consecutive days) in order to improve the management of green spaces responding to oxidative stress. At the end of exposure, plants do not exhibit any foliar symptoms. Profiles related to membrane injury, photosynthetic gas exchange, chlorophyll a fluorescence, pigment content and enzymes/metabolites linked to the synthesis of phenylpropanoids show a vulnerability in terms of: (i) change in the ionic and solute reactions of the membrane cells (maximum value of +34%, 28 days after the beginning of fumigation, compared to controls); (ii) inhibition of the carbon assimilation (?41%), mostly as a consequence of non-stomatal limitation of assimilation rate; (iii) progressive inhibition of the Calvin cycle, as demonstrated by decreases in Rubisco carboxylation efficiency and regeneration capacity (?35 and ?21%, respectively, at the end of exposure), quantum yield of electron transfer at PSII and in the fraction of energy passively dissipated as heat and fluorescence (?34% and ?31%); (iv) senescence process (decline in demand for reducing power and energy at the end of treatment); (v) damage to the chlorophyll pigment system. However, the activation of xanthophylls cycle and phenylpropanoid metabolism, that can be considered a strategy in plants in order to regulate light absorbed energy and to scavenge reactive oxygen species does not preserve PSII photochemistry from impairment at the end of exposure. 相似文献

The squid pen (gladius) from the Loligo vulgaris was used for preparation of β-chitin materials characterized by different chemical, micro- and nano-structural properties that preserved, almost completely the macrostructural and the mechanical ones. The β-chitin materials obtained by alkaline treatment showed porosity, wettability and swelling that are a function of the duration of the treatment. Microscopic, spectroscopic and synchrotron X-ray diffraction techniques showed that the chemical environment of the N-acetyl groups of the β-chitin chains changes after the thermal alkaline treatment. As a consequence, the crystalline packing of the β-chitin is modified, due to the intercalation of water molecules between β-chitin sheets. Potential applications of these β-chitin materials range from the nanotechnology to the regenerative medicine. Normative regulations on benzene in fuels and urban management strategies are expected to improve air quality. The present study deals with the application of self-organizing maps (SOMs) in order to explore the spatiotemporal variations of benzene, toluene, ethylbenzene, and xylene levels in an urban atmosphere. Temperature, wind speed, and concentration values of these four volatile organic compounds were measured after passive sampling at 21 different sampling sites located in the city of Trieste (Italy) in the framework of a multi-year long-term monitoring program. SOM helps in defining pollution patterns and changes in the urban context, showing clear improvements for what concerns benzene, toluene, ethylbenzene, and xylene concentrations in air for the 2001–2008 timeframe. 相似文献

This paper describes and evaluates the mortality associated with the potential stressor effect induced by application of artificial photoperiods in rainbow trout (Oncorhynchus mykiss). After application of artificial photoperiods, high mortalities of trout subjected to this management were recorded (36% in LD 14:10 and 25% in LD 24:0) as compared to controls (7%). The recorded signology related to diseased trout included darkening, lethargy and erratic swimming. Among the morphological changes, skin lesions and ulcerative-type necrosis could be observed. Blood tests, histopathology and isolation of pathogens identified as Flavobacterium psychrophilum, associated with Aeromonas, Pseudomonas and Saprolegnia sp., were performed. The application of artificial photoperiods suggests that fish may be more susceptible to opportunistic diseases; thus, it is advisable to emphasize preventive measures in fish subjected to this management. 相似文献

In this study, we analysed the effect of population density and food concentration on the fecundity of a Mediterranean strain of Acartia tonsa to maximize egg production. During 4‐day feeding experiments, egg hatching success and faecal pellet production were also followed. The algae Rhinomonas reticulata was supplied at different concentrations corresponding to 250, 500, 1000, 1500, 2000 and 3000 μg C L?1 day?1 at the following adult copepod density: 40, 80 and 160 ind. L?1. Our results show a positive relationship between algal concentration and egg production under all experimental conditions confirming that the quantity of food strongly limits A. tonsa fecundity. Maximum egg production (57 eggs per female) was reached at the lowest density and at the maximum food concentration. Percentage of egg hatching success was not dependent on the quantity of food used. At the same food concentration, an increase in population density from 40 to 80 ind. L?1 induced an increase in faecal pellet production per couple which did not correspond to an increase in egg production, suggesting that higher energetic costs were shifted to swimming activity. Productivity of the A. tonsa Mediterranean strain is mainly limited by the quantity of food rather than by crowding conditions. 相似文献

Lysozyme from equine neutrophil granulocytes was isolated in a pure form by fast performance liquid chromatography, i.e. ion-exchange chromatography and reversed-phase chromatography. The lysozyme lysed Micrococcus luteus, Bacillus subtilis and Staphylococcus lentus and was also bactericidal against the Gram-negative bacteria Escherichia coli, Klebsiella pneumoniae, Bordetella bronchiseptica, and Serratia marcescens. Staphylococcus aureus and Staphylococcus epidermidis were not lysed. The lysozyme was only very slightly bactericidal for S. epidermidis and S. aureus. Equine neutrophil lysozyme was found to be bactericidal for Gram-positive as well as for Gram-negative bacteria without further treatment. Equine and chicken egg white lysozymes were found to be immunologically related when examined using specific antisera against each of them. Alpha 2-beta 1-glycoprotein may be found free in horse serum or complexed with alpha-1-proteinase inhibitor to form pre-alpha 2-elastase inhibitor. There has been little information published concerning alpha 2-beta 1-glycoprotein and its possible tissue sources in horses. A peroxidase-antiperoxidase technique was used to identify alpha 2-beta 1-glycoprotein in buffy coat and bone marrow neutrophils of healthy horses. Macrophages and neutrophils in bronchoalveolar lavage samples from clinically normal horses and from horses with chronic pulmonary disease also were positive for alpha 2-beta 1-glycoprotein. Alpha 2-beta 1-glycoprotein was identified in some instances in normal equine hepatocytes of formalin-fixed liver sections. In formalin-fixed lung sections from horses with chronic, small-airway disease and chronic bronchointerstitial pneumonia, alpha 2-beta 1-glycoprotein was observed in some airway secretions and in macrophages. 相似文献
