Triploidization is an interesting tool to produce sterile fish. In the yellowtail tetra, Astyanax altiparanae, this can be applied for aquaculture and surrogate technologies. In this study, we compared the efficacy of cold (2 C) or heat shock (38 C, 40 C, and 42 C) on triploid induction in the yellowtail tetra. The eggs were treated with cold or heat shock, 2 min postfertilization (30 min in cold shock or 2 min in heat shock). Intact embryos served as the control group. Ploidy status was confirmed by karyotyping, flow cytometry, and nuclear diameter of erythrocytes. The hatching rate decreased after cold shock (12.69 ± 15.76%) and heat shock at 42 C (0.35 ± 0.69%) in comparison with the control group (63.19 ± 16.82%). At 38 C and 40 C, hatching rates (61.29 ± 17.73% and 61.75 ± 22.1%, respectively) were not decreased. Only one triploid arose at 38 C (1/80). At 40 C, a high number of triploids arose (72/78). At 42 C, very few embryos developed into the hatching stage. A large number of haploid individuals arose after cold shock (61/75), with only one triploid. Our results indicate that heat shocking of embryos at 40 C is optimum for triploid production in the yellowtail tetra. 相似文献

ABSTRACT We describe a method to evaluate the virulence of Mycosphaerella fijiensis, the causal agent of black leaf streak disease (BLSD) of banana and plantain. The method is based on the delivery of weighed slurries of fragmented mycelia by camel's hair brush to 5-by-5-cm areas on the abaxial surface of banana leaf blades. Reliable BLSD development was attained in an environmental growth chamber with stringent lighting and humidity controls. By localizing inoculum onto small areas of large leaves, we achieved a dramatic increase in the number of strains that can be tested on each leaf and plant, which is critical for comparing the virulence of numerous strains concurrently. Image analysis software was used to measure the percentage of each inoculated leaf section showing BLSD symptoms over time. We demonstrated that the level of disease of four isolates was correlated with the weight of the mycelium applied and relatively insensitive to the degree of fragmentation of hyphae. This is the first report demonstrating that weighed mycelial inoculum, combined with image analysis software to measure disease severity, can be used to quantitatively assess the virulence of M. fijiensis under rigorously controlled environmental conditions. 相似文献

Secondary metabolites and host defense compounds were shown to occur in xylem sap, and leaves of Vitis vinifera cv. Italia and cv. Matilde naturally infected by the esca-associated fungi Phaeomoniella chlamydospora (Pch), Togninia minima (Tmi) and Fomitiporia mediterranea (Fme). Samples of xylem sap and leaves were collected from healthy vines and from vines showing severe symptoms of brown wood-streaking caused by Pch and Tmi, or from vines with symptoms of both brown wood-streaking and white rot caused by Fme. Xylem sap collection was carried out during the early spring of 2003 and 2004, corresponding to the phenological phases: (A) cotton bud; (B) green tip; (C) leaves out; (D) stretched out leaves; and (E) visible clusters. In the present work we have studied the accumulation of biomolecules (pentaketides and α-glucans), host defense compounds (benzaldehydes, benzoic acid and cinnamic acid derivatives, flavonols, flavanols, flavan-3-ol derivatives and stilbenes) at different stages of grapevine development. Accumulation and changes in total phenolics and recurring phenolics, and of three phytotoxic secondary metabolites (scytalone, isosclerone and pullulan) were analyzed by HPLC. We conducted a five-year survey (2011–2015) of barley and wheat fields in Paraná state, Brazil, obtaining 754 Fusarium isolates from spikes with fusarium head blight (FHB)-symptoms. Multilocus genotyping and TEF-1α gene sequence analyses confirmed the dominance of the F. graminearum species complex (FGSC, 75.7%), but F. poae (11.5%), as well as F. avenaceum and related members of the F. tricinctum species complex (FTSC, 8.1%) appeared as substantial contributors to FHB. Within the FGSC, F. graminearum of the 15-ADON genotype was dominant (63%), followed by F. meridionale of the NIV genotype (23.1%), F. cortaderiae of the NIV (7%) or 3-ADON (2.6%) genotypes, and F. austroamericanum (3.8%) of the 3-ADON genotype. Substantial variation in pathogen composition was observed across years, with F. poae and F. meridionale frequencies significantly elevated in some years. Most F. poae strains produced DAS, diANIV, and butenolide, but not neosolaniol, T-2, or HT-2. All FTSC species produced moniliformin. Enniatin production was widespread among FTSC species, with the single F. acuminatum strain found to be the strongest producer of enniatins. Our findings confirm FGSC as a major contributor to FHB and expand considerably our knowledge of the presence, frequency, and conditions under which other pathogens may emerge, altering the spectrum of toxins that may accumulate in grain. 相似文献

Stem rot caused by Lasiodiplodia theobromae is an important postharvest disease of papaya in Brazil, responsible for reducing the quality and quantity of fruits. Fungicide
use is one of the main disease management measures. However, there are no estimates available of pathogen sensitivity to commonly
employed fungicides. Therefore, the EC50 from 120 isolates of L. theobromae from northeastern Brazil, representative of six populations of the pathogen, was estimated in vitro for fungicides of the
methyl benzimidazole carbamates—MBC (benomyl and thiabendazole) and demethylation-inhibiting—DMI (imazalil, prochloraz, tebuconazole)
groups. Mycelial growth on fungicide-free media and virulence on papaya fruits of the MBC-sensitive and non-sensitive isolates
were compared. For MBCs, 8.4% of isolates were non-sensitive to fungicides. For the remaining 91.6%, the mean EC50 ranged from 0.002 to 0.13 μg ml−1 and 0.36 to 1.27 μg ml−1 for benomyl and thiabendazole, respectively. For DMIs, the mean EC50 range for imazalil was 0.001 to 2.27 μg ml−1, 0.04 to 1.75 μg ml−1 for prochloraz, and 0.14 to 4.05 μg ml−1 for tebuconazole. The EC50 values of non-sensitive isolates were significantly (P≤0.05) higher those for the sensitive isolates for each of the DMI fungicides. Differences (P≤0.05) were found in the levels of sensitivity to DMI fungicides among the isolate populations associated with orchards. The
populations from two orchards were less sensitive to DMIs. No solid evidence was found for fitness costs relating to MBC non-sensitive
isolates because mycelial growth in fungicide-free media and virulence on papaya fruits were similar to those of sensitive

Environmental conditions influence phenology and physiological processes of plants. It is common for maize and sorghum to be sown at two different periods: the first cropping (spring/summer) and the second cropping (autumn/winter). The phenological cycle of these crops varies greatly according to the planting season, and it is necessary to characterize the growth and development to facilitate the selection of the species best adapted to the environment. The aim of this study was to characterize phenological phases and physiological parameters in sorghum and maize plants as a function of environmental conditions from the first cropping and second cropping periods. Two parallel experiments were conducted with both crops. The phenological characterization was based on growth analyses (plant height, leaf area and photoassimilate partitioning) and gas exchange evaluations (net assimilation rate, stomatal conductance, transpiration and water-use efficiency). It was found that the vegetative stage (VS) for sorghum and maize plants was 7 and 21 days, respectively, longer when cultivated during the second cropping. In the first cropping, the plants were taller than in the second cropping, regardless of the crop. The stomatal conductance of sorghum plants fluctuated in the second cropping during the development period, while maize plants showed decreasing linear behaviour. Water-use efficiency in sorghum plants was higher during the second cropping compared with the first cropping. In maize plants, in the second cropping, the water-use efficiency showed a slight variation in relation to the first cropping. It was concluded that the environmental conditions as degree-days, temperature, photoperiod and pluvial precipitation influence the phenology and physiology of both crops during the first and the second cropping periods, specifically cycle duration, plant height, leaf area, net assimilation rate, stomatal conductance and water-use efficiency, indicating that both crops respond differentially to environmental changes during the growing season. 相似文献

In a third experiment, the expression and the segregation of the well known R1 gene was influenced by an additional genetic factor. In that case there were indications for a dominant suppressor. This was established by the selection of susceptible plants carrying a RFLP allele of probe GP21 closely linked to R1. In three of the four F1 populations, resulting from crosses between such susceptible plants and susceptible tester plants, resistnat progenies were found. The resistance appeared to be R1-specific. The quality of the potato has been found to vary, when grown under different agricultural and environmental conditions, such as the level of fertilization. Consequently these factors may influence the acrylamide formation during the preparation of French fries. These assumptions were studied on three varieties: Bintje, Ramos, and Saturna from the harvest of 2003. Decreasing N fertilization caused increases in the reducing sugar concentration from 60% up to 100% on DM for all varieties studied. Due to a high correlation between the reducing sugar content and the generation of acrylamide during frying, this resulted in a parallel increase in the acrylamide concentration of the French fries. Thus by lowering the amount of N fertilizer, an increase of 30-65% of the acrylamide generation during frying could be observed. It seems of extreme importance to find an appropriate balance between the level of N fertilizer in order to diminish acrylamide formation but on the other hand to obtain an acceptable tuber and to consider the environmental impact. All results reported should be seen in the perspective of the warm growing season of 2003. 相似文献

Protons and helium nuclei are the most abundant components of the cosmic radiation. Precise measurements of their fluxes are needed to understand the acceleration and subsequent propagation of cosmic rays in our Galaxy. We report precision measurements of the proton and helium spectra in the rigidity range 1 gigavolt to 1.2 teravolts performed by the satellite-borne experiment PAMELA (payload for antimatter matter exploration and light-nuclei astrophysics). We find that the spectral shapes of these two species are different and cannot be described well by a single power law. These data challenge the current paradigm of cosmic-ray acceleration in supernova remnants followed by diffusive propagation in the Galaxy. More complex processes of acceleration and propagation of cosmic rays are required to explain the spectral structures observed in our data. 相似文献
