rhBMP-2 solution on a collagen sponge was placed along the diaphysis of an atrophicradius, which had a history of recurring fractures. Two months after rhBMP-2 treatment, new mineralized bone was present, which significantly increased the diameter of the radius and allowed the removal of the external skeletal fixator (ESF). This study compared vitamin A, vitamin E, selenium (Se), and L-lactate in blood and synovial fluid in 2 groups of 6 dogs; a control group (without OA) and an osteoarthritic group with spontaneous cranial cruciate ligament rupture and OA. Concentrations of vitamin E were significantly higher in serum than in synovial fluid in both OA (P = 0.006) and control (P = 0.0008) groups. Vitamin E concentration in synovial fluid was significantly higher in the OA group than in the control group (P = 0.009). Concentrations of Se were significantly higher in serum than in synovial fluid in both OA (P = 0.003) and control (P = 0.0006) groups. There were no significant differences in levels of Se, vitamin A, and L-lactate between the 2 groups. This is the first study to show an increased concentration of vitamin E in the synovial fluid of dogs with OA compared with dogs that did not have OA. 相似文献
Johne's disease is an enteric mycobacterial infection of ruminants that has significant global economic impact. The classic host reaction is one of an early T-cell mediate immune response, with predominant interferon gamma (IFNγ) activity; there is subsequent lowering of this response as animals reach the terminal stages of disease. Interleukin (IL)-10, which can suppress Th1-type and enhance Th2-type cytokine production, is considered to play a role in the later stages of Johne's disease. To determine the role of IL-10 throughout the course of Johne's disease we studied groups of sheep with either no Johne's disease (n = 10), natural infection (n = 30) or experimental infection (n = 58). Disease status of the animals was comprehensively assessed by culture of Mycobacterium avium subsp. paratuberculosis (Mptb), histopathology and serology. Antigen-specific IL-10 secretion in peripheral blood of sheep exposed to Mptb was significantly higher than in control animals (P < 0.001) as early as 4 months post-inoculation, and increased progressively. In ileal and jejunal lymph node cells, IL-10 secretion was also significantly higher in animals that were exposed to Mptb compared to controls (P < 0.05). The early IL-10 response seen in peripheral blood cells may be a reflection of early responses at sites of Mptb infection. IL-10 secretion from ileal and jejunal lymph node cells was significantly higher in exposed animals with no lesions or with paucibacillary lesions when compared to animals with multibacillary lesions. These novel findings demonstrate that increased IL-10 activity commences soon after exposure to the causative mycobacterium and may play a role in determining disease outcome. 相似文献
The aim of this study was to compare the development of Glasser's disease in sow-reared and colostrum-deprived piglets. Ninety piglets from a commercial pig farm in Spain were used. The farm was positive for Haemophilus parasuis. Fifty-two pigs were sow-reared (SR) and 38 were colostrum-deprived (CD) piglets. The animals were intratracheally inoculated with H. parasuis serovar 5 and sacrificed at 1, 2 and 3 days post-infection. To assess the development of disease, antibody titers, clinical signs, pathological lesions, microbiological isolation and PCR amplification were compared between the groups. Inoculation of SR pigs did not cause clinical signs or lesions of Glasser's disease. In SR pigs, H. parasuis isolation and specific PCR amplification from tissues showed a very low number of positive samples. In contrast, in CD pigs, inoculation resulted in the typical signs and lesions of Glasser's disease. Positive microbiological isolation and specific PCR products were obtained from the majority of the tissues tested, and no antibodies against H. parasuis were detected. The experimental infection using CD pigs describes a successful method to study this microorganism and confirms the important role that maternal antibodies play in protection against clinical signs and disease. 相似文献
1.?An experiment determined the effect of 6 different lysine concentrations (40, 52, 58, 65, 90 and 130 g/kg CP) in two protein concentrations (210 and 260 g/kg) on the efficiency of energy utilisation of broiler chickens and turkeys. Ninety-six male Ross 308 broiler chicks and 96 male BUT Big 6 turkey poults were fed at 90% of ad libitum during a 12 d bioassay period. At the end of the bioassay, birds were slaughtered and carcase crude protein, fat and gross energy were measured and nutrient metabolisability determined.
2.?Broiler chickens had lower AME and AMEN than turkeys. AME was higher in the 260 g/kg than in the 210 g/kg protein diet; however, no difference was evident in AMEN between any of the diets in both species. A significant species × lysine interaction indicated that the decrease in AME and AMEN with increasing lysine was greater in turkeys than in broiler chickens.
3.?Turkeys had lower weight gains and efficiencies of energy retention than broiler chickens. The effects of different lysine concentrations on growth and the efficiency of energy retention were significant and growth responses were quadratic but there were negative linear responses in the efficiency of energy retention in both species at both crude protein concentrations. Conjugated linoleic acids (CLA), 9-cis:11-trans and 10-trans:12-cis, have been shown to be able to modify some immune cells parameters and plasma lipids in a variety of experiment models. Since lymphocytes and polymorphonuclear cells (PMNC) have a large spectrum functions in the immune response, the knowledge in this field has to be expanded. Beagle dogs were fed a control diet or a CLA supplemented diet for nine months. Blood was collected for biochemical analysis and lymphocyte and PMNC isolation. PMNC were assayed for lysosome content, phagocytic activity and superoxide anion production. A lymphocyte proliferation capacity assay was done. The CLA fed dogs had a 34% reduction in total cholesterol (P < 0.05), 28% in LDL (P < 0.05) and 28% non-HDL-cholesterol (P < 0.05). Neither of the PMNC parameters evaluated demonstrated significant alteration. Lymphocytes from CLA group increased by 45% their mitotic capacity (P < 0.05). Our study demonstrates that CLA can successfully modify the lipid profile of dogs (monogastrics) when fed at reasonable levels, but did not significantly alter inflammatory function as would generally predicted. Ex situ collections offer the potential to reduce extinction risks, affording option to society in maintaining future breeding opportunities for productivity and heritage traits. However, how much should we be seeking to collect and conserve in gene banks, and where? We developed a mathematical model to optimize logistical decisions of breed conservation choices and to evaluate alternative scenarios for efficiently re‐allocating genetic materials currently stored in different European gene banks, allowing for cross‐country collections, cost and cryogenic capacity differentials. We show how alternative allocations for the breeds that are currently stored in 11 European gene banks could reduce overall conservation costs by around 20% by selecting cryogenic banks that have relatively lower combination of fixed and collection costs, and are geographically closer to collection regions. Our results show that centralizing collections in one gene bank would double the costs, relative to collective European collections approaches. We also calculate marginal costs of collections and show that increasing diversity within the gene banks implies in higher costs per conserved breed. 相似文献
Glässer’s disease is a re-emerging swine disease characterized by a severe septicaemia. Vaccination has been widely used to control the disease, although there is a lack of extended cross-protection. Trimeric autotransporters, a family of surface exposed proteins implicated in host-pathogen interactions, are good vaccine candidates. Members of this family have been described in Haemophilus parasuis and designated as virulence-associated trimeric autotransporters (VtaA). In this work, we produced 15 recombinant VtaA passenger domains and looked for the presence of antibodies directed against them in immune sera by immunoblotting. After infection with a subclinical dose of H. parasuis Nagasaki, an IgG mediated antibody response against 6 (VtaA1, 5, 6, 8, 9 and 10) of the 13 VtaA of the Nagasaki strain was detected, indicating that they are expressed in vivo. IgA production against VtaA was detected in only one animal. VtaA were more likely to be late antigens when compared to early (Omp P5 and Omp P6) and late (YaeT) defined antigens. Antibody cross-reaction with two orthologs of Nagasaki’s VtaA5 and 6, VtaA15 and 16 of strain HP1319, was also detected. No antibodies against VtaA were detected in the sera of animals immunized with a bacterin of the Nagasaki strain, suggesting poor expression in the in vitro conditions used. Taken together, these results indicate that VtaA are good candidate immunogens that could be used to improve H. parasuis vaccines. However, their capacity to confer protective immunity needs to be further studied. 相似文献
