Cell wall degradation is a crucial process within the malting process of barley. Therefore, the haplotype diversity of genes for two cell wall degrading enzymes, (1 → 3),(1 → 4)-β-d-Glucan-4-glucanohydrolase and (1 → 4)-β-Xylan-endohydrolase 1, was investigated and associations to malting quality parameters were performed. The (1 → 3),(1 → 4)-β-d-Glucan-4-glucanohydrolase gene glb2 had two major haplotypes defined by three SNPs and one INDEL, which explained 8.9 and 9.5% of the total variation of malt extract content and viscosity in the spring barley gene pool, respectively. The most significant associations of (1 → 4)-β-Xylan-endohydrolase 1 gene X-1 were found for diastatic power, saccharification VZ45 and soluble nitrogen with 18, 12 and 8% of the total variation explained by SNP3 in the spring barleys. Over a period of a few years, Pepino mosaic virus (PepMV) has become one of the most important viral diseases in tomato production worldwide. Infection by PepMV can cause
a broad range of symptoms on tomato plants, often leading to significant financial losses. At present, five PepMV genotypes
(EU, LP, CH2, US1 and US2) have been described, three of which (EU, LP and US2) have been reported in Europe. Thus far, no
correlation has been found between different PepMV genotypes and the symptoms expressed in infected plants. In this paper,
the genetic diversity of the PepMV population in Belgian greenhouses is studied and related to symptom development in tomato
crops. A novel assay based on restriction fragment length polymorphism (RFLP) was developed to discriminate between the different
PepMV genotypes. Both RFLP and sequence analysis revealed the occurrence of two genotypes, the EU genotype and the CH2 genotype,
within tomato production in Belgium. Whereas no differences were observed in symptom expression between plants infected by
one of the two genotypes, co-infection with both genotypes resulted in more severe PepMV symptoms. Furthermore, our study
revealed that PepMV recombinants frequently occur in mixed infections under natural conditions. This may possibly result in

BACKGROUND: Clinical studies investigating platelet function in dogs have had conflicting results that may be caused by normal physiologic variation in platelet response to agonists. OBJECTIVES: The objective of this study was to investigate platelet function in clinically healthy dogs of 4 different breeds by whole-blood aggregometry and with a point-of-care platelet function analyzer (PFA-100), and to evaluate the effect of acetylsalicylic acid (ASA) administration on the results from both methods. METHODS: Forty-five clinically healthy dogs (12 Cavalier King Charles Spaniels [CKCS], 12 Cairn Terriers, 10 Boxers, and 11 Labrador Retrievers) were included in the study. Platelet function was assessed by whole-blood aggregation with ADP (1, 5, 10, and 20 microM) as agonist and by PFA-100 using collagen and epinephrine (Col + Epi) and Col + ADP as agonists. Plasma thromboxane B(2) concentration was determined by an enzyme immunoassay. To investigate the effect of ASA, 10 dogs were dosed daily (75 or 250 mg ASA orally) for 4 consecutive days. RESULTS: A higher platelet aggregation response was found in CKCS compared to the other breeds. Longer PFA-100 closure time (Col + Epi) was found in Cairn Terriers compared to Boxers. Plasma thromboxane B(2) concentration was not statistically different between groups. Administration of ASA prolonged the PFA-100 closure times, using Col + Epi (but not Col + ADP) as agonists. Furthermore, ASA resulted in a decrease in whole-blood platelet aggregation. CONCLUSIONS: Platelet function is influenced by breed, depending upon the methodology applied. However, the importance of these breed differences remains to be investigated. The concept of degree of compactness (DC), referred to as field bulk density (BD) as a percentage of a reference bulk density (BDref), was developed to characterize compactness of soil frequently disturbed, but for undisturbed soil such as under no-tillage critical degree of compactness values have not been tested. The objective of this study was to compare methods to determine BDref and limits of DC and BD for plant growth under no-tillage in subtropical soils. Data from the literature and other databases were used to establish relationships between BD and clay or clay plus silt content, and between DC and macroporosity and yield of crops under no-tillage in subtropical Brazil. Data of BDref reached by the soil Proctor test on disturbed soil samples, by uniaxial compression with loads of 200 kPa on disturbed and undisturbed soil samples, and 400, 800 and 1600 kPa on undisturbed soil samples, were used. Also, comparisons were made with critical bulk density based on the least limiting water range (BDc LLWR) and on observed root and/or yield restriction in the field (BDc Rest). Using vertical uniaxial compression with a load of 200 kPa on disturbed or undisturbed samples generates low BDref and high DC-values. The standard Proctor test generates higher BDref-values, which are similar to those in a uniaxial test with a load of 1600 kPa for soils with low clay content but lower for soils with high clay content. The BDc LLWR does not necessarily restrict root growth or crop yield under no-tillage, since field investigations led to higher BDc Rest-values. A uniaxial load greater than 800 kPa is promising to determine BDref for no-tillage soils. The BDref is highly correlated to the clay content and thus pedotransfer functions may be established to estimate the former based on the latter. Soil ecological properties are affected before compaction restricts plant growth and yield. The DC is an efficient parameter to identify soil compaction affecting crops. The effect of compaction on ecological properties must also be further considered. 相似文献
There is a growing interest in highly fermentable dietary fibers having the potential to reduce risks of disease through the production of short-chain fatty acids (SCFA). Recently a digestion-resistant retrograded maltodextrin (RRM), classified as type 3 resistant starch was developed. Systematic work to determine its molecular and physiological properties was carried out to determine (1) the fraction resistant to digestion in vitro and in vivo, (2) its postconsumption effect on blood glucose in healthy volunteers, and (3) its in vitro fermentation pattern, at different ages, by use of pooled fresh human fecal inoculum. RESULTS: The digestion resistant fraction obtained in vivo from ileostomy patients (59.4%) is similar to that obtained by the AOAC method for measuring retrograded resistant starch (59.7%). The relative glycemic response after consumption of 50 g of RRM was 58.5% compared to glucose set as 100%. When exposed to colonic microbiota, in vitro obtained indigestible fractions behave similarly to those obtained in vivo in ileostomy patients. Fermentation of RRM and production of butyric acid is negligible during the first months of life but develops subsequently during weaning. In adults, RRM fermentation results in a high yield of SCFA, with butyrate representing 21-31 mol % of total SCFA. The high yield of SCFA during colonic fermentation, observed from weaning age on, as well as the potential to help reduce glycemic load may be of benefit to a number of health-related functions in the host. The extent and persistence of the effect of soil compaction in a system with annual ploughing were investigated in 21 long-term field experiments in Sweden with a total of 259 location-years. Crop yield, soil physical properties and plant establishment were determined. All experiments had two common treatments: control (no extra traffic) and compacted (350 Mg km ha−1 of experimental traffic in the autumn prior to ploughing), using a tractor and trailer with traditional wheel equipment and an axle load restricted to 4 Mg. During the rest of the year, both treatments were conventionally and equally tilled. The compaction was repeated each autumn for at least 7 years, and the yield was determined each year until 5 years after the termination of the compaction treatment.
Compaction decreased the porosity and the proportion of large pores and increased the tensile strength of dry aggregates. On clay and loam soils, it decreased the proportion of fine aggregates in the seedbed and the gravimetric soil water content in the seedbed.
The yield in the compacted treatment declined compared with the control during the first 4 years, after which it reached steady state. During this steady state, the compaction treatment caused a yield loss of 11.4%, averaged over 107 location-years. Within 4–5 years after the termination of the compaction treatment, the yield returned to the control level. The average yield loss at individual sites increased with increasing clay content.
Results from additional treatments indicated that yield loss was linearly correlated with the amount of traffic up to 300–400 Mg km ha−1. With greater ground contact pressure or a greater soil water content at time of traffic, there was a greater yield loss.
Soil compaction effects on yield were similar for all spring-sown crops, and the percentage yield loss seemed to be independent of the yield. In a few location-years with winter wheat there was on average no yield decrease.
There were 5.1% less plants in the compacted treatment than in the control. The yield decrease was significantly correlated with the number of plants.
Between years results were highly variable, and no consistent correlations between yield loss and soil water content at the time of traffic or the weather conditions during the growing period were found. Soil compaction affected yield during years with good as well as poor conditions for crop growth. 相似文献
Using in-growth soil cores in cylindrical mesh bags, the effects of 3 soil compaction treatments on growth of crop roots were studied in a sandy soil. The bags were inserted after crop emergence in holes (70 mm diameter; 60 cm depth) augered in the soil in crop row interspaces. In 1984 (with rapessed), at all sampling dates, root biomass in the inserted cores decreased with increased compaction of the plough layer (0–25 cm) as well as the subsoil (25–60 cm). Root biomass in the subsoil was low. In 1985 (with wheat), the effects of compaction in the subsoil were similar, although root biomass was greater than in 1984. However, in the plough layer there were significant differences in root biomass on only one sampling date. The mesh bag technique should be a useful complement to other field methods in studies of relations between physical soil characteristics or tillage treatments and root growth. 相似文献
A single nucleotide polymorphism in the equine herpesvirus 1 (EHV-1) DNA polymerase gene (ORF30 A2254 to G) has been associated with clinical signs of equine herpes myeloencephalopathy (EHM). The purpose of our study was to determine the odds ratio for this genetic marker and EHM using a panel of field isolates from North America collected over the past twenty-three years. EHV-1 isolates cultured at the Cornell University Animal Health Diagnostic Laboratory from 1984 to 2007 were retrieved along with their clinical histories. DNA was extracted from these EHV-1 cultures and allelic discrimination was performed using real-time PCR. The results were confirmed by sequencing of the target region in ORF30. PCR and sequencing were in 100% agreement and showed that 19 out of the 176 isolates had the ORF30 G2254 allele (11%), of which16 were EHM cases and 3 respiratory or abortion cases. The odds of having neurologic disease with the ORF30 G2254 genotype were computed as 162 times greater than those with the opposite allele ORF30 A2254 (95% confidence interval: 35–742). Despite this strong statistical significance, 24% (5/21) of horses with neurologic disease in our study population harbored the “non-neurologic” form of the allele (ORF30 A2254), suggesting that other factors may also contribute to the onset of EHM. 相似文献
