Stripe rust of wheat caused by the fungal pathogen is a destructive foliar disease of wheat. Thus, it is crucial step to characterize the resistant germplasm for stripe rust in a diverse germplasm pool for their ultimate utilization in efficient crop rust resistance breeding. In the present study, we followed two pronged strategies involving integrated phenotypic and molecular characterization of 440 diverse wheat germplasm lines for rust resistance. The germplasm panel was extensively evaluated in field epiphytotic conditions during two consecutive years. After rigorous screening, 72 accessions were successfully revealed as resistant to moderately resistant to stripe rust. Subsequently, entries were then evaluated for their field agronomicperformances, considering prerequisites for serving as a donor germplasm,particularly for yield and 33 potential rust-resistant accessions were identified. Furthermore, to detect the sources of resistance, accessions were molecular characterized for potential race-specific resistance genes Yr5, Yr10,Yr15, and effective adult plant resistance (APR) gene Lr34/Yr18/pm38. We identified the 22 accessions possessing one or more single resistance genes and two accessions were observed with at least three of them. Moreover, Lr34/Yr18/pm38 was determined to confer resistance when observed along with any of the race-specific genes. Thus, the study not only provides proof of concept methodology to identify candidate resistant sources from large germplasm collections but simultaneouslyconfirmed the contribution of combining race-specific andnon-specific APR genes. The finding could further assist in the potential deployment of resistant genes directly into the stripe rust breeding program by involving marker-assisted approaches.  相似文献

Summary Methods of chemical kinetics are used to study stress relaxation behaviour of wood blocks made fromPinus sylvestris and compressed parallel to grain at 20°C and 65% relative humidity. The effect of stress levels on stress behaviour is investigated. It is found that both activation volume and activation energy decrease with increasing stress level. The results prove the validity of the chemical kinetics approach in describing stress relaxation of wood blocks in compression. Suggestive explanations are given for the variations in activation energy and activation volume with stress level.Part of this work was carried out at the School of Agriculture and Forestry Sciences, University of North Wales, Bangor, UK through an ODA TCT fellowship granted to one of the authors (SPSR). An experiment was conducted for 120 days to evaluate optimum dietary lipid requirements for gonadal maturation of Cyprinus carpio fed with varying dietary lipid levels under biofloc‐based systems (BFT). About 180 fingerlings (22 g ± 0.05) were randomly distributed in 15 tanks (300 L) at the rate of 48 no./m2 and fed with varying lipid levels (T1—4% lipid with BFT, T2—6% lipid with BFT, T3—8% lipid with BFT, T4—10% lipid with BFT, control—10% lipid without BFT). The C/N ratio of 20:1 was maintained using tapioca flour as carbon source. Biofloc units supplemented with 8% dietary lipid (T3) showed advanced maturation in terms of absolute fecundity (9,913 ± 7.62), relative fecundity (229.0/g ± 11.92), gonadosomatic index (24.47% ± 1.27), hepatosomatic index (1.97% ± 0.07), condition factor (0.02 g/cm3 ± 0.00) compared with control (clear water with 10% lipid) (p < .05). Histological observations of gonads also revealed that the biofloc groups with supplementation of 8% dietary lipid promoted gonadal maturation for female oocyte and 6% dietary lipid promoted maturation for males, compared with control (clear water with 10% lipid). The results obtained in this experiment elucidate that the biofloc improves gonadal maturation of common carp broodstock at a dietary supplementation with 8% lipid compared with conventional system of broodstock management.  相似文献

A late blight forecasting model “INDO-BLIGHTCAST” has been developed at the Central Potato Research Institute, Shimla, India using meteorological data and late blight appearance dates at four locations in the Indo-Gangetic plains. The model involves computation of physiological days (P-days) and mean relative humidity (RH) of the night, accrued over seven consecutive days. Late blight was predicted to appear within 15 days if moving cumulative effective temperature (P-days) and RH exceeded 52.5 and 525, respectively for seven consecutive days. The developed model was also tested against independent data sets at three locations in the plains, two in plateau region and three in the hills. Statistical comparisons of observed and predicted dates of late blight appearance showed that the mean absolute error was 10.48, while the residual mean square error was only 13.17 indicating that the errors were quite low. The Willmott D index was 0.84 which is quite close to unity thus indicating high accuracy of model predictions and its applicability across regions and seasons. Receiver operating characteristic analysis also confirmed the superiority of this combination (accuracy of 73.6%, AUC of 0.725) to predict late blight appearance as well as its non-appearance in unfavourable years.  相似文献

Background

While vascular and immune abnormalities are common in juvenile and adult dermatomyositis (DM), the molecular changes that contribute to these abnormalities are not clear. Therefore, we investigated pathways that facilitate new blood vessel formation and dendritic cell migration in dermatomyositis.

Methods

Muscle biopsies from subjects with DM (9 children and 6 adults) and non-myositis controls (6 children and 7 adults) were investigated by immunohistochemistry using antibodies that recognize existing (anti-CD146) and newly formed blood vessels (anti-αVβ3) and mature dendritic cells (anti-DC-LAMP). Blood vessel quantification was performed by digitalized image analysis. Additional muscle biopsies from subjects with adult DM and non-myositis controls were used for global gene expression profiling experiments.

Results

A significant increase in neovascularization was found in muscle biopsies of DM patients; neovascularization (αVβ3 positive capillaries and vessels per muscle fiber) was much higher in juvenile than in adult DM patients (control vs juvenile DM: Mean ± SE: 0.06 ± 0.01 vs 0.6 ± 0.05; p < 0.0001 and control vs adult DM: Mean ± SE: 0.60 ± 0.1 vs 0.75 ± 0.1; p = 0.051). Gene expression analysis demonstrated that genes that participate not only in angiogenesis but also in leukocyte trafficking and the complement cascade were highly up regulated in DM muscle in comparison to age matched controls. DC-LAMP positive dendritic cells were highly enriched at perivascular inflammatory sites in juvenile and adult DM patients along with molecules that facilitate dendritic cell transmigration and reverse transmigration (CD142 and CD31).

Conclusion

These results suggest active neovascularization and endothelial cell activation in both juvenile and adult DM. It is likely that close association of monocytes with endothelial cells initiate rapid dendritic cell maturation and an autoimmune response in DM.

Green tea manufacture was standardized with respect to the inactivation of polyphenol oxidase (PPO), rolling, and drying for quality manufacture. Inactivation of PPO by parching, steaming, microwave heating, and oven heating was monitored in tea shoots. The inactivated shoots were rolled under regimens of high and low pressures and dried by microwave heating, oven heating, or sun-drying; total phenols and catechins were estimated. Parched and sun-dried teas contained the lowest levels of total phenols and catechins, and their infusions were dull in color with a slightly burnt odor. Microwave-inactivated and-dried teas showed the highest levels of total phenols and catechins, and their infusions were bright in color and sweet in taste with a subtle pleasant odor. In steam-inactivated and oven/microwave-dried teas, total phenol and catechin contents were intermediate between parched and sun-dried teas and microwave-inactivated and microwave-dried teas, and their infusions were bright with a umami taste.  相似文献

The crude ethanolic extract of aerial parts of Leucas aspera was tested for its acaricidal properties against Rhipicephalus (Boophilus) annulatus. The per cent adult mortality, inhibition of fecundity and hatching of laid ova were studied at concentrations of 1.56, 3.13, 6.25, 12.5, 25, 50 and 100mg/ml. Adult tick mortality was significant at the highest concentration tested. Inhibition of fecundity of treated groups differed significantly from control and was concentration dependent. L. aspera extract also produced complete failure of eclosion of eggs from the treated ticks even at lower dilutions of the extract.  相似文献
