Xanthomonas campestris pv. campestris (Xcc) is the causal agent of black rot in Brassicaceae. It is widespread in Italy and severe outbreaks occur under conditions that favour disease development. In this study a multilocus sequence typing approach (MLST) based on the partial sequence of seven loci was applied to a selection of strains representative of the main areas of cultivation and hosts. The aim was to investigate whether the long tradition of brassica crops in Italy has influenced the evolution of different Xcc populations. All loci were polymorphic; 14 allelic profiles were identified of which 13 were unique to Italian strains. Based on the seven loci, the most common genotype within the Italian Xcc strains (AP1) was also the most representative genotype found in worldwide Xcc strains. This genotype was included in a new clonal complex in addition to three other clonal complexes already identified in Xcc populations. The phylogenetic reconstruction using a concatenated dataset of four conserved protein-coding genes, dnaK, fuyA, gyrB and rpoD, showed that the Italian strains belonged to two genetic groups. Physiological races were also investigated for the first time in Italy. The race structure of Xcc was determined by inoculating eight differential Brassica lines belonging to five species and showed that, in Italy, race 4 is the most widespread, followed by races 1 and 6. No correlation was found between allelic profiles, host of isolation, geographical origin and races, although a prevalent race was identified within the same clonal complex. 相似文献

An unknown Phytophthora species was discovered in the central Peruvian Andes on blighted foliage of the native South American plant species Urera lacineata. Urera is a genus of native flowering shrubs in the nettle family Urticaceae. This new taxon Phytophthora urerae sp. nov. is herewith formally described based on extensive morphological analysis, phylogenetic analysis of nuclear and mitochondrial loci, and AFLP analysis. Phytophthora urerae sp. nov. is a close relative of the Irish famine pathogen, Phytophthora infestans, and only the third clade 1c taxon described from South America to date. In contrast to the clade 1c taxon Phytophthora andina, first described in South America as a hybrid, P. urerae does not appear to be a hybrid based on cloning and sequencing nuclear loci. Findings of new species in South America may provide novel insights into the origin and evolutionary history of clade 1c Phytophthora species. 相似文献

One of the economically important diseases of lettuce is lettuce big-vein disease (LBVD), which leads to severe yield losses. LBVD is associated with a complex of two viruses, Lettuce big-vein associated virus (LBVaV) and Mirafiori lettuce big-vein virus (MLBVV). These viruses are transmitted by motile zoospores of Olpidium spp. fungi, which persist in the soil for decades through resting spores. In greenhouse and field experiments, this study tested whether changing plant and soil temperatures together with fungicide application would have a significant effect on controlling LBVD in lettuce. Soil fumigation with metam sodium was not effective at controlling the disease, as opposed to treatment with chloropicrin and methyl bromide. Moreover, the fungicides carbendazim and fluazinam were effective in reducing the incidence of Olpidium virulentus. Nevertheless, control of the fungal vector did not seem to be sufficient to control the disease due to the transition ability of the virus under low vector abundance. Crop covers, which affect the favourable environmental conditions for the viruses by lowering soil temperature and raising air temperature, reduced the disease symptoms. Combining fungicides with crop cover had a synergistic effect on reducing disease symptoms, thus providing a sustainable solution for LBVD. 相似文献

Brown spot needle blight (BSNB), a disease of pine trees caused by the fungus Lecanosticta acicola, has been known in Slovenia since 2008 and in Croatia since 1975. Recent outbreaks in Slovenia prompted this study to compare L. acicola populations in these two neighbouring European countries. Sixty-nine isolates collected from three pine species (Pinus mugo, P. halepensis and P. nigra) were used to determine the phylogenetic relationships, genetic structure, and reproductive strategy of the pathogen. EF1-α sequences showed that Slovenian and Croatian isolates share a common ancestry with individuals from central and northern Europe. Population structure analysis revealed four distinct population clusters of L. acicola in these two countries, generally corresponding to their respective geographic location and host. An unequal ratio of mating types and a low overall genetic diversity in the population indicated a strong influence of asexual reproduction. Although some of the oldest recorded European occurrences of BSNB are from Croatia, this study provided no evidence that the population studied in Croatia was the source of the sampled outbreaks in Slovenia. A programme of field trials for the study of the winter barley–Rhynchosporium commune pathosystem is reported. The associated seedborne disease rhynchosporium leaf scald is regarded as having an important impact on barley yields. The analysis in this study relates to the impact of the seed source (commercial or farm-saved seed) on disease incidence and to the spatial pattern of rhynchosporium leaf scald disease incidence. Disease incidence data were calculated from field data recorded as disease severity. Mean disease incidence was higher in the crops grown from farm-saved seed than in those grown from commercial seed, although great agronomic significance cannot be attached to this result. The spatial pattern of rhynchosporium leaf scald disease incidence was characterized in terms of the binary power law (BPL) and was indicative of an aggregated pattern. Programme-wide BPL results were described using a novel phytopathological application of a random coefficients model. These results have application in field sampling for rhynchosporium leaf scald disease. 相似文献

Cotton blue disease (CBD) is the most important disease present in cotton crops in South America and cotton leafroll dwarf virus (CLRDV) is the causal agent. The disease has been controlled by sowing cotton varieties resistant to CLRDV. However, in the 2009/10 growing season, an outbreak due to an atypical CLRDV isolate (CLRDV-at) occurred in northwest Argentina. Although CLRDV and CLRDV-at genomes are very closely related, the symptoms they produce in cotton plants are quite different. P0 is the most divergent protein between the isolates and in CLRDV is a silencing suppressor protein. This work characterized the silencing suppressor activity of the P0 protein encoded by CLRDV-at (P0CL-at) and evaluated its role in Cbd-resistance break in cotton plants. It was demonstrated that P0CL-at, despite having a mutation in the consensus of the F-box-like motif, was able to suppress local RNA silencing, but displayed lower activity than P0CL. P0CL and P0CL-at showed no differences in the interaction with Gossypium hirsutum SKP1 orthologue (GSK1) and Nicotiana benthamiana SKP1 and both P0 proteins triggered destabilization of ARGONAUTE1. However, when the ability to enhance PVX symptoms was evaluated, P0CL-at was shown to be a weaker pathogenicity factor than P0CL in N. benthamiana. Interestingly, trans-expressed P0CL-at enabled CLRDV to systemically infect CBD-resistant plants, and a chimeric CLRDV-P0CL-at infectious clone succeeded in establishing infection in CBD-resistant cotton varieties with symptoms resembling those produced by CLRDV-at. These results strongly suggest that P0CL-at is the avirulence (Avr) determinant involved in breaking cotton Cbd gene-based resistance. 相似文献

The impact of increasing temperatures on the population dynamics of the soil-dwelling nematode Globodera pallida, a persistent and economically important pest of potatoes, was investigated. The reproductive factor (final population⁄initial population) and length of life cycle were found to be temperature sensitive. Pot experiments performed over 4 months allowed comparison of the effect on development of G. pallida of two temperature regimes: an average temperature comparable to current field conditions (14.3 °C) and an average temperature above current field conditions (17.3 °C). A larger second generation of juveniles was observed at 17.3 °C compared to 14.3 °C. Multiplication of G. pallida at field sites in Shropshire and East Lothian (average soil temperatures of 15.5 and 14.1 °C, respectively, during potato cropping) was also examined. A quantitative PCR assay and visual examination of roots were used to monitor the dynamics of the G. pallida populations in both field sites at 4-weekly intervals. Four cultivars, Desirée, Cara, Maris Piper and Estima, were grown with and without nematicide treatments. Nematicide treatments suppressed population increases at both sites. Females were observed on the roots of cvs Cara and Desirée at the end of the growing season in Shropshire, but not at East Lothian, and are likely to represent a second generation. 相似文献

The issue of cross‐ or multiple resistance to acetolactate synthase (ALS) inhibitors and the auxinic herbicide 2,4‐D was investigated in Papaver rhoeas L., a common and troublesome weed in winter cereals, in a broad‐scale study across four European countries. A combination of herbicide sensitivity bioassays and molecular assays targeting mutations involved in resistance was conducted on 27 populations of P. rhoeas originating from Greece (9), Italy (5), France (10) and Spain (3). Plants resistant to the field rate of 2,4‐D were observed in 25 of the 27 populations assayed, in frequencies ranging from 5% to 85%. Plants resistant to ALS‐inhibiting herbicides (sulfonylureas) were present in 24 of the 27 populations, in frequencies ranging from 4% to 100%. Plants resistant to 2,4‐D co‐occurred with plants resistant to sulfonylureas in 23 populations. In four of these, the probability of presence of plants with cross‐ or multiple resistance to 2,4‐D and sulfonylureas was higher than 0.5. ALS genotyping of plants from the field populations or of their progenies, identified ALS alleles carrying a mutation at codon Pro197 or Trp574 in 2,4‐D‐sensitive and in 2,4‐D‐resistant plants. The latter case confirmed multiple resistance to 2,4‐D and ALS inhibitors at the level of individual plants in all four countries investigated. This study is the first to identify individual plants with multiple resistance in P. rhoeas, an attribute rarely assessed in other weed species, but one with significant implications in designing chemical control strategies. 相似文献

Increasingly, weeds have been taking on global distributions. With the proliferation of invasive weeds has come the challenge of managing these species over broad geographical regions, with diverse habitats and political jurisdictions. Here, we review the management of Mikania micrantha Kunth (Asteraceae; mile‐a‐minute) throughout its invaded range, extending through most of the Pacific islands and southern and south‐east Asia. Context matters when determining the best course of action for managing M. micrantha, as it has invaded a large variety of agricultural and natural systems. In Queensland, Australia and Florida, USA, M. micrantha has been targeted in relatively successful eradication campaigns, highlighting the importance of early detection and rapid response methods, while elsewhere in its invaded range, populations are either still increasing or showing limited signs of decline. An inter‐regional approach to research and management should incorporate successful management strategies employed throughout the invaded range including, but not limited to, chemical and cultural control practices, manual and mechanical control, classical biological control using the rust fungus Puccinia spegazzinii, plant–plant competition and integrated approaches utilising two or more control methods concurrently. Additional knowledge of M. micrantha genetics is required to determine if management approaches could be fine‐tuned for particular populations. Countries bordering the Mekong River formed a network in 2011 to co‐ordinate the management of invasive species such as M. micrantha. Expanding such a collaborative approach to other regions could further reduce populations of M. micrantha and limit its spread. 相似文献
