Summary Lucerne was irrigated for three years on a slowly permeable, duplex soil, with saline water up to 2.4 dS m–1 without significant yield decline. Irrigation water of 4.5 dS m–1 significantly reduced yield. Lucerne yield was most closely related to the soil ECe of the 0–15 cm depth, rather than the total rootzone, and was described by; Relative yield=100–6.5 (ECe-2.1). While lucerne roots reached depths of at least 150 cm, approximately 80% of total root length was located in the 0–60 cm depth.Increasing salinity increased the plant concentrations of sodium and chloride, however, these changes were not closely related to changes in yield.Soil salinity increased with increasing salinity of the applied water. However, during the irrigation season water penetration and the accumulation of salt within the profile was predominantly restricted to the 0–60 cm depth. No portion of the applied irrigation water was available as a leaching fraction. Any leaching of salts to the watertable, particularly below 120 cm, was due to winter rainfall rather than the application of summer irrigation water.Ripping the soil to a depth of 75 cm increased water infiltration and resulted in increased crop yields, but did not significantly affect the crop relative yield-soil ECe relationship.From the results it is proposed that on the slowly permeable duplex soils, when watertable depth is controlled, management strategies for lucerne irrigated with saline water should be based on controlling the salinity of the shallow soil depths, to 60 cm. 相似文献
Two white clover (Trifolium repens L.) genotypes, identified by their differing ability to increase in biomass in response to added phosphorus (P) (high P responder, low P responder) were subjected to P deprivation and examined for differences in root growth, root surface acid phosphatase activity, and soluble and ionically‐bound root cell wall isoform profile. As leaf P levels declined, the high P responder showed a greater increase in biomass allocated to the roots, and highest root surface acid phosphatase activity. However, these differences were not statistically significant. The onset of P deprivation enhanced the intensity of several acid phosphatase isoforms in the P deprived root extracts from both genotypes. After 11 days, one basic isoform (resolved atpH4.5) was enhanced in the cell wall extract, an enhancement that was also observed after 26 days. However, for the soluble isoforms, a temporal separation of response was observed in both genotypes. After 11 days, there was no discernable enhancement of the major staining basic isoform or in the major staining acidic isoform (resolved at pH 8.8) in extracts from P‐deprived roots. After 26 days, these isoforms were enhanced in extracts from P‐derived root tissues. Further, a second acidic isoform could now be discerned as a major staining enzyme in P‐derived root extracts from both genotypes. 相似文献
Methylselenocysteine (MeSeCys) is an amino acid derivative that possesses potent anticancer activity in animals. Plants that can tolerate growth on soils with high Se content, known as Se hyperaccumulators, do so by converting inorganic Se to MeSeCys by the enzyme selenocysteine methyltransferase (SMT). A cDNA encoding the SMT from a Se hyperaccumulator was overexpressed in tomato (Solanum lycopersicum). Transgenic plants were provided with selenite or selenate to the roots during fruit development, and liquid chromatography-mass spectrometry was used to show that MeSeCys accumulated in the fruit but not in the leaves. Depending on the transgenic line and Se treatment, up to 16% of the total Se in the fruit was present as MeSeCys. MeSeCys was produced more effectively from selenite on a percentage conversion basis, but greater accumulation of MeSeCys could be achieved from selenate due to its better translocation from the roots. Denitrification has the potential to remove excess nitrogen from groundwater passing through riparian buffers, thus improving water quality downstream. In regions with markedly seasonal precipitation, transient stream flow events may be important in saturating adjacent floodplain soils and intermittently providing the anaerobic conditions necessary for denitrification to occur. In two experiments we characterised the denitrification potential of soils from two contrasting floodplains that experience intermittent saturation. We quantified under controlled laboratory conditions: 1) potential rates of denitrification in these soils with depth and over time, for a typical period of saturation; and 2) the influences on rates of nitrate and organic carbon. Treatments differed between experiments, but in each case soil-water slurries were incubated anaerobically with differing amendments of organic carbon and nitrate; denitrification rates were measured at selected time intervals by the acetylene-block technique; and slurry filtrates were analysed for various chemical constituents. In the first experiment (ephemeral tributary), denitrification was evident in soils from both depths (0-0.3 m; 0.3-1.1 m) within hours of saturation. Before Day 2, mean denitrification rates at each depth were generally comparable, irrespective of added substrates; mean rates (Days 0 and 1) were 5.2 ± 0.3 mg N kg dry soil−1 day−1 (0-0.3 m) and 1.6 ± 0.2 mg N kg dry soil−1 day−1 (0.3-1.1 m). Rates generally peaked on Days 2 or 3. The availability of labile organic carbon was a major constraint on denitrification in these soils. Acetate addition greatly increased rates, reaching a maximum in ephemeral floodplain soils of 17.4 ± 1.8 mg N kg dry soil−1 day−1 on Day 2: in one deep-soil treatment (low nitrate) this overcame differences in rates observed with depth when acetate was not added, although the rate increase in the other deep-soil treatment (high nitrate) was significantly less (P ≤ 0.01). Without acetate, peak denitrification rates in this experiment were 6.9 ± 0.4 and 2.8 ± 0.2 mg N kg dry soil−1 day−1 in surface and deep soils, respectively. Differences in rates were observed with depth on all occasions, despite similar initial concentrations of dissolved organic carbon (DOC) at both depths. Levels of substrate addition in the second experiment (perennial stream) more closely reflected natural conditions at the site. Mean denitrification rates were consistently much higher in surface soil (P ≤ 0.001), while the source of water used in the slurries (surface water or groundwater from the site) had little effect on rates at any depth. Mean rates when all treatments retained nitrate were: 4.5 ± 0.3 mg N kg dry soil−1 day−1 (0-0.3 m depth); 0.8 ± 0.3 mg N kg dry soil−1 day−1 (0.3-1.0 m); and 0.6 ± 0.1 mg N kg dry soil−1 day−1 (1.8-3.5 m). For comparable treatments and soil depths, denitrification potentials at both sites were similar, apart from higher initial rates in the ephemeral floodplain soils, probably associated with their higher DOC content and possibly also their history of more frequent saturation. The rapid onset of denitrification and the rates measured in these soils suggest there may be considerable potential for nitrate removal from groundwater in these floodplain environments during relatively short periods of saturation. 相似文献
The spotted-wing drosophila, Drosophila suzukii Matsumura, is an invasive pest causing significant damage to soft skinned fruits. Control of D. suzukii is critical since there is no tolerance for infested fruit in the market. While most insecticides control one or more D. suzukii life-stages (e.g., egg, larvae, and adult), the impact of insecticides that are toxic to immature stages is unclear on the subsequent generation of a field population. Insecticides were applied at field recommended rates on cherries and blueberries in the laboratory to determine immature D. suzukii mortality. Spinetoram, cyantraniliprole, malathion, methomyl, spinosad, and phosmet resulted in relatively high mortality of all immature life stages. Zeta-cypermethrin, cyclaniliprole, and fenpropathrin resulted in lower mortality of egg and all larval instars. Malathion was also applied to lowbush blueberries with different fruit sizes (small, medium, and large) in the laboratory and there was no statistical difference in mortality rates depending on fruit sizes. Mortality data from the laboratory experiments were used to parameterize a refined D. suzukii population model. The model revealed that the timing and order of different insecticide classes are important to control D. suzukii population. Model runs that included early applications of more effective insecticides resulted in high immature mortality and greater reduction of D. suzukii populations compared to treatments applied later.
Eucalyptus spp. are among the most widely planted exotic trees in Ethiopia. Several damaging leaf pathogens are known from Eucalyptus spp. worldwide. Of these, Mycosphaerella spp. are among the most important, causing the disease known as Mycosphaerella leaf disease (MLD). Characteristic symptoms of MLD include leaf spot, premature defoliation, shoot and twig dieback. Recent disease surveys conducted in Ethiopian Eucalyptus plantations have revealed disease symptoms similar to those caused by Mycosphaerella spp. These symptoms were restricted to E. globulus trees growing in several localities in south, south western and western Ethiopia. The aim of this study was to identify the fungi associated with this disease. This was achieved by examining ascospore germination patterns, anamorph associations and sequence data from the Internal Transcribed Spacer (ITS) region of the rRNA operon, for representative isolates. Several different ascospore germination patterns were observed, suggesting that more than one species of Mycosphaerella is responsible for MLD on E. globulus in Ethiopia. Analysis of sequence data showed that three Mycosphaerella spp., M. marksii, M. nubilosa and M. parva were present. This is the first report of these three species from Ethiopia and represents a valuable basis on which to build further studies in the region. 相似文献
Phytoremediation is an emerging technology that uses plants and their associated microbes to clean up pollutants from the soil, water, and air. In order to select the plant growth-promoting rhizobacteria(PGPR) for phytoremediation of heavy metal contamination, 60 bacterial strains were isolated from the rhizosphere of two endemic plants, Prosopis laevigata and Spharealcea angustifolia, in a heavy metal-contaminated zone in Mexico. These rhizobacterial strains were characterized for the growth at different pH and salinity, extracellular enzyme production, solubilization of phosphate, heavy metal resistance, and plant growth-promoting(PGP) traits, including production of siderophores and indol-3-acetic acid(IAA). Overall, the obtained rhizobacteria presented multiple PGP traits. These rhizobacteria were also resistant to high levels of heavy metals(including As as a metalloid)(up to 480 mmol L(-1)As(V), 24 mmol L(-1)Pb(Ⅱ), 21 mmol L(-1)Cu(Ⅱ), and 4.5 mmol L(-1)Zn(Ⅱ)). Seven rhizobacterial strains with the best PGP traits were identified as members of Alcaligenes, Bacillus, Curtobacterium, and Microbacterium, and were selected for further bioassay.The inoculation of Brassica nigra seeds with Microbacterium sp. CE3R2, Microbacterium sp. NE1R5, Curtobacterium sp. NM1R1,and Microbacterium sp. NM3E9 facilitated the root development; they significantly improved the B. nigra seed germination and root growth in the presence of heavy metals such as 2.2 mmol L(-1)Zn(Ⅱ). The rhizobacterial strains isolated in the present study had the potential to be used as efficient bioinoculants in phytorremediation of soils contaminated with multiple heavy metals. 相似文献
Natural clay minerals can play an important role in crude remediation of wastewater polluted with the heavy metals (HMs) Cu,Zn and Ni.The presence and timing of addition of natural dissolved organic matter (DOM) have a significant effect on the HM removal by clay mineral sorbents.However,the influence of the presence of DOM on the remediation of the used clay mineral sorbents once saturated with HMs is largely unknown.To resolve this,clay mineral-rich soil column of varying composition,loaded (i) with Cu,Zn and Ni only,(ii) first with DOM followed by Cu,Zn and Ni,or (iii) with DOM,Cu,Zn and Ni simultaneously,was used in a set of desorption experiments.The soil columns were leached with 0.001 mol L-1 CaCl2 dissolved in water as control eluent and 0.001 mol L-1 CaCl2 dissolved in DOM as treatment eluent.During the preceding loading phase of the sorbent,the timing of DOM addition (sequential or concurrent with HMs) was found to have a significant influence on the subsequent removal of the HMs.In particular when the column was loaded with DOM and HMs simultaneously,largely irreversible co-precipitation took place.Our results indicate that the regeneration potential of clay mineral sorbents in wastewater treatment will be significantly reduced when the treated water is rich in DOM.In contrast,in manured agricultural fields (where HMs enter together with DOM),HM mobility will be lower than expected from interaction dynamics of HMs and clay minerals. 相似文献
A widely used method for analyzing hydrolyzable tannins afer reaction with KIO(3) has been modified to include a methanolysis step followed by oxidation with KIO(3). In the new method, hydrolyzable tannins (gallotannins and ellagitannins) are reacted at 85 degrees C for 20 h in methanol/sulfuric acid to quantitatively release methyl gallate. Dried plant samples can be methanolyzed under the same conditions to convert hydrolyzable tannins to methyl gallate. Oxidation of the methyl gallate by KIO(3) at pH 5.5, 30 degrees C, forms a chromophore with lambda(max) 525 nm, which is determined spectrophotometrically. The detection limit of the method is 1.5 microg of methyl gallate, and with plant samples, relative standard deviations of less than 3% were obtained. 相似文献
