The purpose of this study was to compare concentrations of nitric oxide (NO) in fresh plasma versus frozen plasma, and determine the temporal effects of freezing on jugular venous plasma NO concentrations in clinically healthy ponies. Twenty-eight helminth-naïve ponies, aged from 4 to 6 mo, were raised and maintained under parasite-free conditions. Blood was collected from the jugular vein, centrifuged, and the plasma supernatant was analyzed fresh for NO concentrations using a chemiluminescent method. The remaining samples were aliquoted into 12 samples and stored at −70°C until they were analyzed for NO concentration. Plasma NO concentration was measured at monthly intervals for 12 mo. There were significant differences in the plasma NO concentration across time compared with the baseline value at 1, 3, 4, 6, 7, and 11 mo. However, these values remained within the range for clinically healthy equids compared with concentrations of NO from horses with increased NO from diseased states.  相似文献

A study was carried out in order to compare two techniques for culture of strongyle larvae: the usual fecal culture and a new one where eggs were extracted from feces and larvae were grown in a more defined medium (agar plate fortified with Earle's medium and yeast extract). The efficiency of "on-agar" cultures was better than that of the fecal cultures with a difference of 26% for Trichostrongylus colubriformis and 32% for Ostertagia circumcincta. The variability observed between the number of larvae collected from each culture was low (8% on average) in comparison with that of fecal cultures (15% on average) thus demonstrating a better reproducibility. On the other hand growth of the larvae was similar to that obtained in fecal medium. This new technique avoids the difficulty of controlling the conditions found in fecal cultures (particularly moisture) and could become a more accurate method for diagnosis than the conventional methods. A morphometric study of infective larvae derived from the two methods of culture was carried out. Despite the more defined conditions of development in the "on-agar" cultures, it was not possible to obtain sufficiently homogeneous populations with regard to their measurements so as to distinguish larvae merely with the help of these criteria.  相似文献

Three molecular typing methods were used to investigate genetic diversity among Xanthomonas campestris pv. campestris isolates obtained in Israel and others previously obtained from different geographical locations (collection isolates). Using pulsed-field gel electrophoresis (PFGE), amplified fragment length polymorphism (AFLP) and repetitive sequence-based PCR (rep-PCR), 22 different isolates were divided into 11, 12 and 13 differentiated genotypes, respectively. All collection isolates yielded different genotypes and, among the isolates from Israel, several new genotypes were found. These findings not only support the observed heterogeneity within X. campestris pv. campestris , but also suggest that variability at the genomic level in this pathovar is higher than previously estimated. Moreover, while previous studies suggested that PCR patterns obtained with integron-specific primers are conserved in most X. campestris pathovars, PCR patterns of this element yielded four different types among the X. campestris pv. campestris isolates tested, thus supporting the relatively high diversity in this pathovar. Although differences in pathogenicity were observed among isolates, assays using cauliflower and radish did not indicate a correlation between pathogenicity and genotype.  相似文献

Extracorporeal shock wave therapy is becoming increasingly used in equine practice to treat musculoskeletal disorders. Although the exact effects of extracorporeal shock waves on tissues still need to be elucidated, transient cutaneous analgesia has been observed within treated areas in humans and horses. The purpose of this study was to determine the onset, magnitude, and duration of cutaneous analgesia after non-focused extracorporeal shock wave (ESW) application by comparing the limb withdrawal reflex latency (LWRL) in treated and untreated areas on the metacarpus of horses, using a focused light source. A single treatment with non-focused ESW was applied to 3 areas over the dorsal aspect of one metacarpus in 12 horses. The LWRL was measured to assess cutaneous sensation in treated and untreated control areas of the treated metacarpus and in an untreated area on the opposite metacarpus, before and at various time points after ESW application. Most treated and control areas revealed a significant decrease in LWRL over time compared with baseline values. Although the results of our study do not suggest cutaneous analgesia after ESW application to the equine metacarpus, we advise cautious use of this treatment modality for horses in training or before competition until further research is done.  相似文献

Leaf-cutting ants consume up to 10% of canopy leaves in the foraging area of their colony and therefore represent a key perturbation in the nutrient cycle of tropical forests. We used a chronosequence of nest sites on Barro Colorado Island, Panama, to assess the influence of leaf-cutting ants (Atta colombica) on nutrient availability in a neotropical rainforest. Twelve nest sites were sampled, including active nests, recently abandoned nests (<1 year) and long-abandoned nests (>1 year). Waste material discarded by the ants down-slope from the nests contained large concentrations of nitrogen and phosphorus in both total and soluble forms, but decomposed within one year after the nests were abandoned. Despite this, soil under the waste material contained high concentrations of nitrate and ammonium that persisted after the disappearance of the waste, although soluble phosphate returned to background concentrations within one year of nest abandonment. Fine roots were more abundant in soil under waste than control soils up to one year after nest abandonment, but were not significantly different for older sites. In contrast to the waste dumps, soil above the underground nest chambers consistently contained lower nutrient concentrations than control soils, although this was not statistically significant. We conclude that the ‘islands of fertility’ created by leaf-cutting ants provide a nutritional benefit to nearby plants for less than one year after nest abandonment in the moist tropical environment of Barro Colorado Island.  相似文献

Bread doughs of various compositions were prepared and studied by computed X-ray microtomography (XRT) with high resolution (5 μm). Their cellular structure was followed during the last stage of proofing, from 40 min to 180 min, to focus on the films separating bubbles and their possible coalescence. Image analysis allowed to determine the porosity, the connectivity index, the bubble size and the cell walls thickness distributions which were fitted by usual mathematical functions. Images showed that different compositions led to different cellular structures. The kinetics of porosity and connectivity had sigmoid shapes with 0.7 and 1 as asymptotic values, respectively. For an average formula, mean cell width grew from 410 μm to 675 μm and the mean cell wall thickness grew from 200 μm to 230 μm. At the end of proofing, most bubbles were interconnected, i.e. they were separated by films of thickness lower than 5 μm. Changes of recipe led to different structures, which were discussed through the influence of the liquid fraction and of the sugar content on the coalescence of bubbles. These results suggested that at the end of fermentation, the dough could be considered as a three phase co-continuous medium: gas/liquid/viscoelastic matrix.  相似文献

Attempts have been made to predict Chopin alveograph or French bread-making tests, using tree-based models and PLS regressions. Data came from three sets of trials, involving 130, 214 and 103 different genotypes, which were described for HMW-GS, LMW-GS and small-scale tests currently used in breeding programs. Segmentation trees and PLS regressions indicated that HMW-GS and LMW-GS were not sufficient to explain alone the variability of bread wheat quality. This could be partly due to “allele × environment” and “locus × locus” interactions. For HMW-GS, Glu-B1 was the predominant locus for alveograph and French bread-baking, and some differences in the alleles hierarchy were demonstrated according to the end-use parameter considered. For LMW-GS, Glu-B3 seemed to be preponderant, with alleles b′, c and g being favourable and allele c′ unfavourable. Joint use of the information brought by glutenin subunits and technological tests did not enable to predict satisfactorily, neither the different parameters of French bread-baking, nor the extensibility L of alveograph. Only the prediction of the strength W proved reliable, and robust PLS equations were proposed for this alveograph parameter. These prediction equations could be of interest to select for high values of W in the mid generations of breeding.  相似文献

  • ? Liming is a forestry practice used to counteract forest decline in acidic soils. It consists of direct Ca and Mg input to forest soil, which restores tree mineral nutrition, but also modifies microbial communities in soil. The aim of this study was to assess the effects of liming on both belowground (ectomycorrhizal root tips) and aboveground (epigeous sporocarps) fungal communities.

Results showed that the modification of soil chemical properties (pH, and Ca-Mg contents versus total free Al and Fe concentrations) was a stronger factor of ECM community structuring than tree host. The species appearing in limed plots were ubiquist or known as good competitors and replaced acidophilic and stress species.
  • ? At the sporocarp level, tree host was a stronger factor of community structuring than soil chemical properties associated with liming. On the whole, there was a shift in the community composition from a typical acidophilic forest fungal community of medium altitude in the untreated plots to a less typical one, with the reduced dominance of acidophilic fungi while many late-stage forest species appeared.

    In the climate change discussion, the possibility of carbon sequestration of forests plays an important role. Therefore, research on the effects of environmental changes on net primary productivity is interesting. In this study we investigated the influence of changing temperature, precipitation and deposition of sulphur and nitrogen compounds on forest growth. The database consisted of 654 plots of the European intensive monitoring program (Level II plots) with 5-year growth data for the period 1994–1999. Among these 654 plots only 382 plots in 18 European countries met the requirements necessary to be used in our analysis. Our analysis was done for common beech (Fagus sylvatica), oak (Quercus petraea and Q. robur), Scots pine (Pinus sylvestris) and Norway spruce (Picea abies). We developed an individual tree growth model with measured basal area increment of each individual tree as responding growth factor and tree size (diameter at breast height), tree competition (basal area of larger trees and stand density index), site factors (soil C/N ratio, temperature), and environmental factors (temperature change compared to long-term average, nitrogen and sulphur deposition) as influencing parameters. Using a mixed model approach, all models for the tree species show a high goodness of fit with Pseudo-R2 between 0.33 and 0.44. Diameter at breast height and basal area of larger trees were highly influential variables in all models. Increasing temperature shows a positive effect on growth for all species except Norway spruce. Nitrogen deposition shows a positive impact on growth for all four species. This influence was significant with p < 0.05 for all species except common beech. For beech the effect was nearly significant (p = 0.077). An increase of 1 kg N ha−1 yr−1 corresponds to an increase in basal area increment between 1.20% and 1.49% depending on species. Considering an average total carbon uptake for European forests near 1730 kg per hectare and year, this implies an estimated sequestration of approximately 21–26 kg carbon per kg nitrogen deposition.  相似文献
