Glycated proteins are considered as one of the factors involved in the pathogenesis of diabetic complications, including nephropathy. These proteins are formed endogenously under conditions of hyperglycemia, as well as being provided with food containing sugars, which was subjected to high temperature. Examples are egg products. One of the proteins found in eggs in a relatively high concentration is chicken cystatin (ovocystatin). It is now believed that some proteins can passage the intestinal epithelium by transcytosis directly into the bloodstream. Thus, glycated protein present in food can be an additional source of glycotoxins. The aim of this study was to compare the affinity of native and glycated cystatin to the brush border membranes of rat kidney. Kinetic analysis was performed with surface plasmon resonance technique using sensor chip L1. Dissociation constants for native and glycated cystatin (Kd) were 2.76 μmol/L and 3.82 μmol/L, respectively. The results of our study indicate that glycation only slightly affects binding of cystatin to brush border membranes. This suggests that glycated cystatin and other glycated proteins may also be efficiently taken up in the kidney proximal tubule. The observation may be important for understanding the mechanisms involved in the development of diabetic nephropathy. 相似文献

Scaling is widely recognized as a central issue in ecology. The associated cross-scale interactions and process transmutations make scaling (i.e. a change in spatial or temporal grain and extent) an important issue in understanding ecosystem structure and functioning. Moreover, current concepts of ecosystem stewardship, such as sustainability and resilience, are inherently scale-dependent. The importance of scale and scaling in the context of forest management is likely to further increase in the future because of the growing relevance of ecosystem services beyond timber production. As a result, a consideration of processes both below (e.g. leaf-level carbon uptake in the context of climate change mitigation) and above (e.g. managing for biodiversity conservation at the landscape scale) the traditional focus on the stand level is required in forest ecosystem management. Furthermore, climate change will affect a variety of ecosystem processes across scales, ranging from photosynthesis (tree organs) to disturbance regimes (landscape scale). Assessing potential climate change impacts on ecosystem services thus requires a multi-scale perspective. However, scaling issues have received comparatively little attention in the forest management community to date. Our objectives here are thus first, to synthesize scaling issues relevant to forest management and second, to elucidate ways of dealing with complex scaling problems by highlighting examples of how they can be addressed with ecosystem models. We have focused on three current management issues of particular importance in European forestry: (1) climate change mitigation through carbon sequestration, (2) multi-functional stand management for biodiversity and non-timber goods and services and (3) improving the resilience to natural disturbances. We conclude that taking into account the full spatiotemporal heterogeneity and dynamics of forest ecosystems in management decision-making is likely to make management more robust to increasing environmental and societal pressures. Models can aid this process through explicitly accounting for system dynamics and changing conditions, operationally addressing the complexity of cross-scale interactions and emerging properties. Our synthesis indicates that increased attention to scaling issues can help forest managers to integrate traditional management objectives with emerging concerns for ecosystem services and therefore deserves more attention in forestry. 相似文献

Few tree size/leaf area correlations have been produced for hardwoods, where the extrapolation from individual branches to
the whole tree is less straightforward than in conifers with more regular branching patterns. We used randomized branch sampling
to estimate leaf area of European beech (Fagus sylvatica L.) trees of different stands, ages and areas in Austria. Cross-sectional areas (CSA) predicted 87–92% of leaf area variation,
the best predictor being the sum of branch CSA. Leaf area was somewhat better correlated with CSA at breast height than at
the base of the crown, and using sapwood instead of total CSA made little difference. While there was no effect of growth
area, a stepwise regression model showed that dominant trees in pole-stage had, for unclear reasons, significantly higher
leaf area/CSA relationships. A comparison with regressions produced from smaller beech trees in other parts of Europe suggests
that the leaf area/basal area regression is generally valid for beech in central Europe. 相似文献

BACKGROUND: Although the toxins of Bacillus thuringiensis Berliner (Bt) are frequently used to control lepidopteran pests, the tolerance or resistance of some lepidopteran moth strains may limit Bt applications. In this study, insecticidal cocktails consisting of sublethal doses of Cry1Ab toxin and additive compounds were tested for their suppressive effect on larval relative growth rate (RGR) in Ephestia kuehniella Zeller under laboratory conditions.RESULTS: In the first step, the suppressive effect of diflubenzuron, soybean trypsin inhibitor (STI) and chitinase on RGR was confirmed. In the second step, these compounds were incorporated into a mixture of crushed kernels of Bt maize hybrid MON 810-YieldGard((R)) and its isoline with concentrations of Cry1Ab toxin ranging from 0.011 to 0.091 microg g(-1) diet. An additive effect on the suppression of larval RGR in E. kuehniella was found in a combination of diflubenzuron, STI and STI + chitinase as secondary compounds in insecticidal cocktails. Chitinase showed no additive effect on RGR.CONCLUSION: The highest suppression level was found in cocktails with STI + chitinase as a secondary compound. It is hypothesized that the protease inhibitor (STI) protects both chitinase and Cry1Ab proteins from endogenous proteases in the larval midgut and prolongs their insecticidal activities. Dinoflagellates of the genus Ostreopsis are known to cause (often fatal) food poisoning in tropical coastal areas following the accumulation of palytoxin (PLTX) and/or its analogues (PLTX group) in crabs, sea urchins or fish. Ostreopsis spp. occurrence is presently increasing in the northern to north western Mediterranean Sea (Italy, Spain, Greece and France), probably in response to climate change. In France, Ostreopsis. cf. ovata has been associated with toxic events during summer 2006, at Morgiret, off the coast of Marseille, and a specific monitoring has been designed and implemented since 2007. Results from 2008 and 2009 showed that there is a real danger of human poisoning, as these demonstrated bioaccumulation of the PLTX group (PLTX and ovatoxin-a) in both filter-feeding bivalve molluscs (mussels) and herbivorous echinoderms (sea urchins). The total content accumulated in urchins reached 450 μg PLTX eq/kg total flesh (summer 2008). In mussels, the maximum was 230 μg eq PLTX/kg (summer 2009) compared with a maximum of 360 μg found in sea urchins during the same period at the same site. In boreal ecosystems, an increase in soil temperature can stimulate plant growth. However, cambium phenology in trees was better explained by air than soil temperature, which suggested that soil temperature is not the main limiting factor affecting xylogenesis. Since soil temperature and snowmelt are correlated to air temperature, the question whether soil temperature directly limits xylogenesis in the stem will remain unresolved without experiments disentangling air and soil temperatures. This study investigated the effects of an increase of 4 °C in soil temperature and a consequent 1-week earlier snowmelt on growth of black spruce [Picea mariana (Mill.) BSP] in the boreal forest of Quebec, Canada. The soil of two natural stands at different altitudes was warmed up with heating cables during 2008-2010 and cambial phenology and xylem production were monitored weekly from April to October. The results showed no significant effect of the treatment on the phenological phases of cell enlargement and wall thickening and lignification. The number of cells produced in the xylem also did not differ between control and heated trees. These findings allowed the hypothesis of a direct influence of soil temperature on stem growth to be rejected and supported the evidence that, in the short term, air temperature is the main limiting factor for xylogenesis in trees of these environments. 相似文献

Although the heterogeneity of wood could preclude assessment of the performances of adhesion, the within-tree variations in its thermodynamic properties still remain unexplored. This study analyzed the surface free energy of wood (γ W ) calculated by the geometric mean approach along the radius and at different sampling heights in black spruce and balsam fir with a statistical model, employing apparent contact angles of three testing liquids. Sampling height and species affected the apparent contact angles, and consequently the polar ( $ \gamma_{W}^{P} $ ) and dispersed ( $ \gamma_{W}^{D} $ ) components of γ W , but no significant difference was observed along the radius. On average, γ W was 43.77?mJ?m?2 at the stem base and gradually decreased, attaining 27.19?mJ?m?2 at 14?m of height. $ \gamma_{W}^{P} $ was markedly higher than $ \gamma_{W}^{D} $ with an opposite pattern along the tree height. These findings demonstrated the huge variability in γ W along the stem, which could be related to the structural or chemical features of wood. 相似文献
