This study aimed to evaluate the effect of five salt solutions in the maintenance of morphological features of cortical alveolus, hydration and fertilization capacity of Prochilodus lineatus oocytes. For this purpose, five saline solutions were tested: Ringer's solution, Ringer's lactate solution, Hank's balanced salt solution (HBSS), Hank's balanced salt solution without calcium (HBSS without calcium) and solution for salmonid eggs. Oocytes were maintained for 2 hr in saline solution with controlled temperature subsequently evaluated for hydration, cortical activation and fertilization ability. In the evaluation of the fertilization ability, two controls were used: C1—fertilized oocytes after extrusion—and C2—oocytes kept in ovarian fluid and fertilized after 2 hr. There was a significant reduction in the viability of oocytes C2 (28.8% ± 12.9%) compared to C1 (65.3% ± 26.7%), and no significant differences were found between treatments HBSS and HBSS without calcium and C2. Only HBSS and HBSS without calcium maintained the non‐activated state of the gametes, with a fertilization rate of 16.4% ± 6.7% and 5.6% ± 2.3%, respectively; however, they did not extend the viability of oocytes, such that they continued to undergo degradation during the storage period, similar to oocytes retained only in ovarian fluid.  相似文献

We examined 65 lodgepole pine (Pinus contorta Dougl. ex Loud. var. latifolia Engelm.) sites in Alberta using a dendrochronological approach in order to examine the relationships between climate and growth of lodgepole pine across elevational ecoregions and diameter size classes. The 4 elevational ecoregions sampled included the Boreal Highlands (BH: 13 sites); the Foothills (FH: 36 sites); a grouping of the montane and subalpine zones of the southern Rocky Mountains (RM: 12 sites); and the montane zone of the Cypress Hills (CH: 4 sites). The first diameter size class was comprised of the three largest (top) diameter trees at each site. The tree list of each plot was ranked, then divided into three groups of equal basal area and the tree at the midpoint of each group (small, medium, and large) was selected for the other three size classes. Annual growth in basal area of lodgepole pine was generally sensitive to heat and moisture stress in late summer of the previous year, the degree of winter harshness, and the timing of the start of the growing season. Growth was inhibited by low temperature in all winter months at the most northern BH sites which had the coldest winters, but this effect was interrupted in some of the midwinter months in the more southerly sites in the RM, and we postulated this was due to the damaging influences of Chinook winds. Interannual growth patterns were strongly correlated between top diameter trees and the other classes, and trees of all diameter classes generally responded to climate in the same way, which indicated that it is sufficient to sample only the largest diameter trees in a stand to provide insight into growth–climate relationships. The forecasted growth estimates indicate that future climate warming will negatively impact the productivity of lodgepole pine in the FH, the heart of lodgepole distribution and productivity in Alberta.  相似文献

Asobara japonica (Hymenoptera: Braconidae), Ganaspis brasiliensis and Leptopilina japonica (Hymenoptera: Figitidae) are Asian larval parasitoids of spotted wing drosophila, Drosophila suzukii (Diptera: Drosophilidae). This study evaluated these parasitoids’ capacity to attack and develop from 24 non-target drosophilid species. Results showed that all three parasitoids were able to parasitize host larvae of multiple non-target species in artificial diet; A. japonica developed from 19 tested host species, regardless of the phylogenetic position of the host species, L. japonica developed from 11 tested species; and G. brasiliensis developed from only four of the exposed species. Success rate of parasitism (i.e., the probability that an adult wasp successfully emerged from a parasitized host) by the two figitid parasitoids was low in hosts other than the three species in the melanogaster group (D. melanogaster, D. simulans, and D. suzukii). The failure of the figitids to develop in most of the tested host species appears to correspond with more frequent encapsulation of the parasitoids by the hosts. The results indicate that G. brasiliensis is the most host specific to D. suzukii, L. japonica attacks mainly species in the melanogaster group and A. japonica is a generalist, at least physiologically. Overall, the developmental time of the parasitoids increased with the host’s developmental time. The body size of female A. japonica (as a model species) was positively related to host size, and mature egg load of female wasps increased with female body size. We discuss the use of these parasitoids for classical biological control of D. suzukii.

Food products that are high in fiber and low in glycemic impact are healthier. Amylose is a form of resistant starch that mimics dietary fiber when consumed. A durum wheat (Triticum durum) line was created that lacks starch synthase IIa (SSIIa) activity, a key enzyme in amylopectin biosynthesis, by identifying a null mutation in ssIIa‐B following mutagenesis of a line that has a naturally occurring ssIIa‐A null mutation. Our objective here was to compare seed, milling, pasta, and nutritional characteristics of the SSIIa null line with a wild‐type control line. The SSIIa null line had increased amylose and grain protein with lower individual seed weight and semolina yield. Refined pasta prepared from the SSIIa null semolina absorbed less water, had increased cooking loss, had a shorter cook time, and was considerably firmer even after overcooking compared with the wild‐type line. Color of the SSIIa null cooked and uncooked pasta was diminished in brightness compared with the wild type. Nutritionally, the SSIIa null pasta had increased calories, fiber, fat, resistant starch, ash, and protein compared with the control line, along with reduced total and available carbohydrates. Pasta made from high‐amylose durum wheat provides a significant nutritional benefit along with enhanced end‐product quality via firmer pasta that resists overcooking.  相似文献

The negative effects of oxygen on white wine quality and the various factors which influence it (including temperature, dissolved oxygen, pH, and free SO(2)) are well documented both at the sensory and compositional levels. What is less defined is the quantitative relationship between these parameters and the kinetics of the development of the negative effects of oxidation. The experiment presented here attempts to generate data which can be used to predictively model the oxidative degradation of white wines. Bottled wines were submitted to extreme conditions (45 degrees C temperature, O(2) saturation) during 3 months witth samples taken every 15 days for both sensorial and chemical analysis (GC-O/FPD/MS, 420 nm). The synergistic effects of increasing temperature and O(2) at lower pH are evident, both on the decrease in levels of terpene alcohols and norisoprenoids (which impart floral aromas), and on the development of off-flavors such as "honey-like", "boiled-potato", and "farm-feed" associated with the presence of phenylacetaldehyde, methional, and 1,1,6-trimethyl-1,2-dihydronaphthalene.  相似文献

Understanding the relationship between soil biodiversity and ecosystem functioning is critical to predicting and monitoring the effects of ecosystem changes on important soil processes. However, most of Earth's soils are too biologically diverse to identify each species present and determine their functional role in food webs. The soil ecosystems of Victoria Land (VL) Antarctica are functionally and biotically simple, and serve as in situ models for determining the relationship between biodiversity and ecosystem processes. For a few VL taxa (microarthropods, nematodes, algae, mosses and lichens), species diversity has been intensively assessed in highly localized habitats, but little is known of how community assemblages vary across broader spatial scales, or across latitudinal and environmental gradients. The composition of tardigrade, rotifer, protist, fungal and prokaryote communities is emerging. The latter groups are the least studied, but potentially the most diverse. Endemism is highest for microarthropods and nematodes, less so for tardigrades and rotifers, and apparently low for mosses, lichens, protists, fungi and prokaryotes. Much of what is known about VL diversity and distribution occurs in an evolutionary and ecological vacuum; links between taxa and functional role in ecosystems are poorly known and future studies must utilize phylogenetic information to infer patterns of community assembly, speciation, extinction, population processes and biogeography. However, a comprehensive compilation of all the species that participate in soil ecosystem processes, and their distribution across regional and landscape scales is immediately achievable in VL with the resources, tools, and expertise currently available. We suggest that the soil ecosystems of VL should play a major role in exploring the relationship between biodiversity and ecosystem functioning, and in monitoring the effects of environmental change on soil processes in real time and space.  相似文献

beta-Carotene and six xanthophylls (lutein, neoxanthin, violaxanthin, luteoxanthin, cryptoxanthin, and echinenone) have been identified and semiquantitatively or quantitatively determined in musts and port wines for the first time. An HPLC method was developed and compared with that of one based on thin layer cromatography with scanning densitometry. The most abundant carotenoids present in red grape varieties are beta-carotene and lutein. In wines, significant quantities of violaxanthin, luteoxanthin, and neoxanthin were found. This study was done with berries (skin and pulp), musts, and fortified wines. Some experiments were performed to follow carotenoid content from grapes to wines. Although the levels of beta-carotene and lutein found in fortified wines were lower than those found in musts, other xanthophylls, such as neoxanthin, violaxanthin, and luteoxanthin, exist in appreciable amounts in young ports.  相似文献
