Abstract

There is a large number of hill people in northern Thailand, who practices shifting cultivation. In order to analyze the soil ecological problems involved in the transition from traditional shifting cultivation to more intensive upland farming, the authors carried out comparative studies on the dynamics of organic matter and its related properties in soils both in the traditional shifting cultivation systems adopted by Karen people and more intensive upland farming practiced by Thai and Hmong people in the area. The contents of organic matter and available N in the surface 10 cm layers of soil from the fields continuously cultivated were lower than those in soils under prolonged fallow (more than 10 y) or natural forest. Based on the rate of soil respiration, the amount of organic matter decomposed within 1 y was estimated to reach nearly 10% of that stored in the upper 50 cm layers of the soil profile in the upland crop fields. These results indicate that the organic matter-related resources markedly decreased under continuous cropping. The contents of C, N, and P in the microbial biomass of the surface 10 cm layers of soil ranged from 0.37 to 2.09 mg C g?l soil, from 22.7 to 188 µg N g?l soil, and from 6.1 to 65.7 µg P g?l soil, respectively. Since the contents of microbial C, N, and P in the surface soils were generally higher under prolonged fallow and natural forests than in the fields continuously cultivated, the microbial activity and/or the amounts of C, N, and P available for biological activity seemed to have declined under continuous upland farming. The incubation experiment to assess the N mineralization pattern showed two remarkable characteristics: 1) there was an initial time lag until active mineralization of N occurred in the soils from young fallow forest and 2) the soil burning effect was observed after burning in the fields under prolonged fallow. The active process of nitrification after N mineralization was always associated with a sharp fall in soil pH, suggesting that soil acidification was promoted and basic cations were lost from the soils. In conclusion, rapid deterioration of the soil organic matter-related properties in cropping fields can be considered to be one of the ecological reasons why upland fields must be returned to fallow again a few years after forest reclamation in traditional shifting cultivation systems. Therefore, in alternative farming systems with more intensive land use, it is essential to apply organic materials into soils to decrease the rate of soil degradation, or to improve the soil fertility, in avoiding soil acidification along with nitrification.  相似文献

The water penetration into a rice grain during soaking was monitored by three-dimensional gradient echo magnetic resonance imaging, which has the advantage of a short measurement time and a sufficient sensitivity for low-moisture samples. The water penetration route was compared between the milled and brown rice grains of two japonica cultivars. In normal milled rice grains of Koshihikari, water first penetrated the ventral side surface and the embryo attachment site of the endosperm, then migrated along the central line and transverse cracks, and finally diffused to all parts of the endosperm. In milled rice grains of Yamadanishiki with a white core, water quickly infiltrated into the cracks or chalky parts on the dorsiventral line and then diffused to the lateral side. In brown rice grains of both cultivars water penetrated extremely slowly and did not infiltrate directly into transverse cracks or the white core due to inhibition by the pericarp and seed coat. The central part of every grain allowed more water to penetrate. The route, pattern and speed of water penetration were determined by the morphological structure, crack formation and hardness distribution associated with the packing of the starch granules in the grain.  相似文献

The inner cellular structure of microconidia of Magnaporthe oryzae was examined using fluorescent probes and electron microscopic techniques. The volume of the nucleus relative to the cell was significantly larger in microconidia than in macroconidia or vegetative hyphae, similar to observations for spermatia of other fungi. Selective fluorescent staining revealed that cytosolic RNA was less abundant in microconidia than in macroconidia and germ tubes, suggesting that general metabolic activity in microconidia is low. Consistently, GFP expression driven by the TrpC promoter was highly active during the formation of phialides and microconidia but gradually decreased as the microconidia matured. Such data suggest that microconidia are in a quiescent or dormant state.  相似文献

ABSTRACT Suppressive effects of soil amendment with residues of 12 cultivars of Brassica rapa on damping-off of sugar beet were evaluated in soils infested with Rhizoctonia solani. Residues of clover and peanut were tested as noncruciferous controls. The incidence of damping-off was significantly and consistently suppressed in the soils amended with residues of clover, peanut, and B. rapa subsp. rapifera 'Saori', but only the volatile substance produced from water-imbibed residue of cv. Saori exhibited a distinct inhibitory effect on mycelial growth of R. solani. Nonetheless, disease suppression in such residue-amended soils was diminished or nullified when antibacterial antibiotics were applied to the soils, suggesting that proliferation of antagonistic bacteria resident to the soils were responsible for disease suppression. When the seed (pericarps) colonized by R. solani in the infested soil without residues were replanted into the soils amended with such residues, damping-off was suppressed in all cases. In contrast, when seed that had been colonized by microorganisms in the soils containing the residues were replanted into the infested soil, damping-off was not suppressed. The evidence indicates that the laimosphere, but not the spermosphere, is the site for the antagonistic microbial interaction, which is the chief principle of soil suppressiveness against Rhizoctonia damping-off.  相似文献

The development of appressoria by germinating Blumeria graminis conidia depends on its germ tubes making contact with the host surface. Low angle, low temperature scanning electron microscopy showed that 80% or more of first-formed germ tubes of f. spp. hordei, tritici and avenae conidia emerged from close to the host leaf surface, and so made contact with it allowing them to become functional primary germ tubes. Light microscopy of f. sp.hordei confirmed this result and, in addition, showed that germ tubes frequently emerged close to, and contacted, various hydrophobic and hydrophilic artificial substrata. Geometric models of conidium-substratum interfaces were developed and a “surface point method” was derived to predict the frequency with which contact would result if germ tube emergence was a random phenomenon. However, observed contact frequencies were far higher (c. three to eight times) than predicted. Thus, the germ tube emergence site was determined as a response to substratum contact. In part, this appeared to be a non-specific response. Nevertheless, germ tube contact frequencies were greater on the curved surface of leaf epidermal cells than on planar surface, suggesting that specific recognition of leaf surface characteristics may promote directional emergence. The area of contact required to stimulate directional germ tube emergence was very small: contact with a microneedle tip or with a spiders’ suspension thread was sufficient for many conidia. Similarly, on leaves, the only contact is between the tips of a limited number of conidial wall projections and the edges of epicuticular leaf wax plates. Micromanipulation to roll conidia so that their original site of contact with a leaf was rotated away from it, led to the majority of first-formed germ tubes growing away from the leaf, i.e. emerging close to the site of original contact. The experiments indicated that the site of germ tube emergence is determined within 1 min of deposition. This implicated the release of conidial extracellular materials in recognition of the conidium-leaf surface contact site.  相似文献

Pseudomonas fluorescens FPT9601, a plant growth-promoting rhizobacterium (PGPR) isolated from tomato rhizosphere, can protect tomato (Lycopersicon esculentum Mill) from bacterial wilt disease caused by Ralstonia solanacearum. This strain produces antibiotics 2,4-diacetylphloroglucinol (2,4-DAPG) and hydrogen cyanide (HCN). It also produces proteases and uncharacterized siderophores (Sid). A mutant strain SM2214, obtained by Tn5 insertion, did not produce 2,4-DAPG, HCN or proteases, but overproduced Sid. Marker-exchange mutagenesis confirmed that a single transposon insertion caused the multiple phenotypic changes of this mutant. Complementation of the mutant with a 1.3-kb DNA fragment that was amplified from genomic DNA of the wild-type P. fluorescens strain by PCR could restore the lost functions of the mutant strain. Nucleotide sequencing revealed that the fragment contained a 642-bp open reading frame (ORF) highly homologous to the regulator responser gene gacA. The in vitro anti-bacterium test and plant protection experiment under greenhouse conditions indicated that the gacA gene played an important role in the suppression of tomato bacterial wilt disease. An endophytic actinomycete, Streptomyces sp. R-5, which had been isolated from a field-grown rhododendron plant, was used to protect rhododendron seedlings in tissue culture from Pestalotia disease caused by Pestalotiopsis sydowiana. R-5 had intense antagonistic activity against P. sydowiana without adversely affecting the seedlings in glass flasks. A suspension of R-5 was spread on the surface of the multiplication medium in glass flasks in which seedlings were growing. Ten days later, the 4th upper leaf of seedlings was inoculated with P. sydowiana and incubated for 14 days. In controls untreated with R-5, substrate mycelia of P. sydowiana grew on all leaves and stems above and below the 4th leaf within 2–3 days of inoculation. Such growth resulted in the wilting death of 54% of seedlings by 14 days. In contrast, only the inoculated leaves turned brown in ca. 90% of seedlings growing on medium treated with R-5. None of these seedlings died. Thus, treatment of the medium surface with R-5 efficiently protects the seedlings from infection by P. sydowiana. Scanning electron microscopy revealed that substrate mycelia of R-5 grew on and beneath the cuticle of leaves of the treated seedlings. The effects of chlordimeform on rectus abdominis muscle of frog were investigated. Chlordimeform (10?3M) caused a slow contraction, and at lower concentration (10?5–10?3M) it inhibited the acetylcholine-induced contraction in noncompetitive manner. When chlordimeform was applied to the muscle of Rana catesbiana, K+-induced contraction was also inhibited in noncompetitive manner. Whereas it had no effect on caffeine-induced contraction.Chlordimeform-induced contraction was not affected by respective addition of d-tubocurarine (10?4M), procaine (10?3M), or eserine (0.3 mM), which results were same as that of K+-induced contraction. Chlordimeform, at lower concentration (10?5–10?3M), inhibits the acetylcholine- and K+-induced contractions probably owing to depression of not only the sensitivity of endplate but also the excitability of cell membrane.  相似文献

This study was conducted to examine the utility of vitrification for bovine embryos with low‐quality grade, and simple cryoprotectants dilution method for practitioners. In Experiment 1, survival of frozen embryos was compared with that of vitrified embryos using minimum volume cooling (MVC). Then, vitrified embryos were used to confirm the optimum sucrose concentration in Experiment 2. The survival rates of embryos that had been vitrified following diluted cryoprotectants with the one‐step in‐straw method were compared with those of fresh control embryos in Experiment 3. Frozen‐thawed or vitrified‐warmed blastocysts were cultured with TCM‐199 supplemented with 100 μmol/L beta‐mercaptoethanol +5% fetal bovine serum at 38.5°C in an atmosphere of 5% CO2 in air, their survival after 24 hr were compared. The development to term of fair quality in vivo embryos after vitrification was examined in Experiment 4. Results show that survival rates of frozen‐thawed embryos were lower (< .05) than that of vitrified‐warmed ones. When vitrified embryos were warmed in 0.3 mol/L sucrose in straws, their survival rate was 100%. The total cell numbers of vitrified‐warmed embryos were comparable to those of fresh control embryos. The six calves from 13 vitrified embryos were delivered in Experiment 4. These results indicate that MVC vitrification following one‐step cryoprotectants dilution is utilized to preserve low‐quality bovine embryos.  相似文献
