在已掌握甘蓝型油菜―萝卜附加系染色体构成,并构建萝卜分子连锁图谱的基础上,以甘蓝型油菜―萝卜附加系为材料,利用(dp)RAPD分子标记技术,筛选附加系中外源萝卜9条染色体(A~I)的分子标记,为进一步研究萝卜染色体与连锁群对应关系奠定基础.试验筛选了576个随机引物(组合),有413个能从附加系中扩增出萝卜染色体的(dp)RAPD标记,平均有效引物(组合)占71.7%,其中RAPD反应有效引物率67.7%,dpRAPD反应有效引物率73.2%.试验共得到899个萝卜染色体(dp)RAPD标记,包括354个RAPD标记和545个dpRAPD标记,覆盖萝卜整个基因组的全部9条染色体,不同染色体的标记数目变化范围在41~160,其中最多的是染色体C,最少的是染色体I.  相似文献

Insects in childish play and education There is reported on nursery-rhymes on lady-bird and cockchafer, on using beetles or butterflies flattered by aband as play works, holding crickets, katylids, anf cicades in cages, hunting katylids and dragonflies, laters by means of spiderwebs in aring like bound willow twig, and other plays of children in whole the world. The childhood memoirs of some German poets on insects are stimulating to fight against noxious insects, in collecting insects and researching their life, and in hindering from shaking on death insects without reason. Such education is very need for understanding pest control and environment protection actions. Therefore it is an important task for applied entomology.  相似文献

The INHAND Proposal (International Harmonization of Nomenclature and Diagnostic Criteria for Lesions in Rats and Mice) has been operational since 2005. A Global Editorial Steering Committee (GESC) manages the overall objectives of the project and the development of harmonized terminology for each organ system is the responsibility of the Organ Working Groups (OWG), drawing upon experts from North America, Europe and Japan.Great progress has been made with 9 systems published to date – Respiratory, Hepatobiliary, Urinary, Central/Peripheral Nervous Systems, Male Reproductive and Mammary, Zymbals, Clitoral and Preputial Glands in Toxicologic Pathology and the Integument and Soft Tissue and Female Reproductive System in the Journal of Toxicologic Pathology as supplements and on a web site – www.goreni.org. INHAND nomenclature guides offer diagnostic criteria and guidelines for recording lesions observed in rodent toxicity and carcinogenicity studies. The guides provide representative photo-micrographs of morphologic changes, information regarding pathogenesis, and key references. During 2012, INHAND GESC representatives attended meetings with representatives of the FDA Center for Drug Evaluation and Research (CDER), Clinical Data Interchange Standards Consortium (CDISC), and the National Cancer Institute (NCI) Enterprise Vocabulary Services (EVS) to begin incorporation of INHAND terminology as preferred terminology for SEND (Standard for Exchange of Nonclinical Data) submissions to the FDA. The interest in utilizing the INHAND nomenclature, based on input from industry and government toxicologists as well as information technology specialists, suggests that there will be wide acceptance of this nomenclature. The purpose of this publication is to provide an update on the progress of INHAND.  相似文献

Barley has for feeding purposes a shortage of essential amino acids, especially lysine, threonine, and methionine, and an excess of proline and glutamine. In the present study, we have introduced into barley an antisense construct against C-hordeins, the storage protein with the lowest nutritional quality. SDS-PAGE and reverse phase HPLC revealed a relative reduction in the amounts of C-hordeins and relative increases in the content of the other storage proteins. The five different lines analyzed had lower amounts of proline, glutamic acid/glutamine, and phenylalanine (up to 12%, 6%, and 9% reductions), while the lysine, threonine, and methionine content was increased with up to 16%, 13% and 11%. It is concluded that antisense mediated suppression of C-hordein synthesis may be a promising approach for improving the nutritional value of barley as a feed crop while at the same time reducing the environmental nitrogen load.  相似文献

Sulfur (S) fertilization has been long-known to influence the amounts of total free amino acids in plants. To determine the impact of S deficiency in wheat on the concentration of, in particular, free asparagine, the spring wheat cultivar 'Star' was grown in a laboratory scale (5 L pot) at five different levels of S fertilization. After maturity, the kernels were milled into white flours (1-5) and analyzed for their contents of total S and total nitrogen as well as for free amino acids and glucose, fructose, maltose, and sucrose. Extremely high concentrations of free asparagine (Asn; 3.9-5.7 g/kg) were determined in flours 1 and 2 (30 and 60 mg of S), whereas much lower amounts (0.03-0.4 g/kg) were present in flours grown at higher S levels. The amounts of the reducing carbohydrates were, however, scarcely affected by S fertilization. In agreement with the high amount of Asn in flours 1 and 2, heating of both flours led to the generation of very high amounts of acrylamide (1.7-3.1 mg/kg) as well as of 3-aminopropionamide (40-76 mg/kg). Similar concentrations were measured in crispbread prepared from both flours. Application of rheological measurements on doughs prepared from each flour and a determination of the loaf volume of bread baked therefrom clearly indicated that flours 1 and 2 would be excluded from commercial bread processing due to their poor technological properties. Two commercial flours showed relatively low concentrations of acrylamide after a thermal treatment.  相似文献

The South American tomato pinworm, Tuta absoluta (Meyrick, 1917) (Lepidoptera: Gelechiidae), is heavily targeted by insecticide applications. Methoxyfenozide is highly effective against T. absoluta with reduced side effects to natural enemies. This work aimed to (1) genetically and biochemically characterize resistance of the T. absoluta GBN population to methoxyfenozide, (2) establish cross resistance profiles with other insecticide groups and (3) monitor resistance in populations with the goal of improving T. absoluta insecticide resistance management (IRM). Methoxyfenozide resistance was completely recessive, polyfactorial and autosomal. Effective dominance revealed that 10 mg methoxyfenozide/L would be enough to eliminate susceptible homozygotes and heterozygotes, thus used to diagnose resistance in field populations. The synergism of methoxyfenozide toxicity in the resistant population for PBO (SR?=?95×), DEF (SR?=?51×) and DEM (SR?=?45×), suggested monooxygenases, esterases and glutathione S-transferases as resistance mechanisms. However, only monooxygenase activity appeared to be involved in methoxyfenozide resistance. Resistance ratio for methoxyfenozide (2352-fold) after selection and cross-resistance ratios of a lab-selected GBN strain (“GBN-Sel”) were significant relative to a susceptible strain “JDR1-Sus” for tebufenozide (656-fold), cartap hydrochloride (10.68-fold), deltamethrin (4.70-fold), abamectin (2.65-fold), lufenuron (2.22-fold) and indoxacarb (1.92-fold), with negative cross-resistance to spinetoram (0.32-fold). Evidence of control failures was observed in 10 field populations of T. absoluta (mortalities between 13 and 76%), and all populations showed frequencies of resistant phenotypes (percentage survivorship ranging between 4 and 96%). A rational basis for managing resistance to bisacylhydrazines is discussed, along with details of recommended T. absoluta resistance management tactics.

An 11‐year‐old female spayed Labrador Retriever was presented for a draining, painful subcutaneous mass palpated over a previously implanted pacemaker generator. Infection was suspected and the mass was removed surgically. On cut surface, the mass was friable and mottled tan to brown with firm pale tan nodules, surrounding the pacemaker lead wire adjacent to the pacemaker generator. Cytologic interpretation of impression smears was consistent with a sarcoma, and suggestive of a rhabdomyosarcoma due to the presence of strap‐like cells. On histopathologic examination, a highly invasive nodular mass surrounded the pacemaker lead, composed of pleomorphic round, spindle and strap cells, and multinucleated giant cells. The population exhibited microscopic invasion into the deep portion of the fibrous capsule surrounding the pacemaker generator. There were tumor emboli within small to medium subcutaneous veins adjacent to the mass. Immunohistochemically, the neoplastic cells stained positive for α‐sarcomeric actin and vimentin, and negative for α‐smooth muscle actin, consistent with a rhabdomyosarcoma arising at the site of the pacemaker generator. To our knowledge, this is the first report of a rhabdomyosarcoma associated with the lead wire of a pacemaker generator in a dog.  相似文献

Sleepy cod Oxyeleotris lineolatus is a species of freshwater goby in demand in Australian markets by consumers of Asian origin. It is related to marble goby Oxyeleotris marmoratus , the most expensive freshwater food fish in Asia, which is cultured throughout southeast Asia in ponds and cages. The performance of sleepy cod in culture conditions was investigated to assess the viability of farming them in northern Australia. Sleepy cod fingerlings (62.8 ± 0.8 mm total length and 2.56 ± 0.095 g) were stocked into experimental ponds at 32,857 fish/ha, and grown out for 8 mo. Shelter was provided in each of three replicate ponds and was absent in three control ponds. The provision of shelter in juvenile growout was found to be of no benefit, although fish in ponds provided with shelter weighed slightly more per unit length than fish in ponds without shelter. Cannibalism was not a problem in growout, and survival was close to 100%. After the shelter trial was completed, fish were graded into large and small classes (three replicates of each), and grown out without shelter at the same density for 158 d. Following that, fish were again graded, and the largest 30% retained from growout at a density of 8,857 fish/ha (large, 198 ± 6.44 g) or 10,000 fish/ha (small, 48.9 ± 1.27 g). These were grown out for 188 d. Growth of selected stock at low densities was slower than earlier growth rates, although smaller fish gained weight more rapidly than larger fish. Growth rates were better than the only published data for marble goby. Further investigation into high density culture and different genotypes of sleepy cod needs to be undertaken to determine the viability of pond culture.  相似文献

Triploidy in American oysters (Crassostrea virginica) was induced by blocking polar body formation with cytochalasin B. Triploid oysters created by treatment during meiosis I grew faster during the first 3 years of life than did diploid siblings. Triploid oysters created during meiosis II grew at the same rate as their controls. Because heterozygosity was higher in oysters created during meiosis I than in other groups, the increased growth must be due to heterozygosity rather than to triploidy per se.  相似文献
