Sorption–desorption of the fungicide triadimefon in field‐moist silt loam and sandy loam soils were determined using low‐density supercritical fluid extraction (SFE). The selectivity of SFE enables extraction of triadimefon from the soil water phase only, thus allowing calculation of sorption coefficients (Kd) at field‐moist or unsaturated conditions. Triadimefon sorption was influenced by factors such as soil moisture content and temperature; sorption increased with increased moisture content up to saturation, and decreased with increased temperature. For instance, Kd values for triadimefon on the silt loam and the sandy loam soils at 40 °C and 10% water content were 1.9 and 2.5 ml g−1, respectively, and at 18% water content, 3.3 and 6.4 ml g−1, respectively. Isosteric heats of sorption (ΔHi) were −42 and −7 kJ mol−1 for the silt loam and sandy loam soils, respectively. Sorption–desorption was also determined using an automated accelerated solvent extraction system (ASE), in which triadimefon was extracted from silt loam soil by 0.01 M CaCl2. Using the ASE system, which is basically a fast alternative to the batch equilibration system, gave a similar ΔHi value (−29 kJ mol−1) for the silt loam soil (Kf = 27 µg1 − 1/n ml1/n g−1). In order to predict transport of pesticides through the soil profile more accurately on the basis of these data, information is needed on sorption as a function of soil water content. © 2000 Society of Chemical Industry 相似文献

The aim of this study was to explore whether anti-Ostertagia ostertagi antibody levels measured by ELISA in individual milk samples hold promise as a decision parameter for either herd-level decisions or selective anthelmintic treatments by investigating (1) the relationship between individual and bulk-tank milk ELISA results; (2) the relationships of individual milk ELISA results with non-parasitic cow factors and (3) the relationship between individual milk ELISA results and the milk production response after anthelmintic treatment. Twelve farms were randomly allocated to a whole-herd treatment with eprinomectin or a placebo in October 2004 and individual milk samples and a bulk-tank milk sample were collected 1 month before and 1 month after treatment. Linear mixed models were used to investigate the associations of ELISA results with (a) breed, actual milk production, lactation stage, somatic cell count, age and sampling month and (b) the milk production response after anthelmintic treatment. There was a reasonable correlation between the mean individual and bulk-tank milk ELISA results (r = 0.72). Individual cow ELISA results increased with higher lactation number and were lower in November than September. The associations with the other non-parasitic factors were weak and not significant. Milk yield responses to anthelmintic treatment were greater when treatment was given in early lactation and increased with the pre-treatment ELISA result and cow age. However, these latter two interaction terms were not significant when they were put in the model together. We conclude that (1) O. ostertagi ELISA results from individual milk samples may provide more information on the herd's parasitic status than a single bulk-tank milk result; (2) lactation number should be taken into account when interpreting ELISA results from individual milk samples and (3) the value of the O. ostertagi antibody level in individual cow milk samples to predict individual production responses after anthelmintic treatment remains equivocal. 相似文献

Botrytis cinerea causes gray mold disease and affects hundreds of plant species, including tomato (Lycopersicon esculentum). The wild nightshade, Solanum lycopersicoides, is cross compatible with tomato and is more resistant to B. cinerea, thus representing a potential source for crop improvement. Tests involving droplet inoculation of detached leaves and spray inoculation of entire seedlings demonstrated that resistance to B. cinerea varies among S. lycopersicoides accessions, with S. lycopersicoides LA2951 being the most resistant accession tested. Expression of resistance in the intergeneric hybrid (L. esculentum cv. 'VF36' × S. lycopersicoides LA2951) suggested that resistance is at least partially dominant in tomato. A green fluorescent protein-tagged B. cinerea strain was used for confocal microscopic comparison of infection in leaves of S. lycopersicoides and tomato. Even though S. lycopersicoides supported spore germination, there was evidence for hyphal lysis and death 3 days after inoculation, at a time when lesions were expanding on susceptible tomato plants. The reduced frequency of B. cinerea lesion spread on S. lycopersicoides explains why this fungus produced fewer spores in this wild nightshade than in tomato. 相似文献

Soil fungi and oomycetes (syn. peronosporomycetes) are the most common causes of pea diseases, and these pathogens often occur in complexes involving several species. Information on the dynamics within this complex of pathogens, and also between the complex of pathogens and other fungi in the development of root disease is limited. In this study, next-generation sequencing of nuclear ribosomal internal transcribed spacer-1 was used to characterize fungal communities in agricultural soils from nine pea fields, in which pea roots showed different degrees of disease. Fungal species richness, diversity, and community composition were analyzed and compared among the different pea soils. After filtering for quality and excluding non-fungal sequences, 55,460 sequences clustering into 434 operational taxonomic units (OTUs), were obtained from the nine soil samples. These sequences were found to correspond to 145–200 OTUs in each soil. The fungal communities in the nine soils were strongly dominated by Ascomycota and Basidiomycota. Phoma, Podospora, Pseudaleuria, and Veronaea, at genus level, correlated to the disease severity index of pea roots; Phoma was most abundant in soils with diseased plants, whereas Podospora, Pseudaleuria, and Veronaea were most abundant in healthy soils. No correlation was found between the disease severity index and the abundance of some of the other fungi and oomycetes normally considered as root pathogens in pea. 相似文献

The landscape matrix is suggested to influence the effect of habitat fragmentation on species richness, but the generality
of this prediction has not been tested. Here, we used data from 10 independent studies on butterfly species richness, where
the matrix surrounding grassland patches was dominated by either forest or arable land to test if matrix land use influenced
the response of species richness to patch area and connectivity. To account for the possibility that some of the observed
species use the matrix as their main or complementary habitat, we analysed the effects on total species richness and on the
richness of grassland specialist and non-specialist (generalists and specialists on other habitat types) butterflies separately.
Specialists and non-specialists were defined separately for each dataset. Total species richness and the richness of grassland
specialist butterflies were positively related to patch area and forest cover in the matrix, and negatively to patch isolation.
The strength of the species-area relationship was modified by matrix land use and had a slope that decreased with increasing
forest cover in the matrix. Potential mechanisms for the weaker effect of grassland fragmentation in forest-dominated landscapes
are (1) that the forest matrix is more heterogeneous and contains more resources, (2) that small grassland patches in a matrix
dominated by arable land suffer more from negative edge effects or (3) that the arable matrix constitutes a stronger barrier
to dispersal between populations. Regardless of the mechanisms, our results show that there are general effects of matrix
land use across landscapes and regions, and that landscape management that increases matrix quality can be a complement to
habitat restoration and re-creation in fragmented landscapes. 相似文献

Today, office work in western societies is more sedentary and more mentally demanding than ever before. This way of working plays a role in the increase in lifestyle diseases related to sedentary routines and stress. Green outdoor environments have been found to motivate physical activity and reduce stress levels. In this study, we therefore considered these environments to have a positive affect on work force health and wellbeing. However, research on the workplace outdoor environment is still limited. To investigate how and why the workplace outdoor environment is used by office workers and the impact of these environments on office workers’ health and wellbeing, five research questions were formulated. The questions addressed the use of the outdoor environments; the outdoor activities; encouragement and impediments for going outdoors; characteristics of the outdoor environment, and the impact of workplace outdoor environments on employees’ health, job satisfaction and work performance. In order to answer these research questions, we conducted a web-based questionnaire, completed by (N) 402 office workers from six Danish companies. The results show that 37.8% of the respondents spent time outdoors during their working day, and that the proportion of men spending time outdoors was higher than the proportion of women. Eight outdoor activities were identified, and significant relationships between the outdoor activities and perceived sensory dimensions (PSD) of the outdoor environment were found, indicating that the presence of the dimension ‘serene’ increased the odds for respondents going outdoors and that five of the eight activities would be performed. Only few significant relationships between outdoor environment use and employee health, job satisfaction and work performance were identified. 相似文献

Skin lesions on pigs inflicted by humans compromise animal welfare and are the subject of increased public and political attention in Denmark. Systematic surveillance of such skin lesions was enforced in April 2010 at all Danish pig abattoirs, through the recording of meat inspection Code 904 for the presence of skin lesions suspected to be human inflicted. Weeds constitute major constraints for farmers by reducing crop yield and quality. However, weeds are managed effectively using herbicides, but this may cause harmful effects on human health and the environment. In an experiment on weed seed predation, we tested the biological control potential of carabid beetles to combat weeds in the absence of herbicides. Seeds from three common weed species were placed in cereal fields on conventional and organic farms located along a landscape complexity gradient (area annual crops within 1 km) in two distinct regions in Sweden. Carabid beetles were sampled in the same fields using pitfall traps. Neither carabid species richness nor seed removal was related to organic farming. Seed removal was significantly related to carabid species richness and both carabid species richness and seed removal was higher in landscapes with large total area of annual crops, although the evenness of the carabid communities was lower. The carabid genera with strongest positive relationship to seed removal differed between regions (Trechus in Uppland and Pterostichus in Scania), as did the preference for the different weed seed species. This study concludes that carabid species richness contributes to weed seed predation and that large scale landscape context explains more variation in the carabids’ responses than local farming practices. 相似文献

The actions of many extracellular stimuli are elicited by complexes of cell surface receptors, heterotrimeric guanine nucleotide-binding proteins (G proteins), and mitogen-activated protein (MAP) kinase complexes. Analysis of haploid yeast cells and their response to peptide mating pheromones has produced important advances in our understanding of G protein and MAP kinase signaling mechanisms. Many of the components, their interrelationships, and their regulators were first identified in yeast. Current analysis of the pheromone response pathway (see the Connections Maps at Science's Signal Transduction Knowledge Environment) will benefit from new and powerful genomic, proteomic, and computational approaches that will likely reveal additional general principles that are applicable to more complex organisms. 相似文献
