The purpose of this prospective study was to investigate platelet function using in vitro tests based on both high and low shear rates and von Willebrand factor (vWf) multimeric composition in dogs with cardiac disease and turbulent high-velocity blood flow. Client-owned asymptomatic, untreated dogs were divided into 4 groups: 14 Cavalier King Charles Spaniels (Cavaliers) with mitral valve prolapse (MVP) and no or minimal mitral regurgitation (MR), 17 Cavaliers with MVP and moderate to severe MR, 14 control dogs, and 10 dogs with subaortic stenosis (SAS). Clinical examinations and echocardiography were performed in all dogs. PFA100 closure times (the ability of platelets to occlude a hole in a membrane at high shear rates), platelet activation markers (plasma thromboxane B2 concentration, platelet surface P-selectin expression), platelet aggregation (in whole blood and platelet-rich plasma with 3 different agonists), and vWf multimers were analyzed. Cavaliers with moderate to severe MR and dogs with SAS had longer closure times and a lower percentage of the largest vWf multimers than did controls. Maximal aggregation responses were unchanged in dogs with SAS but enhanced in Cavaliers with MVP (regardless of MR status) compared with control dogs. No significant difference in platelet activation markers was found among groups. The data suggest that a form of platelet dysfunction detected at high shear rates was present in dogs with MR and SAS, possibly associated with a qualitative vWf defect. Aggregation results suggest increased platelet reactivity in Cavaliers, but the platelets did not appear to circulate in a preactivated state in either disease. 相似文献

OBJECTIVE: To evaluate the ability of a questionnaire regarding the nonphysical aspects of quality of life (QOL) to differentiate sick and healthy dogs. ANIMALS: 120 dogs. PROCEDURE: The questionnaire was administered by telephone to owners of 120 dogs with appointments at a veterinary teaching hospital. A QOL score was calculated for each dog on the basis of questions relevant to the dog during the 7 days before the interview. Scores were recorded as bar graphs, and linear regression was used to examine the effect of health status and other variables on QOL score. Certain questions were eliminated post hoc, on the basis of defined criteria, and the analyses were repeated. RESULTS: Scores were similar for sick (range, 670% to 93.8%) and healthy (range, 68.0% to 89.8%) dogs. Environment (suburban vs rural) and duration of ownership were significant explanatory variables and accounted for 10.5% of the variation in the QOL score. Eleven questions were eliminated post hoc. The scores derived from the 2 versions of the questionnaire were highly correlated (r = 0.92). CONCLUSIONS AND CLINICAL RELEVANCE: There was no evidence that the QOL questionnaire could differentiate healthy dogs from sick dogs; environmental and owner factors appeared to be more important. 相似文献

The majority of Halicephalobus gingivalis-infections in horses have been fatal and are usually not diagnosed before necropsy. Therefore, knowledge about the nematode and the pathogenesis of infection in horses is limited. This has resulted in an on-going discussion about the port of entry and subsequent dissemination of H. gingivalis within the host. The present case of H. gingivalis-infection in a horse was diagnosed ante mortem. Post mortem findings, the distribution pattern of H. gingivalis nematodes in the brain, a high prevalence of inflammation in close relation to blood vessels, and the presence of the nematode in multiple organs with a disseminated pattern of distribution strongly suggested a haematogenous spread of the nematode in the horse. 相似文献

Background
The fox tapeworm Echinococcus multilocularis has foxes and other canids as definitive host and rodents as intermediate hosts. However, most mammals can be accidental intermediate hosts and the larval stage may cause serious disease in humans. The parasite has never been detected in Sweden, Finland and mainland Norway. All three countries require currently an anthelminthic treatment for dogs and cats prior to entry in order to prevent introduction of the parasite. Documentation of freedom from E. multilocularis is necessary for justification of the present import requirements.Methods
The probability that Sweden, Finland and mainland Norway were free from E. multilocularis and the sensitivity of the surveillance systems were estimated using scenario trees. Surveillance data from five animal species were included in the study: red fox (Vulpes vulpes), raccoon dog (Nyctereutes procyonoides), domestic pig, wild boar (Sus scrofa) and voles and lemmings (Arvicolinae).Results
The cumulative probability of freedom from EM in December 2009 was high in all three countries, 0.98 (95% CI 0.96-0.99) in Finland and 0.99 (0.97-0.995) in Sweden and 0.98 (0.95-0.99) in Norway.Conclusions
Results from the model confirm that there is a high probability that in 2009 the countries were free from E. multilocularis. The sensitivity analyses showed that the choice of the design prevalences in different infected populations was influential. Insulin-like growth factor-1 (IGF-1), which mediates most effects of growth hormone, has effects on cardiac mass and function, and plays an important role in the regulation of vascular tone. In humans, an inverse relationship between degree of heart failure (HF) and circulating IGF-1 concentrations has been found in several studies. In dogs with HF, few studies have focused on IGF-1. We examined circulating IGF-1 concentrations in dogs with mitral regurgitation (MR) caused by myxomatous mitral valve disease. Study 1 included 88 Cavalier King Charles Spaniels (CKCSs) with a broad range of asymptomatic MR (median serum IGF-1: 76.7 microg/L; 25-75 percentile, 59.8-104.9 microg/L). As expected, standard body weight and percentage under- or overweight correlated directly with IGF-1. MR (assessed in 4 different ways) did not correlate with IGF-1. In study 2, 28 dogs with severe MR and stable, treated congestive HF had similar serum IGF-1 concentrations (median, 100.8 g/L; 25-75 percentile, 74.9-156.5 microg/L) as 11 control dogs (79.6 microg/L; 25-75 percentile, 64.1-187.4 microg/L; P = .84). In study 3, the plasma IGF-1 concentration of 15 untreated CKCSs with severe MR was 16.4 +/- 24.2 microg/L lower (P = .02) at the examination when decompensated HF had developed (80.8 +/- 30.9 microg/L) than at a visit 1-12 months earlier (97.2 +/- 39.8 microg/L), possibly in part due to an altered state of nutrition. The studies document that circulating IGF-1 concentrations are not altered before development of congestive HF in dogs with naturally occurring MR, but decrease by approximately 20% with the development of untreated HE In treated HF, circulating IGF-1 concentrations apparently return to within the reference range. 相似文献

The objective of this experiment was to determine the standardized ileal digestibility(SID) of amino acids(AA) in 3sources of distillers dried grains with solubles(DDGS) with different concentrations of fat.Twelve growing barrows(initial body weight:76.1 ± 6.2 kg) were randomly allotted to a replicated 6×4 Youden square design with 6 diets and 4 periods.The fat content of the 3 sources of DDGS were 11.5,7.5,and 6.9%respectively.Diets contained 60%DDGS and fat concentration of the diets were 7.5,52,and 5.2%,respectively.Two additional diets containing the 2sources of DDGS with 75 and 6.9%fat were also formulated,and corn oil was added to these diets to increase the concentration of fat in the diets to levels that were calculated to be similar to the diet containing conventional DDGS with 11.5%fat.A N-free diet was also formulated to calculate endogenous losses of crude protein(CP) and AA from the pigs.Pigs were fed experimental diets during four 7-d periods.The first 5 d of each period were an adaptation period and ileal digesta were collected on d 6 and 7 of each period.The apparent ileal digestibililty(AID) and SID of CP and all indispensable AA,except AID Pro and SID of Trp,were greater(P 〈 0.01) in conventional DDGS than in the 2 sources of DDGS with reduced fat.Adding oil to the diets containing the 2 sources of DDGS with reduced fat did not consistently increase SID of AA.In conclusion,conventional DDGS has greater SID values for most AA compared with DDGS that contains less fat and inclusion of additional oil to diets containing low-fat DDGS does not increase AID or SID of AA.The lower AA digestibility in low-fat DDGS could not be overcome by the inclusion of additional fat to the diets. 相似文献

In this paper, multilevel logistic models which take into account the multilevel structure of multi-site pig production were used to estimate the variances between pigs produced in Danish multi-site pig production facilities regarding seroconversion to Actinobacillus pleuropneumoniae serotype 2 (Ap2) and Mycoplasma hyopneumoniae (Mh). Based on the estimated variances, three newly described computational methods (model linearisation, simulation and linear modelling) and the standard method (latent-variable approach) were used to estimate the correlations (intra-class correlation components, ICCs) between pigs in the same production unit regarding seroconversion. Substantially different values of ICCs were obtained from the four methods. However, ICCs obtained by the simulation and the model linearisation were quite consistent. Data used for estimation were collected from 1161 pigs from 429 litters reared in 36 batches at six Danish multi-site farms chronically infected with the agents. At the farms, weaning age was 3-4.5 weeks, after which batches of pigs were reared using all-in/all-out management by room. Blood samples were collected shortly before: weaning, transfer from weaning-site to finishing-site, and sending the first pigs in the batch for slaughter (third sampling). Few pigs seroconverted at the weaning-sites, whereas considerable variation in seroconversion was observed at the finishing-sites. Multilevel logistic models (initially including four levels: farm, batch, litter, pig) were used to decompose the variation in seroconversion at the finishing-site. However, there was essentially no clustering at the litter level-leading to the use of three-level models. In the case of Ap2, clustering within batch was so high that the data eventually were reduced to two levels (farm, batch). For seroconversion to Ap2, ICC between pigs within batches was approximately 90%, whereas the ICC between pigs within batches for Mh was approximately 40%. This indicates that the possibility for Mh to spread between pigs within batches is lower than for Ap2. The diversity in seroconversion between batches within the same farm was large for Ap2 (ICC approximately 10%), whereas there was a relative strongly ICC (approximately 50%) between batches for Mh. This indicates that the transmission of Mh is more consistent within a farm, whereas the presence of Ap2 varies between batches within a farm. 相似文献

The strength and fracture behavior of dowel-type connections with stiff dowels loaded perpendicular to grain was studied by nonlinear 3D finite element (FE)-analysis. A cohesive zone model was used to model the perpendicular to grain fracture of the wood, i.e., failure by wood splitting along the grain. The influence of load eccentricity and dowel-to-loaded-edge distance was studied for a plate type of geometry loaded in tension and for a simply supported beam loaded in bending. The strength found from the FE-analysis is compared to strength from experimental tests with centric loading, showing overall good agreement. Numerical results for centric loading are further compared to strength predictions according to the linear elastic fracture mechanics (LEFM)-based design criterion present in Eurocode 5 (EN 1995-1-1:2004). The comparison showed good agreement regarding the relative influence of connection geometry, but the design criterion appears, however, to yield unconservative strength predictions. The results of the FE-analyses regarding dowel load eccentricity showed that such loading conditions may yield significantly lower strengths compared to centric loading. An approximate engineering method to account for the strength reduction due to load eccentricity is, furthermore, presented. 相似文献

In this study, we quantified the effects of local neighbourhood competition, light availability, and proximity to skid trails on the growth of sugar maple (Acer saccharum Marsh.) trees following selection harvest. We hypothesized that growth would increase with decreasing competition and increasing light availability, but that proximity to skid trails would negatively affect growth. A total of 300 sugar maples were sampled ∼10 years after selection harvesting in 18 stands in Témiscamingue (Québec, Canada). Detailed tree and skid trail maps were obtained in one 0.4 ha plot per stand. Square-root transformed radial growth data were fitted to a linear mixed model that included tree diameter, crown position, a neighbourhood competition index, light availability (estimated using the SORTIE light model), and distance to the nearest skid trail as explanatory variables. We considered various distance-dependent or -independent indices based on neighbourhood radii ranging from 6 to 12 m. The competition index that provided the best fit to the data was a distance-dependent index computed in a 6 m search radius, but a distance-independent version of the competition index provided an almost equivalent fit to data. Models corresponding to all combinations of main effects were fit to data using maximum likelihood, and weighted averages of parameter estimates were obtained using multimodel inference. All predictors had an influence on growth, with the exception of light. Radial growth decreased with increasing tree diameter, level of competition and proximity to skid trails, and varied among crown positions with trees in suppressed and intermediate positions having lower growth rates than codominants and dominants. Our results indicate that in selection managed stands, the radial growth of sugar maple trees depends on competition from close (≤6 m) conspecific neighbours, and is still affected by proximity to skid trails ∼10 years after harvesting. Such results underscore the importance of minimizing the extent of skid trail networks by careful pre-harvest planning of trail layout. We also conclude that the impact of heterogeneity among individual-tree neighbourhoods, such as those resulting from alternative spatial patterns of harvest, can usefully be integrated into models of post-harvest tree growth. 相似文献
