In Khorezm, a district of Uzbekistan situated in the Aral Sea Basin, soil salinization is an important driver of soil degradation in irrigated agriculture. The main objective of this study was to identify techniques that enable rapid estimation of soil salinity. Therefore, bulk electrical conductivity of the soil (ECa-meas) was measured with three different devices (2P, 4P, and CM-138) and electrical conductivity of the soil paste (ECp-meas) was measured with the so-called 2XP device. These measurements were compared with independent estimates of ECa-calc and ECp-calc based on laboratory measurements of the saturated extract, ECe, of soil samples from the same sites. Soil salinity could be assessed satisfactorily with all four devices. ECp-meas could be well reproduced by the 2XP device (R 2 = 0.76), whereas ECa-meas estimates using 2P, 4P, and CM-138 in the field were less accurate (R 2 < 0.50). The sensitivity of all devices to the main ions Cl and Ca2 + suggests that the measuring principles are similar for all instruments. The devices can therefore be used interchangeably. Field assessment of soil salinity was considerably enhanced by the use of CM-138, because large areas can be quickly assessed, which may be desirable in spite of the lower accuracy.  相似文献

Habitat fragmentation is a major cause for species loss, but its effect on invertebrates with low active dispersal power, like terrestrial gastropods, has rarely been studied. Such species can not cross a hostile habitat matrix, for which the predictions of island theory, such as positive relations between species richness and patch size, should apply. In order to test this prediction, we studied gastropod species diversity by assessing gastropod assemblage characteristics from 35 sites in 19 fragments of deciduous old-growth forests in the Lower Rhine Embayment, Germany. Assemblages differed between larger (≥700 ha) and smaller forests (<400 ha), those of large forests held a higher percentage of forest species. Although α-diversity was similar between the two forest size classes, small forests often comprised matrix species, resulting in a higher β-diversity. Edge effects on the species richness of matrix species were noticeable up to 250 m into the forest. Hierarchical partitioning revealed that distance to disturbances (external edge, internal edges like roads) explained most assemblage variables, whereas forest size and woodland cover within a 1 km radius from the sites explained only a few assemblage variables. Densities of two forest-associated species, Discus rotundatus and Arion fuscus, decreased with forest size. Yet, forest size was positively correlated with richness of typical forest species and densities of Limax cinereoniger. The latter species seems to need forests of >1,000 ha, i.e., well above the size of most fragments. In conclusion, the prediction is valid only for forest species. This study determined the relationship between the amount of irrigation water applied and the yield and quality of trickle-irrigated chile peppers (Capsicum annuum L.). From 1977 through 1983, field experiments were conducted near Las Cruces, NM. Field trickle lines were buried 5–8 cm below the soil surface and the amount of applied water was monitored with water meters. Meteorological data were gathered at a nearby weather station for irrigation scheduling. A linear relation, Y = ?14.11 + 0.56Diw with a R2 = 0.69 was found for green chiles where Y is the green chile yield (t/ha) and Diw is the depth of irrigation water applied (cm), including rain. Maximum yields occurred between 80 and 95 cm of water applied and varied from year to year. Maximum red chile production demands the same irrigation regime as does the green chile, but the influence of fall weather may overshadow the irrigation effects. No water can be saved by stressing the plants during any period of the growing season. The pungency of the green chile peppers shows a clear trend with the irrigation treatments; the drier the treatment, the more pungent the pepper. However, other factors also play a role and may reduce the irrigation effects completely. The effect of harvest date on yield was significant, but this effect can be levelled off by harvesting twice. An interaction effect was found between row spacing and total amount of irrigation water applied.  相似文献

With the commercial release in Australia in 2004 of a vaccine against feline immunodeficiency virus (FIV; Fel‐O‐Vax FIV®), the landscape for FIV diagnostics shifted substantially. Point‐of‐care (PoC) antibody detection kits, which had been the mainstay for diagnosing FIV infection since the early 1990s, were no longer considered accurate to use in FIV‐vaccinated cats, because of the production of vaccine‐induced antibodies that were considered indistinguishable from those produced in natural FIV infections. Consequently, attention shifted to alternative diagnostic methods such as nucleic acid detection. However, over the past 5 years we have published a series of studies emphasising that FIV PoC test kits vary in their methodology, resulting in differing accuracy in FIV‐vaccinated cats. Importantly, we demonstrated that two commercially available FIV antibody test kits (Witness? and Anigen Rapid?) were able to accurately distinguish between FIV‐vaccinated and FIV‐infected cats, concluding that testing with either kit offers an alternative to PCR testing. This review summarises pertinent findings from our work published in a variety of peer‐reviewed research journals to inform veterinarians (particularly veterinarians in Australia, New Zealand and Japan, where the FIV vaccine is currently commercially available) about how the approach to the diagnosis of FIV infection has shifted. Included in this review is our work investigating the performance of three commercially available FIV PoC test kits in FIV‐vaccinated cats and our recommendations for the diagnosis of FIV infection; the effect of primary FIV vaccination (three FIV vaccines, 4 weeks apart) on PoC test kit performance; our recommendations regarding annual testing of FIV‐vaccinated cats to detect ‘vaccine breakthroughs’; and the potential off‐label use of saliva for the diagnosis of FIV infection using some FIV PoC test kits. We also investigated the accuracy of the same three brands of test kits for feline leukaemia virus (FeLV) diagnosis, using both blood and saliva as diagnostic specimens. Based on these results, we discuss our recommendations for confirmatory testing when veterinarians are presented with a positive FeLV PoC test kit result. Finally, we conclude with our results from the largest and most recent FIV and FeLV seroprevalence study conducted in Australia to date.  相似文献

The aim of this study was to investigate the effects of temperature, light and oxygen on the germination of Amaranthus caudatus (var. viridis) seeds. At the highest temperature studied (35°C), seed germination was found to be most rapid and highest in both the dark and the light. Germination was inhibited by continuous white light. The lower the temperature, the greater the inhibitory effect of light. The far-red and blue spectral regions were the most effective in light inhibition. The light effect was also dependent on the percentage of oxygen in the atmosphere around the seeds; it was less marked in O2-enriched atmospheres. Light/ oxygen dependency resulted from the action of the seed coat and endosperm. Scarified seeds were much less sensitive to light and hypoxia than non-scarified seeds. Le but de ce travail était d'étudier les effets de la température, de la lumiere et de I'oxygène sur la germination des graines d'Amaranthus caudatus (var. viridis). A la temperature la plus élevée utilisée (35°C), la germination était la plus rapide et la plus complète tant à la lumière qu'à 1'obscuritè. La germination était inhibée par la lumière blanche continue. Plus la temperature était basse, plus 1'effet inhibiteur de la lumière était important. Les radiations rouge sombre et bleues du spectre étaient les plus in-hibitrices. L'effet de la lumière dépendait aussi de la teneur de 1'atmosphère en oxygène; il était moins marqué dans des atmosphères enrichies en oxygène. L'interaction lumière/oxygène provenait du tégument et de 1'albumen. Les graines scarifiées étaient beaucoup moins sensibles à la lumière et a 1'hypoxie que les graines non scarifiées. Die Wirkung des Lichts hing auch vom Sauerstoffgehalt der die Samen umgebenden Luft ab: Bei O25-Anreiche-rung war sie weniger deutlich. Die Licht/ Sauerstoff-Wechselwirkung wurde von der Samenschale und dem Endosperm beeinfluβt: Geritzte Samen waren gegenüber Licht und O2-Mangel viel weniger empfindlich als ganze.  相似文献

Freshly harvested mature Bromus rubens L. seeds are dormant. This dormancy corresponds to the inability of seeds to germinate in the dark at temperatures above 15°C, but is practically not apparent at temperatures between 5 and 15°C. Continuous white light markedly inhibits germination of dormant seeds, even at low irradiances, and reinforces their sensitivity to oxygen deprivation. However, prolonged illumination with white light does not alter subsequent germination in the dark. Breaking of dormancy during dry storage at 20°C results in a widening of the temperature range within which a high rate of germination occurs, and in almost complete disappearance of sensitivity of seeds to light. The structures surrounding the embryo (glumellae, caryopsis envelopes and possibly endosperm) are mainly responsible for dormancy and photosensitivity of whole seeds, probably because they limit the oxygen supply to the embryo. However, the embryo itself appears to play a role in dormancy. The results obtained are discussed in relation to dormancy of cereal seeds, and in relation to field establishment of B. rubens as a weed. La dormance des semences de Bromus rubens L. en relation avec les effets de la température, de la lumière et de l'oxygène Les semences mûres de Bromus rubens L. fraîchement récoltées sont dormantes. Leur dormance correspond à une inaptitude à germer à l'obscuritéà des températures supérieures à 15°C, mais elle ne se manifeste pratiquement pas entre 5 et 15°C. La lumière blanche continue inhibe nettement la germination des semences dormantes, même à de faibles énergies, et renforce leur sensibilitéà la privation d'oxygène. Cependant, un éclairement prolongé n'a aucune conséquence sur la germination ultérieure à l'obscurité. L'élimination de la dormance pendant la conservation au sec, à 20°C, se traduit par un élargissement de la gamme thermique qui assure une bonne germination et par la disparition presque complète de la sensibilité des semences à la lumière. Les structures qui entourent l'embryon (glumelles, enveloppes du caryopse et peut-être albumen) sont, en grande partie, responsables de la dormance et de la photosensibilité des semences entières, sans doute parce qu'elles limitent l'apport d'oxygène à l'embryon. Toutefois, l'embryon lui-même semble participer à la dormance. Les résultats obtenus sont discutés par comparaison avec la dormance des semences des céréales et en relation avec l'implantation, dans les cultures, de B. rubens en tant que mauvaise herbe. Das Brechen der Dormanz während der trockenen Lagerung bei 20 °C führt zu einem vergrößerten Temperaturbereich, in dem die Samen gut keimen, und läßt ihre Lichtempfindlichkeit fast vollständig verschwinden. Die Strukturen um den Embryo (Spelzen, Schale der Caryopse und möglicherweise Endosperm) sind hauptsächlich für die Dormanz und die Lichtempfindlichkeit des ganzen Samens verantwortlich, vermutlich deshalb, weil sie die Sauerstoffversorgung des Embryos einschränken. Aber auch der Embryo selbst scheint bei der Dormanz eine Rolle zu spielen. Die Untersuchungsergebnisse werden im Vergleich zur Dormanz von Getreidesamen und hinsichtlich des Vorkommens von Bromus rubens als Unkraut diskutiert.  相似文献

The μ-opioid receptor (MOR) is expressed in bovine, human, equine and canine oocytes, and in seasonal breeders, it is expressed with higher intensity during the anoestrous phase. Supplementation of in vitro maturation (IVM) medium with opioid agents, agonists or antagonists, was shown to affect oocyte maturation in several species such as rat, bovine and equine. This study reports the effects of supplementing IVM medium with naloxone (Nx), an opioid antagonist, on nuclear and cytoplasmic maturation rate of oocytes recovered from anoestrous bitches. Cytoplasmic maturation was examined in terms of mitochondrial (mt) distribution. In order to confirm the receptor-mediated action of Nx, in oocytes of anoestrous bitches, MOR expression was analyzed by Western blot. Cumulus–oocyte complexes, recovered from the ovaries of bitches in anoestrous, were cultured in vitro and Nx was added at the concentrations of 1 × 10−6, 1 × 10−8 and 1 × 10−10  m . The rate of oocytes resuming meiosis after culture in presence of 1 × 10−6  m Nx (29%) was significantly higher than that of oocytes of control group (12%; p < 0.05). However, treatment with Nx did not affect mt distribution pattern. In denuded oocytes and in corresponding cumulus cells, a doublet of 65 and 50 kDa was observed. We conclude that, in oocytes of anoestrous bitches, MOR is expressed and Nx significantly improves nuclear maturation rate. Further studies should be performed to elucidate the expression of other opioid receptors, such as δ and κ, and possible interactive effects of their antagonists on canine oocyte maturation.  相似文献

OBJECTIVE: To determine effectiveness of a pour-on formulation of doramectin against Damalinia bovis, Haematopinus eurysternus, Linognathus vituli, Solenopotes capillatus, Chorioptes bovis, Sarcoptes scabiei, Hypoderma bovis, and Hypoderma lineatum. ANIMALS: Cattle of various ages with naturally acquired or artificial infestations with 1 or more species of lice, mites, or grubs. PROCEDURE: In 10 louse and 6 mite studies, cattle were treated with doramectin (500 microg/kg, topically) on day 0, and parasite counts were performed approximately weekly from days 0 to 35. In 6 grub studies, cattle expected to harbor Hypoderma spp were treated before emergence of warbles. After warbles began to emerge, they were counted every 2 weeks, and grubs were collected and identified by species. RESULTS: Burdens of D bovis, H eurystemus, L vituli, and S capillatus on doramectin-treated cattle were 0 by 28 days after treatment. Burdens of C bovis and S scabiei decreased to 0 in naturally infested cattle and approximately 0 in artificially infested cattle by day 14 to 15. In grub studies, 107 of 136 control cattle had warbles, whereas 2 of 136 doramectin-treated cattle had 1 warble each, which represented a cure rate of 98.5%. CONCLUSION AND CLINICAL RELEVANCE: One topical application of doramectin was highly efficacious against common species of lice, mites, and grubs known to affect performance, health, and appearance of cattle.  相似文献

Soybean lipoxygenase (LOX) inactivation [0.4 mg/mL in Tris-HCl buffer (0.01 M, pH 9)] was studied quantitatively under constant pressure (up to 650 MPa) and temperature (-15 to 68 degrees C) conditions and kinetically characterized by rate constants, activation energies, and activation volumes. The irreversible LOX inactivation followed a first-order reaction at all pressure-temperature combinations tested. In the entire pressure-temperature area studied, LOX inactivation rate constants increased with increasing pressure at constant temperature. On the contrary, at constant pressure, the inactivation rate constants showed a minimum around 30 degrees C and could be increased by either a temperature increase or decrease. On the basis of the calculated rate constants at 102 pressure-temperature combinations, an iso-rate contour diagram was constructed as a function of pressure and temperature. The pressure-temperature dependence of the LOX inactivation rate constants was described successfully using a modified kinetic model of Hawley.  相似文献

The use of exogenous gonadotrophins in puberty inducement and ovulation synchronization is a technique that has a positive influence on the management of swine. The purpose of this study was to verify the effects of a second gonadotrophin treatment [equine chorionic gonadotrophin (eCG) and luteinizing hormone (LH), intramuscularly (i.m.)] upon the second oestrus synchronization and fertility in gilts. Seventy-one NAIMA (Pen Ar Lan) gilts had their first oestrus (puberty inducement) induced by a hormonal treatment (eCG and LH). Then, they were randomly distributed into two treatments, with (T1) and without (C) gonadotrophin treatment at the second oestrus. The animals were fed with a single ration (16% of crude protein and 3286.73 kcal ME/kg), and timed artificial insemination performed at the second oestrus. Gilts were slaughtered for embryo recovery and ovary examination about 5 days after insemination. There was no evidence of a difference in the percentage of the second oestrus expression (T1 - 90.90% and C - 86.84%), the duration of the oestrus cycle (T1 - 19.62 +/- 0.82 days and C - 19.67 +/- 4.14 days), the percentage of follicular cysts (T1 - 15.15% and C - 18.42%) and number of ovulations (T1 - 14.60 +/- 5.7 and C - 13.23 +/- 4.8) between treatments (p > 0.05). However, the hormonal treatment (T1) showed minor oestrus dispersion and embryo viability (T1 - 8.4 +/- 5.6 and C - 11.2 +/- 4.6) (p < 0.05). These results indicate that the better synchronization and expression of the second oestrus when using gonadotrophins (eCG and LH) is followed by a lower embryo viability, which is probably the consequence of the heterogeneous follicle recruitment during the injection of eCG.  相似文献
