The hypothesis was that forage species and ensiling conditions have an impact on the formation of biogenic amines and the feed choice of goats in short‐term preference trials. At ensiling, lucerne (Medicago sativa L., first cut), red clover (Trifolium pratense L., first cut) and Italian ryegrass (Lolium multiflorum Lam ., second cut) were treated differently to obtain a range of fermentation qualities. Six treatments of each forage species were prepared and included different dry‐matter concentrations, chemical and biological silage additives, and additions of soil. Silages were sampled for chemical analyses (proximate constituents, fermentation products and other volatile compounds, crude protein fractions and biogenic amines) and stored anaerobically in vacuum‐sealed plastic bags for use in preference trials (one trial for each forage species) with Saanen‐type wethers (= 8 or 6). Each possible two‐way combination of the six silage treatments and a standard hay (= 21 combinations) was offered for ad libitum intake for 3 hr. Data were analysed using multidimensional scaling, analysis of variance and correlation analysis between silage characteristics and dry‐matter intake (DMI). For each forage species, fermentation characteristics and crude protein fractions revealed only small differences among treatments. Although the degree of proteolysis, as measured by non‐protein nitrogen, of all silages was high, biogenic amine and butyric acid concentrations were low. The different treatments apparently had no direct influence on the formation of biogenic amines and feed choice. The preference behaviour within one forage species was strongly divergent, but DMI rankings of the three species were very similar.  相似文献

The demosponge Suberites domuncula has been described to contain high levels of a proteinaceous toxin, Suberitine, that displays haemolytic activityIn the present study this 7-8 kDa polypeptide has been isolated and was shown to exhibit also cytotoxic effects on cells of the same species. Addition of retinal, a recently identified metabolite of β-carotene that is abundantly present in S. domuncula was found to reduce both the haemolytic and the cell toxic activity of Suberitine at a molar ratio of 1:1. Spectroscopic analyses revealed that the interaction between β-carotene and Suberitine can be ascribed to a reversible energy transfer reaction. The enzyme that synthesises retinal in the sponge system is the β,β-carotene-15,15'-dioxygenase [carotene dioxygenase]. In order to clarify if this enzyme is the only β-carotene-metabolizing enzyme a further oxygenase had been identified and cloned, the (related) carotenoid oxygenase. In contrast to the dioxygenase, the carotenoid oxygenase could not degrade β-carotene or lycopene in Escherichia coli strains that produced these two carotenoids; therefore it had been termed related-carotenoid oxygenase. Exposure of primmorphs to light of different wavelengths from the visible spectrum resulted after 3 days in a strong upregulation of the dioxygenase in those 3D-cell aggregates that had been incubated with β-carotene. The strongest effect is seen with blue light at a maximum around 490 nm. It is concluded that the toxin Suberitine is non-covalently modified by retinal, the cleavage product from β-carotene via the enzyme carotene dioxygenase, a light inducible oxygenase. Hence, this study highlights that in S. domuncula the bioactive metabolite, retinal, has the property to detoxify its homologous toxin.  相似文献

High year-to-year variability in the incidence of Fusarium spp. and mycotoxin contamination was observed in a two-year survey investigating the impact of maize ear rot in 84 field samples from Germany. Fusarium verticillioides, F. graminearum, and F. proliferatum were the predominant species infecting maize kernels in 2006, whereas in 2007 the most frequently isolated species were F. graminearum, F. cerealis and F. subglutinans. Fourteen Fusarium-related mycotoxins were detected as contaminants of maize kernels analyzed by a multi-mycotoxin determination method. In 2006, a growth season characterized by high temperature and low rainfall during anthesis and early grain filling, 75% of the maize samples were contaminated with deoxynivalenol, 34% with fumonisins and 27% with zearalenone. In 2007, characterized by moderate temperatures and frequent rainfall during the entire growth season, none of the 40 maize samples had quantifiable levels of fumonisins while deoxynivalenol and zearalenone were detected in 90% and 93% of the fields, respectively. In addition, 3-acetyldeoxynivalenol, 15-acetyldeoxnivalenol, moniliformin, beauvericin, nivalenol and enniatin B were detected as common contaminants produced in both growing seasons. The results demonstrate a significant mycotoxin contamination associated with maize ear rots in Germany and indicate, with regard to anticipated climate change, that fumonisins-producing species already present in German maize production may become more important.  相似文献

An autoregressive procedure to predict the hydraulic conductivity — Comparison of measured and predicted results An instantaneous profile method was used to measure the unsaturated hydraulic conductivity. Relatively new techniques involving undisturbed soil samples instrumented with minitensio-meters and Time-Domain-Reflectometry (TDR) mini-probes were used for the experiments. The laboratory method allows a high spatial and temporal resolution. Laboratory measurements were carried out for 40 soil horizons with a wide spectrum of texture and bulk density. In addition, retention curves were measured using the standard pressure plate apparatus. Using this homogeneous set of data, an autoregressive model was developed which allows a stepwise calculation of the hydraulic conductivity for a water potential range of —30 up to —600 hPa. This model was developed for loamy sands, sandy, silty and clayey soils in conjunction with data from the retention curves. The calculation procedure starts with the determination of an initial unsaturated conductivity (k) close to field capacity, i.e., for water potential from —60 hPa up to —100 hPa. This first value is then used to predict other conductivity values using appropriate changes in soil water content corresponding to a defined range of the soil water potential. Subsequently, the hydraulic conductivities for higher and lower potentials were estimated considering the k value of the previous step in combination with the data of the retention curve of the next water potential range. The advantage of this empirical model is the indirect consideration of soil structure, in contrast to the closed-form van Genuchten-Mualem (vGM) model. To demonstrate these effects on different fitting procedures, the vGM model was also used to describe soil hydraulic functions. The accuracy of both, the vGM model and the autoregressive one, were compared for various fitting procedures and soils.  相似文献

In einem dreijährigen Gefäßversuch wurde der Einfluß unterschiedlicher C‐Gehalte von Böden aus dem “Thyrower Bodenfruchtbarkeitsversuch”; auf den Ertrag von Roggen und Ölrettich sowie deren Boden‐ und Düngerstickstoffaufnahme geprüft. Bei Roggen führte höherer Humusgehalt nur bei Unterlassung einer mineralischen Stickstoffdüngung zu höheren Erträgen, beim nachgebauten Ölrettich auch in der gedüngten Variante. Eine bessere Ausnutzung des Düngerstickstoffs bei höherem Humusgehalt und eine Mehrmobilisation von Bodenstickstoff durch mineralische Stickstoffdüngung konnte nicht festgestellt werden. Von dem nach Versuchsabschluß im Boden verbleibenden Düngerstickstoff wurden zwischen 70 ... 90 % als Bestandteil von Humusfraktionen ermittelt.  相似文献

The antioxidant capacity of methanol and ethanol seed extracts from Euterpe oleracea Mart. (a?aí) against the reactive oxygen species (ROS) peroxyl radicals, peroxynitrite, and hydroxyl radicals was studied with the total oxidant scavenging capacity (TOSC) assay in a modified and automated version. Cold methanol digestion was the most efficient extraction method with respect to the antioxidant capacity. The extracts exhibit good antioxidant capacity against peroxyl radicals, similar to the capacity of the pulp. The antioxidant capacity against peroxynitrite and hydroxyl radicals is even higher. The main antioxidants identified by HPLC-MS and HPLC-CEAD are five different procyanidins (di- through pentamers); furthermore, protocatechuic acid and epicatechin were identified as minor compounds. Determination of TOSC values of HPLC seed extract fractions indicates that the procyanidins contribute substantially to the overall antioxidant capacity. In addition, however, other compounds that have not yet been identified are responsible for a large part of the observed antioxidant capacity.  相似文献

Wettability of soil affects a wide variety of processes in soils like infiltration, percolation, preferential flow, and surface runoff. Even though efforts have been made to determine contact angles and surface tension or energy of smooth surfaces, the determination on granular materials like soil particles remains unsolved. One objective of this study was to test the consistency of contact angles (CA) measured with the newly modified and easy‐to‐apply Wilhelmy plate method by using solid particles and liquids with defined variations of surface tension. A second objective was to derive basic physical surface properties for the irregularly shaped and chemically heterogeneous soil particles. Advancing contact angles were determined by using model soils varying in texture from clay to coarse sand to check the impact of grain size on the CA measurements. Varying the solid‐surface tension with dichlorodimethlylsilane treatments provided for pure‐water wetting stages ranging from wettable to extremely hydrophobic. The surface tensions of the liquids were varied from 72 mN m–1 to 25 mN m–1 by using water or water‐ethanol mixtures. The surface tensions of the model soils were determined with the zero degree–contact angle method following Zisman's critical–surface tension concept. Results show that the measured CA varied continuously with the variations of the surface tension of the liquid and the solid phase, respectively. A general interpretation of the results is possible by using the concept of the Equation of State Approach.  相似文献

Chemical investigation of the ethyl acetate extract from the marine-derived Streptomyces sp. isolate B1848 resulted in three new isoquinolinequinone derivatives, the mansouramycins E–G (1a–3a), in addition to the previously reported mansouramycins A (5) and D (6). Their structures were elucidated by computer-assisted interpretation of 1D and 2D NMR spectra, high-resolution mass spectrometry, and by comparison with related compounds. Cytotoxicity profiling of the mansouramycins in a panel of up to 36 tumor cell lines indicated a significant cytotoxicity and good tumor selectivity for mansouramycin F (2a), while the activity profile of E (1a) was less attractive.  相似文献

Nmim content in the soil, N-fertilization and N uptake of winter wheat in the international organic nitrogen long-term fertilization experiment (IOSDV) Berlin-Dahlem During the 9th and 10th year of the long-term IOSDV field experiment micro plots were put in three treatments. Labelled15 N (160 resp. 110 kg/ha N as ammonium sulfate) was fertilized to winter wheat subdivided into three portions. Nmin in soil was determined five times during the season, plant biomass was harvested at different growth stages and N uptake was calculated. Using the15 N-technique permitted a discrimination between fertilizer-N and soil-N. Preferential uptake of fertilizer-N by the wheat crop but also immobilisation in soil were observed until June. Subsequently the immobilized N was remineralized and assimilated by wheat. But the native Nmin of soil was minimaly utilized during the initial growth of wheat. Therefore the absolute amount of soluble N temporarily increased caused by a mineralisation of the organic matter.  相似文献
