Citrus Variegated Chlorosis (CVC) is currently present in approximately 40% of citrus plants in Brazil and causes an annual loss of around 120 million US dollars to the Brazilian citrus industry. Despite the fact that CVC has been present in Brazil for over 20 years, a relationship between disease intensity and yield loss has not been established. In order to achieve this, an experiment was carried out in a randomized block design in a 3 × 2 factorial scheme with 10‐year‐old Natal sweet orange. The following treatments were applied: irrigation with 0, 50 or 100% of the evapotranspiration of the crop, combined with natural infection or artificial inoculation with Xylella fastidiosa, the causal agent of CVC. The experiment was evaluated during three seasons. A negative exponential model was fitted to the relationships between yield versus CVC severity and yield versus Area Under Disease Progress Curve (AUDPC). In addition, the relationship between yield versus CVC severity and canopy volume was fitted by a multivariate exponential model. The use of the AUDPC variable showed practical limitations when compared with the variable CVC severity. The parameter values in the relationship of yield–CVC severity were similar for all treatments unlike in the multivariate model. Consequently, the yield–CVC intensity relationship (with 432 data points) could be described by one single model: y = 114·07 exp(?0·017 x), where y is yield (symptomless fruit weight in kg) and x is disease severity (R2 = 0·45; P < 0·01). 相似文献

Antioxidant capacities of vetiver (Vetiveria zizanioides) oil were evaluated by two different in vitro assays: the DPPH* free radical scavenging assay and the Fe2+-metal chelating assay. Results showed that the vetiver oil (VO) possessed a strong free radical scavenging activity when compared to standard antioxidants such as butylated hydroxytoluene (BHT) and alpha-tocopherol. However, its metal chelating capacity was relatively weak. VO (10 microL/mL) dissolved in methanol exhibited approximately 93% free radical scavenging activity in the DPPH* assay and approximately 34% Fe2+ chelating activity in the metal chelating assay. By contrast, 10 mM BHT and 0.1 mM alpha-tocopherol exhibited 93 and 89% free radical scavenging activities in the DPPH* assay, respectively, and 1 mM EDTA exhibited approximately 97% activity in the metal chelating assay. Among the complex constituents in the crude VO, beta-vetivenene, beta-vetivone, and alpha-vetivone, which had shown strong antioxidant activities, were isolated and identified using various chromatographic techniques including silica gel open column chromatography, silica HPLC, and GC-MS. Selected seafood including rabbitfish (Siganus canaliculatus), brown-spotted grouper (Epinephelus areolatus), clam (Tapes philippinarum), oyster (Ostrea rivularis), shrimp (Penaeus japonicus), and crab (Charybdis feriatus), commonly found in the Hong Kong wet market, was monitored for their distribution and seasonal variations of their bromophenol contents. Specifically, 2-bromophenol (1). 4-bromophenol (2). 2,4-dibromophenol (3). 2,6-dibromophenol (4). and 2,4,6-tribromophenol (5). were monitored due to their flavor impact to seafood. All samples of marine origin contained bromophenols throughout a year. Crab had the highest concentration of total bromophenol content throughout the season. Concentrations of compounds 1, 4, and 5 in the local seafood were generally higher than that found in the literature values to provide characteristic flavor, but lower than that required to cause off-flavor. Variations of the flavor impact of bromophenols in seafood during a season could be better shown by their flavor values. Distribution and seasonal variations of bromophenol content in seafood coincided well with the seasonal growth cycle of the bromophenol synthesizing seaweeds, e.g. brown algae, in the region suggesting the abundant source of bromophenols in the environment might have a high impact on the quantity of bromophenols found in seafood. 相似文献

During a three‐year project from 2003 to 2006, two models have been developed to predict leaf rust (Puccinia recondita and P. triticina) occurrence and to simulate disease incidence progress curves on the upper leaf layers of winter rye (PUCREC) and winter wheat (PUCTRI). As input parameters the models use air temperature, relative humidity and precipitation. PUCREC and PUCTRI firstly calculate daily infection favourability and a cumulative infection pressure index and, in a second step, disease incidence is estimated. An ontogenetic model (SIMONTO) is used to link disease predictions to crop development. PUCREC and PUCTRI have been validated with data from 2001 to 2005. Both models give satisfactory results in simulating leaf rust epidemics and forecasting dates when action thresholds for leaf rust control are exceeded. 相似文献

We describe a fibrosarcoma in a 12-year-old Quarterhorse × Arabian gelding as a sequela to equine influenza vaccination. Shortly after the second vaccination, swelling at the site was noticed by the owner and it continued to increase in size over the following 6 months. Biopsy of the mass indicated a fibrosarcoma had developed at the vaccination site. It was approximately 20 cm in diameter and elevated well above the level of the skin. There was no clinical evidence of metastases to the lungs or local lymph nodes. Surgical resection of the mass was performed and the wound healed by first and second intention. Histopathological examination and immunohistochemical staining confirmed a myofibroblastic fibrosarcoma with multifocal osseous metaplasia. To the authors' knowledge, this is the first equine case of a vaccine-associated fibrosarcoma. 相似文献

We describe two structures of the intact bacterial ribosome from Escherichia coli determined to a resolution of 3.5 angstroms by x-ray crystallography. These structures provide a detailed view of the interface between the small and large ribosomal subunits and the conformation of the peptidyl transferase center in the context of the intact ribosome. Differences between the two ribosomes reveal a high degree of flexibility between the head and the rest of the small subunit. Swiveling of the head of the small subunit observed in the present structures, coupled to the ratchet-like motion of the two subunits observed previously, suggests a mechanism for the final movements of messenger RNA (mRNA) and transfer RNAs (tRNAs) during translocation. 相似文献

Background
Studies have reported that heparin may be unsuitable as an anticoagulant in human plasma samples when quantifying cytokines using multiplex bead array assays. For mouse samples, multiplex assays have been validated for serum and EDTA-plasma, but it remains to be elucidated whether heparin influences the quantification of cytokines, and if so – to what extent. Furthermore, laboratory mice are often anesthetized for blood sampling, which causes acute stress that may influence circulating cytokine concentrations and thus bias experimental results. The objectives of the present study were to identify whether specific cytokine concentrations varied between heparin-plasma, serum, and EDTA-plasma, and whether short isoflurane anesthesia would influence the concentrations of these cytokines in the circulation. Twenty-three acute phase and pro-inflammatory cytokines were quantified in matched serum, EDTA-plasma, and heparin-plasma samples from anesthetized and unanesthetized male NMRI mice using a multiplex assay. In addition, samples from unanesthetized mice were spiked with three levels of heparin.Results
The concentrations of five out of 23 cytokines were significantly different between sample types, but only one cytokine (IL-17A) differed between heparin-plasma and serum. When further spiking the heparin-plasma with increasing concentrations of heparin, there was a significant effect on 11 cytokines, where the cytokine recovery could be correlated to the heparin concentration for ten of these cytokines. Anesthesia resulted in lower concentrations of G-CSF, but had no significant impact on the concentrations of the other 22 cytokines.Conclusion
In mice, heparin seems like a suitable anticoagulant for obtaining plasma for multiplex assays for the cytokines IL-1α, IL-1β, IL-2, IL-6, IL-9, IL-12p40, IL-12p70, IL-13, G-CSF, GM-CSF, IFN-γ, KC, MCP-1, MIP-1α, MIP-1β, RANTES and TNFα, but an effect of heparin in high concentrations should be considered for the cytokines IL-9, IL-12p40, IL-12p70, KC, MCP-1, MIP-1β and RANTES. Rwt4 (synonym of Rmg1), a temperature-insensitive gene for resistance to Avena isolates of Magnaporthe oryzae, was identified in wheat cultivar Norin 4 in a seedling assay. The significance of Rwt4 was evaluated using flag leaves of wheat cultivars. At high temperature, Norin 4 was completely resistant to Avena isolate Br58, while Chinese Spring, a noncarrier of Rwt4, was susceptible. Genetic analysis of F2 plants derived from Norin 4 × Chinese Spring indicated that the resistance of flag leaves of Norin 4 to the Avena isolate is conditioned by a single major gene. Segregation analysis of F3 seedlings derived from the F2 plants showed that the major gene is actually Rwt4. These results suggest that Rwt4 is effective against Avena isolates throughout the growth stages. Furthermore, screening of Pyricularia isolates from various hosts suggested that Panicum isolates are possible carriers of the corresponding avirulence gene, PWT4. Segregation analyses of F2 and F3 seedlings showed that Panicum isolates actually carry PWT4, and, therefore, that Rwt4 is also effective against Panicum isolates. On the other hand, none of the Oryza, Setaria, Triticum, and Lolium isolates tested was a carrier of PWT4.
