The main objective of the present work was to clarify the causal relationship between leaf apoplastic pH increase and Fe chlorosis under alkaline growth conditions. It has been shown that nitrate supply in contrast to ammonium supply induced a pH increase in the apoplast of young green leaves of Helianthus annuus which was followed within 12 hours by leaf yellowing. Hence nitrate nutrition is the primary cause of a high leaf apoplastic pH which induces Fe deficiency chlorosis and not the impaired provision of ATP for plasmalemma H+ pumps in yellow leaves. Supply of bicarbonate in physiological concentrations had virtually no influence on leaf apoplastic pH. Spraying leaves with diluted acids (citric acid, sulphuric acid) resulted in a decrease of apoplastic pH followed by leaf re-greening. Interestingly, the Fe concentrations remained the same in the yellow control leaves and in the sprayed green leaves. From this it follows that Fe efficiency in leaves is mainly related to the Fe distribution between apoplast and symplast. It was demonstrated that Fe chlorosis induced by nitrate nutrition begins from the base of the youngest leaves, presumably from growing interveinal microsites showing high nitrate uptake rates. Leaf yellowing spread gradually from the leaf base to the tip and after seven days of nitrate supply the leaf was almost completely yellow (98%). Leaf yellowing was measured by means of a video imaging technique. Leaf apoplastic pH recordings were conducted after loading the fluorescent dye FITC-Dextran (4000 D) into the leaf apoplast of intact plants thus simulating in vivo conditions. To improve water use efficiency on drylands it is essential to understand the mechanisms affecting water balance partitioning in arable land ecosystems. A field experiment was conducted, from October 2001 to October 2004, to evaluate the effects of field management regimes on water balance and water use efficiency in a winter wheat (Triticum aestivum L.) system in the Loess Plateau, China. The field management regimes tested were: (i) conventional agricultural management (winter wheat followed by a ploughed summer fallow); (ii) conventional management plus a fallow crop used as green manure; (iii) application of wheat straw mulch (0.8 kg m−2), which was retained throughout the year (no summer ploughing). A process-oriented ecosystem model (CoupModel) was calibrated with field measurements of soil water contents, leaf area indices, plant heights and crop yields, then used to generate comparative simulations of the water balance partitioning under the wheat straw mulch, fallow crop and conventional management regimes. The simulations indicated that during the experimental period mulching increased soil water storage by 5–8%, decreased soil evaporation by 11–13%, and increased wheat transpiration by 2–5% compared with the conventional management regime, thus increasing the wheat yield and water use efficiency. Furthermore, water reached deeper horizons under mulching than under conventional practice, resulting in 15% more deep percolation in a wet year. The simulation results also indicated that growing green manure during the fallow period decreased soil water storage, leading to lower wheat yields. Mulching proved to be an efficient measure for increasing yields, and possibly contributed to groundwater recharge.  相似文献

Near-infrared reflectance spectroscopy (NIRS) is known for its inexpensiveness, rapidity and accuracy and may become a useful tool for the assessment of soil quality. Objectives were (i) to evaluate the ability of NIRS to predict several chemical and biological properties of organically managed arable soils as well as the properties of grain yield from winter cereals for a closed population and (ii) to test whether the use of field-moist and pre-treated (quick-freezing followed by freeze-drying and grinding) samples will generate similar results. One hundred and sixteen soil samples from nine organically managed farms from Germany sampled in 2005 and 2006 were used for this investigation. Spectra of the near-infrared region (including the visible range, 400–2500 nm) from field-moist (<2 mm) or pre-treated soil samples were recorded. A modified partial least-square regression method and cross-validation were used to develop an equation over the whole spectrum (first–third derivation). For the pre-treated soils, good predictions were obtained for pH, contents of organic C, total N, plant-available P (Olsen) and exchangeable K (calcium-acetate-lactate (CAL)), contents of microbial biomass C and N (Cmic and Nmic) and ergosterol, basal respiration, metabolic quotient, the ratio of organic C/total N, the grain yield of winter cereals and grain nitrogen uptake. The RSC (the ratio of standard deviation of laboratory results to standard error of cross-validation) was greater than 2.0, the correlation coefficients (r) of a linear regression (measured against predicted values) were greater than or equal to 0.9 and the regression coefficients (a) ranged from 0.9 to 1.1. Similar good predictions were obtained if field-moist samples were used, with the exception of P (Olsen), K (CAL), metabolic quotient, grain yield of winter cereals and grain nitrogen uptake (satisfactory predictions) and ergosterol content (unsatisfactory prediction). Good predictions of the contents of Mg (CaCl2) and microbial biomass P (Pmic) were achieved for field-moist but not for pre-treated samples. Despite sample preparation, only satisfactory predictions were obtained for the ratios of Cmic/Nmic and ergosterol/Cmic and grain nitrogen content (1.4RSC2.0, r0.8 and 0.8a1.2). However, unsatisfactory predictions for field-moist and pre-treated samples were achieved for the content of P (CAL), the nitrogen mineralisation rate and the ratios of Cmic/Pmic and basal respiration/nitrogen mineralisation rate. Our results demonstrate that biological soil properties can be predicted with NIRS for closed populations in both sample states. The pre-treatment should be used if samples have to be stored prior to infrared measurements for periods longer than a month.  相似文献

The objectives of the study were to identify principal hydrological pathways and source areas of N and P losses by multi-scale monitoring and to estimate total nutrient losses from the catchment. An agricultural catchment with rain-fed agriculture and irrigated paddy fields in subtropical China was monitored with regularly sampling, together with intensive sampling during and after rain storms. Regular weekly sampling showed that the N concentrations in the overland flows from the upland and paddy fields were higher than those from the streams, but lower than those in the subsurface waters. The N concentration, on average, was 10.0 mg L− 1 in the well and 1.7 mg L− 1 in the spring water, the former was 10.2 times as high as that in the stream waters (1.0–1.5 mg L− 1). Nitrogen and P in the overland flows originated dominantly in particulate forms from the uplands (over 70%) and in dissolved forms from the paddy fields. Inorganic N and P dominated in the streams and subsurface waters. The intensive sampling allowed us to establish flow-nutrient concentration relationships and to extrapolate nutrient losses during rainstorms without regular sampling. The extrapolation increased the estimated nutrient losses by about 30% to 50%. The average total nutrient losses within three water years were estimated as 21 kg N ha− 1 yr− 1 and 1 kg P ha− 1 yr− 1, accounting for 9.5% and 1.4% of chemical N and P fertilizers applied to the catchment after subtracting the nutrient inputs with irrigation and rainfall. The estimation showed that paddy fields were as important as the uplands in terms of nutrient losses. These results suggest that control of soil erosion and excessive irrigation could be effective to reduce nutrient export through overland flow and subsurface flow.  相似文献

在奥地利及其他多数欧洲国家英国紫杉濒临灭绝,被列为濒危树种.基于不同地理位置的自然群落的比较,调查了奥地利两块基因资源保育林内英国紫杉的空间结构、更新状况及可采取的保育方法等.每块样地圆杆材分布变化较大.林地Stiwollgraben内紫杉单位面积植株总数,平均胸径和平均株高分别为492 n·hm-2, 8.8 cm and 6.3 m;Leininger Riese仅为45 n·hm-2, 16.3 cm and 7.6 m.Stiwollgraben群落紫杉长势较好,健康植株79%以上,而Leininger Riese群落的健康植株低于49%.两块样地内紫杉更新模式差异较大,与其各自的树龄动态分布相一致.1年龄幼树相比较,Stiwollgraben林地内紫杉密度为13019株/公顷,而Leininger Riese仅为1368株/公顷.Leininger Riese林地内未发现树高51-150cm的幼树,而Stiwollgraben林地内树高30-150cm的幼树鲜能见到.这种情况下,英国紫杉林的保育需要管理完善的保护区、采伐期间采取长期轮伐,同时还要防止牲畜啃食和减轻种间竞争,以此来增强红豆杉的生存能力.图4表4参41.  相似文献

The aim of the study was to investigate the serological reactions of pregnant sows to vaccination with Hyoresp. Further investigations were performed in the offspring of these sows to follow the dynamics of maternal antibodies and the reaction to vaccination at different points in time. The study was conducted in three farrow-to-finish herds endemically infected with M. hyopneumoniae. A total of 30 gilts and 31 sows were vaccinated 8 and 4 weeks ante partum with Hyoresp (Merial GmbH) or given phys. saline solution as a placebo. The offspring was divided into three groups receiving Hyoresp at 1 and 4 or at 4 and 8 weeks of age. The control group was treated with phys. saline solution at 1 and 4 weeks of age. Before vaccination, antibodies against M. hyopneumoniae were detected in 85% of the gilts and 68% of the sows, confirming the endemic infection of the herds. Vaccination of the sows induced a significant increase in the antibody concentration in serum within four weeks and enhanced the concentration of antibodies in the colostrum. As expected, significantly enhanced levels of antibodies were also detected during the first four weeks of life of the offspring of vaccinated sows. The piglets' serological reaction to vaccination at 1 and 4 weeks of age showed marked interferences with maternal antibodies, so that a reaction could be demonstrated only at 8 weeks of age. The serological reaction of piglets vaccinated at 4 and 8 weeks of age was much stronger than that of piglets vaccinated earlier. Surprisingly, the vaccination status of the sow had no effect on the serological response of the piglets in either vaccination scheme. Maternal antibodies are known to reduce the risk of M. hyopneumoniae infections in piglets. Vaccinating the sows against M. hyopneumoniae may thus be an option for farrowing-to-finish herds with an enhanced risk for infections due to ineffective separation of different age groups, poor gilt acclimatisation or high gilt replacement rates.  相似文献

We compared immunogenicity in pigs of whole cell lysate proteins (WCP) with murein-associated proteins (MAP) obtained from a virulent serotype 2 strain of Streptococcus (S.) suis grown at 32 or 42 degrees C. Protein fractions were tested for their ability to induce antibodies in 3-week-old piglets by enzyme-linked immunosorbent assay and Western blot analysis. We found a significant increase in the antibody levels in all sera irrespective of the preparation used for immunization. However, alpha-WCP sera showed higher reactivities than alpha-MAP sera, and piglets immunized with 32 degrees C preparations (alpha-32 sera) showed higher responses than those immunized with 42 degrees C preparations (alpha-42 sera). Western blot analysis revealed that alpha-WCP sera in part reacted with different proteins when compared with alpha-MAP sera. Furthermore, some proteins were only detected by alpha-32 but not by alpha-42 sera. In conclusion, the results demonstrate the immunogenicity of cell wall MAP of S. suis, and highlight the importance of considering growth conditions in the preparation of subunit vaccines.  相似文献

Data from the Swedish Forest Inventory was used to calculate mass balances for base cations Ca, Mg and K for Swedish forests. Using lysimeter and forest survey soil analyses to estimate present base cation leaching from the root zone reveals that weathering plus base cation deposition is not sufficient to support both, the present base cation leaching rate and the present rate of uptake caused by stem growth. Calculations suggest that 96% of the productive forested area may have higher rates of removal than supply for one or more base cation. Under a best-case scenario, assuming less pollution, the present growth rate and 100% efficiency in uptake of available nutrients, the area with more removal than supply would still be at least 30% of the total area. Forest soils are being depleted at a rate where the exchangeable reservoirs have high risk of being severely depleted in the next few decades in central and southern Sweden. During 1983–1985 the depletion rate is calculated to be, on the average, 0.33 keq ha−1 year−1. The weathering rate and present base cation deposition can sustain growth at a level where (80–85)×106m3 stemwood year−1 can be harvested. Any harvested growth beyond this volume must be sustained by artificial means.

For whole-tree harvesting without base cation return, the calculations indicate that it would significantly increase the base saturation depletion rate to an average of 0.62 keq ha−1 year−1, and risk depletion of the soil in less than one-to-two rotation periods almost anywhere in Sweden.

The calculations stress the importance that sustainable forest management must include the management of nutrient fluxes and reservoirs.  相似文献
