Antimicrobial agents are used extensively off‐label in mink, as almost no agents are registered for this animal species. Pharmacokinetic (PK) and pharmacodynamic (PD) data are required to determine antimicrobial dosages specifically targeting mink bacterial pathogens. The aims of this study were to assess, in a PKPD framework, the empirical dosage regimen for a combination of trimethoprim (TMP) and sulfadiazine (SDZ) in mink, and secondarily to produce data for future setting of clinical breakpoints. TMP and SDZ PK parameters were obtained experimentally in 22 minks following IV or oral administration of TMP/SDZ (30 mg/kg, i.e. 5 mg/kg TMP and 25 mg/kg SDZ). fAUC/MIC with a target value of 24 hr was selected as the PKPD index predictive of TMP/SDZ efficacy. Using a modeling approach, PKPD cutoffs for TMP and SDZ were determined as 0.062 and 16 mg/L, respectively. By incorporating an anticipated potentiation effect of SDZ on TMP against Escherichia coli and Staphylococcus delphini, the PKPD cutoff of TMP was revised to 0.312 mg/L, which is above the tentative epidemiological cutoffs (TECOFF) for these species. The current empirical TMP/SDZ dosage regimen (30 mg/kg, PO, once daily) therefore appears adequate for treatment of wild‐type E. coli and S. delphini infections in mink. 相似文献

Red clover (Trifolium pratense) and lucerne (Medicago sativa) are among the most common legume species used for ruminant feeding systems. Besides the greater nutritive quality and first-year ley yield compared to lucerne, red clover decreases its yield over the years. However, under unfavourable soil conditions the persistency of lucerne can be compromised, not performing as expected. This study compared yield and chemical composition of eight lucerne cultivars and one tetraploid red clover cultivar, with similar growth capacity as lucerne, replicated over two years. In the second year, four lucerne cultivars and the red clover cultivar were analysed for crude protein (CP) fractions, dry matter and neutral detergent fibre (NDF) digestibility. A randomized block design was used with three field blocks per cultivar for the multiple comparisons within each year. There were differences in DM yield, concentrations of CP and NDF and CP fractions among the lucerne cultivars, but the differences were inconsistent between years and between growth cycles within year. The red clover cultivar had lower CP and NDF concentrations than a majority of the lucerne cultivars, and greater in vitro organic matter digestibility compared to all of the lucerne cultivars. Different packaging conditions of salt-cured cod fillets were studied during 2-years chilled storage. Yield and quality of salt-cured Atlantic cod fillets packed in sealed polystyrene boxes or in traditional cardboard boxes were compared. After 4 months, yield decreased by approximately 10% and decreased further when packed in cardboard boxes. Lipid oxidation was low after 12 months, and no microbiological spoilage was registered. The total amount of bacteria in the fish reduced with storage time, independent of packaging. The reddish color of halophilic bacteria was detected after 7–8 weeks at 18°C, but not if packed with MAP. 相似文献

Background
Quantitative multi-elemental analysis by inductively coupled plasma (ICP) spectrometry depends on a complete digestion of solid samples. However, fast and thorough sample digestion is a challenging analytical task which constitutes a bottleneck in modern multi-elemental analysis. Additional obstacles may be that sample quantities are limited and elemental concentrations low. When microbial strains compete for the same limiting nutrient in continuous culture, resource-based competition theory predicts that only one strain will survive and all others will die out. The surviving strain expected from theory will be the one with the smallest subsistence or "break-even" concentration of the limiting resource, a concentration defined by the J parameter. This prediction has been confirmed in the case of auxotrophic bacterial strains competing for limiting tryptophan. Because the value of J can be measured on the strains grown alone, the theory can predict the qualitative outcomes of mixed-growth competition in advance of actual competition. 相似文献

Sorghum ergot produces dihydroergosine (DHES) and related alkaloids, which cause hyperthermia in cattle. Proportions of infected panicles (grain heads), leaves and stems were determined in two forage sorghum crops extensively infected 2 to 4 weeks prior to sampling and the panicles were assayed for DHES. Composite samples from each crop, plus a third grain variety crop, were coarsely chopped and half of each sealed in plastic buckets for 6 weeks to simulate ensilation. The worst-infected panicles contained up to 55 mg DHES/kg, but dilution reduced average concentrations of DHES in crops to approximately 1 mg/kg, a relatively safe level for cattle.
