The agricultural landscape of central Europe has changed dramatically in recent decades due to intensified cultivation, bringing many of its characteristic species to the brink of extinction. We investigated whether landscape structure affects the genetic structure and diversity of remnant populations of the two arable plant species Adonis aestivalis and Consolida regalis. We used dominant amplified fragment length polymorphism markers (AFLPs) and compared populations from six regions of 5 km² in central Germany. These regions represent two different classes of landscape structural complexity: intensively used, homogeneous landscapes (>95% of area covered by arable land with low extent of field margins) or heterogeneous regions (<60% of area covered by arable land with large extent of field margins). Contrary to expectations, within‐population diversity levels did not significantly differ between homogeneous and heterogeneous landscapes. No significant isolation‐by‐distance was found for either species, regardless of landscape structure, and genetic structures may still mirror more continuous conditions before large‐scale restructuring commenced in Germany's arable landscapes from the 1950s onwards. These results suggest that current landscape complexity, as such, is not as important for local‐level genetic structure for the species studied. Thus, homogenised and intensively used landscapes may also be important for the conservation of arable plant diversity and should not be neglected.  相似文献

Background: Many in‐house tests for the diagnosis of feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV) infection are licensed for use in veterinary practice. A new test with unknown performance has recently appeared on the market. Objectives: The aims of this study were to define the efficacy of a new in‐clinic test system, the Anigen Rapid FIV Ab/FeLV Ag Test, and to compare it with the current leading in‐clinic test, the SNAP Kombi Plus FeLV Antigen/FIB Antibody Test. Methods: Three‐hundred serum samples from randomly selected healthy and diseased cats presented to the Clinic of Small Animal Medicine at Ludwig Maximilian University were tested using both the Anigen Rapid Test and the SNAP Kombi Plus Test. Diagnostic sensitivity, specificity, and positive and negative predictive values were calculated for both tests using Western blot as the gold standard for verification of FIV infection and PCR as the gold standard for FeLV infection. Results: The presence of antibodies against FIV was confirmed by Western blot in 9/300 samples (prevalence 3%). FeLV DNA was detected by PCR in 15/300 samples (prevalence 5%). For FIV infection the Anigen Rapid Test had a sensitivity of 88.9%, specificity of 99.7%, positive predictive value of 88.9%, and negative predictive value of 99.7%. For FeLV infection, the Anigen Rapid Test had a sensitivity of 40.0%, specificity of 100%, positive predictive value of 100%, and negative predictive value of 96.9%. Diagnostic accuracy was similar to that of the SNAP Kombi Plus Test. Conclusion: The new Anigen Rapid FIV Ab/FeLV Ag Test performed very well and can be recommended for use in veterinary practice.  相似文献

Two alpacas from a herd in southwest Switzerland died for unknown reasons. Necropsy revealed chronic weight loss and pale mucous membranes. Infection with hemotropic mycoplasmas was suspected and subsequently confirmed by molecular methods. In order to investigate the epidemiological situation in this herd, a real-time TaqMan(?) qPCR assay for the specific detection and quantification of hemoplasma infection in South American camelids was developed. This assay was based on the 16S rRNA gene and amplified 'Candidatus Mycoplasma haemolamae' DNA, but not DNA from other hemoplasmas or non-hemotropic mycoplasma species. The lower detection limit was one copy/PCR, and the amplification efficiency was 97.4%. In 11 out of 24 clinically healthy herd mates of the two infected alpacas, 'Candidatus M. haemolamae' infection was confirmed. No correlation was found between bacterial load and clinical signs or anemia. The assay described herein enables to detect and quantify 'Candidatus M. haemolamae' and may be used in future studies to investigate the prevalence, pathogenesis and treatment follow-up of hemoplasma infections in South American camelids.  相似文献

Habitat fragmentation has been generally regarded detrimental to the persistence of many species, especially those with limited dispersal abilities. Yet, when exactly habitat elements become functionally disconnected very much depends on the dispersal ability of a species in combination with the landscape’s composition in which it occurs. Surprisingly, for many small and ground-walking generalists knowledge at what spatial scale and to what extent landscape structure affects dispersal is very scarce. Because it is flightless, the bush cricket Pholidoptera griseoaptera may be regarded susceptible to fragmentation. We applied habitat occupancy surveys, population genetic analyses and movement modelling to investigate the performance of P. griseoaptera in an agricultural mosaic landscape with suitable habitat patches of varying size and isolation. Despite its presumed dispersal limitation we could show that P. griseoaptera occupied the majority of suitable habitats, including small and isolated patches, showed a very low and non-significant genetic differentiation (F ST = 0.0072) and, in the model, managed to colonize around 73% of all suitable habitat patches within one generation under weak and strong landscape-effect scenarios. We conclude that P. griseoaptera possesses the behavioural attributes (frequent inter-patch dispersal) necessary to persist in this landscape characterized by a patchy distribution of habitat elements. Yet, sound recommendations to landscape planning and conservation require more research to determine whether this represents a general behaviour of the species or a behavioural adaptation to this particular landscape.  相似文献

Striga hermonthica is a major constraint to smallholder subsistence agriculture production in the sub-Saharan African region. Low soil fertility and overall environmental degradation has contributed to the build-up of the parasitic weed infestation. Improved cropping systems have to be introduced to address the interrelated problems of S. hermonthica and soil fertility decline. Thus, the effects of improved fallow with leguminous shrub Sesbania sesban on maize yields and levels of S. hermonthica infestation on farm land in the bimodal highlands of western Kenya were investigated. The experimental treatments were arranged in a phased entry, and randomized complete block scheme were six months Sesbania fallow, 18 months Sesbania fallow, six months natural fallow consisting of regrowth of natural vegetation without cultivation, 18 months natural fallow, continuous maize cropping without fertilizer application, and continuous maize cropping with P and N fertilization. Results show that Sesbania fallows significantly (p<0.05) increase maize yield relative to continuous unfertilized maize. S. hermonthica plant populations decrease in continuous maize between the first season (mean = 428 000 ± 63 000 ha−1) and second season (mean=51 000 ± 15 000 ha−1), presumably in response to good weed management. S. hermonthica seed populations in the soil decrease throughout the duration of the experiment in the continuous maize treatments. Short-duration Sesbania fallows can provide modest yield improvements relative to continuous unfertilized maize, but short-duration weedy fallows are ineffective. Continuous maize cultivation with good weed control may provide more effective S. hermonthica control than fallowing.  相似文献

Climate change with increasing temperature and ocean acidification (OA) poses risks for marine ecosystems. According to Pörtner and Farrell [1], synergistic effects of elevated temperature and CO2-induced OA on energy metabolism will narrow the thermal tolerance window of marine ectothermal animals. To test this hypothesis, we investigated the effect of an acute temperature rise on energy metabolism of the oyster, Crassostrea gigas chronically exposed to elevated CO2 levels (partial pressure of CO2 in the seawater ~0.15 kPa, seawater pH ~ 7.7). Within one month of incubation at elevated Pco2 and 15 °C hemolymph pH fell (pHe = 7.1 ± 0.2 (CO2-group) vs. 7.6 ± 0.1 (control)) and Peco2 values in hemolymph increased (0.5 ± 0.2 kPa (CO2-group) vs. 0.2 ± 0.04 kPa (control)). Slightly but significantly elevated bicarbonate concentrations in the hemolymph of CO2-incubated oysters ([HCO 3]e = 1.8 ± 0.3 mM (CO2-group) vs. 1.3 ± 0.1 mM (control)) indicate only minimal regulation of extracellular acid-base status. At the acclimation temperature of 15 °C the OA-induced decrease in pHe did not lead to metabolic depression in oysters as standard metabolism rates (SMR) of CO2-exposed oysters were similar to controls. Upon acute warming SMR rose in both groups, but displayed a stronger increase in the CO2-incubated group. Investigation in isolated gill cells revealed a similar temperaturedependence of respiration between groups. Furthermore, the fraction of cellular energy demand for ion regulation via Na+/K+-ATPase was not affected by chronic hypercapnia or temperature. Metabolic profiling using 1H-NMR spectroscopy revealed substantial changes in some tissues following OA exposure at 15 °C. In mantle tissue alanine and ATP levels decreased significantly whereas an increase in succinate levels was observed in gill tissue. These findings suggest shifts in metabolic pathways following OA-exposure. Our study confirms that OA affects energy metabolism in oysters and suggests that climate change may affect populations of sessile coastal invertebrates such as mollusks.  相似文献
