The detection of Mycobacterium paratuberculosis organisms in bovine faeces by isolation was compared with that by the microscopical examination of Ziehl-Neelsen stained faecal smears for the presence of clumps of acid-fast M. paratuberculosis organisms. Faeces were obtained from cattle naturally or experimentally infected with M. paratuberculosis as well as from uninfected cattle. Microscopical examination was an unreliable method for the detection of M. paratuberculosis organisms, since the organisms were only detected in 99 (=55.9%) of 177 culturally positive faecal samples. 1111 addition, clumps of acid-fast organisms indistinguishable from M. paratuberculosis were also observed iin three of 18 samples from cattle free from Johne's disease and in 18 of 37 culturally negative samples from paratuberculous cattle. When M. paratuberculosis organisms were added to faeces from an uninfected cow, results showed that isolation attempts should be positive when 15 or more M. paratuberculosis organisms per gram of faeces are present. 相似文献

We describe the development of solar water-splitting cells comprising earth-abundant elements that operate in near-neutral pH conditions, both with and without connecting wires. The cells consist of a triple junction, amorphous silicon photovoltaic interfaced to hydrogen- and oxygen-evolving catalysts made from an alloy of earth-abundant metals and a cobalt|borate catalyst, respectively. The devices described here carry out the solar-driven water-splitting reaction at efficiencies of 4.7% for a wired configuration and 2.5% for a wireless configuration when illuminated with 1 sun (100 milliwatts per square centimeter) of air mass 1.5 simulated sunlight. Fuel-forming catalysts interfaced with light-harvesting semiconductors afford a pathway to direct solar-to-fuels conversion that captures many of the basic functional elements of a leaf. 相似文献

The detection and therapy of canine vector-borne diseases in imported dogs are of major importance in small animal practice. Over the last years, the import of dogs from the Mediterranean region and Southeast Europe has increased, countries often endemic for a variety of vector-borne diseases such as babesiosis, hepatozoonosis, leishmaniosis, dirofilariosis or ehrlichiosis. This retrospective study presents the evaluation of data from our diagnostic laboratory on vector-borne infections in imported dogs from the years 2004-2008. Specific antibodies were detectable in 20.5% of all samples with individual detection rates of 8.9%, 9.6% and 10.8% for Babesia canis ssp., Leishmania spp. and/or Ehrlichia canis. A total of 5.5% of all samples tested by direct methods were positive. Up to 1.1% of Giemsa-stained blood/buffy coat smears were positive for B. canis ssp., Rickettsia spp. or Hepatozoon spp. Microfilariae were detectable by the Knott's Test in 6.4% and heartworm antigen was detectable using the DiroChek-ELISA in 3% of the examined samples. EDTA-blood samples were positive for Leishmania spp.-(14.9%), E. canis- (5.3%) and A. phagocytophilum-DNA (5.0%) by PCR. Therewith, imported dogs have a high chance of being carriers of pathogens. As some diseases may also be of a zoonotic concern, in case of the availability of competent vectors, the key focus in the future should be aimed at the prevention of importing infected dogs or at compulsory diagnostic screening and treatment of infected dogs at the time of import. 相似文献

Advances in magnetic resonance (MR) imaging and three-dimensional (3D) modeling software provide the tools necessary to create sophisticated, interactive anatomic resources that can assist in the interpretation of MR images of extremities, and learning the structure and function of limb musculature. Modeling provides advantages over dissection or consultation of print atlases because of the associated speed, flexibility, 3D nature, and elimination of superimposed arrows and labels. Our goals were to create a diagnostic atlas of pelvic limb muscles that will facilitate interpretation of MR images of patients with muscle injury and to create a 3D model of the canine pelvic limb musculature to facilitate anatomic learning. To create these resources, we used structural segmentation of MR images, a process that groups image pixels into anatomically meaningful regions. The Diagnostic Atlas is an interactive, multiplanar, web-based MR atlas of the canine pelvic limb musculature that was created by manually segmenting clinically analogous MR sequences. Higher resolution volumetric MR and computed tomography (CT) data were segmented into separately labeled volumes of data and then transformed into a multilayered 3D computer model. The 3D Model serves as a resource for students of gross anatomy, encouraging integrative learning with its highly interactive and selective display capabilities. For clinicians, the 3D Model also serves to bridge the gap between topographic and tomographic anatomy, displaying both formats alongside, or even superimposed over each other. Nitrate-N (NO3 --N) pollution of water resources is a widely recognized problem. Water and nitrogen fertilizer are the two most important factors affecting NO3 --N movement to surface and groundwater. Field trials were conducted from 1998 to 2000 growing seasons to investigate the combined impacts of water table management (WTM) and N fertilization rate on NO3 --N concentration in the soil profile and in drain discharge. There were two water table treatments: free drainage (FD) with open drains at a 1.0 m depth from the soil surface and subirrigation (SI) with a target water table depth of 0.6 m below the soil surface, and two N fertilizer rates: 120 kg N ha-1 (N120) and 200 kg N ha-1 (N200) in a split-plot design. Compared to FD, SI reducedNO3 --N concentration in the soil by up to 50% averaged over the two N rates. Concentrations of NO3 --N in drainage water fromSI plots were lower than those from FD by 55 to 73%. These findings suggest that SI can be used as a means of reducing soil NO3 --N pollution and drainage water NO3 --N concentrations. 相似文献

There is a growing interest in the adoption of conservation tillage systems [no-till (NT) and reduced tillage (RT)] as alternatives to conventional tillage (CT) systems. A 2-year study was conducted to investigate possible environmental consequences of three tillage systems on a 2.4-ha field located at Macdonald Research Farm, McGill University, Montreal. The soil was a sandy loam (0.5 m depth) underlain by a clay layer. Treatments consisted of a factorial combination of CT, RT, and NT with the presence or absence of crop residue. Soil NO3--N concentrations tended to be lower in RT than NT and CT tillage treatments. Denitrification and N2O emissions were similar among tillage systems. Contrary to the popular assumption that denitrification is limited to the uppermost soil layer (0–0.15 m), large rates of N2O production were measured in the subsurface (0.15–0.45 m) soil, suggesting that a significant portion of produced N2O may be missed if only soil surface gas flux measurements are made. The N2O mole fraction (N2O:N2O+N2) was higher in the drier season of 1999 under CT than in 2000, with the ratio occasionally exceeding 1.0 in some soil layers. No-tillage systems contribute to physical, chemical and biological changes in the soil. The effects of different tillage practices and phosphorus (P) fertilization on soil microbial biomass, activity, and community structure were studied during the maize growing season in a maize–soybean rotation established for 18 years in eastern Canada. Soil samples were collected at two depths (0–10 and 10–20 cm) under mouldboard plow (MP) and no-till (NT) management and fertilized with 0, 17.5, and 35 kg P ha?1. Results show that the duration of the growing season had a greater effect on soil microbiota properties than soil tillage or P fertilization at both soil depths. Seasonal fluctuations in soil microbial biomass carbon (SMB-C) and nitrogen (SMB-N), in dehydrogenase and alkaline phosphomonoesterase activities, and in total phospholipids fatty acid (PLFA) level, were greater under NT than MP management. The PLFA biomarkers separated treatments primarily by sampling date and secondly by tillage management, but were not significantly affected by P fertilization. The abundance of arbuscular mycorrhizal fungi (AMF; C16:1ω5) and fungi (C18:2ω6,9) was lower under NT than MP at the 10–20-cm soil depth in July. Phosphorus fertilization increased soil microbial biomass phosphorus (SMB-P) and Mehlich-3 extractable P, but had a limited impact on the other soil properties. The inclusion of legumes in semi‐arid native grasslands may promote the productivity and nutritive value of forage. This study was designed to assess the effect of legumes (the introduced legume Medicago sativa or the native legume Dalea purpurea) and soil P fertility (addition of 0, 50, or 200 P2O5 kg/ha at seeding) on the dry matter and nutrient content of native grasses mixtures, compared with the commonly used introduced forage grass Bromus biebersteinii grown with M. sativa. Plant harvests were performed in September 2008, July 2009 and September 2009. Plants nutrient content, δ15N value and dry matter were analysed. Results show that the M. sativa enhanced the N and P concentrations of native grass mixtures early in the summer, as well as the N concentration in Bouteloua gracilis in late summer of the driest year, 2009. The higher AM fungal diversity promoted by M. sativa was positively correlated with the dry matter and nitrogen uptake of M. sativa and with the P concentration of native grasses, in early summer. Overall, this study shows that M. sativa promoted beneficial AM fungal taxa and improved forage production in the semi‐arid prairies. 相似文献
