We quantified the resistance levels of transgenic rice plants, expressing Myxococcus xanthus protoporphyrinogen oxidase (PROTOX) in chloroplasts and mitochondria, to PROTOX inhibitors, acifluorfen, oxyfluorfen, carfentrazone-ethyl, and oxadiazon. We also determined whether active oxygen species-scavenging enzymes are involved in the resistance mechanism of transgenic rice. The transgenic rice line M4 was about >200-fold more resistant to oxyfluorfen than the wild-type (WT). M4 was also resistant to acifluorfen, carfentrazone-ethyl, and oxadiazon, but did not show multiple resistance to imazapyr and paraquat, which have different target sites. Acifluorfen, oxyfluorfen, carfentrazone-ethyl, and oxadiazon reduced the chlorophyll content in leaves of WT, but had minimal or no effect on M4. The PROTOX inhibitors also caused significant lipid peroxidation in the treated leaves of WT rice. However, the malondialdehyde production in M4 was not affected by these herbicides. The WT rice had higher activities of superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase than M4 after treatment with PROTOX inhibitors. A similar response was observed in all cases of antioxidant isozyme profiles analyzed. However, the induction in antioxidant activity in WT was not enough to overcome the toxic effects of a PROTOX inhibitor so the plant eventually died.  相似文献

Previous studies reported that omega-3 fatty acid and α-linolenic acid are important compounds that prevent cardiovascular disease and cancer in humans. Soybean [Glycine max (L.) Merr.] oil typically contains ~8 % α-linolenic acid (ALA). Elevated (~15 %) ALA content in seed oil is a trait of wild soybeans (G. soja Sieb. and Zucc.). Decreasing the ratio of linoleic acid (LA) to ALA to 4:1 or lower (compared to the ratio of 6 or 7:1 found in commercial soybean seed) should have health benefits for humans. This study was conducted to determine the environmental stability of elevated ALA acid recombinant inbred lines (RILs) derived from a cross of PI 483463 (wild soybean with 15 % ALA) and Hutcheson (cultivar with 9 % ALA). The fatty acid profile analysis from nine environments showed that the content of ALA for the RILs 156, 159 and 166 ranged from 10.7 to 15.7, 14.0 to 15.8, and 14.8 to 15.8, and averaged 13.9, 14.9, and 15.2 % respectively. The contents of ALA from these RILs and the wild soybean parent showed consistently higher than cultivated check soybeans. Two of the three RILs with elevated ALA content and ratios of <4:1 LA to ALA were as stable in ALA content as the high and low linolenic acid parents across growing environments. This indicates that lines with elevated ALA content developed from wild soybean PI 483463 are stable in ALA across environments and would be useful in improving soybeans with lower LA to ALA ratios.  相似文献

中国城镇化进程的快速推进,使得各种建设活动对土地资源的需求日益增加,引发一系列生态环境问题,特别是在生态脆弱区,土地资源不合理的开发利用加剧了区域生态安全风险。因此,为探究区域生态安全格局优化的途径方法,保障区域生态安全和土地资源合理利用,该研究通过解译判读怀来县1993年、2003年和2013年3期遥感影像,运用土地利用变化信息图谱及构建建设用地和生态用地竞争优势模型,分析了怀来县土地利用变化热点区域及建设用地与生态用地竞争优势分布区,划分了建设用地与生态用地优势分布临界线,同时,基于景观生态学理论,构建了怀来县综合生态安全格局,并将建设用地与生态用地竞争优势分布区应用于生态安全格局优化。结果表明:怀来县综合生态安全格局面积为1 470.59 km2,占怀来县总面积的82.51%,基本安全格局分布在临界线外围,满意安全格局外围主要沿临界线分布、理想安全格局主要分布在临界线内部。基本生态安全格局受到建设用地扩张的压力相对较小,满意生态安全格局主要位于生态优势区,在一定程度可以起到对建设用地扩张的限制作用,作为怀来县重要的生态缓冲区,理想安全格局受到建设用地扩张的压力较大,其内部应注重生态廊道网络建设,引导自然斑块以廊道或小型斑块的形式分散地融入建设用地中。研究方法和结果可以为区域生态规划和城乡布局等提供决策参考。  相似文献

The objective of this study was to determine if the South American catfish (Rhamdia quelen) is suitable to be farmed in polyculture with rainbow trout (Oncorhynchus mykiss) in intensive systems during the juvenile phase to maximize feed efficiency. Juveniles of rainbow trout (3.94?±?0.11 g) and South American catfish (2.07?±?0.04 g) were distributed in 16 tanks (100 L) with continuous water renewal at the density of 50 fish/tank. The experimental design was completely randomized with four treatments (proportions between species) and four replicates. The treatments were 100% trout (100T), 70% trout and 30% catfish (70T30C), 50% trout and 50% catfish (50T50C), and 100% catfish (100C). Fish were fed twice daily with pelleted commercial feed (45% crude protein) during an experimental period of 56 days. No feed was provided for the catfish in polyculture. The weight gained by the trout was higher in polyculture. Fish survival did not differ among the treatments. The average survival of the trout in all tanks was 99.6?±?1.0%, while the survival of the South American catfish was 97.9?±?2.7%. The total feed conversion ratio was lower in the 70T30J treatment, followed by the 100T treatment. Rainbow trout and South American catfish are compatible species for farming together in the first phase of their juvenile development. The different spaces occupied by these species inside tanks probably prevent competition or agonistic behavior. Catfish eat the non-ingested leftover diet from the trout, which improves feed conversion and increases sustainability.  相似文献

Nine Monochoria vaginalis Pres1 accessions from Chonnam province, Korea were tested for resistance to the sulfonylurea herbicide, imazosulfuron, in whole-plant response bioassay. All accessions were confirmed resistant (R) to imazosulfuron. The GR50 (imazosulfuron concentration that reduced shoot dry weight by 50%) values of R accessions were 1112-3172 (accession #9) times higher than that of the standard susceptible (S) accession. Accession #9 exhibited cross-resistance to other sulfonylurea herbicides, bensulfuron-methyl, cyclosulfamuron and pyrazosulfuron-ethyl, but not to the imidazolinone herbicides, imazapyr and imazaquin. The R biotype could be controlled by other herbicides with different modes of action, such as mefenacet and pyrazolate, applied to soil at recommended rates. Foliar-applied herbicides, 2,4-D and bentazone, also controlled both the R and S biotypes. Sulfonylurea-based mixtures, except ethoxysulfuron plus fentrazamide, did not control resistant M. vaginalis. Rice yield was reduced 70% by resistant M. vaginalis that escaped pyrazosulfuron-ethyl plus molinate, compared with hand weeding in direct-seeded rice culture. In contrast, rice yield was reduced 44% by resistant M. vaginalis that survived the pyrazosulfuron-ethyl plus molinate treatment, compared with pyrazolate plus butachlor in transplanted rice culture. In vitro acetolactate synthase (ALS) activity of the R biotype was 183, 35, 130 and 31 times more resistant to imazosulfuron, bensulfuron-methyl, cyclosulfamuron and pyrazosulfuron-ethyl, respectively, than the S biotype. Imidazolinone herbicides, imazapyr and imazaquin had similar effect on in vitro ALS activity of the R and S biotypes. The in vivo ALS activity of the R biotype was also less affected than the S biotype by the sulfonylurea herbicides imazosulfuron and pyrazosulfuron-ethyl. Results of in vitro and in vivo ALS assays indicate that the resistance mechanism of M. vaginalis to sulfonylurea herbicides may be due, in part, to an alteration in the target enzyme, ALS. Since the level of resistance in the enzyme assay was much lower than that in the whole-plant assay, other mechanisms of resistance, such as herbicide metabolism, may be involved.  相似文献

Pyribenzoxim, benzophenone O‐[2,6‐bis(4,6‐dimethoxypyrimidin‐2‐yloxy)benzoyl]oxime, is a new post‐emergence herbicide providing broad‐spectrum weed control in rice fields. [14C]Pyribenzoxim was used to study the pharmacokinetics of the compound after oral administration of a dose of 1000 mg kg?1 to male Sprague–Dawley rats. The material balance ranged from 97.3 to 99.7% of the administered dose and urinary and fecal recovery accounted for 97.1%, with the majority of radioactivity recovered in feces (88.6%) by 168 h after treatment. Elimination as volatile products or as carbon dioxide was negligible. The following values were obtained for the compound in the blood: AUC0–168h, 28400 µg equiv h g?1; Tmax, 12 h; Cmax, 372 µg equiv g?1; half‐life, 53 h. Radioactivity in tissue decreased from 96.1% of applied radiocarbon at 6 h to 0.4% at 168 h and the highest concentration of radioactivity among the tissues was observed in liver while the lowest residues were found in brain. The elimination half‐lives of radioactivity from tissues was in the range of 7 to 77 h and Tmax values of 12, 24 and 12 h were observed for blood, liver and kidney, respectively. 本文研究了紫胶树脂在漂白和脱氯过程中分子量、热寿命、酸价、碘价、羟基值和氯含量的变化情况。结果证明:漂白过程中,氯主要以自由基加成的方式进入紫胶树脂,同时还有氯的自由基取代和树脂上醛基、伯羟基的氧化反应。在脱氯过程中,漂白胶树脂上大部分危害其贮存性的结合氯被除去,这些氯通过消去和取代反应脱离树脂,同时产生双键和羟基。脱氯后仍残留在紫胶树脂上的氯相当稳定,难以自行脱落,经过脱氯处理,漂白胶具备了片胶所具备的优良性质。  相似文献

This report describes the diagnosis of porcine circovirus 2 (PCV2)-associated enteritis in 6 weaned pigs without postweaning multisystemic wasting syndrome by histopathology, virus isolation, and in situ hybridization. The most unique lesions were granulomatous inflammation affecting Peyer's patches, characterized by infiltrates of epithelioid macrophages and giant multinucleated cells. Large, multiple, basophilic or amphophilic, grape-like intracytoplasmic inclusion bodies were often seen in the cytoplasm of histiocytic cells and giant multinucleated cells. No microscopic lesions were observed in the lymphoid tissue, such as lymph node, spleen, and tonsil. A strong hybridization signal for PCV2 was detected in the cytoplasm of histiocytes and giant multinucleated cells in Peyer's patches. Porcine circovirus 2 was isolated from homogenates of the small and large intestines in 2 weaned pigs. The presence of diarrhea and granulomatous enteritis, and abundant PCV2 DNA associated with the microscopic lesions is suggestive of PCV2-associated enteritis. Thus, PCV2-associated enteritis could be a distinct clinical manifestation of PCV2.  相似文献
