Grasslands and agroecosystems occupy one-third of the terrestrial area, but their contribution to the global carbon cycle remains uncertain. We used a set of 316 site-years of CO2 exchange measurements to quantify gross primary productivity, respiration, and light-response parameters of grasslands, shrublands/savanna, wetlands, and cropland ecosystems worldwide. We analyzed data from 72 global flux-tower sites partitioned into gross photosynthesis and ecosystem respiration with the use of the light-response method (Gilmanov, T. G., D. A. Johnson, and N. Z. Saliendra. 2003. Growing season CO2 fluxes in a sagebrush-steppe ecosystem in Idaho: Bowen ratio/energy balance measurements and modeling. Basic and Applied Ecology 4:167–183) from the RANGEFLUX and WORLDGRASSAGRIFLUX data sets supplemented by 46 sites from the FLUXNET La Thuile data set partitioned with the use of the temperature-response method (Reichstein, M., E. Maximum values of the quantum yield (α=75 mmol · mol?1), photosynthetic capacity (Amax=3.4 mg CO2 · m?2 · s?1), gross photosynthesis (Pg,max=116 g CO2 · m?2 · d?1), and ecological light-use efficiency (εecol=59 mmol · mol?1) of managed grasslands and high-production croplands exceeded those of most forest ecosystems, indicating the potential of nonforest ecosystems for uptake of atmospheric CO2. Maximum values of gross primary production (8 600 g CO2 · m?2 · yr?1), total ecosystem respiration (7 900 g CO2 · m?2 · yr?1), and net CO2 exchange (2 400 g CO2 · m?2 · yr?1) were observed for intensively managed grasslands and high-yield crops, and are comparable to or higher than those for forest ecosystems, excluding some tropical forests. On average, 80% of the nonforest sites were apparent sinks for atmospheric CO2, with mean net uptake of 700 g CO2 · m?2 · yr?1 for intensive grasslands and 933 g CO2 · m?2 · d?1 for croplands. However, part of these apparent sinks is accumulated in crops and forage, which are carbon pools that are harvested, transported, and decomposed off site. 为了建立敏感、稳定、特异的PCR检测体系,用于实验动物弓形虫感染的检测。采用B1基因设计引物,建立常规PCR,用P30基因设计引物,建立巢式PCR;用两种PCR方法检测实验感染弓形虫小鼠血液和腹腔波中的DNA动态变化;用巢式PCR检测自然状态下的普通级豚鼠、教学和科研用兔的弓形虫感染率,并和常规PCR检到结果比较。结果,巢式PCR可检测到1fgDNA含量,比常规PCR敏感lOO倍;对其他微生物DNA无交叉现象,特异性强;对同一样品重复检测3次,阴、阳性结果一致,稳定性好。小鼠感染弓形虫2d后,巢式PCR对小民腹腔液的阳性检出率为83.3%,对血液的阳性检出率为33.3%;感染3d后,腹腔液阳性检出率为100%;而常规PCR在小鼠感染3d和4d后才能在腹腔液和血液中检测到,检出率各为16.7%。受检普通级豚鼠没有感染弓形虫,教学和科研用兔的弓形虫总感染率为14.3%。结论认为,巢式PCR方法可用于实验动物弓形虫早期感染的检测,具有敏感性高、特务性强、稳定性好的特点。 相似文献

为了探索特异性免疫学诊断方法对鸡骨型白血病(OP)的检测效果。用双盲法分批采样同时进行羽髓琼脂扩散试验和胫骨X线摄片检查对比,共检834只鸡(433羽属天然自发OP鸡群,401羽属人工诱发实验鸡群)。结果羽髓琼扩试验的OP阳性检出率为3.48%(29/834:)。阳性病理符合率为75.86%(22/29)。阳性误诊(假阳性)率24.13%(7/29),阴性误诊(假阴性)率36.15%(291/805)。而X线摄片检查的OP阳性检出率为38.37%(320/834)。X线阳性完全可获病理组织学证实,不存在假阳性或假阴性。把禽白血病羽髓琼扩试验试用于检测鸡骨型白血病尚属初次报道,其阳性检出率低而误诊率高,尚不宜作OP检测手段。放射摄片诊断的阳性检出率高而误诊率低,可快速诊断,故适用于OP检疫。 相似文献

本文以EAV(禽内源性反转录病毒片段 )为探针 ,EcoRI为限制性内切酶 ,对萧山鸡、SR92A系及其杂交后代进行了DNA指纹图谱的研究。结果表明 :EAV探针适合于鸡的DNA指纹分析 ,能产生清晰可辩的图谱。平均条带总数为11.384± 0 .84 4 ,两品种间、公母间条带总数均无显著差异。萧山鸡和SR92A系品种内相似系数分别为 0 .5 81、0 .5 11,高于品种间相似系数 (0 .2 31)。杂交后代相似系数为 0 .30 1。 相似文献

The objective of this experiment was to evaluate the effects of replacement of whole‐plant corn with oat and common vetch on the fermentation quality, chemical composition and aerobic stability of total mixed ration (TMR) silage in Tibet. Four TMR that varied in the forage sources on dry matter basis were used: (i) 52% whole‐plant corn (Control); (ii) 43% oat + 12% common vetch (OC3.6); (iii) 38% oat + 18% common vetch (OC2.2); and (iv) 33% oat + 23% common vetch (OC1.5). Silos were opened on day 45 and then subjected to an aerobic stability test for 12 days. The results showed that all silages were well preserved with low pH and NH3‐N, and high lactic acid and V‐scores. With the increasing proportion of common vetch, crude protein, ether extract increased (P < 0.05), and neutral detergent fiber and acid detergent fiber decreased (P < 0.05). Under aerobic conditions, treated silages were more stable than the control silage as indicated by lower (P < 0.05) pH and yeast population. It was concluded that replacement of whole‐plant corn with oat and common vetch had no unfavorable effects on the fermentation quality and improved crude protein content and aerobic stability of TMR silage.
