This study had 2 objectives: 1) to determine the involvement of Mycoplasma hyopneumoniae in respiratory outbreaks in herds of pigs, with the use of a nested polymerase chain reaction (nPCR) and an enzyme-linked immunosorbent assay (ELISA); and 2) to determine if the dynamics of M. hyopneumoniae infection differ between 3-site versus 1- or 2-site production systems (in which at least farrowing/gestation and nursery pigs are on the same site). Animals of different ages from 12 Spanish farms with respiratory problems were randomly sampled. Blood samples and nasal swabs were collected in a single farm visit, and ELISA and nPCR tests, respectively, were performed. All the farms demonstrated M. hyopneumoniae. According to the proportions of infected animals and the appearance of clinical signs in the different age groups, the farms were divided into 2 groups: farms in which M. hyopneumoniae probably played an important role in the observed respiratory outbreak and farms in which M. hyopneumoniae was not the main agent involved in the outbreak. Although seroconversion occurred in most herds in the finishing units, the number of seropositive pigs in the first group of farms was greater than the number in the second group. Statistically significant differences (P < 0.0001) between farms with a 1- or 2-site production system versus those with a 3-site production system were detected in nPCR results but not in rates of seroconversion. The farm effect also had a great influence on both controlled parameters: the pathogen’s DNA and antibody detection. Thus, although M. hyopneumoniae was present in all the studied farms, there were significant differences in the infection dynamics and clinical implications according to the type of production system, and M. hyopneumoniae colonization and seroconversion were greatly influenced by the effect of the individual farm.  相似文献

Euphorbia heterophylla populations endued with resistance to both imazethapyr (an acetolactate synthase inhibitor) and fomesafen (a protoporphyrinogen oxidase inhibitor) were first detected in Brazil in 2004. The objectives of this study were to determine the number and dominance of genes conferring imazethapyr and fomesafen resistance in E. heterophylla and to establish the types of inheritance of the resistance traits. It was hypothesised that two dominant nuclear genes control this herbicide resistance and each gene confers resistance to a single herbicide. We crossed a susceptible (S) and a multiple-resistant (R) E. heterophylla biotype and generated F1 families which, in turn, were self-fertilised to produce F2 families. Backcrosses of the F1 families with the S- and R parental biotypes resulted in BCs and BCr families respectively. The F1, F2, BCs and BCr families and the S and R parental biotypes were subjected to imazethapyr and fomesafen wherein each herbicide was used as a selecting agent. F1 plants and the S- and R parental biotypes were used to evaluate the dominance of the resistance trait in multiple-dose bioassays with imazethapyr and fomesafen. The observed frequencies of resistant plants in the F1, F2, BCs and BCr families did not significantly differ from the expected frequencies for a resistance trait regulated by two dominant genes in which each gene confers resistance to a single herbicide. The lack of difference in herbicide response between the F1 family and the R parental biotype was taken as evidence for completely dominant resistance to imazethapyr and fomesafen. Herbicide resistance in E. heterophylla presents with dominant monogenic nuclear inheritance for each herbicide mode of action. Our findings underscore the necessity and urgency of adopting integrated strategies to control E. heterophylla and to inhibit the evolution of new herbicide-resistant strains.  相似文献

Unravelling the effect of climate variability on species biology has been one of the main goals of ecological studies. Environmental factors such as river discharge and temperature have being proposed as triggers of reproductive cycle in fish. In temperate climates, fish reproduction is affected mainly by temperature, while is influenced by flood pulses in large tropical rivers. We evaluated the influence of temperature and river discharge as triggers of Iheringichthys labrosus’ reproduction. We studied the following reproductive variables: gonadosomatic index (GSI), hepatosomatic index (HSI) and condition factor (K). Females of I. labrosus were examined in a time series of ten years along three sites located in Lower Uruguay River at the beginning (last week of November-first week of December, spring) and by the end of the reproductive period (middle-end of April, autumn). Generalised linear models detected a positive effect of mean winter temperature and site on the spring mature female GSI. Site was significant in addition to the interaction between temperature and site. Moreover, the effect of winter temperature was stronger than water discharge in triggering reproductive timing of I. labrosus, contrary to the model proposed for other species in tropical systems. Additionally, a literature review suggested a latitudinal gradient on reproduction, occurring earlier and prolonged with decreasing latitude (and increasing temperature). In this study, we stated the effect of temperature on the life history in a Neotropical fish using a multifaceted approach, particularly important in the current context of global climate warming.  相似文献

We analyzed the tolerance of Copaifera lucens seeds to sub- mersion in water to assess the use of this species for direct seeding in riparian forest restoration programs. Seeds were submerged in water for 2, 4, 8, 16 and 32 days or not submerged (control = 0 days of submer- gence). For the control and at the end of each period of submersion, germination and seedling vigor tests were carried out. For germination tests, seeds were sown in plastic pots containing sand and kept in labora- tory conditions. The percentage of seed germination, the germination rate and the average germination time were analyzed. For seedlings, total biomass, leaf area, leaf mass per area and leaf area ratio were analyzed. Submersion time drastically affected the dissolved oxygen content and seed germination. Between 4 and 8 days of submersion there was a de- crease from 83.8% to 15.6% in the germination percentage. No seed germination occurred after 16 days of submersion. Although there was a significant decrease in the percentage of seed germination between 4 and 8 days of submersion, seedling vigor was not affected. Seeds of this species were partially tolerant to submersion in water, suggesting that C. lucens is a promising species for direct seeding in riparian forest restora- tion projects.  相似文献

RNA interference (RNAi) of target genes is triggered by double-stranded RNAs (dsRNAs) processed by conserved nucleases and accessory factors. To identify the genetic components required for RNAi, we performed a genome-wide screen using an engineered RNAi sensor strain of Caenorhabditis elegans. The RNAi screen identified 90 genes. These included Piwi/PAZ proteins, DEAH helicases, RNA binding/processing factors, chromatin-associated factors, DNA recombination proteins, nuclear import/export factors, and 11 known components of the RNAi machinery. We demonstrate that some of these genes are also required for germline and somatic transgene silencing. Moreover, the physical interactions among these potential RNAi factors suggest links to other RNA-dependent gene regulatory pathways.  相似文献

Current yeast interactome network maps contain several hundred molecular complexes with limited and somewhat controversial representation of direct binary interactions. We carried out a comparative quality assessment of current yeast interactome data sets, demonstrating that high-throughput yeast two-hybrid (Y2H) screening provides high-quality binary interaction information. Because a large fraction of the yeast binary interactome remains to be mapped, we developed an empirically controlled mapping framework to produce a "second-generation" high-quality, high-throughput Y2H data set covering approximately 20% of all yeast binary interactions. Both Y2H and affinity purification followed by mass spectrometry (AP/MS) data are of equally high quality but of a fundamentally different and complementary nature, resulting in networks with different topological and biological properties. Compared to co-complex interactome models, this binary map is enriched for transient signaling interactions and intercomplex connections with a highly significant clustering between essential proteins. Rather than correlating with essentiality, protein connectivity correlates with genetic pleiotropy.  相似文献

ABSTRACT

Crop genotypes that make best use of potassium (K) fertilizers can promote agricultural sustainability. However, screening germplasm collections for responses to K fertilizers is often laborious and expensive. To reduce costs, the number of K fertilizer treatments required to identify better genotypes should be minimized. This might be achieved by exploiting the mathematical relationships between biomass, plant K content, and K supply. This study employed 14 barley (Hordeum vulgare L.) genotypes growing in a hydroponics system that allowed the K supply to roots to be controlled through the K concentration in a flowing solution. It sought to determine the minimal number of treatments required to model the relationships between (a) shoot biomass and K supply, (b) plant K content and K supply, and (c) shoot biomass and plant K content. The relationships between (a) shoot biomass and K supply and (b) plant K content and K supply for any given genotype could be fitted by Michaelis–Menten equations and each of these could be estimated from data obtained at two, appropriately-chosen, rates of K supply. The relationship between shoot biomass and plant K content could be estimated from these relationships. However, the optimum K supply required for accurate estimates differed between genotypes and whether shoot biomass or plant K content was to be estimated. It is, therefore, suggested that the relationships between shoot biomass, plant K content, and K supply might best be determined from measurements of biomass and K content at three, carefully-selected, rates of K supply.  相似文献

Bleached Kraft mill effluent was treated in an activated sludgereactor followed by an allophanic soil adsorption system (ASAS). Under aerobic conditions, removal efficiencies of biological oxygen demand (BOD5) and chemical oxygen demand (COD) varied between 57.7–96.5% and 30.3–57.0%, respectively, depending on the hydraulic retention time (HRT). On the other hand, tannin-lignin and phenolic compounds removal efficiencies attained values between 13.2–51.2 and 3.6–33.5%,respectively. An allophanic soil adsorption system was designed for color and phenolic compounds removal. Three different types of soils were used: Natural allophanic soil as the control compared, with calcinated and acidified allophanic soil. The initial removal efficiencies for phenolic compounds varied between 72 an 87% for activated soils, while color initial removal efficiencies were between 95 and 99%. Moreover, COD and tannin-lignin initial removal efficiencies reached maximum values of 74 and 87%, respectively, for calcinated soil. Design parameters show that there is an enhancement factor in adsorption capacities for both activated soils. In fact, phenolic compounds breakpoint adsorption capacity increased 5.3 times for calcinated soil and 17.6 times for acidified soil, while saturation capacity increased between 2.2 and 3.2 times. In addition, color breakpoint adsorption capacity increased 2.8 times for calcinated soil and 10.4 times for acidified soil, while saturation capacity increased between 3.2 and 5.5 times.  相似文献
