Gray mold, caused by Botrytis cinerea, is the main postharvest decay of table grapes. It can develop in the vineyard and spread rapidly among berries after harvest, during long distant transport, cold storage and shelf-life. In conventional agriculture, bunches are sprayed with fungicides after flowering, at pre-bunch closure, at veraison, and later, depending on the time of harvest. Harvested bunches are usually stored in the presence of sulfur dioxide. However, the use of synthetic fungicides and of sulfur dioxide is not allowed on organic grapes and the study of alternative methods to control postharvest decay has developed over several decades, along with the demand for safer storage methods. This review summarizes the results published in the field within the last 5 years (2006-2010). We can group these approaches as follows: (i) biocontrol agents; (ii) natural antimicrobials; (iii) GRAS type decontaminating agents; and (iv) physical means. Two biocontrol agents, Muscodor albus and Hanseniaspora uvarum, have shown equal or better effectiveness than conventional methods to control gray mold of table grapes in laboratory scale experiments. Currently, the bottleneck for the commercial use of biocontrol agents is that the registration process is comparable to that of fungicides, with similar costs but often with a narrower market. This delays their transition from experimental to practical use. Natural antimicrobials, such as salts, chitosan, and plant extracts, have demonstrated good results and often have been applied in various scales. Several GRAS-classified sanitizers have been tested to extend postharvest storage of table grapes, including acetic acid, electrolyzed oxidizing water, ozone, and ethanol. Physical technologies involving variations in temperature, UV-C irradiation, pressure or changing atmospheric composition, are all postharvest practices which require significant adaptation by an industry which is accustomed to minimal intervention during harvest. Overall, the use of ozone and of calcium chloride are two promising examples of treatments that are beginning to be adopted on a commercial scale. The requirements for the optimal treatment of grapes against gray mold before harvest or during storage are summarized.  相似文献

The eight vitamers of vitamin E (alpha-, beta-, gamma-, and delta-tocopherols and -tocotrienols) have different antioxidant and biological activities and have different distributions in foods. Some cereals, especially oat, rye, and barley, are good sources of tocotrienols. A fast procedure for the determination of tocopherols and tocotrienols (tocols) in cereal foods was developed. It involves sample saponification and extraction followed by normal phase high-performance liquid chromatography (HPLC). The results have been compared with those found by direct extraction without saponification. The method is sensitive and selective enough to be tested on a wide variety of cereal samples. The highest tocol levels were found in soft wheat and barley ( approximately 75 mg/kg of dry weight). beta-Tocotrienol is the main vitamer found in hulled and dehulled wheats (from 33 to 43 mg/kg of dry weight), gamma-tocopherol predominates in maize (45 mg/kg of dry weight) ), and alpha-tocotrienol predominates in oat and barley (56 and 40 mg/kg of dry weight, respectively).  相似文献

Although Bois noir is one of the main phytoplasma diseases of grapevine, the gene expression and enzyme activities that underlie physiological changes occurring in symptomatic and recovered (with spontaneous or induced symptom remission) plants are mostly unknown. Bois noir symptomatic leaves (September 2006, 2007) and symptomless leaves from infected symptomatic plants (September 2007) of Sangiovese (moderately susceptible) and Chardonnay (highly susceptible) cultivars were collected. Moreover, leaves from infected symptomless plants of both cultivars were harvested in June 2007. Leaves from recovered plants were also collected in the same periods. In recovered plants of both cultivars, class III chitinase and almost every time phenylalanine ammonia-lyase and chalcone synthase expression were increased for all collection periods. In symptomatic leaves of both cultivars, the expressions of the same genes were up-regulated and also those of β-1,3-glucanase and flavanone 3-hydroxylase. The activities of chitinase, phenylalanine ammonia-lyase, β-1,3-glucanase, and superoxide dismutase generally correlated with gene expression. For the moderately susceptible Sangiovese, the defense genes were generally up-regulated in both symptomatic and symptomless leaves (for all collection periods). This behavior was not observed in the highly susceptible Chardonnay, in which changes in gene expression were linked to evident symptom display. Therefore, the physiological response of the plants to this pathogen infection appear to be the reason for the resistance of the cultivar to the disease.  相似文献

The ammonia flux from a green manured field was measured at a site in southern Italy. The green manure was provided by ploughing in about 66 and 78 t ha−1 of broad beans at the flowering stage during the spring in 1995 and 1996. The NH3 flux was estimated by means of the micrometeorological simplified aerodynamic method with the ammonia concentration measured at two levels above soil surface. Ammonia concentration was measured over a 2–3 h sampling period by trapping air NH3 in a H2SO4 solution and analysing this sample in the laboratory with the colorimetric method using the Nessler solution. About 70% of the total amount of ammonia was emitted in the first two days immediately after manuring. The NH3 flux was regulated by the same mechanism as the water vapour transport. Seventy percent of the daily ammonia was emitted around noon. The ammonia emission stopped during rainy and cold days and started again as soon as solar radiation, soil and air temperature increased, following plant decomposition. The total amount of ammonia emitted in the experimental period (28 days in 1995 and 18 days in 1996) was about 13 kg ha−1 for both years.  相似文献

Maize is used as an alternative to wheat to elaborate foodstuffs for celiac patients in a gluten-free diet. However, some maize prolamins (zeins) contain amino acid sequences that resemble the wheat gluten immunodominant peptides and their integrity after gastrointestinal proteolysis is unknown. In this study, the celiac IgA-immunoreactivity to zeins from raw or nixtamalized grains, before and after peptic/tryptic digestion was evaluated and their possible immunogenicity was investigated by in silico methods. IgA from some celiac patients with HLA-DQ2 or DQ8 haplotypes recognized two alpha-zeins even after peptic/tryptic proteolysis. However, digestion affected zeins after denaturation, reduction, and alkylation, used for identification of prolamins as alpha-zein A20 and A30 by MS/MS sequencing. An in silico analysis indicated that other zeins contain similar sequences, or sequences that may bind even better to the HLA-DQ2/DQ8 molecules compared to the already identified ones. Results concur to indicate that relative abundance of these zeins, along with factors affecting their resistance to proteolysis, may be of paramount clinical relevance, and the use of maize in the formulation and preparation of gluten-free foods must be reevaluated in some cases of celiac disease.  相似文献

Vegetable crops are frequently infected by fungal pathogens, which can include seedborne fungi. In such cases, the pathogen is already present within or on the seed surface, and can thus cause seed rot and seedling damping‐off. Treatment of vegetable seeds has been shown to prevent plant disease epidemics caused by seedborne fungal pathogens. Furthermore, seed treatments can be useful in reducing the amounts of pesticides required to manage a disease, because effective seed treatments can eliminate the need for foliar application of fungicides later in the season. Although the application of fungicides is almost always effective, their non‐target environmental impact and the development of pathogen resistance have led to the search for alternative methods, especially in the past few years. Physical treatments that have already been used in the past and treatments with biopesticides, such as plant extracts, natural compounds and biocontrol agents, have proved to be effective in controlling seedborne pathogens. These have been applied alone or in combination, and they are widely used owing to their broad spectrum in terms of disease control and production yield. In this review, the effectiveness of different seed treatments against the main seedborne pathogens of some important vegetable crops is critically discussed. © 2013 Society of Chemical Industry  相似文献

The official methods for the quantification of aflatoxin M1 in dairy products (cheese and yogurt) include extraction into dichloromethane or chloroform, evaporation of the solvent, partitioning of the reconstituted residue with hexane, and subsequent analysis. To secure a rapid and inexpensive screen for aflatoxin M1 contamination, a sensitive competitive ELISA, using a rabbit polyclonal antibody, was developed for measuring aflatoxin M1 in milk and used in a comparative study for measuring the extraction efficiency of aflatoxin M1 in aqueous or organic solvent buffers using yogurt samples. An aqueous sodium citrate solution was found to be suitable for extracting aflatoxin M1, thus eliminating the need for organic solvents. The citrate extraction proved to be efficient (recovery ranged from 70 to 124%) in fortified samples of very different kinds of dairy products, including yogurt and six types of cheese. Fourteen yogurt and cheese samples were extracted with citrate solution and analyzed by ELISA. A good correlation was observed (y=0.95x-0.59, r2=0.98) when the data were compared with those obtained through the official method, across a wide range of aflatoxin M1 contaminations (10-200 ng/kg).  相似文献

BACKGROUND: Emamectin benzoate is a novel macrocyclic lactone insecticide derived from naturally occurring avermectin molecules isolated by fermentation from the soil microorganism Streptomyces avermitilis Kim & Goodfellow. The present study aims to evaluate the toxicity of emamectin benzoate to codling moth, Cydia pomonella (L.), and oriental fruit moth, C. molesta (Busck), under laboratory and semi‐field conditions. RESULTS: Dose response bioassays showed that emamectin benzoate had a high level of intrinsic toxicity to early‐stage larvae of both species, and that contact activity might contribute significantly to mortality. In the semi‐field trials, residual toxicity lasted for more than 1 week. Ovicidal activity was recorded only for C. pomonella (approximately 30%), irrespective of the concentrations tested. Field trials confirmed the efficacy of emamectin benzoate on codling moth when applied at 7 day intervals. Fruit damage, both from the first and second generations, was comparable with that on treatment with chlorpyrifos‐ethyl, used as a chemical reference. CONCLUSION: Emamectin benzoate may be considered a valuable tool for the control of codling moth as a component of an IPM programme. Its collective advantages are: high efficacy, lack of cross‐resistance with currently used products, control of secondary pests such as oriental fruit moth and selective toxicity that spares beneficials.
