Arrhythmias and transient changes in cardiac function after topical administration of one drop of phenylephrine 10% in an adult cat undergoing conjunctival graft

HistoryA 2-year-old, entire female, Somali cat weighing 3.8 kg was admitted for a conjunctival graft on the right eye, for treatment of an acute descemetocele. Medetomidine 4.2 μg kg^?1 and methadone 0.2 mg kg^?1 were administered by intramuscular injection as preanaesthetic medication. Anaesthesia was induced using diazepam 0.26 mg kg^?1 and propofol 4 mg kg^?1 administered by intravenous (IV) injection. Following endotracheal intubation, anaesthesia was maintained with isoflurane delivered in oxygen (1 L minute^?1) and nitrous oxide (2 L minute^?1) via a non-rebreathing system. Twenty minutes after induction of anaesthesia, one drop of a 10% phenylephrine hydrochloride solution was administered topically to the right eye.Physical examinationAfter phenylephrine administration, a decrease in heart rate (from 95 to 80 beats minute^?1) and an increase in arterial blood pressure occurred. The pulse then became difficult to palpate manually and multifocal ventricular premature contractions were observed on the electrocardiogram.ManagementNitrous oxide was discontinued and the isoflurane vaporizer setting was decreased from 1.5% to 0.5%. Lidocaine 1 mg kg^?1 IV was administered, this resulted in ventricular bigeminy. The quality of the femoral pulse improved and was regular in rhythm and character. Surgery was completed as fast as possible. The bigeminy progressively disappeared and before disconnecting the cat from the breathing system, there was a normal sinus rhythm with a heart rate of 85 beats minute^?1.Follow-upEchocardiography was performed during recovery and showed mitral and aortic valve insufficiency and dilation of the left ventricle, suggesting a reduction in systolic function. Echocardiography was repeated the following day and was normal.ConclusionsIn order to diminish the potential for cardiovascular sequelae associated with systemic absorption of ocular phenylephrine, less concentrated solutions, smaller drop size or different instillation techniques should be considered for topical use in small patients.     

Multinucleated giant cells with an osteoclast phenotype derived from caprine peripheral blood mononuclear cells

Formation of multinucleated giant cells (MGCs) by macrophage fusion is a typical cytopathic effect of lentiviral replication in caprine monocytes and MGC formation from cultured caprine peripheral blood mononuclear cells (PBMCs) has been considered to be diagnostic for small ruminant lentivirus (SRLV) infection. In this study, formation of MGCs was observed after 7–14 days when PBMCs were cultured from healthy goats free from SRLV infection. These MGCs expressed tartrate-resistant acid phosphatase, calcitonin receptor, integrin αVβ3, cathepsin K and matrix metalloproteinase 9 and were able to resorb bone in vitro in the absence of RANKL and macrophage colony stimulating factor, consistent with an osteoclast phenotype.     

Effect of selection under cultivation on morphological traits and yield of Ladino white clover landraces

One reason for the relatively low genetic advance realized in forage legumes may be the possible selection, under the severe intraspecific competition occurring naturally during cultivation, of a few high-yielding genotypes that would be of similar value in a broadly-based landrace or old cultivar and a narrowly-based, modern variety. The effect of selection under cultivation was assessed in two Ladino white clover farm landraces. Genotypes collected from each farm after a three-year crop cycle (survivor populations), or obtained from the seed used to establish each meadow (original populations), were multiplied clonally and evaluated in isolation and monoculture conditions for one year. The occurrence of directional selection was observed only in the landrace featured by greater intra-population variation in favour of genotypes with longer petioles (+11%), larger leaf size (+13%), less stolons (–14%) and lower production of heads (–16%). Selection for the first two traits would imply an increase of competitive ability, whilst that for the fourth trait appeared mainly as a correlated response. The trade-off between leaf gigantism and head production could act as a buffering mechanism against rapid change in genotype frequencies following random mating. A slight trend towards directional selection for higher yield (+6%) in the same landrace was not statistically significant. No disruptive or stabilizing selection was observed for any trait in either landrace.     

A modified nitrogen oxide-ozone photolytic cycle

Nitrogen Oxide-0₃ photolytic cycle is dependent upon the presence of NO in ambient air. From our air monitoring observations we have found that the NO concentration in the atmosphere is often zero (0 to 5 ppb). This suggests a need to modify/change the photolytic cycle. Linear regression analyses on the O₃ and NO₂ concentrations in ambient air, measured in real time were carried out. The predicted values of O₃ and NO₂ from these regression equations were close to the observed values. On the basis of these two observations a modified photolytic cycle, including some other components (such as HONO₂) is discussed.     

Determination of high molecular mass polycyclic aromatic hydrocarbons in a typical Italian smoked cheese by HPLC-FL

High-performance liquid chromatography with fluorescence detection was used for the analysis of polycyclic aromatic hydrocarbons (PAHs), in "Diavoletto" smoked cheese. Such cheese is typically produced in the Sorrento peninsula, and it is smoked commonly with different materials of vegetable origin. The importance of the smoking generation material is proven by the attention that the EU is paying in indicating the list of wood that may be used to produce smoking flavor agents. The PAHs considered are classified as "probable human carcinogens" by the U.S. Environmental Protection Agency for sufficient data from animal bioassays. The smoked samples contained high molecular mass PAHs with different levels ranging from 0.12 to 6.21 microg/kg. The determination was carried out also on liquid smoking flavor agents, smoke-flavored cheese, and nonsmoked cheese to measure the level of contamination before the treatment.     

Behavior of a fenhexamid photoproduct during the alcoholic fermentation of Saccharomyces cerevisiae

The fungicide fenhexamid [N-(2,3-dichloro-4-hydroxyphenyl)-1-methylcyclohexanecarboxamide] degraded rapidly by UV or sunlight irradiation, yielding 7-chloro-6-hydroxy-2-(1-methylcyclohexyl)-1,3-benzoxazole (CHB) as a main photoproduct. CHB was isolated, and its effect on alcoholic fermentation of Saccharomyces cerevisiae was studied. The results indicate that the presence of CHB does not affect the extent of alcohol production. After 12 days, the amount of CHB in the fermentation medium decreased by ca. 65%. Only 25% of the missing CHB was recovered unchanged from yeasts, most likely because it was adsorbed on the yeast wall cell. The remaining part degraded during the fermentation process. Glucan and chitin, two potential adsorbents, which constitute yeast cell walls, exhibited affinity for CHB.     

Isotope ratios of lead in Italian wines by inductively coupled plasma mass spectrometry

Lead and its isotope ratios (IRs) in 83 Italian wines from 42 different administrative provinces produced on laboratory scale in 2000 were measured by inductively coupled plasma mass spectrometry. Lead had a median of 19.0 microg/L, ranging from 10.0 up to 149 microg/L and was higher in the samples from northwestern Italy. The values of the IRs were (mean +/- standard deviation) 1.171 +/- 0.011 for (206)Pb/(207)Pb, 2.071 +/- 0.017 for (208)Pb/(206)Pb, 2.425 +/- 0.014 for (208)Pb/(207)Pb, 17.84 +/- 0.32 for (206)Pb/(204)Pb, 15.24 +/- 0.31 for (207)Pb/(204)Pb, and 37.31 +/- 0.52 for (208)Pb/(204)Pb. These ratios agree with those in the literature for several European regions. (206)Pb/(207)Pb distinguished the northwestern from the northeastern and southern Italian wines, and the southern samples were distinguishable only from the northwestern ones. (208)Pb/(206)Pb distinguished the northwestern from the southern Italian samples. Nevertheless, the lead IRs do not seem to be a very effective tool for the origin authentication of wines from different Italian regions.     

Phenolic constituents and antioxidant properties of Xanthosoma violaceum leaves

An extract of Xanthosoma violaceum leaves was subjected to a polyphenol profile determination, including total polyphenols, and antioxidant activity evaluation. Analysis of the extract resulted in the isolation of a new flavone C-glycoside, apigenin 6-C-beta-D-glucopyranosyl-8-C-beta-D-apiofuranoside (1), as well as known flavone C-glycosides, including vitexin (2), isovitexin (3), isovitexin 4'-O-rhamnopyranoside (4), apigenin 6-C-[beta-D-glucopyranosyl-(1-->6)-beta-D-glucopyranoside] (5), and apigenin 6,8-diC-beta-D-glucopyranoside (6). The antioxidant activity of the extract was assessed by means of two different in vitro tests: bleaching of the stable 1,1-diphenyl-2-picrylhydrazyl radical (DPPH test) and peroxidation induced by the water-soluble radical initiator 2,2'-azobis(2-amidinopropane) hydrochloride, on mixed dipalmitoylphosphatidylcholine/linoleic acid unilamellar vesicles (LP-LUV test). In both tests used, the extract and a fraction II showed a significant antioxidant/free-radical scavenging effect (fraction II, EC(50) = 11.6 microg/mL) in comparison to alpha-tocopherol (EC(50) = 10.1 microg/mL).
