Influence of watercontent and salts on the nitrification in samples of a Dystric Cambisol Samples of a Dystric Cambisol from a beech site produced nitrate but autotrophic nitrifying microorganisms could not be detected. Net nitrification of the humic layer and the upper 5 cm of the mineral soil during incubation at 22°C was investigated. Nitrification rate increased with increasing water content of the soil. Additions of ammonium or peptone did not increase the nitrification in the humic layer. Supply of (ammonium-)sulphates and chlorides with concentrations higher than 2 mMol per kg soil inhibited nitrification totally. This could not be ascribed to pH-changes. Additions of phosphates, lime or alkali to the soil samples increased nitrification.  相似文献

The development of C-distribution on functional groups in soil organic matter (SOM) was investigated in whole soil samples and organomineral particle-size fractions from the 34-year-old pot experiment Hu3-Loamy marl at Rostock. CPMAS-13C-NMR spectra of grass roots and whole soil samples indicated the enrichment of carboxyl-C, aliphatic C and aromatic C in SOM, Only minor changes were observed in the SOM-C distribution on functional groups between 13th and 34th experimental year. The investigation of organomineral clay and silt-size fractions showed a specific retention of aliphatics in clay. The most obvious changes in organic matter composition in size-fractions were the increase of carbohydrate-C and decrease of aromatic C.  相似文献

To assess the effects of Fusarium infection on the polysaccharides of winter wheat grain (Triticum aestivum L.), grain samples obtained from plants artificially inoculated with Fusarium culmorum were analyzed. Microscopy revealed obvious damage to the starch granules in the seriously infected samples. The Fusarium infection had no analytically detectable influence on the starch and total insoluble dietary fiber content of the wheat grain. There were significantly positive relationships between alpha-amylase activity, cellulase activity, total soluble dietary fiber content, pentosan content, and degree of infection quantified by an enzyme-linked immunosorbant assay, which would indicate the importance of fungal enzymes. A distinct higher Hagberg falling number (FN) was determined in the seriously infected samples, while the viscosity and sucrose content of the flour decreased. However, the addition of a liquid medium contaminated with F. culmorum led to a significant decrease in the FN. Depending on the type of buffer used, the alpha-amylase of F. culmorum demonstrated its maximum activity between pH 5.5 and pH 7.0 at 30-50 degrees C. Remarkably, this fungal alpha-amylase showed a thermostable characteristic and was active over a wide range of temperatures, from 10 to 100 degrees C. This type of thermostability suggests that the alpha-amylase of F. culmorum may damage starch granules throughout the processing of wheat flour, thereby inducing weak dough properties and unsatisfactory bread quality.  相似文献

Translocation and dispersion of chloride, bromide, nitrate and sulfate tracers in two typical Fluvi-Eutric Cambisols Chloride, bromide, nitrate and sulfate were applied as tracers on two weakly to moderately acidic Fluvi-Eutric Cambisols. The soil solution was sampled by ceramic suction cups down to a soil depth of 180 cm. At definite time intervals also soils material was taken from different depth to produce the soil saturation extract. The concentrations of the non-adsorbed anions chloride, bromide and nitrate were very similar in the suction cup solutions and in the saturation extracts, whereas the concentrations of the weakly adsorbed sulfate partially showed considerable differences. Obviously both methods extract in acid soils different fractions of the dissolved sulfate. By comparison of marked sulfate peaks with those of chloride, bromide or nitrate, which occur at the same time in different soil depth, a Kd value of 0.16 1/kg can be calculated for the Ap horizon and of 0.02 1/kg for the subsurface horizons of one Fluvi-Eutric Cambisol. Values of the dispersivity of the different horizons were estimated by varying the values of the dispersivity reported in the literature until the best fit of the simulated anion depth distribution to the measured values was found. The estimated dispersivities varied between 1.0 cm in the Ap horizon and 1.8 and 10.0 cm in the subsurface horizons.  相似文献

In Asian medicine the fruit of the okra plant, Abelmoschus esculentus (L.) Moench., is used as a mucilaginous food additive against gastric irritative and inflammative diseases. To find a rational basis for its use against these diseases, several crude and purified carbohydrate-containing fractions from immature okra fruits were isolated and analyzed, and their effects against Helicobacter pylori in an in situ adhesion model on sections of human gastric mucosa were determined. Pretreatment of the bacteria with a fresh juice preparation inhibited the bacterial adhesion almost completely. Lyophilization and reconstitution of an extract solution led to a reduction of this effect. A crude polysaccharide (RPS) isolated from the fresh juice by ethanolic precipitation showed strong inhibitory effects. Further fractionation of RPS revealed a purified, highly acidic subfraction (AF III) with high antiadhesive qualities. Carbohydrate analysis revealed the presence of rhamnogalacturonans with a considerable amount of glucuronic acid, whereas other inactive subfractions contained little glucuronic acid or were glucuronic acid-free. After heat denaturation of the fresh juice or protein precipitation with 5% TCA the antiadhesive activity of the fresh extract was reduced, indicating that besides polysaccharides, protein fractions also exhibited antiadhesive properties. SDS-PAGE analysis of the precipitate revealed several bands of glycosylated proteins between 25 and 37 kDa that were almost diminished in the nonactive supernatant. Preincubations of gastric tissue with any of the active fractions did not lead to reduced bacterial binding. The antiadhesive activity is therefore due to the blocking capacity of specific Helicobacter surface receptors that coordinate the interaction between host and bacterium. Neither of the active fractions showed inhibitory effects on bacterial growth in vitro. The antiadhesive qualities of okra were assumed to be due to a combination of glycoproteins and highly acidic sugar compounds making up a complex three-dimensional structure that is fully developed only in the fresh juice of the fruit.  相似文献

Nineteen Sesbania accessions were investigated, in a total of six species, to characterize the accessions based on morphological and agronomic data using multivariate methods, to identify a core set of morphological and agronomic attributes to be used in germplasm characterization of Sesbania spp., and to evaluate the forage potential of these accessions. Plots, in randomized block design, consisted of single 16 m-long rows of eight plants, spaced at 2 m between plants and 3 m between rows. Morphological data were obtained from the first four plants in each plot. The four remaining plants, submitted to four cuts at a height of 1 m, provided agronomic data. Principal component analysis indicated that variance accumulated by the first two components for morphological and agronomic data was 74.4% and 77.0%, respectively. Two large groups of accessions were formed in the cluster analysis for morphological data, one consisting of S. virgata, subgenus Daubentonia and the second group comprising the other five species. Cluster analysis using agronomic data classified higher yielding perennial species in three groups, and the annual species, which stood out principally in relation to seed production, in three other groups. Accession n° 8 of S. sesbanwas classified in an isolated group due to its forage superiority. A character discard resulted in the selection of eight characters: floral initiation, pod and leaf length, number of leaflet pairs, plant regrowth height, leaf dry matter yield, seed production and vigour, indicated as a core set of attributes for Sesbania germplasm characterization.  相似文献

Products made from Herba Santa (Eriodictyon californicum (H. & A.) Torr.) have been used as bitter remedies for some pharmaceutical applications for many years, but they are actually too aromatic to be useful for many food or pharmaceutical applications. In sensory studies flavanones homoeriodictyol (1), its sodium salt (1-Na), sterubin (2), and eriodictyol (4) could significantly decrease the bitter taste of caffeine without exhibiting intrinsic strong flavors or taste characteristics. Further investigations on 1-Na elicited a broad masking activity between 10 and 40% toward different chemical classes of bitter molecules (e.g. salicin, amarogentin, paracetamol, quinine) but not toward bitter linoleic acid emulsions. For caffeine and amarogentin, dose-response studies were performed; the masking activity toward bitter taste for both compounds reached a plateau at higher concentrations of 1-Na. Due to these facts, homoeriodictyol sodium salt (1-Na) seems to be a very interesting new taste modifier for food applications and pharmaceuticals.  相似文献
