To obtain structure-function information of a range of carbohydrates, which are available only in very small quantities, an in vitro fermentation method using 7 mg of carbohydrate, 0.7 mL of basal medium, and 1% (w/v) of fecal bacteria was validated against a pH-controlled batch culture with 150 mL of basal medium and 1.5 g of test carbohydrate. This method was used to determine the influence of different glycosidic linkages and monosaccharide compositions of disaccharides on the selectivity of microbial fermentation. A prebiotic index (PI) was calculated for each disaccharide. Generally, disaccharides with linkages of 1-2, 1-4, and 1-6 generated a high PI score, with kojibiose and sophorose showing the greatest values (21.62 and 18.63, respectively). Apart from 6alpha-mannobiose, mannose-containing disaccharides gave a low PI due to low numbers of bifidobacteria and lactobacilli and an increase in bacteroides. The structure-function information obtained in this study may lead to a predictive understanding of how specific structures are fermented by the human gut microflora.  相似文献

An analysis of the polar extracts from sweet potato Ipomoea batatas (Convolvulaceae) led to the isolation of seven unknown aminoacyl sugars. On the basis of 1D, 2D NMR, and mass spectrometry data, the structures of the compounds were elucidated as: beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-valyl]-glucopyranoside (1), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-tyrosyl]-glucopyranoside (2), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-threonyl]-glucopyranoside (3), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-hystidyl]-glucopyranoside (4), 2-beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-alanyl]-glucopyranoside (5), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-tryptophanyl]-glucopyranoside (6), and beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-glycyl]-glucopyranoside (7).  相似文献

The aim of this study was to determine if plastic mulch films may have positive effects on bell pepper plants grown under shade house conditions. The experimental design was split-plot, where large plots contained four plastic mulch films (black, aluminum, silver, and white) compared to bare soil and small plots contained two bell pepper cultivars. Plant shoot dry weight was higher in plastic mulch treatments than in bare soil. The net photosynthesis rate in the cultivar SWG-46 exceeded in net photosynthesis with respect to the cultivar SWG-42 in one of three sampling dates. Nutrients in leaf showed a similar behavior in plastic mulch treatments than the control but the total content of nitrogen (N), potassium (K), and sulfur (S) was increased. The total yield was higher in black plastic mulch than in the other plastic mulch colors and lowest in the control.  相似文献

This paper reports a fully automatic respirometric technique with low maintenance costs that allows the routine determination of the specific oxygen uptake rate (SOUR), as well as the cumulative oxygen demand at 20 h, (OD20). The procedure involves the measurement of the dissolved oxygen concentration in an aqueous suspension of compost that is supplied with various nutrients, none of which contains organic carbon. The proposed method was used to monitor composting in a compost pile consisting of 65% organic matter obtained by selective collection of municipal solid waste (MSW), 30% sewage sludge and 5% plant trimming residues. Based on the results, the maximum SOUR value reached, (SOURmax), decreases gradually with time, to values near 1 mgO2/gVS.h after 130 days; indicating that the final product is stable. Linear correlations between OD20 and the absorbances at 465 and 665 nm of aqueous extracts of compost, and its soluble chemical organic demand (CODsol), and soluble organic carbon (TOCsol) are significant at the p ≤ 0.01 level. However, the use of such variables instead of respirometric data requires prior experimentation with the type of waste to be composted in order to identify the absolute values at which biological activity ceases.  相似文献

Summary Fusarium solani f.sp. eumartii Carp. Snyder and Hansen (Fusarium eumartii) is a soil inhabitant that induces the so-called Potato Wilt and Stem End Rot disease. Prior to wilting, the pathogen induces peculiar small bronze spots on the leaflets. Failure to isolate F. eumartii from infected leaflets suggests the involvement of a toxin in the disease. The fungus was grown in liquid Richard's medium and thereafter a filtrate was obtained dialyzing (MW cutoff 12,000–14,000) and sterilizing the culture by filtration (0.22 m). Potato leaves treated with both the pathogen or the filtrate showed symptoms of bronze spots and significantly higher electrolyte leakage when compared to controls. Tomato leaves showed neither bronze spots nor electrolyte leakage after plant inoculation with the pathogen or with the filtrate treatment. Both, the absence of visible symptoms and the lack of electrolyte leakage in tomato could be associated to a certain degree of host specificity of the F. eumartii filtrate towards potato. The filtrate also induced symptoms similar to infections by F. eumartii in adult plants and in vitro plantlets of cultivars Huinkul MAG and Kennebec. Callus responses to the filtrate were related to responses of the cultivars to the pathogen in greenhouse. These results show the potential of the culture filtrate of F. eumartii for use in screening for wilting resistance.  相似文献

Summary The invasive potential of a species can be assessed by propagule pressure, which measures the chances for propagules of a species to find a suitable habitat for establishment and reproduction. Seeds, fruits, and vegetative structures that contribute to the propagule pressure are morphologically, physiologically and genetically different from one another, thus each kind should have a specific way of contributing to a successful invasion. In this paper we review plant traits that contribute to the propagule pressure. Seed production provides an estimate of the potential multiplication rate of the weed. However, it is gap-sensing mechanisms of seeds based on dormancy termination and germination requirements, which significantly contribute to the naturalization and invasion processes assuring a successful seedling establishment in environments of high competition. Dispersal of propagules reduces competition, mating with a sibling, and subsequent inbreeding depression, and increases colonization opportunities and range of expansion. Some of those benefits can be achieved in a population by existence of dormancy mechanisms and thus, the existence of a seed bank. Finally, vegetative propagation may ensure expansion of local populations when seedling establishment is low. Broadening the scope of traits that are considered in the breeding programs aimed at commercial production of plant propagules, to include those related to propagule pressure, is essential for adequate evaluation of invasive potential.  相似文献

The inheritance of 16 important agronomic traits and its relationship were studied for four years in a population of 167 almond [P. dulcis (Mill.) D.A. Webb] seedlings obtained from a cross between the French selection ‘R1000’ (‘Tardy Nonpareil’ × ‘Tuono’) and the Spanish cultivar ‘Desmayo Largueta’. For some traits (blooming and leafing date) descendants segregated between the value of both progenitors, meanwhile for others the mean of the offspring was lower (bloom density, productivity and ripening date) or higher (in-shell/kernel ratio and double kernels). As expected, kernel bitterness and self-incompatibility behaved as monogenic traits. Some important correlations between traits were detected. The implications of the transmission and the correlation of these traits in the breeding programmes are discussed.  相似文献
