Subclinical low-prevalent Aujeszky's disease (AD) serological test reactors test reactors are defined as those few swine within a qualified AD virus (ADV)-negative herd that have antibodies to wild type virus. However, clinical signs of the associated diease are not observed in these putatively infected swine or elsewhere in the herd. Twelve such animals, including 7 previously vaccinated with a genetically modified ADV, were identified in Illinois (USA) during a 2.5 year period. The humoral immune responses of the 5 nonvaccinated swine were assessed by an enhanced virus neutralization test and a radioimmunoassay. Anti-ADV antibodies were determined to be present in the serum from 4 of these swine. Attempts to isolate ADV by in vitro and in vivo inculations of cell cultures and weanling mice, respectively, of tonsillar and trigeminal nerve ganglionic tissue preparations from each animal (vaccinated and nonvaccinated) were unsuccessful. Tonsillar and trigeminal nerve ganglionic tissues of each animal were screened for the presecence of wild type and/or vaccine viral genomes by a soluble polymerase chain reaction (PCR) coupled with Southern hybridization. Unique PCR primers were used distinguish between wild type and vaccine viral DNAs. Additional PCR procedure, which amplifiers a portion of the essential and highly conserved viral gp50 gene, also was employed in an effort to detect viral genomes. Wild type viral DNA was found in the tissues from at least 5 of the vaccinated and 3 of the nonvaccinated swine. These results indicate that such animals should be considered as being infected with ADV. Further, these findings emphasize the need to develope highly specific and sensitive antemortem testing methods for accurate assessment of ADV infection in herds containing such subclinical, yet serologically positive, swine.  相似文献

The objective of this study was to measure and compare the serum concentrations of dexamethasone after oral and transdermal administration using pluronic lecithin organogel in six healthy cats. The study was designed as a crossover, in which the cats were randomly assigned to two groups. The cats received a single dose (0.05 mg kg(-1)) of dexamethasone either orally or transdermally on the inner pinna. Blood samples were taken at 0, 5, 15, 30, 60, 90 and 120 min, and 3, 4, 6, 8, 12 and 24, 48 and 72 h post dexamethasone administration. A mean peak serum concentration of 30.1 ng mL(-1) was detected 15 min after oral administration. Serum concentrations were below detection limits by 24 h. In contrast, there was no significant increase in serum concentrations of dexamethasone after transdermal administration. In cats, transdermal administration of a single dose of dexamethasone did not result in significant serum concentrations compared to oral administration.  相似文献

Objective To determine survival over time of infectious equine herpesvirus‐4, feline herpesvirus‐1, and feline calicivirus in three commercially available and commonly used ophthalmic solutions (eyewash, fluorescein, and proparacaine HCl). Sample population Viruses used in this study were originally isolated from eyes of animals referred to the University of Illinois. Equine herpesvirus‐4 was propagated in MDBK cells and feline herpesvirus‐1 and feline calicivirus in CRFK cells. Procedure After separately inoculating a designated solution with a specific titer of an individual virus, solutions were incubated per manufacturer's recommendations, either at 4 °C or 25 °C. Virus titers within solutions were subsequently measured at 1, 8, and 24 h and 3, 5 and 7 days post inoculation using either plaque or TCID50 assays. Results Equine herpesvirus‐4, feline herpesvirus‐1, and feline calicivirus were present in eyewash for 7 days, 5 days, and 7 days, respectively. Eyewash did not decrease survival time of any virus when compared to controls. Equine herpesvirus‐4 and feline herpesvirus‐1, both enveloped viruses, were not recovered at any time ≥ 1 h post inoculation in fluorescein. Feline calicivirus, a nonenveloped virus, was present in fluorescein for 7 days. Equine herpesvirus‐4 and feline herpesvirus‐1 did not remain infectious in proparacaine at any time ≥ 1 h post inoculation, but feline calicivirus was recovered at up to 24 h post inoculation. Conclusions Equine herpesvirus‐4, feline herpesvirus‐1, and feline calicivirus may be readily transmissible via the eyewash solution used in this study. Risk of iatrogenic transmission of the three viruses used in this study was significantly reduced in both fluorescein and proparacaine solutions. Feline calicivirus, the only nonenveloped virus evaluated, remained viable longer in both fluorescein and proparacaine solutions.  相似文献

Rapidly fatal mediastinal hemorrhage in a previously healthy dog is described. The lesion was manifest radiographically by widening of the dorsal mediastinum. Although the exact origin of hemorrhage was not identified, it is hypothesized that one or more dorsal intercostal arteries were ruptured when the dog ran to an abrupt stop at the end of a long chain.  相似文献

Netting surveys of lakes varying in pH (4.4–7.1) showed that lake trout (Salvelinus namaycush) populations fail to recruit at pH <5.5 and are lost from lakes with pH<5.2. Brook trout (S. fontinalis) were extirpated in lakes with pH <5.0. In regional chemical surveys of Ontario lakes, it was found that 2% of sampled brook trout lakes and 2.5% of lake trout lakes were acidified (alkalinity <0 uEq L?1). Threshold pH levels determined from fisheries assessments were used to estimate that 1% of lake trout and brook trout populations have been lost due to acidification.  相似文献

Habitat fragmentation is thought to create a barrier to individual movements particularly for area-sensitive species which, by definition, prefer to breed in large tracts of forest. For two breeding seasons, we radio-tracked an area-sensitive species, the scarlet tanager Piranga olivacea, in a fragmented landscape in northeastern PA. We found that scarlet tanagers made extensive and frequent movements among fragments. Paired males were less likely to leave their capture fragment, and traveled shorter distances. Unpaired males in fragments had two distinct tactics we labeled: “Sedentary” and “Mobile”. Sedentary males stayed at one fragment and sang at high rates, while mobile males spent 58% of the total time tracked off their capture fragment and traveled over a kilometer away from the capture site over open fields and through forests. Mobile males were not floaters per se because they were territorial (i.e., singing) in multiple sites. Habitat structure of a fragment did not correlate with the percent time a male spent off the capture fragment. Males in fragments experienced lower pairing success and were more likely to be first time breeders compared to males in continuous forest. Our results suggest that movements by scarlet tanagers in fragmented landscapes are not restricted during the breeding season, and that these movements are related directly to pairing status and indirectly to population density.  相似文献

High-resolution chemostratigraphy reveals an episode of enrichment of the redox-sensitive transition metals molybdenum and rhenium in the late Archean Mount McRae Shale in Western Australia. Correlations with organic carbon indicate that these metals were derived from contemporaneous seawater. Rhenium/osmium geochronology demonstrates that the enrichment is a primary sedimentary feature dating to 2501 +/- 8 million years ago (Ma). Molybdenum and rhenium were probably supplied to Archean oceans by oxidative weathering of crustal sulfide minerals. These findings point to the presence of small amounts of O2 in the environment more than 50 million years before the start of the Great Oxidation Event.  相似文献

Much of the attention by social scientists to the rapidly growing organic agriculture sector focuses on the benefits it provides to consumers (in the form of pesticide-free foods) and to farmers (in the form of price premiums). By contrast, there has been little discussion or research about the implications of the boom in organic agriculture for farmworkers on organic farms. In this paper, we ask the question: From the perspective of organic farmers, does “certified organic” agriculture encompass a commitment to “sustainability” that prioritizes social goals? Specifically, we aim to broaden our understanding of the relationship between social sustainability and organic agriculture by drawing attention to issues affecting farmworkers, whose labor and contribution tends to elude most discussions of organic agriculture. We present findings from a survey of organic farmers in California about the possible incorporation of social standards into organic certification criteria. Our findings suggest that, at best, lukewarm support for social certification within organic agriculture exists among certified organic farmers in California. They also question expectations that organic agriculture necessarily fosters social or even economic sustainability for most of the farmers and farmworkers involved. However, we also find exceptions to the patterns evidenced in our survey. In-depth interviews with select organic farmers demonstrate that there are individuals whose practices are atypical and demonstrate that, under some circumstances, an organic production system can be at once environmentally, economically, and socially sustainable.  相似文献
