In the Vosges Mountains (NE of France), integrated plot-catchment studies have been carried out since 1985 in the Strengbach basin to study the influence of acid atmospheric inputs on surface water quality and element budgets. In this paper, available mid-term time series (1985–1991) have been considered to detect obvious trends, if any, in surface water chemistry and element budgets. Air quality data showed a slight decline for SO2, whereas NO2 slightly increased over the period, but these trends are not very significant. This is in agreement with increased N concentration (mainly as NH 4 + ) and with the stability of SO 4 2? in open field precipitation. Because of a significant decrease in rainfall amount over the period, only inputs of NH 4 + increased significantly whereas H+ and SO 4 2+ inputs declined. In spring and streamwaters, pH and dissolved Si concentration increased mainly as a result of a reduced flow. Na+, K+, Cl? and HCO-3~? concentrations remained stable whereas Ca2+, Mg2+ and SO 4 2+ concentrations declined significantly. Only NO 3 ? concentration increased significantly in springwaters. The catchment budgets revealed significant losses of base cations, Si and SO 4 2? . These losses decreased over the period. Nitrogen was retained in the ecosystem. However, a longer record is needed to determine whether or not changes in surface water chemistry have resulted from short-term flow reductions or long-term changes in input-output ion budgets. This is specially true with N because the decline in SO 4 2? output was accompanied by N accumulation.  相似文献

Site-directed mutagenesis of the large fragment of DNA polymerase I (Klenow fragment) yielded two mutant proteins lacking 3',5'-exonuclease activity but having normal polymerase activity. Crystallographic analysis of the mutant proteins showed that neither had any alteration in protein structure other than the expected changes at the mutation sites. These results confirmed the presumed location of the exonuclease active site on the small domain of Klenow fragment and its physical separation from the polymerase active site. An anomalous scattering difference Fourier of a complex of the wild-type enzyme with divalent manganese ion and deoxythymidine monophosphate showed that the exonuclease active site has binding sites for two divalent metal ions. The properties of the mutant proteins suggest that one metal ion plays a role in substrate binding while the other is involved in catalysis of the exonuclease reaction.  相似文献

Commercial laying hens were examined microbiologically at necropsy 31 or 42 weeks after aerosol vaccination with the F strain of Mycoplasma gallisepticum (MG). Mycoplasma isolates were studied in Western blots probed with polyclonal antiserum raised in rabbits to F strain immunogen. The persistence of the vaccine strain was demonstrated by detection of a 75-kilodalton immunoreactive protein, which was present in all MG isolates and thought to be a unique marker of the F strain. Use of PCA-F to probe Western blots allowed simultaneous identification of non-MG isolates, non-F strains of MG, and the F strain of MG.  相似文献

Lyme borreliosis     
Despite more than 25 years' experience with Lyme borreliosis, much remains to be learned about this complex zoonosis. Practicing veterinarians, particularly those in the northeastern and upper midwestern states, where Lyme borreliosis is highly endemic, should be familiar with the ecologic features and typical clinical signs of Lyme borreliosis. Interpretation of signs and serologic test results should be made with consideration of the regional prevalence of Lyme borreliosis and the animal's opportunity for exposure to infected Ixodes spp. The availability of recently marketed topical acaracides is a valuable adjunctive measure in prevention of Lyme borreliosis. A maximally effective prevention strategy should include consideration of environmental modification, activity restrictions, routine examinations for ticks, prompt removal of attached ticks, and vaccination. Technologic advances, such as the C6 EIA and the Osp A recombinant vaccine, offer the promise of additional tools for the clinical management and prevention of this tick-borne zoonosis.  相似文献

A 24-year-old female rhesus macaque (Macaca mulatta) presented with a clinical history of chronic heart disease and prolonged recovery from sedation and anesthesia. At necropsy, the heart was markedly enlarged, with thinned ventricular walls, dilated chambers, and severe left atrioventricular valvular insufficiency. The ventricular walls contained numerous narrow, pale, often coalescing streaks that often extended along vessels into the deeper myocardium. Histologically, bands of interstitial fibrosis in the heart were associated with areas of myofiber atrophy, myofibril degeneration and loss, and inflammation. The left adrenal gland contained a 2 x 1 x 0.5 cm tumor with an unusual angiomatous pattern consisting largely of blood-filled sinusoids lined by one to four layers of low cuboidal to polyhedral tumor cells. In most sinusoids, the tumor cells appeared to be in direct contact with blood, although endothelial cells lined some sinusoids. Ultrastructurally, the tumor cells contained abundant electron-dense cytoplasmic granules. These granules were argyrophilic by Sevier-Munger staining and intensely immunoreactive for synaptophysin, chromogranin A, neuron-specific enolase, and S-100. These findings helped confirm the diagnosis of angiomatous pheochromocytoma. The heart lesions match those reported for catecholamine cardiomyopathy in other species.  相似文献

Freshly milled wheat flour has a pale yellow color due to its carotenoids content. Benzoyl peroxide is a bleaching agent typically used to give such flour a better appearance. This free-radical initiator promotes carotenoids oxidation, thereby producing less colored compounds, and benzoic acid is a main final product. Samples of wheat flour were treated with 150 ppm of benzoyl peroxide to begin a bleaching process, and then subjected to ethyl ether extraction at different intervals of time. Benzoyl peroxide and benzoic acid levels in these extracts were monitored by means of HPLC in individual experiences. The resulting concentration of benzoyl peroxide after 9 days of contact with the bleaching agent was 11 ppm, dropping afterward to nondetectable levels. A maximum value for benzoic acid of 16 ppm was found after 12 h of bleaching. Subsequently this level decreased continuously until reaching a residual value of 6 ppm after 3 months.  相似文献
