Genetic Analysis of Resistance to Soil-Borne Wheat Mosaic Virus Derived from Aegilops tauschii. Euphytica. Soil-Borne Wheat Mosaic Virus (SBWMV), vectored by the soil inhabiting organism Polymyxa graminis, causes damage to wheat (Triticum aestivum) yields in most of the wheat growing regions of the world. In localized fields, the entire crop may be lost to the virus.
Although many winter wheat cultivars contain resistance to SBWMV, the inheritance of resistance is poorly understood. A linkage
analysis of a segregating recombinant inbred line population from the cross KS96WGRC40 × Wichita identified a gene of major
effect conferring resistance to SBWMV in the germplasm KS96WGRC40. The SBWMV resistance gene within KS96WGRC40 was derived
from accession TA2397 of Aegilops taushcii and is located on the long arm of chromosome 5D, flanked by microsatellite markers Xcfd10 and Xbarc144. The relationship of this locus with a previously identified QTL for SBWMV resistance and the Sbm1 gene conferring resistance to soil-borne cereal mosaic virus is not known, but suggests that a gene on 5DL conferring resistance to both viruses may be present in T. aestivum, as well as the D-genome donor Ae. tauschii. 相似文献

Allocation of biomass and nutrient elements including Nitrogen to above and belowground compartments of beech seedlings (Fagus sylvatica L.) treated by labeled nitrogen fertilizer in the form of 15NH4 and 15NO3 were investigated at the end of two successive growing seasons. Pot cultured beech seedlings were grown at a green house on intact soil cores sampled from three adjacent stands including beech, Norway spruce and mixed beech-spruce cultures of Solling forest, Germany. Comparing biomass allocation and nutrients concentrations of the seedlings between the control and 15N-fertilized treatments revealed no significant effect of N fertilization on nutrients uptake by seedlings over the experiment. The form of N input influenced its movement into plant pools. It was demonstrated that beech seedlings take up nitrogen mainly in the form of nitrate, which is then reduced in the leaves, although the differences between the retention of NO3 ?-N and NH4 +-N in plants were not statistically significant. Percent recoveries of 15N in trees were typically greater after 15NO3 than after 15NH4 additions. It was indicated that immobilization of 15N tracer in fine roots was a slower process comparing other plant compartments such as stem and coarse roots, but a powerful sink for N during the course of study. 相似文献

The effect of NaCl on growth, biomass and ion relations of two salt-tolerant isolates of Paxillus involutus, MAJ and NAU were investigated. The two Paxillus strains were exposed to the following concentrations of NaCl: 0, 100, 200 and 500 mmol·L^-1. Growth of MAJ and NAU was enhanced by 100 mmol·L^-1 NaCl but severely inhibited at the concentration of 500 mmol·L^-1. NAU exhibited a greater capacity to exclude Na^+ and Cl^- under all salinity levels, whereas the salt-includer MAJ had a higher capacity in nutrient uptake under salt stress. The ratios Na^+/K^+, Na^+/Ca^2+ and Na^+/Mg^2+ in NaCl-treated MAJ and NAU did not increase at levels of 100 and 200 mmol·L^-1 NaCl but markedly increased at 500 mmol·L^-1. This suggests that the two strains, especially MAJ, enhanced nutrient uptake corresponding to the increased Na^+ influx at moderate salinity. We conclude that both MAJ and NAU are able to tolerate 500 mmol·L^-1 NaCl but there are species-specific differences in retaining ionic homeostasis in the two Paxillus strains. NAU is a salt-excluder, MAJ is a salt-includer but retains a high capacity in nutrient selectivity under saline conditions. Their definite mechanisms to enhance salt tolerance of mycorrhizal hosts need further study. 相似文献

Physical properties of the digesta vary along the ruminant digestive tract. They also vary within the forestomach, leading to varying degrees of rumen contents stratification in ‘moose‐type’ (browsing) and ‘cattle‐type’ (intermediate and grazing) ruminants. We investigated the dry matter concentration (DM) and the mean digesta particle size (MPS) within the forestomach and along the digestive tract in 10 goats fed grass hay or dried browse after a standardized 12‐h fast, euthanasia and freezing in the natural position. In all animals, irrespective of diet, DM showed a peak in the omasum and an increase from caecum via colon towards the faeces and a decrease in MPS between the reticulum and the omasum. Both patterns are typical for ruminants in general. In the forestomach, there was little systematic difference between more cranial and more caudal locations (‘horizontal stratification’), with the possible exception of large particle segregation in the dorsal rumen blindsac on the grass diet. In contrast, the typical (vertical) contents stratification was evident for DM (with drier contents dorsally) and, to a lower degree, for MPS (with larger particles dorsally). Although evident in both groups, this stratification was more pronounced on the grass diet. The results support the interpretation that differences in rumen contents stratification between ruminants are mainly an effect of species‐specific physiology, but can be enhanced due to the diet consumed. 相似文献

Taxol (paclitaxel) is a potent anticancer drug first isolated from the Taxus brevifolia Pacific yew tree. Currently, cost-efficient production of Taxol and its analogs remains limited. Here, we report a multivariate-modular approach to metabolic-pathway engineering that succeeded in increasing titers of taxadiene--the first committed Taxol intermediate--approximately 1 gram per liter (~15,000-fold) in an engineered Escherichia coli strain. Our approach partitioned the taxadiene metabolic pathway into two modules: a native upstream methylerythritol-phosphate (MEP) pathway forming isopentenyl pyrophosphate and a heterologous downstream terpenoid-forming pathway. Systematic multivariate search identified conditions that optimally balance the two pathway modules so as to maximize the taxadiene production with minimal accumulation of indole, which is an inhibitory compound found here. We also engineered the next step in Taxol biosynthesis, a P450-mediated 5α-oxidation of taxadiene to taxadien-5α-ol. More broadly, the modular pathway engineering approach helped to unlock the potential of the MEP pathway for the engineered production of terpenoid natural products. 相似文献

The effectiveness of safeguarding and quarantine actions aimed at preventing the introduction and minimizing the impact of exotic plant pests relies on current information about those pests. National Plant Protection Organizations (NPPOs) face the following challenges: staying up to date on continuously emerging new pest information, filtering and evaluating this information, disseminating it effectively and efficiently, applying it toward decision making, and coordinating and tracking the resulting decisions and safeguarding actions. PestLens, the web‐based early‐warning system of the NPPO of the United States, is designed to address all of these challenges. Does forest continuity matter in conservation? - A study of epiphytic lichens and bryophytes in beech forests of southern Sweden 总被引:1,自引:0,他引:1
The effects of forest continuity at local scale for red-listed and indicator species of epiphytic lichens and bryophytes were investigated in 150 Fagus sylvatica stands in southern Sweden. Stands having forest continuity (n = 106) had continuous forest cover more than 350 years, whereas stands lacking continuity (n = 44) had forest cover less than 160 years. Forest continuity was identified by comparing a sequence of historical maps with a modern survey of beech forests. In the field woody beech substrates were searched for the epiphytes of interest. A number of environmental and spatial variables were inventoried and compiled for each stand. In all 64 species (51 lichens, 13 bryophytes) were found in the stands having continuity, and 21 (14 lichens, 7 bryophytes) in the stands lacking continuity. Controlling for the different number of surveyed stands, stands having continuity had significantly more species of lichens, but not of bryophytes. In the stands lacking continuity we did not find lichens associated with the very late succession stage. The quantity of substrates, stand age and forest continuity were the three most important factors explaining species richness as well as composition of studied epiphytes. The effect of continuity was probably due to a combination of a higher substrate quality, mainly old beeches, and a longer time available for colonization. Also, we found strong positive correlations between number of indicator and red-listed epiphyte species. To investigate the effect of silvicultural methods on forest floor C, N and elements stocks an experiment was carried out by sampling the forest floors of a 100-120-years-old species including beech, Norway spruce and mixed beech-spruce at the Solling forest, Germany. While the stocks of carbon and nitrogen in the forest floors of pure beech and spruce were significantly influenced by species specific differences of litter quality (p < 0.001), no significant differences were detected between pure and mixed species stands. Forest floor mass, some elements concentrations and C/nutrient ratios were significantly affected by tree species differences, while no clear dependency between pH and site specific effects was found among pure stands. Acid element concentrations in the forest floors of pure spruce were remarkably higher than the values obtained at beech stand, while the stocks were to some extent modified in mixed silviculture. The base-pump effect of beech significantly controlled variation between mono cultures on calcium stocks, while the acidifying effect of spruce in mixtures resulted in modification of Ca stocks of forest floors. The status of other nutrient elements at mixed species cultures due to variation in nutritional properties and composition of litter compared to pure species were between the range of values observed in mono cultures. 相似文献
