The thermal motion of stiff filaments in a crowded environment is highly constrained and anisotropic; it underlies the behavior of such disparate systems as polymer materials, nanocomposites, and the cell cytoskeleton. Despite decades of theoretical study, the fundamental dynamics of such systems remains a mystery. Using near-infrared video microscopy, we studied the thermal diffusion of individual single-walled carbon nanotubes (SWNTs) confined in porous agarose networks. We found that even a small bending flexibility of SWNTs strongly enhances their motion: The rotational diffusion constant is proportional to the filament-bending compliance and is independent of the network pore size. The interplay between crowding and thermal bending implies that the notion of a filament's stiffness depends on its confinement. Moreover, the mobility of SWNTs and other inclusions can be controlled by tailoring their stiffness.  相似文献

The roles free-living amebae and the parasitic protozoa Entamoeba histolytica and Balantidium coli play as agents of waterborne zoonotic diseases are examined. The free-living soil and water amebae Naegleria fowleri, Acanthamoeba spp., and Balamuthia mandrillaris are recognized etiologic agents of mostly fatal amebic encephalitides in humans and other animals, with immunocompromised and immunocompetent hosts among the victims. Acanthamoeba spp. are also agents of amebic keratitis. Infection is through the respiratory tract, breaks in the skin, or by uptake of water into the nostrils, with spread to the central nervous system. E. histolytica and B. coli are parasitic protozoa that cause amebic dysentery and balantidiasis, respectively. Both intestinal infections are spread via a fecal-oral route, with cysts as the infective stage. Although the amebic encephalitides can be acquired by contact with water, they are not, strictly speaking, waterborne diseases and are not transmitted to humans from animals. Non-human primates and swine are reservoirs for E. histolytica and B. coli, and the diseases they cause are acquired from cysts, usually in sewage-contaminated water. Amebic dysentery and balantidiasis are examples of zoonotic waterborne infections, though human-to-human transmission can occur. The epidemiology of the diseases is examined, as are diagnostic procedures, anti-microbial interventions, and the influence of globalization, climate change, and technological advances on their spread.  相似文献

Biomass represents an abundant carbon-neutral renewable resource for the production of bioenergy and biomaterials, and its enhanced use would address several societal needs. Advances in genetics, biotechnology, process chemistry, and engineering are leading to a new manufacturing concept for converting renewable biomass to valuable fuels and products, generally referred to as the biorefinery. The integration of agroenergy crops and biorefinery manufacturing technologies offers the potential for the development of sustainable biopower and biomaterials that will lead to a new manufacturing paradigm.  相似文献

Current yeast interactome network maps contain several hundred molecular complexes with limited and somewhat controversial representation of direct binary interactions. We carried out a comparative quality assessment of current yeast interactome data sets, demonstrating that high-throughput yeast two-hybrid (Y2H) screening provides high-quality binary interaction information. Because a large fraction of the yeast binary interactome remains to be mapped, we developed an empirically controlled mapping framework to produce a "second-generation" high-quality, high-throughput Y2H data set covering approximately 20% of all yeast binary interactions. Both Y2H and affinity purification followed by mass spectrometry (AP/MS) data are of equally high quality but of a fundamentally different and complementary nature, resulting in networks with different topological and biological properties. Compared to co-complex interactome models, this binary map is enriched for transient signaling interactions and intercomplex connections with a highly significant clustering between essential proteins. Rather than correlating with essentiality, protein connectivity correlates with genetic pleiotropy.  相似文献

Thirty-two barrows were used to determine the effects of supplemental Mg in drinking water on pork quality. Pigs were determined to be free of the halothane and Napole mutations and were individually penned. After a 7-d adjustment period, barrows (111 +/- 1 kg BW) were blocked by BW and allotted randomly within block to 0, 300, 600, or 900 mg of supplemental Mg from Mg sulfate/L of drinking water for 2 d before slaughter. Pigs were not allowed access to feed (0.13% Mg) for 15 h before slaughter but continued to have access to experimental water treatments. Pigs were loaded and transported 110 km (1.75 h) to a commercial abattoir and remained in lairage for 5 h before slaughter. The LM was removed 24 h postmortem. Retail storage was simulated for 8 d, and the remaining LM was vacuum-packaged for 25 or 50 d at 4 degrees C. Plasma Mg concentration increased linearly (P = 0.001) with Mg supplementation; however, Mg concentration of the LM was not affected (P = 0.99) by Mg supplementation. Surface exudate, drip loss, and retail fluid loss of the LM were not affected (P > 0.10) by Mg. Lightness (L*) and redness (a*) of the LM were not affected (P > 0.10) by Mg, with the exception of initial redness (cubic; P = 0.05). Pigs supplemented with 300 or 900 mg of Mg/L had lower yellowness (b*) values of the LM displayed for 0 to 6 d than pigs supplemented with 0 or 600 mg of Mg/L (cubic; P < 0.05). Lightness of the LM after 25 (quadratic; P = 0.03) or 50 (quadratic; P = 0.04) d of vacuum-packed storage was greater at 300 and 600 mg of Mg/L than at 0 or 900 mg/L. Yellowness tended to be greater after 50 d, but not after 25 d, of vacuum-packaged storage for 300 or 600 mg of Mg/L compared with 0 or 900 mg/L (quadratic; P = 0.08). Oxidation of the LM, determined by thiobarbituric acid reactive substances after 4 d of retail storage, increased linearly (P = 0.05) as Mg increased in the drinking water. Furthermore, oxidation of the LM after 8 d of retail storage tended to increase linearly (P < 0.10), primarily because of the high oxidation of LM from pigs supplemented with 900 mg of Mg/L compared with controls (224 vs. 171 +/- 19 microg/kg, respectively). Oxidation of the LM was greater for pigs supplemented with 300 or 900 mg/L compared with 0 or 600 mg of Mg/L (cubic; P < 0.06) after 25 d of vacuum-packed storage. Magnesium did not improve pork quality characteristics of practical significance in pigs without the halothane and Rendement Napole mutations.  相似文献

The crystal structure of biotin synthase from Escherichia coli in complex with S-adenosyl-L-methionine and dethiobiotin has been determined to 3.4 angstrom resolution. This structure addresses how "AdoMet radical" or "radical SAM" enzymes use Fe4S4 clusters and S-adenosyl-L-methionine to generate organic radicals. Biotin synthase catalyzes the radical-mediated insertion of sulfur into dethiobiotin to form biotin. The structure places the substrates between the Fe4S4 cluster, essential for radical generation, and the Fe2S2 cluster, postulated to be the source of sulfur, with both clusters in unprecedented coordination environments.  相似文献

Corridor disease, caused by the tick-borne protozoan parasite Theileria parva, is a controlled disease in South Africa. The Cape buffalo is the reservoir host and uninfected buffalo have become sought-after by the game industry in South Africa, particularly for introduction into Corridor disease-free areas. A real-time polymerase chain reaction (PCR) test for detection of T. parva DNA in buffalo and cattle was developed to improve the sensitivity and specificity of the official diagnostic test package in South Africa. Oligonucleotide primers and hybridization probes were designed based on the 18S ribosomal RNA (rRNA) gene. Amplification of control DNA using Theileria genus-specific primers resulted in detection of T. taurotragi and T. annulata, in addition to T. parva. A T. parva-specific forward primer was designed which eliminated amplification of all other Theileria species, except for Theileria sp. (buffalo); however only the T. parva product was detected by the T. parva-specific hybridization probe set. The real-time PCR assay requires less time to perform, is more sensitive than the other molecular assays previously used in T. parva diagnostics and can reliably detect the parasite in carrier animals with a piroplasm parasitaemia as low as 8.79 x 10(-4)%.  相似文献

Resistance to bacterial speck disease in tomato occurs when the Pto kinase in the plant responds to expression of the avirulence gene avrPto in the Pseudomonas pathogen. Transient expression of an avrPto transgene in plant cells containing Pto elicited a defense response. In the yeast two-hybrid system, the Pto kinase physically interacted with AvrPto. Alterations of AvrPto or Pto that disrupted the interaction in yeast also abolished disease resistance in plants. The physical interaction of AvrPto and Pto provides an explanation of gene-for-gene specificity in bacterial speck disease resistance.  相似文献

Fire in sagebrush rangelands significantly alters canopy cover, ground cover, and soil properties which influence runoff and erosion processes. Runoff can be generated more quickly and in larger volume following fire resulting in increased risk of severe erosion and downstream flooding. The Water Erosion Prediction Project (WEPP) model was developed to predict erosion on cropland, forest, and rangeland. WEPP is a tool that has potential to model the effect of fire on hillslope hydrological processes and help managers address erosion and runoff risks following fire. Experimental results on a steep (35 to 50% slope) sagebrush site suggest that rill erosion is the dominant erosion process following fire and the WEPP parameterization equations related to the rill erosion process need improvements. Rill detachment estimates could be improved by modifying regression-estimated values of rill erodibility. Also, the interactions of rill width and surface roughness on soil shear stress estimates may also need to be modified. In this paper we report the effects of prescribed fire on runoff, soil erosion, and rill hydraulics and compare WEPP estimated erosion for several modeling options with measured erosion.  相似文献

Two new isocoumarins (1 and 2), a new alkaloid (3), and a known N-acetyldopamine dimer (4) were isolated from the ethyl acetate extract of Chinese insect medicine Eupolyphaga sinensis. Their structures were elucidated on the basis of detailed spectroscopic investigations, such as 1D- and 2D NMR spectroscopy, as well as by means of HR-MS. The structure of 1 was firmly confirmed by X-ray crystallography, and the absolute configuration was revealed by experimental and computational optical rotation analyses. Cytotoxicities of 1-4 were measured in vitro against 10 selected cancer cell lines.  相似文献
