Molecular, physiological and pathological characterization of Corynespora leaf spot fungi from rubber plantations in Sri Lanka 总被引:4,自引:0,他引:4
The plant pathogenic fungus Corynespora cassiicola causes a severe leaf spot disease on more than 70 host plant species including Hevea brasiliensis . Genetic variability in 32 isolates of C. cassiicola collected from diverse hosts and locations in Sri Lanka and Australia was assessed using restriction fragment length polymorphism (RFLP) analysis of the internal transcribed spacer (ITS) region of ribosomal DNA and random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) analysis of total fungal DNA. Amplified ITS fragments from all 32 C. cassiicola isolates exhibited an identical size, and restriction analysis with seven different restriction endonucleases revealed identity in all of the detected DNA fragments. This finding of high genetic relatedness was further supported by the cloning and DNA sequencing of the ITS2 region from one Sri Lankan and one Australian isolate. However, RAPD-PCR profiles generated by 15 oligonucleotide decamer primers revealed significant polymorphism between groups of organisms. Genetic relationships among the isolates were determined by cluster analysis of the RAPD-PCR data and seven different RAPD groups were identified. Isolates showed strong correlations between the assigned RAPD group and the location and host plant genotype from which the isolate was collected. The textile industries are characterized as one of the biggest consumers of potable water and chemical products throughout its process, being responsible for the elevated wastewater generation with intense coloration and wide polluting potential. In this context, the present study proposes the development and application of a new coagulant material for textile wastewater treatment. The proposed coagulant (α-Fe2O3-MO) was composed by hematite nanoparticles (α-Fe2O3) obtained by a simple non-pollutant methodology, associated with Moringa oleifera (MO) seeds saline extract compounds. Coagulation/flocculation (CF) efficiency was evaluated by removal of physicochemical parameters such as apparent color, turbidity, and compounds with absorption at UV254nm (UV254nm) through CF tests carried out on Jar test equipment and sedimentation carried out in the presence and absence of external magnetic field (600 k Am?1). Kinetics sedimentation was from 0 to 90 min. The use of this new coagulant allowed the removal of 92.37% for apparent color, 91.43% for turbidity, and 46.09% for UV254nm, indicating that the proposed coagulant association was efficient in the treatment of this type of wastewater under external magnetic field with only 10 min of sedimentation. In addition, the resulting sludge from CF process was tested as base material for a new coagulant synthesis, demonstrating great reuse potential. Therefore, the new proposed coagulant, composed of α-Fe2O3 and the compounds present in the seed extract of MO, has applicability for textile wastewater treatment demonstrating high removal rate for all evaluated parameters with cost reduction in the proposed treatment for this wastewater. 相似文献

Pekin robins (Leiothrix lutea) were once the most widely kept softbills in captivity. As a result of the Convention on the International Trade of Endangered Species of Fauna and Flora (CITES-1997), the worldwide trade of wild-caught pekin robins has been prohibited due to the depletion of native populations of this species. In Brazil, as in other countries, pekin robins imported prior to the enactment of the CITES have disappeared from aviaries because the end of the birds' natural life span has passed, and only very few captive-bred pekin robins now exist. While captive propagation fails to address the primary causes of wild bird population decline, it might help the recovery of populations of this species. This article presents records made over a 10-yr period of a captive colony of pekin robins. Emphasis is placed on the management of the flock, the ailments affecting the birds, and the findings associated with bird losses. The objective of this work was to evaluate rates for applications of water treatment sludge (WTS) as a nutrient source for grasses and leguminous plants cropped in a soil degraded by tin mining in the Amazon Region (Natural Forest of Jamari, Rondonia State, Brazil). The treatments consisted of three rates of nitrogen supplied by WTS (100, 150 and 200 mg kg−1 soil), five combinations of plants, two controls (absolute control, without fertilization; and chemical control, soil+lime+chemical fertilizers). The endosperm proteins of the maize mutants, opaque-2, opaque-7, floury-2, brittle-2, and the double mutant of opaque-2 and brittle-2, were separated into five soluble fractions by the Landry-Moureaux method. As compared to their isogenic normal counterparts, the mutant endosperms had higher concentrations of albumins, globulins, and glutelin-3, and lower concentrations of prolamines. The combination of the opaque-2 and brittle-2 genes enhanced this difference. Although the four mutant genes are located on three different chromosomes, they exert a similar effect on endosperm protein composition. Five other starchmodifying mutants with high lysine content resemble the brittle-2 mutant in endosperm protein composition, when the gene is present either singly or combined with opaque-2. Therefore, the pattern of protein synthesis in all maize mutants with high lysine concentrations may be either identical or very similar. Because no synergistic effect on lysine concentration is obtained when floury-2 is combined with opaque-2, different pathways leading to reduced zein synthesis may exist in the floury and starch-modifying mutants with high lysine concentrations. 相似文献

The antioxidant capacity of propolis from the southern region of Uruguay was evaluated using in vitro as well as cellular assays. Free radical scavenging capacity was assessed by ORAC, obtaining values significantly higher than those of other natural products (8000 μmol Trolox equiv/g propolis). ORAC values correlated well with total polyphenol content (determined by Folin-Ciocalteu method) and UV absorption. Total polyphenol content (150 mg gallic acid equiv/g propolis) and flavonoids (45 mg quercetin equiv/g propolis) were similar to values reported for southern Brazilian (group 3) and Argentinean propolis. Flavonoid composition determined by RP-HPLC indicates a strong poplar-tree origin. Samples high in polyphenols efficiently inhibit low-density lipoprotein lipoperoxidation and tyrosine nitration. In addition, Uruguayan propolis was found to induce the expression of endothelial nitric oxide synthase and inhibit endothelial NADPH oxidase, suggesting a potential cardiovascular benefit by increasing nitric oxide bioavailability in the endothelium. 相似文献
