Type 1 polysaccharide storage myopathy caused by genetic mutation in the glycogen synthase 1 gene is present in many breeds including the Noriker and Haflinger horses. In humans, EMG has already been used to document changes in the muscle activity patterns of patients affected by human glycogen storage disorders. Therefore, the aim of the present study was to describe gluteus muscle activity with surface electromyography (sEMG) in Haflinger and Noriker horses with known GYS1 mutation status during walk and trot. Thirty-two horses (11 Haflinger and 21 Noriker horses) with homozygous non-affected (GG), heterozygous affected (GA) and homozygous affected (AA) status of GYS1 mutation without overt clinical signs of any myopathy were selected for the current study. Using surface electromyography gluteus medius muscle activity at walk and at trot was measured, and muscle activity was described in relation to the maximum observed value at the same sensor and the same gait. In order to further describe the signals in detail comprising both frequencies and amplitudes, the crossings through the baseline and the 25, 50 and 75 percentile lines were determined. The result of the relative muscle activity did not show a consistent difference between affected and non-affected horses. Genetically affected (GA and AA) horses showed significantly less density of muscle activity for both gaits and horse breeds except for the crossings per second at the baseline and 75 percentile at walk in the Haflinger horses and 75 percentile at trot in the Noriker horses. The medians of all calculated density values were significantly lower in the GA Haflingers compared to the GG Haflingers (p = 0.012) and also in the AA Norikers compared to the GG Norikers (p = 0.011). Results indicate that the GYS1 mutation reduces the number of functional muscle fibres detected by sEMG measurements even in the absence of overt clinical signs.  相似文献

Background and aims : Most physiological and biochemical studies on salt stress are NaCl‐based. However, other ions (e.g., K+, Ca2+, Mg2+, and SO 4 2 - ) also contribute to salt stress in special circumstances. In this study, salt stress induced by various salts was investigated for a better understanding of salinity. Methods : Arabidopsis thaliana plants were stepwise acclimated to five iso‐osmotic salts as follows: NaCl, KCl, Na2SO4, K2SO4, and CaCl2. Results and Conclusions : Exposure to KCl and K2SO4 led to more severe toxicity symptoms, smaller biomass, and lower level of chlorophyll than exposure to NaCl and Na2SO4, indicating that Arabidopsis plants are more sensitive to potassium salts. The strongly reduced growth was negatively correlated with the accumulation of soluble sugars observed in KCl‐ and K2SO4‐treated plants, suggesting a blockage in the utilization of sugars for growth. We found that exposure to KCl and K2SO4 suppressed or even blocked sucrose degradation, thus leading to strong accumulation of sucrose in shoots, which then probably inhibited photosynthesis via feedback inhibition. Moreover, K+ was more accumulated in shoots than Na+ after corresponding potassium or sodium salt treatments, thus resulting in decreased Ca2+ and Mg2+ concentrations in response to KCl and K2SO4. However, K2SO4 caused more severe toxicity symptoms than iso‐osmotic KCl, even when the K+ level was lower in K2SO4‐treated plants. We found that Na2SO4 and K2SO4 induced strong accumulation of tricarboxylic acid intermediates, especially fumarate and succinate which might induce oxidative stress. Thus, the severe toxicity symptoms found in K2SO4‐treated plants were also attributed to SO 4 2 - in addition to the massive accumulation of K+.  相似文献

Biochar is increasingly promoted as a beneficial soil conditioner. However, it may contain residues of polycyclic aromatic hydrocarbons (PAHs) as a result of its production by pyrolysis. To date, analytical methods to analyze PAHs in biochar quantitatively are hardly available. This study presents an optimized and validated procedure to quantify the 16 U.S. EPA PAHs in biochar. PAHs were best extracted with Soxhlet for 36 h using 100% toluene. Average absolute recoveries of isotope labeled internal standards used for each analyte from three different biochars ranged from 42% to 72%, and relative recoveries were between 71% and 105%. The limits of detection were biochar-dependent, but on average a factor of >50 lower than quantified PAH concentrations (9-355 mg kg(dry weight)(-1)). The established method prepares the ground for a harmonized protocol for PAH analysis of biochars, a necessity for biochar quality control, registration, and legislation.  相似文献

Purpose To describe a case of an orbito‐nasal cyst in a cat. Procedure An 18‐month‐old male European short‐haired cat was presented to the Ophthalmology service of the Vetsuisse Faculty, University of Zurich for a subcutaneous swelling in the medial canthal region of the right eye (OD). Ophthalmologic, ultrasound and CT examinations, and fine needle aspiration were performed. After lesion excision, the removed tissue was submitted for histopathology. CT examination was repeated 5 months after removal of the cyst. Results Ophthalmologic examination revealed a large fluctuant swelling inferonasal to OD. Despite patent lacrimal puncta, only the first few mm of the lacrimal canaliculi could be cannulated. A normal globe with moderate enophthalmos was present. Ultrasound examination showed a well‐defined lobulated cyst‐like structure in the right orbit, inferonasal and anterior to the eye. CT examination revealed extension of this lesion through the medial orbital wall into the right nasal cavity. Fine needle aspiration confirmed the cystic nature of the lesion. An orbito‐nasal cyst was diagnosed. The orbital part of the cyst was dissected from the surrounding tissue and excised from the periosteum in the medial orbital wall defect. Part of the maxillary bone was removed to allow removal of the cyst from the nasal cavity. Histologically, the cyst wall consisted of a single to multilayered, mostly cuboidal epithelium and surrounding connective tissue. Follow‐up revealed a good functional result and no recurrence 7 months after cyst removal. Conclusions Similar orbito‐nasal cystic structures were reported in dogs but not in cats.  相似文献

ObjectiveTo evaluate the isoflurane-sparing effects of lidocaine administered by constant rate infusion (CRI) during umbilical surgery in calves.Study designRandomized ‘blinded’ prospective clinical study.AnimalsThirty calves (mean 4.7 ± SD 2.5 weeks old) undergoing umbilical surgery.MethodsAfter premedication with xylazine (0.1 mg kg?1, IM), anaesthesia was induced with ketamine (4 mg kg?1, IV) and maintained with isoflurane in O2 administered through a circle breathing system. The calves were assigned randomly to receive a bolus of 2 mg kg?1 lidocaine IV after induction of anaesthesia, followed by CRI of 50 μg kg?1 minute?1 (group L, n = 15) or a bolus and CRI of 0.9% sodium chloride (NaCl, group S, n = 15). End-tidal isoflurane was adjusted to achieve adequate depth of anaesthesia. Heart rate, direct arterial blood pressure and body temperature were measured intraoperatively. Groups were compared by t- tests, anova or Mann–Whitney rank sum test as appropriate.ResultsThe end-tidal concentration of isoflurane (median, IQR) was significantly lower in group L [1.0% (0.94–1.1)] compared to group S [1.2% (1.1–1.5)], indicating a 16.7% reduction in anaesthetic requirement during lidocaine CRI. Cardiopulmonary parameters and recovery times did not differ significantly between groups.Conclusion and clinical relevanceLidocaine CRI may be used as a supplement to inhalation anaesthesia during umbilical surgery in calves in countries where such a protocol would be within the legal requirements for veterinary use in food animals. This study did not show any measurable benefit to the calves other than a reduction in isoflurane requirement.  相似文献

European ash (Fraxinus excelsior) is threatened by Hymenoscyphus pseudoalbidus, the agent of ash dieback disease. Beside ordinary ash dieback symptoms, collar rots have been reported on declining ashes as an additional problem of increasing severity. Since 2009, ash dieback has been surveyed annually in a provenance trial on four different sites in southwest Germany (Metzler et al. in Ger J For Res 183:168–180, 2012). The trial was established in 2005. Data of tree growth, ash dieback symptoms and collar rot prevalence were collected by surveying the trial in summer 2012 and compared with previously published data of the trial. Evaluations revealed a continuous and considerable increase in dieback severity since 2009. The results suggest that the infection process has not come to a standstill yet. Up to 2012, 6 % of the trees remained symptomless, whereas mortality added up to 9 %. There were significant differences in ash dieback severity between the investigated provenances. Collar rot prevalence ranged from 19 to 59 % between study sites. Moreover, high spatial dependency of collar rot prevalence could be detected within sites. Collar rots were more abundant on trees of severe ash dieback intensity, but could also be detected on 15 % of otherwise healthy trees. Mycelium from collar rots could be identified by means of RFLP analyses and sequencing of the ITS region as most likely belonging to Armillaria gallica. The possible roles of Armillaria spp. and H. pseudoalbidus in collar rot formation are discussed.  相似文献
