SUMMARY: Free radical oxidation products, namely conjugated dienes, ultraviolet fluorescence (excitation 325 nm, emission 395 nm) and visible fluorescence (excitation 360 nm, emission 460 nm) were measured in equine synovial fluid exposed to free radicals In vitro and in the plasma and synovial fluids of horses with synovial effusions. The synovial effusions were induced by intra-articularly administered carrageenin (0.3 ml, 1%), which rarely resulted in clinical lameness. The free radicals were generated In vitro by mixtures of iron and ethylene diamine tetra acetate (Fe/EDTA) or mixtures of hypoxanthine and xanthine oxidase (HX/XO). The conjugated diene concentrations and intensity of ultraviolet fluorescence were negligible in plasma and synovial fluid specimens. No increase resulted from incubation of synovial fluids with either a free radical generating system or as a result of the induced inflammation. The intensity of visible fluorescence did not increase in specimens incubated with Fe/EDTA. However, the intensity of visible fluorescence increased in specimens incubated with HX/XO, in synovial effusions induced by carrageenin, in plasma and in synovial fluids aspirated from saline injected controls. The results indicate that the intensity of visible fluorescence of equine synovial fluid increases after exposure to free radicals and during synovitis in the horse, suggesting a possible role for free radicals in the pathogenesis of equine inflammatory joint diseas  相似文献

Angus bulls (n = 24) were selected for either high or low milk EPD, but with similar growth EPD and mated within location (n = 6) at random to Angus cows. Daughters from these matings were bred to calve first at 2 yr of age to common reference sires across locations. Lactation records for 192 daughters were used to evaluate 12-h milk yield, percentage of milk fat and protein, and weaning weight of offspring. Milk production was measured four times during the lactation at regular intervals within location. Dams were separated from their calves the night before milking and milked with a portable milking machine the next morning to estimate 12-h milk yield. A sample of the milk was collected from each cow and analyzed for percentages of milk fat and protein. Data were analyzed as repeated records of the dam. Fixed effects were location, genetic line of sire, gender of calf within location, and milking period, with postpartum interval used as a covariate. Fixed effects and the random effects of sire of dam nested within line, sire of calf, and year were estimated by REML. Genetic line was an important source of variation for milk yield (P < 0.01) and percentage of milk fat (P = 0.03) but not for percentage of milk protein (P = 0.49). Location was significant for all three milk variables (P < 0.01), but the interactions between line and location were not significant. Gender of calf was significant for milk yield (P = 0.04) but not for percentage of milk fat or protein. Line (P = 0.02), location (P = 0.01), calf gender (P = 0.01), and age at weaning (P = 0.01) were significant sources of variation for weaning weight but the interaction of line and location was not (P = 0.69). The correlation coefficient between the sire's milk EPD and 12-h milk yield was significantly different from zero (r = 0.56). The difference between the least squares means for high and low lines for milk yield was 0.66 kg/12 h and the difference was 15.3 kg for weaning weight. The results indicate that there was not evidence for a genotype by environment interaction in milk production for daughters from divergent sires selected for high or low milk EPD.  相似文献

Steers were generated from Angus (A), Beefmaster (BM), Brangus (BA), Gelbray (GB), and Simbrah (SB) sires mated to cows of their breed and to Brahman x Hereford F1 cows (except A) to characterize their carcass traits, composition, and palatability. The 290 steers (48 A, 48 BM, 36 BA, 31 GB, and 46 SB) were slaughtered at an equal fatness end point as determined by real-time ultrasound and visual evaluation. Angus steers had lighter (P < 0.01), more youthful (P < 0.01) carcasses with a higher (P < 0.05) quality grade, more (P < 0.01) fat thickness, and a larger (P < 0.01) longissimus area/100 kg than BM-, BA-, GB-, and SB-sired steers. Angus steers also had a lower (P < 0.01) specific gravity, a higher (P < 0.01) percentage fat and less (P < 0.05) lean in the 9th to 11th rib, and steaks aged for 10 d were more tender (P < 0.01) than steaks from Brahman-derivative sired steers. The BM- and BA-sired steers had lighter (P < 0.01), more youthful (P < 0.05) carcasses, and smaller (P < 0.01) longissimus area than GB- and SB-sired steers. The 9th to 11th rib section from the BM- and BA-sired steers had less lean and more bone (P < 0.01) than GB- and SB-sired steers. The BA-sired steers had more (P < 0.01) marbling and a higher (P < 0.05) quality grade than BM-sired steers. The SB-sired steers had heavier (P < 0.01) carcasses than the GB-sired steers. There were no differences in shear force for steaks aged for 3 d for any of the breed types, but with 10 d of aging, steaks from Angus steers were more tender, possibly indicating that steaks of Brahman-derivative breeds aged at a slower rate than those from Angus.  相似文献

The present study investigated the arrangement and chemical coding of intramural nerve structures supplying the porcine stomach. Tissue samples comprising all layers of the wall of the ventricular fundus were collected from juvenile female pigs (n = 4), which were first deeply anaesthetized and then transcardially perfused with buffered paraformaldehyde. The cryostat sections were processed for double‐labelling immunofluorescence to study the distribution of the intramural nerve structures (visualized with antibodies against protein gene‐product 9.5) and their neurochemical characteristics using antibodies against vesicular acetylcholine transporter (VAChT), nitric oxide synthase (NOS), galanin (GAL), vasoactive intestinal‐polypeptide (VIP), somatostatin (SOM) and substance P (SP). The study confirmed the presence of three distinct nerve plexuses within the wall of the porcine stomach including one myenteric plexus and two, outer and inner, submucous plexuses. The outer and inner submucous plexuses (OSP and ISP, respectively) were similar in respect to the chemical coding of neurons they contained. Most of the neurons expressed immunoreactivity to SP (ISP 58%; OSP 60%) or to VAChT (ISP 56%; OSP 56%), some of them stained for GAL (ISP 18%; OSP 15%) and solitary nerve cells were SOM‐positive (in ISP only). No neurons in the submucous plexuses displayed immunoreactivity to VIP or NOS. In the myenteric plexus, some neurons stained for NOS (20%), VAChT (15%), GAL (10%), VIP (8%) or SP (8%) while no neurons immunoreactive for SOM were encountered. In both submucous and myenteric plexuses, many varicose nerve fibres expressed immunoreactivity to VAChT, GAL or SP, while VIP‐, SOM‐ or NOS‐positive nerve terminals were less numerous. The comparison of the present results with those obtained by other authors has revealed distinct inter‐species differences regarding the arrangement and chemical coding of nerve structures supplying the mammalian stomach.  相似文献

The performance of six groups of six calves each from a previous field experiment was followed from the beginning of housing until turning out seven months later. When housed the groups harboured different levels of infection with Ostertagia ostertagi and had corresponding weight and gain reductions. The most heavily infected groups improved rapidly following housing. Elevated serum pepsinogen levels decreased markedly, and approached normal values at eight weeks. Differences in serum pepsinogen levels between groups diminished considerably, but remained significant over the entire stabling period. The serum albumin level was low in the most seriously affected animals at the time of housing, but normal levels were restored within the first twelve weeks. In general, the number of parasite eggs in the faeces of the animals decreased, but there was considerable fluctuation. Apart from a single sampling date, no significant difference in egg per gram (EPG) could be demonstrated between the experimental groups during the stabling period.The calves were fed according to their age. The live weight differences between most and least affected groups diminished from 64 kg at the time of housing to 37 kg at the end of the stabling period. The reduction took place particularly during the first four weeks of housing.Considering both the grazing and stabling periods, it appeared that anthelmintic treatment twice during the period had no effect on the final live weight, whereas remeated treatments at three-week intervals resulted in an increase of 24 kg. Similarly, moving the animals to a clean pasture in mid-July resulted in an increase of 39 kg at the end of the stabling period when compared to calves which were not moved. Treatment of moved animals did not result in further body weight gain after seven months of housing.No significant correlation was found between parasite EPG of faeces and growth rate during the stabling period. However, a positive correlation was found between serum pepsinogen during the first eight weeks of housing and the weight gain over the entire stabling period. This was in contrast to the correlation experienced during the grazing period.  相似文献
