The classical paradigm for dendritic cell function derives from the study of Langerhans cells, which predominate within skin epidermis. After an encounter with foreign agents, Langerhans cells are thought to migrate to draining lymph nodes, where they initiate T cell priming. Contrary to this, we show here that infection of murine epidermis by herpes simplex virus did not result in the priming of virus-specific cytotoxic T lymphocytes by Langerhans cells. Rather, the priming response required a distinct CD8alpha+ dendritic cell subset. Thus, the traditional view of Langerhans cells in epidermal immunity needs to be revisited to accommodate a requirement for other dendritic cells in this response.  相似文献

Response of russet norkotah clonal selections to nitrogen fertilization   总被引:1,自引:0,他引:1  
The low vine vigor and high N requirement of Russet Norkotah may lead to N loss and groundwater contamination on coarse-textured soils. Recent clonal selections from Texas have produced strains that have larger and stronger vines, which may alter N requirements. This twoyear study examined the N use efficiency (NUE), yield, and quality of Russet Norkotah strain selections fertilized with different N levels on a Hubbard loamy sand in central Minnesota. The selections, Texas Norkotah Strain (TXNS) 112, TXNS 223, and TXNS 278 were grown with standard Russet Norkotah under irrigated conditions and received total N levels of 28, 112, 224, or 336 kg ha-1. Total, marketable, and large (>340 g) tuber yields increased linearly (P>0.05) with rate of N application in 1998 but not in 1997. The genotype main effect was not significant for any of the tuber yield parameters measured based on fresh weight. Vine, tuber, and total dry biomass yields were 116%, 5.8%, and 13.2%, respectively, higher with the selections than Russet Norkotah. Harvest index (HI), or the proportion of total dry matter partitioned to tubers, was 7% greater for Russet Norkotah than the TXNS selections, reflecting the larger vine growth of the selections. The selections accumulated significantly higher N in the vines (0.113 kg kg-1 N) than the standard clone (0.053 kg kg-1 N) as N rate increased from 28 to 336 kg ha-1, and the difference between the selections and the standard clone was larger at higher N rates than at lower N rates. Russet Norkotah partitioned 10% more N to tubers than did the TXNS selections, reflecting the difference in HI between the standard cultivar and its clones. Nitrogen recovered from fertilizer N applied in addition to the 28 kg ha-1 starter N (NUE28) averaged 36% and varied little with genotype, N rate, or cropping year. Biomass accumulation from similar N additions (AUE), however, was significantly higher for the selections than Russet Norkotah at 112 kg N ha-1 in 1997 only. At low N rate (112 kg ha-1), the selections had higher physiological use efficiency (PUE28) (mean 45.9 g g1) than Russet Norkotah (25.9 g g1). Results from this study demonstrate that, although N recovery was similar for the four genotypes, the Texas Norkotah strains produced greater biomass than Russet Norkotah per kg N applied at low rate in 1997 and per kg of fertilizer N absorbed by the plant in both years. However, under the conditions of this study, higher biomass of the selections did not translate into a marketable yield advantage over the standard cultivar.  相似文献

Tetracyclines and bacitracin are used extensively as a growth promotant and a therapeutic, respectively in livestock. In this study, we test whether there is an interaction between bacitracin and tetracycline that can select for an increase in microbial tetracycline sensitivity, using Escherichia coli and Salmonella as a model. We found via in vitro studies that bacitracin at sublethal concentrations preferentially selects for outgrowth of tetracycline-sensitive bacteria from among a population of tetracycline-resistant and tetracycline-sensitive E. coli and Salmonella strains. Most of the bacterial strains employed in our study were shown by PCR to possess the tetracycline resistance gene tet(A) or tet(C), either of whose activity is associated with tetracycline efflux. Bacitracin could be substituted for the lipophylic chelator, fusaric acid, used as a positive selective agent for tetracycline sensitivity. Together, these observations suggest that bacitracin-mediated selection for tetracycline sensitivity alters the function of tetracycline efflux proteins. Using bacitracin and tetracycline simultaneously may improve the effectiveness of the antibiotics, while decreasing the risk of selecting for a population of tetracycline-resistant microorganisms.  相似文献

An outbreak of exudative epidermitis (EE) among piglets in a Swedish SPF-herd initiated a survey for indications as to the cause of disease.The herd was established by caesarean section and has been closed to all new animal material, with the exception of semen for artificial insemination (AI). The study comprised serum samples from the SPF-herd over a 10-year period (n=109) and a close monitoring of animals in the herd during the period after the EE outbreak. Serum samples from conventional boars at the AI-station servicing the herd were also included (n=9). All serum samples were tested for antibodies to porcine circovirus-2 (PCV-2). In addition, 3-week-old piglets from three litters (n=24) farrowed close after the initial EE outbreak were closely monitored for clinical signs of skin disease, sampled for Staphylococcus hyicus, tested for antibodies to porcine parvovirus and in sequentially collected serum samples tested for interferon-alpha (IFN-alpha) and interleukin-6.The PVC-2 serology showed that animals in the herd were sero-negative at least until 2 months prior to the EE outbreak. During the period close after the EE outbreak the animals showed varying levels of antibodies to PCV-2 but all the tested animals had sero-converted 4 months later. The AI boars were also sero-positive to PCV-2 at the time of the EE outbreak. Animals in the SPF-herd remained sero-positive to PCV-2 during the following 7 years. In the monitored litters, one piglet had clinical EE and 15 piglets displayed defined erythemas on the abdomen. Fourteen of the piglets also had IFN-alpha in serum on one or more occasions during the study, indicating viral activity among the animals. S. hyicus was isolated from all of the piglets from the earliest sampling point (3 days of age) and onwards, irrespective of clinical signs. PCV-2 was isolated from lymph node tissue collected from one of the EE affected pigs.Further, increases in the number of stillborn piglets, small litters (<6 piglets) and repeat breeders could be correlated to the time of PCV-2 sero-conversion. Coincidence of active viral infection and sero-conversion to PCV-2 points to the virus as the cause of the EE outbreak and reproductive disturbances.  相似文献
