The mullets of the family Mugilidae, and in particular the species of the genus Mugil, are characterized by highly conserved morphological features, which hinder taxonomic classification considerably. Given this, the present study aimed to evaluate the efficiency of the 5S rDNA gene – a rapid and low cost genotyping marker – for the discrimination of Mugil species. Variations in the banding pattern permitted the reliable differentiation of four species –Mugil curema, Mugil incilis, Mugil sp. and Mugil hospes– although those of three others (Mugil liza, Mugil platanus and Mugil cephalus) were identical. This suggests that these three forms have diverged very recently, on a time scale that has limited changes in the length of the fragments of the 5S rDNA gene. The results of the study confirm the efficiency of this marker as a diagnostic tool for the identification of Mugil species, and its usefulness for population management and pisciculture, an important economic activity involving this group. 相似文献

The concentrations of lactic acid bacteria (LAB) in 42 vaginal samples from healthy and ill bitches were determined. Eight isolates belonging to the genera Lactobacillus and Enterococcus were selected and identified and their in vitro antimicrobial activity against canine pathogens and their ability to adhere to canine vaginal epithelial cells were determined. There was no correlation between the concentrations of vaginal LAB and clinical status, body temperature, vaginal pH, or age. Although the animals were either well or suffering from various illnesses, LAB were found in almost every sample, and the selected isolates showed promising probiotic-related features. These findings are significant for the design of strategies for the modulation of vaginal microbiota by vaginal LAB isolates. 相似文献

The objective of the present work was to determine the effect of Lactobacillus murinus strain LbP2 on canine fecal immunoglobulin A (IgA) levels. Seven dogs were orally treated with a 3-mL suspension of L. murinus LbP2 containing 5 × 109 colony-forming units on alternate days for 2 wk. Six dogs were treated with 3 mL of phosphate-buffered saline as placebo. Fecal samples were taken from the rectal ampulla on days 0 and 16, and the total canine fecal IgA concentration was determined with an immunoperoxidase assay kit. The IgA levels of individual dogs were compared with the nonparametric Wilcoxon test. Differences were considered significant when the P-value was less than 0.05. An increase in the total fecal IgA concentration was observed in the 7 dogs after treatment with L. murinus LbP2 (P = 0.01796). No differences were detected between the initial total fecal IgA values and those obtained at the end of placebo treatment. Thus, after oral administration L. murinus LbP2 showed potential immunomodulatory effects, an important property to assess in a microorganism being considered for use as a probiotic. 相似文献

Decreased fertility in pigs is a common occurrence during summer months. An objective of the current experiments was to evaluate if elevated ambient temperature altered the oocyte plasma membrane including potential receptors for sperm. This would potentially contribute to reduced fertilizability. Treated gilts were exposed in vivo to 32°C for 12 h per day and 20°C for the remaining 12 h per day for 7 days; control gilts were exposed to 22°C for 12 h and 20°C for the remaining 12 h each day. Cumulus–oocyte complexes were also aspirated from ovaries obtained from gilts maintained at thermoneutral ambient temperature and matured in vitro at 38.5°C or 40°C. Relative abundance of a porcine oocyte membrane protein was examined by intensity of immunolabelling of the in vivo and in vitro matured oocytes evaluated with confocal microscopy; fertilizability of the in vitro matured oocytes was evaluated in in vitro fertilization assays. Oocytes obtained from gilts exposed to elevated ambient temperature for 7 days had reduced immunolabelling compared with oocytes from control gilts (p < 0.05). Similarly, oocytes matured in vitro for 44 h at elevated ambient temperature had reduced immunolabelling and reduced fertilizability compared with oocytes matured at 38.5°C (p < 0.01 and p < 0.05). These results suggest porcine oocyte quality is reduced by elevated ambient temperature and immunolabelling of oocytes with antibodies to specific membrane proteins may be effective to evaluate some aspects of oocyte quality. 相似文献

This study was carried out to investigate the effect of supplementing culture medium with different concentrations of taurine and melatonin, on buffalo oocyte in vitro meiotic maturation and embryo development. In experiment 1, oocytes were matured in vitro and the cleaved embryos were cultured in the same following seven culture medium; (i) control (TCM 199 + 10% SS); (ii) control + 0.5 m m taurine; (iii) control + 1 m m taurine; (iv) control + 3 m m taurine; (v) control + 5 μ m melatonin; (vi) control + 10 μ m melatonin and (vii) control + 50 μ m melatonin. In experiment 2, based on the results of experiment 1, to examine the synergistic effect of antioxidants, the oocytes were matured in culture medium (TCM199 + 10% SS), supplemented with both taurine at 1 m m and melatonin at 10 μ m concentration and the cleaved embryos were cultured in the same medium. Supplementation of taurine at 1 m m concentration in the culture medium resulted in a higher (p < 0.05) transferable embryo (TE) yield when compared with control (20.6% vs 14.1%). Supplementation of melatonin at 10 and 50 μ m concentration in the culture medium resulted in a higher (p < 0.05) meiotic maturation rate (90.3% and 88.8% respectively) and TE yield (28.4% and 27.2% respectively), than the other treatments. In experiment 2, the TE yield did not improve by supplementing the culture medium with both taurine and melatonin, when compared with melatonin alone. In conclusion, the results of this study demonstrated that, enriching the culture medium with taurine and melatonin, improves in vitro embryo production efficiency in buffaloes. In Brazil, commercial feedstuffs are an important component in modern animal husbandry, but there is no information available about fungal contamination and ochratoxin A (OTA) production. The aims of this study were to determine the mycoflora incidence in poultry feeds and evaluate OTA production. In addition, the ability to produce OTA by several Aspergillus and Penicillium species was investigated. A total of 96 samples of poultry feeds were collected from four factories in Rio de Janeiro. Samples were examined for total moulds, for Aspergillus and Penicillium spp. occurrence and for their relative densities on dichloran rose bengal chloramphenicol and dichloran 18% glycerol media. The capacity to produce ochratoxin A by selected Aspergillus and Penicillium species was determined by HPLC. Total mould counts were generally higher than 1 x 10(5 )CFU ml(-1). Aspergillus and Penicillium species were isolated in the highest numbers. Aspergillus flovus and Penicillium citrinum were the most prevalent species. There was a high percentage of potential OTA producers (46%). The amount of OTA produced on this substrate was enough to cause adverse effects in animals. Several strains isolated from poultry feeds were able to produce high levels of OTA on chloramphenicol yeast medium. OTA in raw materials needs to be surveyed and storage practices must be investigated to determine occurrence and establish the livestock toxicological risk in poultry feed. 相似文献

The feasibility of substituting soybean meal for fishmeal diets for juvenile white shrimp Litopenaeus schmitti (0.35±0.01 g) was evaluated, and an adequate substitution level was determined. Five diets were evaluated using 46%, 59%, 75%, 88% and 100% substitution levels. Pellet water stability was significantly affected by dietary soybean content (P<0.05). Increased soybean content produced lower pellet stability, ranging from a dry matter loss of 14–22% after a 2‐h immersion, and 20–33% after an 8‐h immersion. After 52 days, significant differences (P<0.05) were found in shrimp weight, feed conversion ratio and protein efficiency ratio. The values were 0.64–1.06 g, 2.8–7.9 and 0.45–1.21, respectively, for the three measurements. Overall, better results were obtained with diets where soybean meal was substituted for fishmeal up to 75%. The 100% soybean meal diet resulted in poor growth performance of shrimp. Survival rates were acceptable for all treatments (90% or higher) and no significant differences were found in survival between treatments. Regression analysis using the broken‐line methodology indicated that 76.5±2% is an optimum soybean substitution level in diets that contained fishmeal and soybean as the major protein sources for grow‐out of juvenile white shrimp. 相似文献

The purpose of the present paper is to develop an easy and quick in vitro method to compare copper availability from breast milk and infant formulas. This study focuses on the differences caused by the use of pH 2.0 (adult gastric pH) or pH 5.0 (newborn gastric pH) in the first stage of the enzymolysis. pH affects Cu solubility, a possible estimator of the availability. Selection of a digestor, times of enzymolysis, centrifugation parameters, and Cu determination by ETAAS were discussed as well. Percentage of Cu solubility was larger from breast milk (gastric pH 2.0, 65.3 +/- 14.0 vs 40.0 +/- 13.9%; gastric pH 5.0, 61.2 +/- 16.5 vs 26.6 +/- 10.3%), but the soluble content was larger from infant formulas for both pHs (gastric pH 2.0, 245.3 +/- 82.1 vs 113.0 +/- 103.4 ng mL(-1); gastric pH 2.0, 169.3 +/- 76.9 vs 75.3 +/- 21.9 vs ng mL(-1)). 相似文献
